한국잔디(Zoysia japonica Steud.)의 분지 발달양상과 광합성기관의 전자 현미경적 구조에 관한 연구 (Studies on the Development of Branch Patterns and the Electronmicroscopic Structure of Photosynthetic organs in Zoysia japonica Steud.)

  • This study was carried out to elucidate the physiological and ecological mechamism of the branch pattern and electronmicroscopic structure of photosynthetic organs in Zoysia japonica Steud. The angles between the internodes of rhizome and the branches of Z.iaponica were changed from 0 to 52 degrees. The rhizomes tended to branch from the 3th node to the 10th node. When the terminal bud was damaged by various environmental factors, the initial branching formation was appeared immediately to the 4th node. The internode growth of the rhizome started from the beginning of April and continued to the end of Octobor. It became evident that there is a division labor between two different kinds of photosynthetic cells of mesophyll and bundle sheath tissues in Z.japonica The thick-walled bundle sheath cells surrounds the leaf vascular bundles as the Kranz type and separates them from the mesophyll cells.

Herbicide Resistant Turfgrass(Zoysia japonica cv. 'Zenith') Plants by Particle bombardment-mediated Transformation

  • Transgenic zoysiagrass (Zoysia japonica cv. Zenith) plants have been obtained by particle bombardment of embryogenic callus with the plasmid pSMABuba, which contains hygromycin resistance (hpt) and bialaphos resistance (bar) genes. Parameters on DNA delivery efficiency of the particle bombardment were partially optimized using transient expression assay of a chimeric $\beta-glucuronidase$(gusA) gene driven by the CaMV 35S promoter. Stably transfarmed zoysiagrass plants were recovered with a selection scheme using hygromycin. Transgenic zoysiagrass plants were confirmed by PCR analysis with specific primer for bar gene. Expression of the transgene in transformed zoysiagrass plants was demonstrated by Reverse transcriptase (RT)-PCR analysis. All the tested transgenic plants showed herbicide BastaR resistance at the field application rate of $0.1\%-0.3\%$.

  • The cycle of heavy metal, copper, was investigated in dynamic grassland ecosystems at a steady state in a Zoysia japonica and a Miscanthus sinensis ecosystem in Mt Kwanak, Korea. Total average storage amounts in Z. japonica and M sinensis grasslands were copper 23.92mg /$m^2$ and 51.82mg /$m^2$ respectively. Estimates of decay constants for based on experimental and mathematical model, were 0.18 in Z. japonica grassland, and 0.30 in M sinensis grassland. Decay half time of copper were 3.85 years in Z. japonica grassland and 2.31 years in A'. sinensis grassland. 95% decay times of initial copper amounts in Z. japonica and A'. sinensis were 16.68 and 9.99 years. Needed times to decay almost all of elements in Z. japonica and M sinensis grassland were 27.80 years and 16.65 years respectively. The copper was losed more rapidly in M. sinensis than in Z. japonica grassland.

  • This research was carried out to elucidate on standing crops , dry matter yields , the nutritive value and in vitro digestihility according to mowing frequency and period during the growing season in the 3cm moving plot in Korean Lawn , Zoysia japonica Sheud . The increasing tendency in the standing crops was showed yield from May to Septemher . In the dry matter yield , there was a tendency of rapid increase from May to August ( maximum level ). Afterwards there appeared a tendency of decrease slight ( Table 3). The nutritive value and in vitro digestihility ; The crude protein and crude fat in the 3cm moving plot showed a tendency of slight decrease as a moving period and frequency . On the other hand , Neutral detergeat fiber , Acid detergent fiber and cellulose showed a tendency of slight increase as mowing period and frequency . in vitro digestibility appeared a rapid decreasing rate as a mowing period and frequency was delayed (table 2.4).

