Class A and class B MADS box genes fro rice flower development

  • We have previously isolated OsMADS4 gene that is a member of the class B MADS box genes from rice. In this study, another member of the class B MADS box genes was isolated from rice flower by the yeast two-hybrid screening method using OsMADS4 as bait. RNA blot analyses revealed that the clone, OsMADS16, was expressed in the second and third whorls, whereas the OsMADS4 transcripts were present in the second, third, and fourth whorls. These expression patterns of the OsMADS16 and OsMADS4 genes are very similar with those of AP3 and PI, the class B genes of Arabidopsis, respectively. In the yeast two-hybrid system, OsMADS4 interacted only with OsMADS16 among several rice MADS genes investigated, suggesting that OsMADS4 and OsMADS16 function as a heterodimer in specifying sepal and petal identities. We have also isolated OsMADS6 gene using OsMADS1 as a probe. Both are members of the AGL2 MADS family. Various MADS genes that encode for protein-protein interaction partners of the OsMADS6 protein were isolated by the yeast two-hybrid screening method. A majority of these genes belong to the AGL2 family. Sequence Homology, expression pattern, and ectopic expression phenotypes indicated that one of the interaction partners, OsMADS14, appears to be homologous to API, the class A MADS gene of Arabidopsis.

SCG10, a Microtubule-Destabilizing Factor, Interacts Directly with Kinesin Superfamily KIF1A Protein in Brain (Kinesin superfamily KIF1A와 결합하는 미세소관 불안정화 단백질 SCG10의 규명)

  • Microtubules, a major cytoskeleton, form parallel arrays in the axon and are oriented with their plus ends toward the cell periphery. Kinesin superfamily proteins (KIFs) are the molecular motors acting in the microtubule-based motilities of organelles in cells. Here, we used the yeast two-hybrid system to identify the protein that interacts with the coiled-coil domain of KIF1A and found a specific interaction with microtubule-destabilizing factor SCG10. SCG10 bound to the amino acid residues between 400 and 820 of KIF1A, but not to other KIFs in the yeast two-hybrid assay. The coiled-coil domain of SCG10 is essential for interaction with KIF1A. In addition, this specific interaction was also observed in the Glutathione S-transferase pull-down assay. An antibody to SCG10 specifically co-immunoprecipitated KIF1A associated with SCG10 from mouse brain extracts. These results suggest that KIF1A motor protein transports SCG10-containing vesicles along microtubules in neurons.

Evaluation of Sesquiterpenoids Content and Growth Characters in Clonal Lines from a Cross between Atractylodes japonica Koidz. ex Kitam. and A. macrocephala Koidz

  • Two Atractylodes species, A. japonica Koidz. ex Kitam. (AJ) and A. macrocephala Koidz (AM) were used in this study. AJ population had higher amounts of Sesquiterpenoids and stronger tolerance to root rot but less vigor of root growth than AM population. Two populations (AJ and AM) were crossed to make interspecific hybrid population. A total of 98 lines propagated clonally were selected from a cross of AJ and AM, and evaluated for contents of sesquiterpenoids, atractylon (ATLN) and atractylenolide III (AT3) using high performance liquid chromatography (HPLC), and growth characters such as plant height, stem number and root weight. HPLC profiles of the hybrids were compared with those of parent plants, and it demonstrated the production of introgression hybrid by crossing between AJ and AM. Of 98 clonal lines,10 lines were selected by 10% level based on the growth vigor and tolerance to root rot, and AJM2102-51 line showed the heaviest root weight (117.1 g/plant) among them. A total of 98 hybrid lines contained on average $0.16\;{\pm}\;0.10\;mg/g$ of $AT3,\;2.00\;{\pm}\;1.37\;mg/g$ of ATLN, and $2.16\;{\pm}\;1.40\;mg/g$ of total sesquiterpenoids, showing high coefficients of variation (above 65%). Ten lines having high contents of sesquiterpenoids were selected, and AJM2101-15 had the highest amount (9.83 mg/g) of ATLN, and showed 40.8 g/plant of root weight similar to mean value (39.9 g/plant) of hybrid lines. The result showed that the introgression of both characters of vigorous growth from AM and high sesquiterpenoids content from AJ could be possible to make new hybrid lines by crossing between AJ and AM.

