Persistence and Distribution of Trunk-Injected Abamectin in Pinus thunbergii and Pinus koraiensis Tissues (수간주입한 아바멕틴의 곰솔과 잣나무 내 분포와 지속성)

  • The residues of abamectin 1.8% EC, resisted for control of pine wood nematode, Bursaphelenchus xylophilus in pine tree were surveyed in tissue of Pinus thunbergii and P. koraiensis after injection of a liquid formulation. Limits of detection of abamectin in tissue of P. thunbergii were $0.05\;mg\;kg^{-1}$ and mean recoveries at $0.5\;mg\;kg^{-1}$ trunk injection were 90.9% and 93.1% respectively in stem and trunk of P. thunbergii. Abamectin 1.8% EC, trunk injected in 15 m height P. thunbergii were detected in all stem (edible part of carrier insect of pine wood nematode, Monochamus alternatus) from 0.29 to $0.73\;mg\;kg^{-1}$ after 150 days injection. Amount of residue of abamectin 1.8% EC in 12.6 cm mean breast height diameter (DBH) P. thunbergii were variable depending on individual trees in natural forest. Amount of residues in lower and middle part of trunk were reduced with the passage of the injection time. In upper part of trunk were detected $1.84\;mg\;kg^{-1}$ on 30 days after injection however $0.65\;mg\;kg^{-1}$ on 15 days after injection and under detection limit on 100 and 180 days after injection in P. thunbergii. Bottom and middle parts of crown were detected $0.183\;mg\;kg^{-1}$ and $0.173\;mg\;kg^{-1}$ respectively on 180 days after injection in P. thunbergii. Mean residues of abamectin in crown and trunk were $0.80\;mg\;kg^{-1}$ and $0.30\;mg\;kg^{-1}$ on 170 days after trunk injection in 20 cm DBH and 9 m height P. koraiensis. Mean residues of abamectin in crown and trunk were $0.67\;mg\;kg^{-1}$ and $0.36\;mg\;kg^{-1}$ on 170 days after trunk injection in 15 cm DBH and 6 m height P. koraiensis.

  • Chestnut(Castanea crenata) were trunk-injected by two methods to check the changes around the injection wound. In September 1993, high concentration of oxytetracycline(OTC) was injected through the injection wound of 1cm diameter and low concentration of OTC through 0.5cm diameter. Trunk diameter of injected trees ranged from 10cm to 20cm. All trees were in their vigorous conditions. Tree reaction was examined in June, 1994. None of them showed any sign of decay by the time. However, under the bark, sapwood was remarkably discolored. But, more severe discoloration was found in 1cm-injection wound than in 0.5cm one. Sapwood of some trees split from the injection wound, and the split was longer in 1cm-injection wounded trees than in 0.5cm trees. From the split, callus grew out and the split was closing. In this kind of trunk injection, the damage was more severe in 1cm-injection wound than in 0.5cm-injection wound.

Selection of Trunk Injection Pesticides for Preventive of Pine Wilt Disease, Bursaphelenchus xylophilus on Japanese Black Pine (Pinus thunbergii) (곰솔(Pinus thunbergii)에서 소나무재선충병 예방을 위한 나무주사용 약제선발)