  • This study was carried out to elucidate the seasonal changes of amylase an nitrate reductase activities in Zoysia japonica Steud. The seasonal changes of the amylase activity AA) in aboveground parts appeared high activity from the beginning of May to the beginning of July and in autumn showed an activity of rapid decreasing tendency. But, the AA in winter(Nov. 15~Feb 28 was not almost detected. The seasonal changes of nitrate reductase activity(NRA) in various organs appeared a very similar figures to the seasonal changes of AA. But, the NRA in the root was not almost detected.

한국잔디에 있어서 Amylase와 Nitrate reductase의 기관별 활성분배에 관한 연구 (A Study on Allocation of Amylase and Nitrate Reductase Acti ities among the organs Zoysia japonica Steud.)

  • Zoysia japonica를 부위별로 나누어서 그들간의 amylase와 nitrate reductase의 활성을 조사한 결과는 다음과 같다. 1. Amylase의 활성은 관부에서 8.36~9.46 unit/mg.rotein/hr.로 가장 높았고 이삭에서 2.04 unit/mg.rotein/hr.로 가장 낮았다. 포복경, 뿌리, 잎에서의 amylase의 활성을 각각 5.42~5.82, 3.76, 2.32~3.16 unit/mg.rotein/hr.나타내었다. 2. Nirate reductase의 활성은 빛을 많이 받는 잎에서 0.35~0.66 n mole/mg.rotein/hr.로 가장 높았고 관부에서 0.06~0.10 n mole/mg.rotein/hr.로 가장 낮았다. 이삭과 포복경에서는 각각 0.31,0.27~0.63 n mole/mg.rotein/hr.를 나타내었다. 이러한 결과로부터 저장기관인 관부나 관부 절간에서 높은 amylase의 활성을 이용하여, 양분을 이삭으로 이동시키고 있음을 알 수 있었다. nitrate reductase의 활성은 chloroplast를 갖지 않는 기관보다 광합성기관에서 더 높았다. 잔디밭에서 같이 사는 크로버와 비교해 보면 amylase의 활성이 Zoysia japonica보다 2배가량 더 높았다. 이러한 결과로부터 잔디밭에서 크로버가 더 생장력이 큼을 알 수 있었다.

  • This study was carried out to investigate the interaction between productivity and light condition and to analyze the material productivity and productive charactaristics under different hours of light in Zoysia japonica. 1. Rate increasement of leaf number and total leaf length was remarkably high at the early growth stage in the control and 9 hour light treatment. The rate gradually decreased as growth proceeded. But 3 hour treatment was very low in the rate from its early growth stage through the whole test period. 2. The increasing rate of leaf area ratio (LAR) in all the experimental plots was remarkably high at the early growth stage after transplanting the grass. The shorter photoperiod resulted remarkable lower increasement of LAR. 3. The rhizome growth rate of the 9 hour photoperiod was high contrast to the 3 hour photoperiod treatment. The increasing rate of node number was also showed similar trend. 4. Chlorophyll content was very high at the 36 days after transplanting and then decreased gradually. Chlorophyll content on 3 hours photoperiod plot severely decreased. The ratio of chlorophyll a to b was decreased significantly at short day treatment. 5. The content of soluble sugar was increased at shorter photoperiod. The highest ratio of sugar content was on August, 28 by HPLC method. Such a result was attributed to accumulation of sugar in spite of low synthesis of ploysacchrides, translocation by low consumption. 6. The increasing rate of standings in all light treatment was high at the early growth stage after transplanting. Short photoperiod treatment by 3hour showed especially low prganic productivity on Zoysia japonica.

제초제 Dicamba의 토양미생물 및 잔디 효소에 의한 분해 (Degradation of the herbicide dicamba by microorganisms isolated from the soil and phosphate extracts of turfgrass, Zoysia Japonica S.)

