The aromatic amino acids, which are composed of $\small{L}$-phenylalanine, $\small{L}$-tyrosine and $\small{L}$-tryptophan, are general components of protein synthesis as well as precursors for a wide range of secondary metabolites. These aromatic amino acids-derived compounds play important roles as ingredients of diverse phenolics including pigments and cell walls, and hormones like auxin and salicylic acid in plants. Moreover, they also serve as the natural products of alkaloids and glucosinolates, which have a high potential to promote human health and nutrition. The biosynthetic pathways of aromatic amino acids share a chorismate, the common intermediate, which is originated from shikimate pathway. Then, tryptophan is synthesized via anthranilate and the other phenylalanine and tyrosine are synthesized via prephenate, as intermediates. This review reports recent studies about all the enzymatic steps involved in aromatic amino acid biosynthetic pathways and their gene regulation on transcriptional/post-transcriptional levels. Furthermore, results of metabolic engineering are introduced as efforts to improve the production of the aromatic amino acids-derived secondary metabolites in plants.
The effect of aromatic amino acids such as phenylalaine, tryptophan and tyrosine on somitogenesis at the early stage of chick embryo has been investigated morphologically using light and electron microscopy. Micrographs of aromatic amino acid injected chick embryo showed that an incomplete somite segmentation occurred and some decremental effect on the nervous system were observed. Somites were poorly developed and their size were variable. Electron micrograph of somatic cells from aromatic amino acid injected chick embryo showed that chromatins were coagulated, some of mitochondria were damaged, and nucleus were transformed considerably in some cases. The protein and nucleic acid levels and some enzyme activities of 15-day chick embryo which received the injection of 1mg of aromatic amino acid in 0.05 ml of saline 24 hours after the incubation were analyzed. Protein, DNA and RNA levels of the test group were not lowered significantly but the activities of enzymes for basic metabolism, such as lactate dehydrogenase, succinate dehydrogenase, malate dehydrogenase and glucose 6-phosphate dehydrogenase were considerably lowered as compared with those of control. From the present expeerimental results, it was tentatively suggested that the administration of amino acid might slow down the yolk granule degradation probably by feed back mechanism resulting in the disturbance of amino acid balance in the cell, which might give rise to impair normal metabolic pattern leading to abnormal somitogenesis to chick embryo at very early stage of development.
An extracellular polymer (exo-polymer) with hepatoprotective properties was produced after a 6-day submerged mycelial culture of Ganoderma lucidum WK-003. The glutamic pyruvic transaminase (GPT) activities in the serum of intoxicated Sprague-Dawley rats were decreased from 871 to 263 by the oral administration of the exo-polymer (20 mg/kg/day) for 4 consecutive days. Rhamnose, arabinose, xylose, mannose, galactose, and glucose were found in the exo-polymer along with aspartic acid, glutamic acid, histidine, serine, glycine, arginine, alanine, tryptophan, valine, phenylalanine, isoleucine, and leucine.
A role for brain serotonin(5-HT) in regulation of the HPA axis has been suggested but remains contoversial and poorly defined. The present experiments were designed to check kinetic parameters of 5-HT turnover in rat hypothalamus and remainder brain areas before and after stress and to test whether using various different pharmacologic approaches to stimulate or eliminate the control serotonergic system have any consistent effect on the stress-induced activation of HPA system. Steady state brain serotonin and 5-HIAA concentrations during 1 min ether stress were significantly elevated without significant rise in the levels of plasma corticosterone, which highly increased 2 minutes after stress. This suggests that the increase in serotonergic neuron activity precede that in HPA activity. Furthermore, during 1 ruin-ether stress or 30 min immobilization stress there is a marked increase in hypothalamic and remainder brain serotonin (5-HT) turnover or synthesis rates assessed by both the pargline/5-HT method and pargyline/5-HIAA method. The stress-induced corticosterone levels were increased by serotonin precursors and serotonin agonist in a dose-related fashion. The stress- induced corticosterone levels were highly elevated by L-tryptophan (100 mg/kg) and Potentiated by monoamine oxidase inhibitor, pargyline or serotonin agonist, 5-MeoDMT. The stress-induced elevation of corticosterone and 5-HT levels in rat brain were not significantly decreased by the administration of 5-HT synthesis inhibitor, PCPA and 5-HT neurotoxin, 5,7-DHT. However, the stress-induced elevation of corticosterone and 5-HT levels were decreased by the destruction of midline raphe nuclei. There was a strong positive correlation between plasma corticosterone and 5-HT concentrations changed by drugs which mainly manipulating 5-HT system in the hyhothalamus and in the remainder of the brain. In conclusion, our present data stongly suggest that 5-HT is an important key neurotransmitter involved in the stress-induced activation of the HPA system.

