Glycolytic and oxidative muscles under acute glucose supplementation differ in their metabolic responses to fatty acyl-CoA synthetase gene suppression

  • Purpose: Skeletal muscles display significant heterogeneity in metabolic responses, owing to the composition of metabolically distinct fiber types. Recently, numerous studies have reported that in skeletal muscles, suppression of genes related to fatty acid channeling alters the triacylglycerol (TAG) synthesis and switches the energy substrates. However, such responses may differ, depending on the type of muscle fiber. Hence, we conducted in vitro and animal studies to compare the metabolic responses of different types of skeletal muscle fibers to the deficiency of fatty acyl-CoA synthetase (Acsl)6, one of the main fatty acid-activating enzymes. Methods: Differentiated skeletal myotubes were transfected with selected Acsl6 short interfering RNA (siRNA), and C57BL/6J mice were subjected to siRNA to induce Acsl6 deficiency. TAG accumulation and expression levels of insulin signaling proteins in response to acute glucose supplementation were measured in immortalized cell-based skeletal myotubes, oxidative muscles (OM), and glycolytic muscles (GM) derived from the animals. Results: Under conditions of high glucose supplementation, suppression of the Acsl6 gene resulted in decreased TAG and glycogen synthesis in the C2C12 skeletal myotubes. The expression of Glut4, a glucose transporter, was similarly downregulated. In the animal study, the level of TAG accumulation in OM was higher than levels determined in GM. However, a similar decrease in TAG accumulation was obtained in the two muscle types in response to Acsl6 suppression. Moreover, Acsl6 suppression enhanced the phosphorylation of insulin signaling proteins (Foxo-1, mTORc-1) only in GM, while no such changes were observed in OM. In addition, the induction ratio of phosphorylated proteins in response to glucose or Acsl6 suppression was significantly higher in GM than in OM. Conclusion: The results of this study demonstrate that Acsl6 differentially regulates the energy metabolism of skeletal muscles in response to glucose supplementation, thereby indicating that the fiber type or fiber composition of mixed muscles may skew the results of metabolic studies.

Relations between Mothers' Responses about Their Preschoolers' Overt and Relational Aggression by Preschoolers' Aggressive Behaviors (유아의 외현적.관계적 공격성에 대한 어머니의 반응과 유아의 공격적 행동 간의 관계)

  • In this study, mothers of 205 4- to 5-year-old preschoolers responded to aggression episodes of Werner et al. (2006); preschoolers' teachers responded to the Preschool Social Behavior Scale (Crick et al., 1997). Results showed, (1) boys exhibited more overt and relational aggression. (2) In overt aggression episodes, mothers used encouragement to boys and rule violation responses to girls; in relational aggression episodes, mothers used encouragement and power assertion responses to girls. (3) Mothers' power assertion about overt aggression related negatively with preschoolers' overt aggressive behaviors; mothers' discussion about relational aggression related positively with preschoolers' overt aggressive behaviors. Implications of these findings for the mothers' responses by aggression types were discussed in order in better understand preschooler's aggressive behaviors.

Teachers' Responses to Curriculum Evaluation of Gifted Education in Visual Art : Case Study of Three Types of Institution for Gifted Education (미술영재교육과정 평가에 대한 교사 인식과 요구 : 영재교육기관 유형별 사례연구)

  • The purpose of this study was to investigate teachers' responses to curriculum evaluation of gifted curriculum in visual arts. Especially, this study examined whether there are differences of the responses among three types of institutions for gifted education (institution for gifted education, community based gifted classroom, and school based gifted classroom). Teachers from these three types of institutions for gifted education were interviewed. The results are as follows. First, teachers viewed that consideration is needed for 3 criteria. Teachers from different institutions showed different responses to 'professionalism of working personnel' and 'utilization of advisory committee'. Second, teachers viewed that 13 criteria need consideration. Teachers from different institutions showed different responses to 'support personnel for gifted education' and 'educational equipment and resource supply'. Third, teachers viewed that consideration is needed for 2 criteria. Teachers from different institutions showed different responses to 'efforts for professional development of instructors'. Fourth, teachers viewed 4 criteria need consideration. Teachers from different institutions showed different responses to 'budget planning for next year'. Based on the findings, implications for developing and implementing the evaluation criteria for the gifted curriculum in visual arts were discussed.