  • 토양미생물 및 잔디 추출액에 의한 제초제 dicamba의 분해성을 평가하기 위하여 본 실험을 수행하였다. Dicamba를 분해하는 토양미생물 5종을 enrichment culture에 의해 토양으로부터 분리하였으며, 이들 균주는 Acidovorax sp., Alcaligenes sp. 및 Variovorax sp.로 동정되었다. Dicamba를 10 ppm 수준으로 처리한 무기배지에서 분리균에 의한 dicamba 분해는 배양 21일 후에 평균 90% 이상이었으며, 분해산물로 5-hydroxydicamba가 검출되었다. 한국잔디 (Zoysia Japonica S.)를 phosphate buffer로 추출하여 추출액에 의한 dicamba 분해를 조사한 결과 dicamba의 반감기는 $2.5{\sim}2.7$일 이었으며, 대사산물로 소량의 4- 및 5-hydroxydicamba가 검출되었다.

Late Fall Nitrogen Application and Turf Cover for Zoysiagrass (Zoysia japonica) Spring Green-up

  • The use of zoysiagrass (Zoysia japonica Steud.) in the transition zone is limited because of a lack of cold hardiness although zoysiagrass has many advantages compared to other warm-season and cool-season grasses. Late-fall N fertilization is often applied for darker green color of turfgrass in early spring and more extensive root growth without rapid top growth. The objective of the study was to evaluate the effects of late fall N application and turf cover for zoysiagrass spring green-up. Clear polyvinyl chloride (PVC) film was used for turf cover. The amount of N applied were 5 and $10g\;N\;m^{-2}$ for the low and high N rate treatments, respectively. Covered zoysiagrass had greater turfgrass color and quality in early spring than non-covered zoysiagrass. The high N rate had 0.6 to 2.3 greater turfgrass quality than the low N rate on 7 of 9 rating dates. Slow-release N as late fall fertilization is more effective for turfgrass color and quality than fast-release N in spring. Turf cover could reduce the period of yellow zoysiagrass, and the earlier time of spring green-up could be advanced by increasing turfgrass quality and growth of zoysiagrass.

답압이 한국잔디의 생육에 미치는 영향 (Effects of Trampling on Growth and Development in Zoysia japonica)

  • 답압스트레스는 골프장 등에 한국잔디(Zoysia japonica)로 조성된 잔디밭에서 인간의 이동, 농기계나 차량의 이동 등에서 종종 발생한다. 본 연구는 잔디의 생육에 답압 스트레스가 미치는 영향을 알아보기 위하여 무답압(0 J $day^{-1}$), 약답압(1,570 J $day^{-1}$), 중답압(4,710 J $day^{-1}$), 강답압(9,420 J $day^{-1}$) 4수준의 답압조건에서 지상부와 지하부 길이, 광합성 능력, 엽수, 건물중, 2-DE 단백질 분석을 실시하였다. 지상부 길이는 답압을 주게 되면 답압을 하지 않은 무처리구 비해 급격하게 줄어들었고, 답압의 충격량이 증가할수록 길이가 짧아지는 경향을 나타냈다. 지하부 길이는 답압을 한 처리구가 답압을 하지 않은 무처리구 보다 길이가 길었고, 그 중 약답압을 한 처리구에서 가장 길었다. 광합성 능력은 답압을 주게 되면 답압의 충격량이 증가할수록 감소하는 경향을 나타내었다. 엽수는 답압을 하지 않은 무처리구에서 가장 많이 나왔고, 답압의 충격량이 증가할수록 감소하는 경향을 나타내었다. 지상부 건물중은 답압이 증가할수록 감소하는 것으로 확인 되었고, 중답압과 강답압에서는 큰 차이를 보이지 않았다. 한국잔디에 답압을 주게 되면 그에 따른 스트레스에 적응하기 위해 반응하는 단백질이 발현되었다. 단백질 변성을 막아주는 heat shock 70 kDa protein 10와 광합성에 관련된 Oxygen-evolving enhancer protein 1이 발현이 증가 되었다. 이상 종합해보면 답압 조건에 따른 한국잔디의 생육에 차이가 있었으며, 일정수준의 답압이 잔디 생육에 있어서 지상부 생육은 답압하지 않은 것보다 저조 하였으나 지하부 생육은 다소 촉진되었다.