A Case of Pellagra Induced by Isoniazid during Treatment of Pulmonary Tuberculosis Pellagra is a disease caused by a deficiency of nicotinic acid or niacin. It is mostly found among people eating corn-based diets in parts of China, Africa and India. It is also induced by drugs, such as isoniazid or 5-fluorouracil. Isoniazid inhibits the conversion of tryptophan to niacin and may induce pellagra, particularly in poorly nourished patients. Pellagra should be suspected whenever tuberculous patients under the treatment with isoniazid develop mental, neurological or gastrointestinal symptoms, even in the absence of typical skin changes. Herein, our experienced of a case of pellagra induced by isoniazid during treatment of pulmonary tuberculosis is reported. The patient was referred due to a skin rash and drowsy mental status. Her skin lesion developed during treatment for pulmonary tuberculosis. Her symptoms were improved after discontinuation of antituberculous agents and on the administration of nicotinamide.
Objective: The objective was to test additivity of digestible energy and nutrient concentrations in the hatchery byproduct mixture fed to nursery pigs. Methods: In the previous studies, energy, phosphorus, calcium, and amino acid digestibility of infertile eggs, unhatched eggs, culled chicks, and a mixture of 3 hatchery byproduct ingredients was determined in nursery pigs (initial body weight = 9.4 to 14.2 kg). An additivity test was conducted using these determined values. Results: No difference was observed between determined and predicted metabolizable energy values in the mixture (3,998 and 3,990 kcal/kg as-is basis, respectively). Measured standardized total tract digestible phosphorus in the mixture was less than the predicted value (4.5 vs 5.3 g/kg as-is basis, respectively; p<0.05). Measured standardized total tract digestible calcium in the mixture was greater compared with the predicted value (40.0 vs 31.7 g/kg as-is basis, respectively; p<0.05). Measured standardized ileal digestible tryptophan in the mixture was greater than the predicted value (3.7 vs 3.1 g/kg as-is basis, respectively; p<0.05) whereas other amino acid values were additive. Conclusion: Energy and most of amino acid concentrations in hatchery byproducts are additive in the mixture fed to nursery pigs.

Purpose: The objective of this study was to provide nutritional information (protein and amino-acid contents) of soybeans (Baktae, Seoritae, Huktae, and Seomoktae) with different cooking methods. Methods: Raw, boiled (in $100{\pm}15^{\circ}C$ of water for 4 hr), and fried (in a pan at $110{\pm}15^{\circ}C$ for $20{\pm}5min$) soybean samples were prepared. Contents of protein and amino acids were determined. Results: Protein content in raw Baktae, Seoritae, Huktae, and Seomoktae soybeans ranged from 361.0 to 386.8 mg/g. Protein contents differed according to cooking methods. They were higher in pan-fried beans (107.9-113.5%) than in raw or boiled soybeans (48.2-49.5%). A total of 18 amino acids were analyzed. Amino acid data sets were subjected to principle component analysis (PCA) to understand their differences according to soybean types and cooking methods. Bean samples could be distinguished better according to cooking method in comparison with bean types by principle component (PC1) and PC2. In particular, fried soybeans contained much higher levels of cystein. Other amino acids were the dominant in raw and boiled ones. On the other hand, the amounts of threonine, histidine, proline, arginine, tyrosine, lysine, tryptophan, and methionine were higher in raw bean samples than in cooked ones. Conclusion: The contents of amino-acids and proteins are more effected by different cooking methods in comparison with soybean types.
