Studies on the Isolation of Phycobiliprotein from S. platensis (S. platensis로부터 phycobiliprotein의 분리에 관한 연구)

  • Isolation of phycobiliprotein from S. platensis was performed by using $30-60{\%}$ ammomium sulfate fractionation, Sephadex G-100 gel filtration and DEAE-Sephacel anionic exchange chromatography. Isolated phycobiliprotein was determined to be a c-phycocyanin with a maximum absorption wavelength at 620 nm. This phycobiliprotein consisted of $({\alpha}$ and $({\beta}$ subunit when analyzed through SDS-PAGE. The molecular weights of $({\alpha}$ and $({\beta}$ subunit were 14.5 kDa and 16 kDa, respectively. The native molecular weight of phycobiliprotein through gel filtration was about 100 kDa. These results show that the structure of phycobiliprotein from S. platensis might be aggregated form of $({\alpha}{\beta})_{3}-trimer$.

Effect of Chitosan-Trimer on the Prevention of Postoperative Intraperitoneal Adhesion Formation in Rats (랫트에서 Chitosan-Trimer가 복강유착에 미치는 영향)

  • This study was performed to investigate the effects of chitosan-trimer (CT) on the prevention of postoperative adhesion formation in the rate model. All animals divided into PBS (control), 1% CT, 3% CT, and chitin treated group. The mean adhesion score in 1% CT group (1.03$\pm$0.63), 3% CT group (0.64$\pm$0.53) and chitin group (1.67$\pm$0.71) was found to be lower than that in control group (2.07$\pm$0.81). More favorable adhesion prevention was achieved in 3% CT group (0.64$\pm$0.53) in comparison with the control group, 1% CT group, and chitin group without any hemorrhagic complications. A statistically significant difference was observed in adhesion formation between control group and 3% CT group (p<0.001). In control group, 44 of 45 sites (97.7%) formed adhesions between the intestine defects. In contrast, 3% CT was effective in reducing the incidence of adhesion formation to 17 to 45 sites (62.2%) (p<0.05). The locations of adhesions were observed in serosa-serosa (60%), serosa-mesentery (13.3%), serosa-connective tissue of testis (10%), omentum-liver (10%), serosa-omentum (3.3%), serosa-cecum (3.3%), and serosa-incision (0%). On the results of histological analysis, grade of inflammation and fibrosis at the sites of postoperative peritoneal adhesion formation were not significantly different in all groups. But, 3% CT showed the lowest score of inflammation and fibrosis. In 3% CT group, the rate of increase of plasma fibrinogen was significantly lower compared with that in control group from pre-operation to 10 days later (p<0.05). There were no appreciable difference in the CBC, leukocyte differential counts and total protein concentrations among four groups. In conclusion, our data suggested that CT should be effective on reducing adhesion formation in experimental rat models. The results also showed that 3% CT does not adversely affect normal wound healing and healthy recovery after operation.

Physical Properties of High-Solid Coatings with Acrylic Resins Containing Acetoacetoxy Group and Allophanate-Trimer (Acetoacetoxy기 함유 아크릴수지와 Allophanate-Trimer에 의한 하이솔리드 도료의 도막물성)

  • Copolymers(HSA-98-20, HSA-98-0, HSA-98+20) which we acrylic resin containing 80% solid content were synthesized by the reaction of monomers, including methyl methacrylate, n-butyl acrylate, and 2-hydroxyethyl acrylate with a functional monomer, acetoacetoxyethyl methacrylate (AAEM), which nay give improvements in cross-linking density and physical properties of films. The physical properties of prepared acrylic resins, containing AAEM, are as follows viscosities, $1420\sim5760cps$ ; number average molecular weight, $2080\sim2300g/mol$; polydispersity index, $2.07\sim2.19$ ; and conversions, $88\sim93%$. In the next step, high-solid coatings (HSA-98-20C, HSA-98-0C, HSA-98+20C) were prepared by the curing reaction between acrylic resins containing 80% solid content and isocyanate at room temperature. Various properties were examined on the film coated with the prepared high-solid coatings. The introduction of AAEM to the coatings enhanced the abrasion resistance and solvent resistance, which indicated the possible use of high- solid coatings for top-coating materials of automobile. Since the curing by viscoelastic measurement occurred in sequence of HSA-98+20C > HSA-98-0C > HSA-98-20C, it was concluded that the curing rates became faster with incresing $T_g$ values.