  • This study was carried out to select effective preventive pesticides against pine wilt disease caused by pinewood nematode (PWN), Bursaphelenchus xylophilus on trunk injection. 1,000 fold aquatic solution of abamectin 1.8% EC and emamectin benzoate 2.15% EC were lower mortality (7.3% and 8.3% respectively) against PWN on 1 day after treatment. However effects of abamectin 1.8% EC, emamectin benzoate 2.15% EC, fosthiazate 30% SL and fenitrothion 30% SL were inhibited the reproduction of PWN over 99.6% in Botrytis cineria media. Effect of trunk injection of abamectin 1.8% EC and emamectin benzoate 2.15% EC at the rate of $10\;m{\ell}$ per 10 cm in diameter of breast height (DBH) on mortality of Japanese black pine, Pinus thungergii by inoculated PWN was 0% and 3.3%, respectively at the applied year however when injection of fosthiazate 30% SL were treated with the rate of $5\;m{\ell}$ per 10 cm tree DBH, mortality of tree was 63.3%. Abamectin 1.8% EC and emamectin benzoate 2.15% EC was showed high preventive efficacy representing >90% against PWN at the following year. PWN preventing efficacy of trunk injection was lower in naturally occurred area (mortality of pine tree in control was 11.7% at the first year) of PWN than artificially infected site (mortality of pine tree in control was >76.7% at the first year), PWN preventing efficacy of trunk injection of abamectin 1.8% EC and emamectin benzoate 2.15% EC at the rate of $10\;m{\ell}$ per 10 cm in DBH was 91.5% and 82.9%, respectively, at the applied year and 89.5% and 82.6% respectively at the following year in PWN naturally occurred site. Control efficacy by trunk injection of abamectin 1.8% EC and emamectin benzoate 2.15% EC was more higher in 10 fold dilution with 10 fold high amount of aquatic solution than no dilution with 10 fold less amount of aquatic solution. The preventive effect of trunk injection of abamectin 1.8% EC and emamectin benzoate 2.15% EC at the rate of $5\;m{\ell}$ per 10 cm in DBH was showed 100% at the applied year in PWN inoculated tree.

Establishment of Injection Protocol of Contrast Material in Pulmonary Angiography using Test Bolus Method and 16-Detector-Row Computed Tomography in Normal Beagle Dogs

  • The aim of this study was to establish an injection protocol of a test bolus and a main bolus of contrast material for computed tomographic pulmonary angiography (CTPA) for visualizing optimal pulmonary arteries in normal beagle dogs. CTPA using a test bolus method from either protocol A or B were performed in each of four normal beagle dogs. In protocol A, CTPA was conducted with a scan duration for around 8 s, setting the contrast enhancement peak of the pulmonary trunk in the middle of the scan duration. The arrival time to the contrast enhancement peak was predicted from a previous dynamic scan using a test bolus (150 mg iodine/kg) injected with the same injection duration using for a main bolus (450 mg iodine/kg). In protocol B, CTPA was started at the predicted appearance time of contrast material in the pulmonary trunk based on a previous dynamic scan using a test bolus injected with the same injection rate as a main bolus. CTPA using protocol A showed the optimal opacification of the pulmonary artery with pulmonary venous contamination. Proper CTPA images in the absence of venous contamination were obtained in protocol B. CTPA with a scan duration for 8 s should be started at the appearance time of contrast enhancement in the pulmonary trunk, which can be identified exactly when a test bolus is injected at the same injection rate used for the main bolus.

Prevention of Oak Wilt by Tree Injection of Culture Suspension of an Antifungal Microorganism, Streptomyces blastmyceticus against Oak Wilt Fungus, Raffaelea quercus-mongolicae

