  • This study was attempted co develope a method for detection of Theileria sergenti infection on the basis of hybridization of parasite DNA with a probe. For construction of a T sergenti genomic library, T sergenti DNA was digested completely with Bam-HI and the fragments were ligated into the Bam-HI site of pUC-19 before transformation of Escherichia colistrain JM83. To detect clones containing the parasite's DNA sequences, a genomic DNA library of T sergenti constructed in pUC-19 was screened by cracking and Southern hybridization. Seven colonies were chosen from 29 colonies which were screened by transformation of Escherichia coli strain JM83. Seven transformants were comfirmed from seven colonies by cracking. The sizes of transformants were about 5Kb, 5.7Kb, 4.3Kb, 7.75Kb, 7.85Kb, 5.8Kb, 3.8Kb, respectively. DNA inserts, T sergenti DNA, and bovine DNA were hybridized with radio-labelled T sergenti DNA. Two($pT_1$, $pT_1$) of the seven inserts and T sergenti DNA reacted strongly but another 5 inserts and bovine DNA showed weak reation. All of the DNA inserts were not reaction, but T sergenti DNA were very weakly and bovine DNA were strongly reacted to hybridization with radio-labelled bovine DNA. Therefore, we obtained total 7 T sergenti DNA fragments in this study.

  • In this study, we try to establish the rapid and specific detection system for Salmonella species. The PhoE gene of Salmonella species was amplified with two specific primers, ST5 and ST8c, using PCR. The probe prepared from the amplified PhoE gene was sequenced and applied for Southern blot analysis. After PCR with ST5 and ST8c primers for PhoE gene, DNA bands of expected size(365bp) from 7 different Salmonella species were detected, but not from 12 enterobacteriaceae and 3 gram positive bacteria. PCR was highly sensitive to detect up to 10fg of purified DNA template and to identify Salmonella species with only 320 heat-lysed bacterial cells. The inhibition of PCR amplification from stool specimen was occurred with 50-fold dilution but disappeared over 100 fold dilution of samples. It was confirmed that the PhoE genes were amplified and cloned with over 97% nacleotide sequence homology of PCR products compared with that of S. typhfmurium LT2. The DNA probe derived from S. typhimurium TA 3,000 showed highly specific and sensitive reaction with PCR products of all tested Salmonella species. These results indicate that PCR was rapid and sensitive detection method for Salmonella species and DNA probe prepared from S. typhimurium TA 3,000 was specific to identify PCR products of different Salmonella species.

  • The heat exchange between the Borehole Heat Exchanger(BHE) and the surrounding ground depends directly on ground thermal conductivity k at the certain site. The k is thus a key parameter in designing BHE and coupled geothermal heat pump systems. Currently, although a thermal hydraulic response test(TRT) is mostly used in practice, the thermal hydraulic TRT needs additional power and is generally time-consuming. A new, simple wireless P/T probe for a hi-speed k determination was introduced in this paper. This technique using a wireless P/T probe is less time-consuming and requires no external source of energy for measurement and predicts local thermal properties by measuring soil temperatures along the depth. Measured temperature data along the depth was analyzed. In order to verify the new technique for the determination of ground thermal conductivity, ground thermal conductivity k that calculated from the measured temperature data using a wireless P/T probe was compared with one obtained from conventional hydraulic TRT. When comparing the average k of two methods, the relative error was approximately 10%. As a result, the electronic TRT can replace the conventional hydraulic TRT method after carrying out the additional research on a lot of sites.

  • The effective thermal conductivity of two-phase materials such as unbonded silica sands saturated with a nanofluid was measured at high temperature using the transient thermal probe method. The nanofluid used in this study was a water-based mixture of 0.1 vol% $Al_2O_3$ nanoparticles with a diameter of 45 nm. The convection problem for fluids was prevented with this measurement method because the fluid was confined to within very small pore spaces. Based on the prediction model for unbonded sands, the thermal conductivities of the saturating nanofluid at high temperatures could be determined with the measured effective thermal conductivities for the two-phase material. In the results, increases in the thermal conductivity ratios of the nanofluid to pure water when temperatures were varied from $30^{\circ}$ to $80^{\circ}C$ were within the range of 4.87%~5.48%.