Post-traumatic stress disorder (PTSD) is a trauma-induced psychiatric disorder characterized by impaired fear extermination, hyperarousal, and anxiety that may involve the release of monoamines in the fear circuit. The reported pharmacological properties of tetramethylpyrazine (TMP) include anti-cancer, anti-diabetic, anti-atherosclerotic, and neuropsychiatric activities. However, the anxiolytic-like effects of TMP and its mechanism of action in PTSD are unclear. This study measured several anxiety-related behavioral responses to examine the effects of TMP on symptoms of anxiety in rats after single prolonged stress (SPS) exposure by reversing the serotonin (5-HT) and hypothalamic-pituitary-adrenal (HPA) axis dysfunction. Rats were given TMP (10, 20, or 40 mg/kg, i.p.) for 14 days after SPS exposure. Administration of TMP significantly reduced grooming behavior, increased the time spent and number of visits to the open arm in the elevated plus maze test, and significantly increased the number of central zone crossings in the open field test. TMP administration significantly reduced the freezing response to contextual fear conditioning and significantly restored the neurochemical abnormalities and the SPS-induced decrease in 5-HT tissue levels in the prefrontal cortex and hippocampus. The increased 5-HT concentration during TMP treatment might be partially attribute to the tryptophan and 5-hydroxyindoleacetic acid mRNA level expression in the hippocampus of rats with PTSD. These findings support a role for reducing the altered serotonergic transmission in rats with PTSD. TMP simultaneously attenuated the HPA axis dysfunction. Therefore, TMP may be useful for developing an agent for treating psychiatric disorders, such those observed in patients with PTSD.
We evaluated the nutritional composition including proximate, amino acid, vitamin, minerals, and the antioxidant activity of buckwheat (Fagopyrum esculentum) pollen grains collected by Apis mellifera bees, to be used as a species of forage plant with Quercus acutissima (acorn) and Actinidia arguta pollen grains. The content of crude protein and fat were found 14.43% and 5.67%, respectively. Eighteen amino acids from buckwheat pollen were found, including 8 essential amino acids. The predominant amino acids were glutamic acid, aspartic acid and lysine, accounting for about 42.7% of total free amino acids. The concentration of vitamin C was the highest value of 13.7 mg/100g, followed by $B_3$ (niacin) and $B_1$ among the detected vitamins. The predominant minerals were potassium (1197.95mg/100g), followed by phosphorus (962.77mg/100g) and magnesium (535.42mg/100g), whereas copper, zinc and manganese were detected as minor elements. Antioxidant activity and total phenolic content accounted for 8.1% at $500{\mu}g/ml$ extract and $2.25{\mu}g/mg$, respectively.
Background: Ischemia-reperfusion injury related to unsuccessful myocardial protection affects postoperative ventricular function and mortality during open-heart surgery. We prospectively compared the effects of administration of histidine-tryptophan-ketoglutarate (HTK) solution and cold blood cardioplegia (CBC) on myocardial protection and clinical outcome in patients undergoing mitral valve surgery. Material and Method: Seventy patients with mitral regurgitation (MR) undergoing mitral valve surgery were randomly divided into the HTK group (n=31) and the CBC group (n=31 ): eight patients were excluded. Perioperative hemodynamics, cardiac medications, pacing, postoperative outcomes and complications were recorded during the hospital stay. All patients received follow-up for at least 6 months postoperatively for morbidity and mortality. Resuか: There were no significant differences in the hemodynamics between the groups during the study period, except for the mean pulmonary artery pressure (MPAP), PCWP and CVP that were lower in the HTK group at 15 min after weaning of CBP. There were no differences for inotropic support and pacing during the 12 hrs postoperatively between the groups. CK-MB values on day 1 and day 2 were $77{\pm}54$ and $41{\pm}23$ for the HTK group and $70{\pm}69$ and $44{\pm}34$ for the CBC group, respectively (p=NS). Postoperative clinical outcomes were similar in both groups for at least 6 months during the follow-up period. Conclusion: These results suggest that the use of HTK solution is as safe as cold blood cardioplegia in terms of myocardial protection.