  • For the control of oak wilt caused by Raffaelea quercus-mongolicae, an antifungal microorganism, Streptomyces blastmyceticus, was used as a potential agent. Culture suspension of S. blastmyceticus was injected into Quercus mongolicae in the research forest of Kangwon National University by $ChemJet^{(R)}$ trunk injection and Macro-infusion at root flare injection. $Alamo^{(R)}$ (a.i., propiconazole 14.5%), a fungicide currently used for the control of oak wilt in USA, was also treated by both methods to compare the efficacy. For preventive efficacy, culture suspension of the pathogen was inoculated at 1 month after injection of either agent. Tested trees were cut at 3 months after treatment, stained with 1% Fuchsin acid, and then non-conductive area (NCA) and re-isolation frequency (RIF) of oak wilt fungus were compared among treatments. While NCA was the highest as 47.3% in pathogen only treatment, it was the lowest as 16.0% in sterilized water treatment by Macro-infusion. NCAs of Alamo treatment by Macro-infusion and ChemJet injection were 25.3% and 32.1%, respectively. NCA of S.blastmyceticus treatment by ChemJet injection was 32.3%, similar with Alamo treatment's by ChemJet injection. All treatments by either injection method showed significantly lower NCA compared to the pathogen only treatment. These results indicate that S. blastmyceticus injection shows the preventive efficacy against oak wilt disease by suppressing the growth of pathogen injected. NCA of Macro-infusion injection of sterilized water was lower as 16.0%, compared to 21.3% of ChemJet injection. It means that Macro-infusion is more effective in translocation of sterilized water than ChemJet injection by even distribution. RIF from wood discs of treated trees showed high in pathogen only treatment, but relatively low in S. blastmyceticus treatment. RIF results were correlated with NCA results. From the above results, it was confirmed that S. blastmyceticus showed preventive efficacy against oak wilt disease by ChemJet trunk or Macro-infusion at root flare injection.

Development of Effective Screening Method for Efficacy Test of Trunk Injection Agents Against Pine Wood Nematode, Bersaphelenchus xylophilus in Japanese Black Pine, Pinus thunbergii (곰솔(Pinus thunbergii)에서 소나무재선충(Bersaphelenchus xylophilus) 나무주사 약제의 효율적인 평가법 개발)

  • Pine wood nematode (PWN), Bursaphelenchus xylophilus is one of the most serious pests of pine tree. Trunk injection of some nematicides in tree is well known as an effective control method. However there are some limitating factors which hindering the efficacy of trunk injection in field or potted tree. In this study we suggested easy and useful alternative screening methods of nematicides against PWN. Reproduction of PWN was influenced by tree twig moisture (high reproduction in high moisture trig) and paraffin coating was 78.6% reduced moisture loss in tested twig. There were no reproduction different in up and down site from infection site of twig at 1 month after inoculation of PWN and also distance (5 and 10 cm) from inoculation site of PWN did not influence the reproduction of PWN. Numbers of reproduced PWN were higher with decreasing diameter of twig. Numbers of reproduced PWN were similar to P. thunbergii and P. densiflora. However reproduction was increased depending on high inoculation density and longer propagation period. When inoculation of PWN on cut twig injected with emamectin benzoate 2.15% EC and morantel tartrate 8% SL in trunk of Pinus thunbergii in the field, PWN number were significantly reduced than untreated control. We suggest this screening method for PWN control agents.

Variation of Soil and Leaf in a 'Wonhwang' Pear Orchard Appled by Selenium Solution (셀레늄 처리방법에 따른 '원황' 배 과원의 토양 및 수체 변화)

  • This study was established on which the selenium (Se), known as one of the functional elements in the human body, treatment was the most effective for the Se uptake in the soil and tree. Se treatments included foliar application, soil fertigation, and trunk injection. Se fertigation and control had similar soil $P_2O_5$, K, and Mg concentrations, and calcium and Se concentrations in the soil were greater on the control and Se fertigation, respectively. Leaf characteristics were not different among the treated trees. No differences were observed for the leaf K and Ca concentrations among the treated trees, and foliar Mg was greater on the Se treated trees than the control. Se foliar application and trunk injection had greater Se concentrations in the leaves and fruits than the Se fertigation and control.