Comparison of Thermal Stress Induced Heat Shock Factor 1 (HSF1) in Goldfish and Mouse Hepatocyte Cultures (붕어와 마우스의 간세포 배양에서 열 스트레스에 의해 유도되는 heat shock factor1 (HSF1)의 비교)

  • Heat shock proteins (HSPs) are induced in response to various physiological or environmental stressors. However, the transcriptional activation of HSPs is regulated by a family of heat shock factors (HSFs). Fish models provide an ideal system for examining the biochemical and molecular mechanisms of adaptation to various temperatures and water environments. In this study, we examined the pattern differentials of heat shock factor 1 (HSF1) and expression of heat shock protein 70 (HSP70) in response to thermal stress in goldfish and mouse hepatocyte cultures by immune-blot analysis. Goldfish HSF1 (gfHSF1) changed from a monomer to a trimer at $33^{\circ}C$ and showed slightly at $37^{\circ}C$, whereas mouse HSF1 (mHSF1) did so at $42^{\circ}C$. This experiment showed similar results to a previous study, indicating that gfHSF1 and mHSF1 play different temperature in the stress response. We also examined the activation conditions of the purified recombinant proteins in human HSF1 (hmHSF1) and gfHSF1 using CD spectroscopy and immune-blot analysis. The purified recombinant HSF1s were treated from $25^{\circ}C$ to $42^{\circ}C$. Structural changes were observed in hmHSF1 and gfHSF1 according to the heat-treatment conditions. These results revealed that both mammal HSF1 (human and mouse HSF1) and fish HSF1 exhibited temperature-dependent changes; however, their optimal activation temperatures differed.

  • NiSi2(111)을 Si(111) 표면 위에 증착하여 그 구조를 STM으로 연구하였다. 실리사이드는 그 표면이 (1$\times$1)의 구조를 띠고 여분의 trimer를 흡착 분자 구조로 가짐을 관찰하였다. 이 trimer로부터 실리사이드의 두 가지 구조 A, B를 구별할 수 있었다. 표면에 관착된 결함으로부터 표면구조는 내부와 같음을 보였다.

  • Oligomeric extracts of poly(ethylene terephthalate-co-ethylene isophthalate)s [(PET/EI] are analyzed by high performance liquid chromatography(HPLC) and nuclear magnetic resonance spectroscopy (NMR). Existence of separated peaks for small cyclics of trimer and tetramer gives the existence of structural isomeric forms. NMR confirms that cyclization of PET/EI occurs more easily at the site of isophthaloyl unit.

  • Alpha-viniferin was isolated from Carex humilis (Cyperaceae), and showed anti-inflammatory effects on carrageenin or histamine-induced paw edema in mice. To understand mode of the anti-inflammatory action. effects of alpha-viniferin on cyclooxygenase (COX)-2, iNOS, oxygen radicals and proinflammatory cytokines have been analyzed. Alpha-viniferin showed selective inhibitory effect with an IC50 value of 5 $\mu\textrm{m}$ on COX-2 activity but showed weak inhibitory effect on the synthesis of COX-2 transcript which was identified by RT-PCR.   • In order to give a silk-like touch to PET fabrics, the PET fabrics were treated with NaOH alkaline solution in various conditions. In alkaline treatment, the liquor flow type pilot weight reduction apparatus with magnetostrictive ultrasonic transducer was used for the study. The effects of ultrasonic application, treatment time and temperature at NaOH 4% and 6"A solution on the decomposition rate of PET fabrics. From the results of the decomposition rate of PET fabrics, the qualitative and quantitative analysis of oligomer after decomposition of PET fabrics carried out by the HPLC. On the other hand, the surface pore characteristics of decomposition PET fabrics measured by porosimetery. The pore characteristics on the surface of treated PET fiber depended on the decomposition rate and did not depend on the ultrasonic cavitation. The pore diameter of alkaline untreated PET fiber were 15A and those of treated PET fibers were 5~6$\AA$ at the maximum pore volume. The average pore sizes of fiber before and after treatment were 141 h and 160h, respectively. Total amount of oligomer of the untreated PET fibers were 1.70wt% and 67.7% of total oligomer occupied with PET cyclic trimer and PET cyclic tetramer. Total amount of oligomer of fiber with 26.9% and 48.0% of weight loss without ultrasonic application were 1.78wt% and 1.79wt%, respectively. Also total amount oligomer of fibers which were reduced 27.7% and 48.2% of weight loss with ultrasonic application were 1.74wt%. This result showed that the removal rate of oligomer in the process of alkaline hydrolysis with ultrasonic higher than that of without ultrasonic application.tion.

Synthesis of Polyacrylates Containing Si, Ge and Sn for High Refractive Index (실리콘, 게르마늄, 주석이 결합된 고굴절률 아크릴 고분자의 합성)

  • New seven acrylic monomers with covalently bonded silicon, germanium, and tin were prepared for high refractive index materials. The monomers were copolymerized with a cross-linkable comonomer (Trimer) to prepare UV-films for optical characterization. The refractive index of the copolymers increased in proportion to the monomer content and extrapolated to determine that of homopolymer. $Ph_3Si$, $Ph_3Ge$, and $Ph_3Sn$ groups contributed to increase the refractive index of acrylic polymer, in which $Ph_3Sn$ was more effective than $Ph_3Ge$. The index increment confidently occurred with $Bu_3Sn$ attachment in comparison with aliphatic acrylic polymers. $Ph_3SnS$-attached acrylate polymer showed a refractive index of 1.671 at 589 nm. The index change was similarly observed at various different wavelengths (656, 830, 1310, and 1550 nm).
