Characteristics and Action Pattern of Alkaline Protease produced from Aspergillus fumigatus (Aspergillus fumigatus이 생산하는 Alkaline protease의 특성과 작용양상)

  • This experiment was conducted to investigate the characteristics of alkaline protease from Aspergillus fumigatus which was isolated from soil as a superior strain for the production of the alkaline protease. The optimum temperature for enzyme activity was $50^{\circ}C$ and optimum pH was 9.0. The enzyme was stable at pH 8.0 to 10.0 and thermal inactivation was shown $30^{\circ}C$. The activity of the enzyme was increased by the addition of $Mn^{++},\;Cu^{++},\;Ba^{++},\;Mg^{++},\;$wheras it was inhibitied by $K^+,\;Fe^{+++},\;Ag^+,\;Pb^{++},\;Na^+,\;Ca^{++},\;Hg^+,\;Zn^{++}$. EDTA. 2, 4-DNP, ${\varepsilon}-amino$ caproic acid did not show inhibitory effect on the proteolytic activity of alkaline protease but P-chloromercuribenzoic acid inhibited the enzyme activity, indicating that reactive sulfhydryl group is required for the enzymatic activity. The reaction of this enzyme followed typical Michael-Menten Kinetics with the Km value of $8.33{\times}10^{-4}mole/{\ell}$ with the Vmax of $47.62{\mu}g/min$. This enzyme had stronger proteolytic activity than trypsin on substrate such as casin and hemoglibin.

Isolation of Leuconostoc mesenteroides subsp. mesenteroides DU-0608 with Antibacterial Activity from Kimchi and Characterization of Its Bacteriocin

  • A bacteriocin-producing strain, DU-0608, was isolated from Kimchi and identified as Leuconostoc mesenteroides subsp. mesenteroides. The bacteriocin from isolate was inhibitory against Listeria monocytogenes, Micrococcus luteus and several strains of lactic acid bacteria. The bacteriocin was inactivated by pepsin, trypsin, $\alpha$-chymotrypsin, protease, $\alpha$-amylase and lipase, but not by catalase or by heating at $100^{\circ}C$ for 60 min. The molecular weight of the bacteriocin was estimated approximately 6 kDa. The inhibitory effect was bactericidal and rapid. Following treatment with isolate bacteriocin, cells of indicator strain (Lactobacillus sake JCM 1157) were damaged at the end regions of the cell wall, whereas the cells treated with nisin were damaged at many places around the cell wall.

  • Biological efficacy assays, including antimicrobial, tyrosinase inhibition and anticancer activities of Chlorella hydrolysate, were carried out. In tyrosinase inhibition assay of Chlorella hydrolysate, $IC_{50}$(inhibitory concentration) was measured as 12%. Chlorella hydrolysate showed high anti-lung cancer activity of 88.2% at a concentration of 0.15%.

  • Several proteolytic activities and the level of and-trypsin in neoplastic tissues of human cervix and liver were compared to those in normal tissues to examine if any correlation exists between malignant behavior of the tumors and the changes in overall proteolytic capacity. Proteolysis against casein and insulin in cervix tumor was increased to 2-to 3-fold while that in liver tumor was reduced to one-tenth to one-half. By contrast, the level of anti-trypsin in cervix tumor was lowered to nearly one-tenth of that in normal tissues while the level rose to about 2-fold in malignant tissues of liver. On the other hand, the activities of plasmin-like protease and plasminogen activator were enhanced 10-20% over the activities in normals. These results suggest that the changes in proteolytic capacity are at least in part due to outbalance in either of proteolytic or its inhibitory activity over the other and occur distinctively to each tumor systems for their malignant behavior.

  • The antioxidative activity of various enzymatic extracts from Sarcodon aspratus (S. aspratus) was evaluated by measuring 1,1-diphenyl-2-picrylhydrazyl (DPPH), and alkyl radical scavenging activity using an electron spin resonance (ESR) spectrometer. For this study, the S. aspratus were enzymatically hydrolyzed by seven carbohydrases (Viscozyme, Celluclast, Dextrozyme, AMG, Promozyme, Maltogenase, and Termamyl) and eight proteases (${\alpha}$-chymotrypsin, Alcalase, Flavourzyme, Neutrase, papain, pepsin, Protamax, and trypsin). The DPPH radical scavenging activities of Viscozyme and pepsin extracts were the highest, and the half maximal inhibitory concentration ($IC_{50}$) values were 0.896 and 0.734mg/mL, respectively. The Celluclast and trypsin extracts showed the highest scavenging activities on alkyl radical, and their $IC_{50}$ values were 0.278 and 0.575mg/mL, respectively. The Celluclast extracts was decreased cell apoptosis in PC-12 cells against $H_2O_2$-induced oxidative damage. The findings of the present study suggest that enzymatic extracts of S. aspratus exhibit antioxidative activity against oxidative stress on PC-12 cells.