Fabrication of Six-port Phase Correlator using Multi-section Power Divider and Coupler (다중결합 Power divider 와 Coupler를 이용한 Six-port 위상 상관기 제작)

  • The general six-port phase correlator is comprised of a Wilkinson power divider and three $90^{\circ}$ hybrid coupler, which has less than 10 % bandwidth. In this paper, the six-port phase correlator using two section power divider has 33 % bandwidth and external matching $90^{\circ}$ hybrid coupler with 15 % bandwidth was designed at the center frequency of 2.5 GHz. The simulation result by ADS2003A indicates that RF port and LO port of proposed six-port phase correlator got wide frequency bandwidth of 14 % for VSWR of 1.5. The fabricated six-port phase correlator has a bandwidth of 12 % similar to the simulation result. The maximum phase discrepancy and insertion loss are $6^{\circ}$ and 2.5 dB over a bandwidth, respectively.

Analyzing the Effectiveness of Education Utilizing Hybrid Model (하이브리드 교수 모델을 이용한 수업 효과 분석)

  • The purpose of this study was to open two classes with the name of $21^{st}$ Leadership, run with two different hybrid teaching styles which are hybrid LZ type and hybrid lz type, and compare them with each other in order to analyze their effectiveness of hybrid model of the subject. The subjects of this study were 64 students who took these classes, and statistical analysis were analyzed through SPSS 21.0 program. As a result of the analysis, first, there were significant development in terms of the knowledge of leadership in both LZ and lz model. but the result of final exam in the group lz was shown only significant development. Second, in the case of unconditional self-acceptance there was significant development only in the group lz. Third, the development of leadership skills was shown only in the group lz. Implications of these results were concluded that the lz is more suitable for the subject of $21^{st}$ Leadership because this model can provide much more opportunities to develope interpersonal relationship skill than LZ model. In addition, suggestions for future research were discussed.

Hybrid Barriers of Iron and Modified-bentonite for the Remediation of Multi-contaminated Water (복합오염물질 제거를 위한 철과 개량 벤토나이트의 복합층에 관한 연구)

  • Hybrid barriers using reduction and immobilization were tested to remediate the groundwater contaminated with multi-pollutants in this study. Iron filings and HDTMA(hexadecyltrimethylammonium)-bentonite were simulated in columns to assess the performance of hybrid barriers for remediation of trichloroethylene(TCE)-contaminated water. TCE reduction rate for the mixture of iron filings and HDTMA-bentonite was about 7 times higher than that for iron filings, only suggesting the reduction of TCE was accelerated when HDTMA-bentonite was mixed with iron filings. TCE reduction rate for the two layers of iron and HDTMA-bentonite was nearly similar to that for iron filings alone, but the partition coefficient($K_d$) for the two layers was 4.5 times higher than for that iron filings only. TCE was immobilized in the first layer with HDTMA-bentonite, and then dechlorinated in the second layer with iron filings. HDTMA-bentonite may contribute to the increase in TCE concentration on iron surface so that more TCE can be reduced. Also, TCE removal in the hybrid barriers was not affected by chromate and naphthalene while the reduction rate of TCE with the co-existing contaminants by iron filings was significantly decreased. Significant TCE removal in this research indicates that the proposed hybrid barrier system has the potential to become the effective remediation alternative during the occurrence of oil shock. Also, if subsurface environments are contaminated with multi-pollutants that contain non-reducible compounds as well as reducible compounds such as TCE, the conventional reactive barriers cannot be applied to this subsurface environment, while the proposed hybrid system can be applied successfully.

Optical and Electrical Properties of Two-Wavelength White Tandem Organic Light-Emitting Diodes Using Red and Blue Materials (적색과 청색 물질을 사용한 2파장 방식 백색 적층 OLED의 광학 및 전기적 특성)

  • We studied optical and electrical properties of two-wavelength white tandem organic light-emitting diodes using red and blue materials. White fluorescent OLEDs were fabricated using Alq3 : Rubrene (3 vol.% 5 nm) / SH-1 : BD-2 (3 vol.% 25 nm) as emitting layer (EML). White single fluorescent OLED showed maximum current efficiency of 9.7 cd/A, and tandem fluorescent OLED showed 18.2 cd/A. Commission Internationale de l'Eclairage (CIE) coordinates of single and tandem fluorescent OLEDs was (0.385, 0.435), (0.442, 0.473) at $1,000cd/m^2$, respectively. White hybrid OLEDs were fabricated using SH-1 : BD-2 (3 vol.% 10 nm) / CBP : $Ir(mphmq)_2(acac)$ (2 vol.% 20 nm) as EML. White single hybrid OLED showed maximum current efficiency of 7.8 cd/A, and tandem hybrid OLED showed 26.4 cd/A. Maximum current efficiency of tandem hybrid OLED was more twice as high as single OLED. CIE coordinates of single hybrid OLED was (0.315, 0.333), and tandem hybrid OLED was (0.448, 0.363) at $1,000cd/m^2$. CIE coordinates in white tandem OLEDs compared to those for single OLEDs observed red shift. This work reveals that stacked white OLED showed current efficiency improvement and red shifted emission than single OLED.