  • A fiber-optic beta/gamma dual detector system with two types of sensing probes was fabricated to detect the beta- and gamma-rays simultaneously. As scintillators of the sensing probe type 1, two different inorganic scintillators, $CaF_2(Eu)$ and LYSO(Ce) crystals, were used to obtain the each scintillating efficiency with respect to beta-and gamma-rays and the inherent energy spectra of radioactive isotopes. In the case of the sensing probe type 2, which is composed of two identical inorganic scintillators and a beta shielding material based on the lead, it could discriminate beta- and gamma-rays using a subtraction method. In conclusion, we demonstrated that the proposed fiber-optic beta/gamma dual detector could measure and discriminate beta- and gamma-rays using both energy spectroscopy and subtraction method.

  • Nowadays the weapon systems are employed air bursting munition (ABM) as smart programmable 40 mm shells which have been developed in order to hit the target with programmed munition that can be air burst after a set distance in the battlefield. In order to improve the accuracy of such a bursting time, by measuring the speed of the munition from the barrel, weapon systems calculate the exact time of flight to the target and then the time information must be inputted to the munition. In this study, we introduce a device capable of detecting a shot at K4 40 mm automatic grenade. The shot is composed of a rotating copper band to convert linear motion into rotary motion when it passes through the barrel, the steel section is exert the effect of fragment and aluminum section to give fuze information. The aluminum section was used to detect munition using eddy current method. To measure muzzle velocity by means of non-contact method, two eddy current probes separated 10 cm was employed. Time interval between two eddy current probe detection times was used as muzzle velocity. The eddy current probe was fabricated U-shape Mn-Zn ferrite core with enamelled copper wire, and 200 kHz alternating current was used to detect inductance change. Measured muzzle velocity using the developed sensor was compared to the Doppler radar system. The difference was smaller than 1%.

  • Bioluminescence imaging (BLI) is the most sensitive animal imaging technique for molecular imaging research. Generally, highly sensitive CCD is used to detect an optical probe introduced in a living mouse. However, in many cases, the light signal emitted from a probe is too small to detect because it is scattered and attenuated by the tissue prior to being detected. The problem is that scattering and attenuation not only inhibit accurate measurement but also make image quality down. Thus we introduced a new method to reduce noise by using property of CCD and method to improve image quality of bioluminescence image by using two steps Gaussian blurring.

  • This paper presents a linear air bearing stage with compensated motion errors by active control of preloads generated by magnetic actuators with combination of permanent and electromagnets. A 1-axis linear stage motorized with a linear motor with 240mm of travel range is built for verifying this design concept and tested its performances. The three motions of the table are controlled with four magnetic actuators driven by current amplifiers and a DSP based digital controller. Three motion errors were measured combined method with laser interferometer and two-probe method with $0.085{\mu}m$ of repeatability for straightness error. The measured motion errors were modeled as functions of the stage position, and compensation were carried out with feedforward control because the characteristics of the motion control with magnetic actuators are linear and independent for each degree-of-freedoms. As the results, the errors were reduced from $1.09{\mu}m$ to $0.11{\mu}m$ for the vertical motion, from 9.42 sec to 0.18 sec for the pitch motion and from 2.42 sec to 0.18 sec for roll motion.

  • Prostate cancer is a malignant tumor occurring in the prostate. Recently, the repetition rate is increasing. Image inspection method which we can check the prostate structure the most correctly is MRI(Magnetic Resonance Imaging), but it is hard to apply it to all the patients because of the cost. So, they use mostly TRUS(Transrectal Ultrasound) images acquired from prostate ultrasound inspection and which are cheap and easy to inspect the prostate in the process of treating and diagnosing the prostate cancer. Traditionally, in the hospital the doctors saw the TRUS images by their eyes and manually segmented the boundary between the prostate and nonprostate. But the manually segmenting process not only needed too much time but also had different boundaries according to the doctor. To cope the problems, some automatic segmentations of the prostate have been studied to generate the constant segmentation results and get the belief from patients. In this study, we propose an average shape model to segment the prostate boundary in TRUS prostate image. The method has 3 steps. First, it finds the probe using edge distribution. Next, it finds two straight lines connected with the probe. Finally it puts the shape model to the image using the position of the probe and straight lines.

  • The sound transmission loss is a key index to evaluate the acoustic performance of a car-body structure at the high frequency range. From this paper, a new validation method for in-situ sound transmission loss is proposed. First, in-situ sound transmission loss is measured by using PU intensity probe on the condition of complete vehicle. Second, validation test, which is consisted of internal, external and total frequency response function test, is performed by using volume acceleration source and microphones. Then, these test results are compared to validate the accuracy of in-situ sound transmission loss. Finally, the test result of in-site method is compared with results of two reverberant room test method and SEA analytical method. The reliability of in-situ method is confirmed by these procedures.