Elementary School Teachers' and Preservice Elementary School Teachers' Actual and Preferred Types of Science Assessment (초등교사와 예비초등교사의 과학평가 실태와 지향)

  • In this study, we investigated elementary and preservice elementary school teachers' actual and preferred types of science assessment focusing on the purpose and the method of assessment. Participants were 75 elementary school teachers and 93 preservice elementary school teachers. The questionnaire adopted from previous relevant studies was administerd in order to examine the types of assessment which the teachers have actually implemented and the preferred types of assessment for hypothetical situations known as the prototypical examples of constructivist assessment. The results revealed that most teachers and preservice teachers tend to possess the summative-oriented purpose of assessment. An affective assessment purpose focused on evaluating students' interest, attitude, and curiosity toward science was newly identified in this study. In analyzing teachers' responses from the viewpoint of the method of science assessment, responses of an informal mode such as observation and portfolio were noticeable. However, most of them were found to correspond to the traditional summative-oriented purpose of assessment. For prototypical constuctivist assessment situations, teachers and preservice teachers were found to prefer the performance and the informal modes than the measurement mode of the assessment methods, whereas their purposes of the assessment were not found to be constructivist.

A nondestructive method for controlling wind loads and wind-induced responses of wooden pagoda

  • High-rise wooden pagodas generate large displacement responses under wind action. It is necessary and wise to reduce the wind loads and wind-induced responses on the architectural heritage using artificial plants, which do not damage ancient architecture and increase greenery. This study calculates and analyzes the wind loads and wind-induced responses on the Yingxian Wooden Pagoda, in China, using artificial plants via the finite element analysis (FEA). A three-dimensional wind-loading field was simulated using a wind tunnel test. Wind loads and wind-induced responses, including the displacement and acceleration of the pagoda with and without artificial plants, were analyzed. In addition, three types of tree arrangements were discussed and analyzed using the score method. The results revealed that artificial plants can effectively control wind loads and wind-induced displacements, but the wind-induced accelerations are enlarged to some extent during the process. The height of the tree significantly affected the shelter effects of the structure. The distance of trees from the pagoda and arrangement width of the tree had less influence on shelter effects. This study extends the understanding of the nondestructive method based on artificial plants, for controlling the wind base loads and structural responses of wooden pagodas and preserving architectural heritage via FEA.

The Effect of Identification Framing as Crisis Response Strategy (위기대응 전략으로서 정체성 프레이밍 효과)

  • The current study challenges to suggest an umbrella strategy applied to different type of crisis, which is different from normative principle in crisis communication. The umbrella or comprehensive strategy in this study is identification framing. Identification framing is strategic message for organizational identification, which is close to social identification. The current study employed experimental design manipulating crisis types, crisis response types, and identification framing. The crisis types were internal versus external crisis, crisis responses were denial versus apology, and using identification framing $2{\times}2{\times}2$ factorial design were used. Two hundreds forty students participated in the experiment. The result showed the significant effectiveness of identification framing in different crisis types and crisis responses.

The Relation between Preservice Teachers' Philosophical Views on Science and Types of Responses to Alternative Hypotheses (예비교사들의 과학철학적 관점과 대안적 가설에 반응하는 유형과의 관계)

  • The purpose of this study was to identify the relation between preservice teachers' philosophical views on science and types of responses to alternative hypotheses. To identify preservice teachers' philosophical views on science, the extraterrestrial impact theory and the volcano-greenhouse theory, alternative hypotheses related to dinosaur extinction were administered to 37 elementary preservice teachers and 52 secondary preservice teachers. Subjects were required to write down their ideas before and after reading the alternative hypotheses, and then the dimensions of responses to initial ideas were analyzed. To analyze, Soh's Philosophical Perspectives Prove(PPP) was used. The results of this study were as follows: (a) elementary and secondary preservice teachers' philosophical views on science corresponds to eclecticism, (b) the main types of responses were partial theory change in elementary preservice teachers and rejection in secondary preservice teachers, (c) preservice teachers' philosophical views on science and types of responses were eclecticism-partial theory change in elementary preservice teachers and eclecticism-rejection in secondary preservice teachers.