Confirmation of $F_1$ Hybridity Using RAPD Markers in Soybean

  • Molecular markers are useful to confirm the hybridity of F1 plant derived from cross of two homozygous parents with similar morphological traits. RAPD markers were used to test F1 hybrid plant obtained from cross of two homozygous soybean (Glycine max) parents. Fl plant for cross I was made from the mating of Hobbit87 (female) and L63-1889 (male) and Fl plant for cross II was obtained from the mating of H1053 (female) and L63-1889 (male). Selfing plant per each cross was also obtained. Among 20 Operon primers used, OPA04 and OPA09 show polymorphism between cross I and II parent. Band in size 1Kb of OPA04 and 2.1Kb of OPA09 primer was polymorphic band. This fragment identified Fl hybrid plant and selfing plant in cross I and II. Female parent Hobbit87 in cross I and H1053 in cross II has no this fragment (recessive allele). However, male parent L63-1889 and Fl hybrid plant in cross I and II has this size of polymorphic band (dominant allele). This indicated that Fl hybrid and selfing plants were detected by RAPD marker before phenotypic marker would be used to identify Fl hybridity. Amplification products of selfing plant for cross I and II were completely same to the those of female parent. When mature, flower color of Fl hybrid plant in cross I and II was purple and flower color of selfing plant in cross I and II was white. Purple flower is dominant trait. Fl hybridity was successfully detected at very early growth stage using RAPD marker. Therefore, RAPD marker can be used broadly to confirm Fl hybridity in many crops.

Species and Hybrid Identification of Genus Coreoleuciscus Species in Hwnag-ji Stream, Nakdong River Basin in Korea (낙동강 상류 황지천에 서식하는 쉬리속(genus Coreoleuciscus) 어류 집단의 종 동정 및 잡종 판별)

  • A natural hybrid of interspecific between the Coreoleuciscus splendidus and C. aeruginos (Cypriniformes: Cyprinidae) was captured in the Hwang-ji Stream, a tributary of the Nakdong River basin in Korea. An interspecific hybrid between C. splendidus and C. aeruginos was genetically identified based on morphological characteristics and the sequence analysis of nuclear recombination activating gene 1 (RAG1) gene (1,334 bp) and mitochondrial cytochrome c oxidase subunit 1 (CO1) gene (1,551 bp). As a result of morphological variations, the natural hybrid appeared to have an intermediate character between two parental species (C. splendidus and C. aeruginos) in three variations of black array (s) on dorsal, caudal and anal fin rays. Phylogenetic analysis inferred from RAG1 and CO1 sequence data revealed that Coreoleuciscus populations from Hwang-ji stream consist of two pure Coreoleuciscus species and a hybrid individual group. The individuals were clearly identified the cross and reciprocal hybrid by CO1 gene analysis. In RAG1 gene, 13 nucleotide variation loci were detected and the hybrid individuals displayed the double peaks of sequence chromatograms at the 9 diagnostic positions. In this study, molecular data and morphological variations were clearly demonstrated that hybridization did occur between C. splendidus and C. aeruginos. However, F2 hybrid generation and reproductive capacity of F1 hybrid individuals were not demonstrated.

A Study on the Implementation of Hybrid Learning Rule for Neural Network (다층신경망에서 하이브리드 학습 규칙의 구현에 관한 연구)

  • In this paper we propose a new Hybrid learning rule applied to multilayer feedforward neural networks, which is constructed by combining Hebbian learning rule that is a good feature extractor and Back-Propagation(BP) learning rule that is an excellent classifier. Unlike the BP rule used in multi-layer perceptron(MLP), the proposed Hybrid learning rule is used for uptate of all connection weights except for output connection weigths becase the Hebbian learning in output layer does not guarantee learning convergence. To evaluate the performance, the proposed hybrid rule is applied to classifier problems in two dimensional space and shows better performance than the one applied only by the BP rule. In terms of learning speed the proposed rule converges faster than the conventional BP. For example, the learning of the proposed Hybrid can be done in 2/10 of the iterations that are required for BP, while the recognition rate of the proposed Hybrid is improved by about $0.778\%$ at the peak.