Effect of Buckwheat Polysaccharides on Digestive Enzyme Activity In Vitro (In vitro에서 메밀의 다당류가 소화효소 활성에 미치는 영향)

  • We examined the effects of crude hemicellulose, alcohol-insoluble hemicellulose, high molecular weight soluble polysaccharide (HMS-P : MW>10 kDa) and low molecular weight souble polysaccharide (LMS-P : MW<10 kDa) fraction isolated from buckwheat (raw, roast and steam) on digestive enzyme activity in vitro. The enzyme activities were measured after the polysaccharides-enzyme mixtures were incubated at $37^{\circ}C$ for 5 min. Crude hemicellulose, alcohol-insoluble hemicellulose and residue lowered ${\alpha}-amylase$ activity, whereas HMS-P and LMS-P had no inhibitory effect. All polysaccharides except LMS-P lowered lipase activity. Crude hemicellulose, alcohol-insoluble hemicellulose, residue and HMS-P showed a marked decrease of trypsin and chymotrypsin activity but LMS-P showed a slight decrease of them.

  • The inhibitory activity of 20 Lactic strains from Kimchi was tested against Escherichia coli and other microorganisms. Of the lactic strains investigated, A7 (Pediococcus cerevisiae) and C4(Leuconostoc spp.) were the most effective in restricting the growth of test organisms. The mixed culture inoculation of each selected lactic strain and Escherichia coli resulted in a drastic reduction in the plate count of Escherichia coli after 24 hours. Similar results were obtained when Staphylococcus aureus and Bacillus cereus were used as test organisms. For all test organisms, the presence of A7 caused a higher death rate constant than that of C4. Addition of catalase in the mixed culture did not prevent inhibition, suggesting that hydrogen peroxide did not cause the inhibition. The filtrate of A7 culture added to Escherichia coli showed identical inhibitory action, however heat treatment of filtrate at $80^{\circ}C$ 30min. destroyed the inhibitory activity. A7 filtrate treated with trypsin substantially lost the inhibitory effect, but not by pepsin. The results imply that the protein-like compound(s) is the principal inhibitor produced by this lactic strain.

  • To obtain further information concerning the nature myofibrillar proteins in a food system, an investigation has been conducted to compare the change in the biochemical property of the myofibril with the changes in the morphological structure of the myofibril. When myofibrils were prepared with 0.16 M KCl-0.04 M Tris-HCl, the band pattern was clear and distinct. There was a uniform thickening of A-band, a sharp appearence of Z-lines and a wide I-band. The band pattern of myofibrils was changed as the composition of extraction solution was changed. Also the ATPase activity of myofibril changed as the length of sarcomere changed. When myofibrils were treated with a low concentration of trypsin, myofibrils turned in the contracted state. With the progress of prolonged trypsin treatment, most of myofibrils exhibited a pattern of alternating light and dark bands, supercontracted pattern. Although myofibrils exhibited a supercontracted band pattern, the ATPase activity of myofibril continued to increase with the progress of trypsin treatment. An assumption was made that tropomyosin may be located in Z-line and that troponin-tropomyosin complex can inhibit the ATPase activity of myofibrils through the structural alternation of myofibril.

Purgative Activities of Seunggitangs

  • The purgative activity of Seunggitang prescriptions (Deseunggitang, Soseunggitang and Joweseunggitang) was measured to compare the laxative potency of these herbal prescriptions. Daeseunggitang and Jowiseunggitang more potently stimulated the transportation of small and large intestine than Rhei Rhizoma alone. However, the small and large intestine transportation activities of Soseunggitang were similar to those of Rhei Rhizoma alone. Soseunggitang inhibited nitrite production in LPS-induced RAW 264.7 cells and trypsin than the other Seunggitangs. The inhibitory activity of Jowiseunggitang was more potent on nitrite production in LPS-induced RAW 264.7 cells than those of Daeseunggitang. These results suggest that Daeseunggitang can be used as a emergent purgative for patients with severe fever and constipation, Soseunggitang can be as a mild purgative for chronic constipation with inflammation and Jowiseunggitang can be as a potent purgative for patients for severe constipation and weak colitis.

  • [$\gamma$-$^{32}$P]ATP was used to test phosphorylation of membrane proteins of mating type a cells of heterobasidiomycetous yeast Rhodosporidium toruloides separated by non-denaturing electrophoresis. The phosphoprotein was observed in the membrane proteins. The phosphorylation was inhibited by the pheromone rhodotorucine A (Rh. A) secreted by mating type A of the yeast. Rh. A didn't inhibit the phosphorylation in the presence of a trigger peptidase (TPase) inhibitor, antipain. Partially digested Rh. A by trypsin maintained the phosphorylation inhibitory activity. These results show that TPase activity plays an important role in the transduction of pheromone signal in the yeast.