Differences of Smooth Pursuit Eye Movement in the Patients with Schizophrenic Disorder in Accordance with Family History and Their Types(Type I and Type II) (정신분열병 환자에 있어서 가족력과 임상유형(Type I, Type II)에 따른 안구추적운동의 차이)

  • Smooth pursuit eye movement, one of the reliable biological markers of schizophrenia, is not always abnormal in schizophrenic patients. Therefore the authors studied it in schizophrenic patients and normal controls and compared the results according to the presence or abscence of family history of psychosis and types (type I and type II). The results are as follows: 1) In the 18 normal control group (8 mole, 10 female), there was no sex difference in the responses of smooth pursuit eye movement. 2) In th 44 schizophrenic group (28 male, 16 female), there also was no sex difference in the responses of smooth pursuit eye movement. 3) In comparison of 44 schizophrenic group to 18 normal control group, there was significantly increased abnormal response in smooth pursuit eye movement in schizophrenic group (P < 0.005). 4) In schizophrenic group, there was no difference in the responses of smooth pursuit eye movement between type I and type II schizophrenia. 5) The presence or abscence of family history of psychosis made no difference in the responses of smooth pursuit eye movement in schizophrenic group. 6) Subdivision of type I or type II in each case of presence or abscence of family history made no difference in the responses of smooth pursuit eye movement in schizophrenic group.

Differential Subcellular Responses in Resistance Soybeans Infected with Soybean Cyst Nematode Races

  • Early nematode development and subcellular responses in resistant soybean lines PI 88788 and PI 437654 infected with races 3 (R3) and 14 (R14) of soybean cyst nematode (SCN), Heterodera glycines Ichinohe, were compared. SCN R14 nematodes penetrated and developed significantly more than R3 at 5-6 days after inoculation. Both races also penetrated and developed more in PI 88788 than in PI 437654. Syncytia, characterized by cell wall dissolution and cellular hypertrophy, were developed more in PI 88788 than in PI 437654 and more by R14 than R3, for which less necrotic responses occurred in the former than the latter. This suggests that the latter two may be more resistant and less virulent than the former two, respectively. A common structural feature found in each of PI 437654 and PI 88788 in relation to SCN-resistance was the formation of prominent cell wall appositions and nuclear degeneration prior to cytoplasmic degradation in syncytial cells, respectively. Necrosis and cell wall apposition are types of hypersensitive responses occurring at early stages of the nematode infection so that these structural modifications indicate the inhibition of initial syncytial development related to the early nematode development. As soybean cultivars and lines with identical or similar genotypes have the same types of structural features related to SCN-resistance, the structural modifications induced by SCN infection may result from the expression of inheritable resistance genes, of which the information can be used for breeding soybean cultivars and lines specifically resistant to SCN races.

Investigation of Seismic Responses of Single- and Bi-Directional Traffic Light Poles (단방향 및 양방향 교통신호등주의 지진응답 분석)

  • The seismic responses of traffic light poles are investigated using a finite element analysis. Among the traffic light poles, single- and bi-directional traffic light poles are considered since such poles are frequently installed on vehicle roads. For a more detailed investigation, three different lengths of the mast arm are considered for each directional pole. For a time-history analysis, six actual and two artificial earthquakes are considered and applied to each direction of the poles (x and y) to investigate which direction input provides more significant responses due to the unsymmetrical structural shape. Herein, the x and y directions are respectively parallel and perpendicular based on the single mast pole case. From the analysis results, the average maximum displacement response is developed with the x-direction input case for both types of light poles. Also, the bi-directional traffic light poles show a 13% larger response than the single-directional traffic light poles. Even though the y-direction input case produces a smaller response, the response difference between the single- and bi-directional light poles considerably increases by about 60%. The average maximum acceleration responses are almost similar for both types of light poles.