Nematicidal Activity of Eclipta prostrata Extract and Terthiophene against Pine Wood Nematode, Bursaphelenchus xylophilus (한련초(Eclipta prostrata) 추출물과 terthiophene의 소나무재선충(Bursaphelenchus xylophilus)에 대한 살선충 효과)

  • Pine wood nematode, Bursaphlenchus xylophilus is a fatal damaging pest, infecting pinewood trees and is one of the serious forest pest in Korea. Trunk injection is one of the most effective method to protect the live pine trees from nematode infection. The aim of this research was to reveal the efficacy of the nematicidal materials from 46 Kyrgyzstan, 88 Vietnam plant materials and 21 active materials (${\alpha}$-terpinene, ${\alpha}$-pinene, ${\beta}$-thujaplicin, cinnamaldehyde, eugenol, emodin, geraniol, limonone, methyl palmitate, matrine, myrcene, methyl gallate, nicotine, quassin, resveratrol, rotenone, thymol, thujaplicin, terthiophene, tuberstemonine, ${\gamma}$-terpinene) isolated from plants for using as trunk injection agents. Eclipta prostrata extract was highly effective against pine wood nematode in plant extracts and terthiophene has the best nematicidal activity from active materials isolated from plants. Foliar spray of terthiophene on Japanese black pine (Pinus thunbergii) tree reduced proliferation of inoculated pine wood nematode at 22 days after treatment, however no efficacy was found over 22 days. Eclipta prostrata extract and terthiophene can be used as nematicide for further nematicidal efficacy test against any other plant parasitic nematodes and trunk injection efficacy against pine wood nematode.

Chemical control of Sycamore Lace Bug, Corythucha ciliata(Say) (버즘나무방페벌레의 약제방제(藥劑防除)에 관(關)한 연구(硏究))

  • For the control of Corythucha ciliata, the insecticidal activity of five commercial pesticides (Monocrotophos 25%SL, Phosphamidon 50%SL, Imidacloprid 20%DC, Thiamethoxam 15%DC and Acetamiprid 20%SL) were examined with trunk injection method in Seoul, Sanbon and Chungju. The insecticidal activity of five commercial pesticides was >88%. The insecticidal activity of Monocrotophos maintained 31 days later after trunk injection when treated leaves were supplied to this pest. This result indicates that one application of trunk injection is enough to control this pest, and this method can save the control cost compared with ground application which should be applied two or three times per year to control this pest. To make the model of treatment dosage at each D.B.H class (cm), $1.0m{\ell}/DBH$, $1.5m{\ell}/DBH$ and $2.0m{\ell}/DBH$ pesticide dosage was used. At <20cm, the insecticidal activity of $1.0m{\ell}$ dosage was >95%, and $1.5-2.5m{\ell}$ dosage was needed for the effective control at 30~50cm. More than $2.5m{\ell}$ dosage was effective at >50cm. Spray method was used in laboratory to select pesticides for ground application. Three commercial pesticides (Ethofenprox 20%EC, Ethofenprox 10%WP and Cyfluthrin 1%EC) were used, and all pesticides showed >95% insecticidal activity.

Occurrence of Jujube Witches' Broom Disease and Control Efficacy of Trunk Injection with Oxytetracycline in Boeun Area from 2018 to 2019 (2018-2019년 보은지역 대추나무 빗자루병 발생 및 옥시테트라사이클린 수간주사 방제 효과)

  • Jujube witches' broom (JWB) disease is a high-risk plant disease that occurred on and devastated jujube orchards throughout South Korea in the 1950s. Recently, as the jujube cultivation area has been significantly increased nationwide, we investigated the disease in jujube orchards in the Boeun region to obtain basic information for proactively evaluating the risk of JWB outbreaks from 2017 to 2019. A field trial was conducted to evaluate the effect of the trunk injection of Sungbocycline (a. i. oxytetracycline calcium alkyl trimethyl ammonium 17%, OTC). The incidence of JWB disease was very low in Boeun, but it was gradually increased mainly in the area and on the farms where it had occurred previously. The trunk injection with OTC was most effective in the early stages of growth. However, even though the trees were treated with OTC, disease symptoms appeared in other healthy branches in severely affected trees. When the tree was first injected with OTC in June, the control efficacy of second injection was higher one month after the first injection than five months. These results suggest that early diagnosis based on various symptoms and early control based on them can be very effective in preventing JWB disease in advance and increasing control efficacy.
