Effects of pH on Purification of GFPuv/Cytochrome c-552 Fusion Protein

  • Fusion gene of GFPuv and Cytochrome c-552 was inserted into the pTrcHis B vector and transferred to E. coli. A fusion protein of GFPuv and Cytochrome c-552 was expressed in BL21. This fusion protein was composed of a His-tag for purification using an immobilized metal affinity chromatography(IMAC). IMAC constitutes a rather facile means of unravelling the principles of recognition and, in particular, of identifying the counterligands on the protein surface, which interact with the ligated and immobilized metal ions. Histidine when present on the surface of a protein molecule under a favorable solvent condition, may serve as electron donors in coordination with the immobilized chelates of some transition metal ions$(Ni^{2+})$.

  • The design and preparation of novel dye rotaxanes have gained much interest recently, since such structure usually exhibits peculiar spectral and optical changes. In spite of the promising results to date, increasing pressure remains to develop novel supramolecular structures based on stimuli-responsive systems. This presentation covers the study of inclusion complexes of cyclodextrins and various chromophores, with an emphasis on our most recent outcome of anisotropic hydrogel. In this system, physical gelation prepared from simple mixture of CD and a azo dye is completed through specific host-guest interaction. The obtained hydrogel exhibits respective morphological transitions based on supramolecular assembly and dissociation, leading to either precipitation or a sol-to-gel transition. It can identify different classes of metal ions, and, among them, naked-eye differentiation of lead ion is possible due to the coordination-induced unthreading of dye molecules. Accompanying structural changes were verified by numerous characterization techniques, including 2D-ROESY, HR-MAS, UV-Visible absorption, small-angle X-ray scattering, and induced circular dichroism measurements. Such properties discussed here will find useful in analytical applications, such as metal ion sensing and removal applications.

Silicidation and Thermal Stability of the So/refreactory Metal Bilayer on the Doped Polycrystalline Si Substrate (Co/내열금속/다결정 Si 구조의 실리사이드화와 열적안정성)

  • Silicide layer structures and morphology degradation of the surface and interface of the silicide layers for he Co/refractory metal bilayer sputter-deposited on the P-doped polycrystalline Si substrate and subjected to rapid thermal annealing were investigated and compared with those on the single Si substrate. The CoSi-CoSi2 phase transition temperature is lower an morphology degradation of the silcide layer occurs more severely for the Co/refractorymetal bilayer on the P-doped polycrystalline Si substrate than on the single Si substrate. Also the final layer structure and the morphology of the films after silicidation annealing was found to depend strongly upon the interlayer metal. The layer structure after silicidation annealing of Co/Hf/doped-poly Si is Co-Hf alloy/polycrystalline CoSi2/poly Si substrate while that of Co/Nb is polycrystalline CoSi2/NbSi2/polycrystalline CoSi2/poly Si.

  • It is observed that an appreciable short-circuit current (Is) flows by the time variation of temperature without applying external field in M1(metal)-P(polymer)-M2(metal)system. In M1-P-M2(A1) system, Is flows in the direction from the electrode(A1) having a lower workfunction to the counter electrode(M1) during heating and its magnitude increases as the thickness of polymer is decreased and as the heating rate is raised. The sign of Is is reversed in lower temperature region (under glass transition temperature) when the direction of temerature variation is changed during heating and cooling. From these experimental results, we can sugest that Is flows in the external short-circuit during the space charge distribution formed around both interfacial surfaces (M1-P and P-M2) is continuously maintained in the non-equilibrium state but not in equilibrium state.

  • Low power bridge type micro gas sensors were fabricated by micro machining technology with TMAH (Tetra Methyl Ammonium Hydroxide) solution. The sensing devices with different heater materials such as metal and poly-silicon were obtained using CMOS (Complementary Metal Oxide Semiconductor) compatible process. The tellurium films as a sensing layer were deposited on the micro machined substrate using shadow silicon mask. The low power micro gas sensors showed high sensitivity to NO with high speed. The pure tellurium film used micro gas sensor showed good sensitivity than transition metal (Pt, Ti) used tellurium film.

  • Laser forming is a promising technology in manufacturing, such as in the shipbuilding, automobile, microelectronics, aerospace and other manufacturing industries. This process forms the sheet metal by utilization of laser-induced thermal stresses. Laser forming process has been studied extensively for rectangular shape geometry. This basic study presents the change in deformation behavior of sheet metal during transition from linear to curved geometries and irradiations as well. A series of experiments have been conducted on a wide range of specimen geometries such as quarter-circular and half circular plate. The reasons for this behavior have been analyzed. Results are compared and analyzed by simulations using ABAQUS. Influence of developed stresses on the bending has been investigated. This study provides the more understanding of forming mechanism influenced by geometry effect.

Analysis of Metal Transfer using Dynamic Force Balance Model in GMAW (동적 힘 평형 모델을 이용한 GMA 용접의 용적이행 해석)

  • A dynamic force balance model is proposed in this work as an extension of the previous static force balance model to predict metal transfer in arc welding. Dynamics of a pendant drop is modeled as the second order system, which consists of the mass, spring and damper. The spring constant of a spherical drop at equilibrium is derived in the closed-form equation, and the inertia force caused by drop vibration is included in the drop detaching condition. While the inertia force is small in the low current range, it becomes larger than the gravitational force with current increase. The inertia force reaches half of the electromagnetic force at transition current, and has considerable effects on drop detachment. The proposed dynamic force balance model predicts the detaching drop size more accurately than the static force balance model.

  • Metal/poly (4,4'-aminotriphen-ylene hexafluoroisopropylidenediphthalimide) (TP6F PI)/metal structure exhibited an electrically volatile phase transition with high (OFF) or low (ON) resistive states when voltage between electrodes swept. Here, we demonstrate a noble set-up in which holes are injected by photoelectron emission process during the voltage sweep instead of direct charge carrier injection via metal electrode, which enables direct investigation into changed electronic structures of TP6F PI both in ON and OFF states using photoemission spectroscopy methods. In the I-V measurement, TP6F PI shows a non-volatile behavior. In spectroscopic results, this non-volatile behavior is leaded from the structural modification of the O=C double bond in phthalimide of TP6F PI by hole injection.

Nanoconfinement effects of MCM-41 on the thermal decomposition of metal borohydrides

  • We used differential scanning calorimetry and a thermogravimetric analysis to investigate the effect of being confined in mesoporous MCM-41 on the decomposition of lithium borohydride and magnesium borohydride when heated. The confinement did not cause a phase transition of the metal borohydrides inside MCM-41, but did lower their decomposition temperature. With the exception of a lowering of the temperature, the decomposition reaction mechanism of the metal borohydrides was nearly the same for both the bulk and confined samples.

  • Nanoporous or nanotubular metal oxide can be fabricated by anodization of metal substrate in fluoride contained electrolytes. The approach allows various transition metals such as Zr, Hf, Nb, Ta to form highly ordered oxide nanostructures. These oxide nanostructures have various advantages such as high surface area, fast electron transport rate and slow recombination in semiconductive materials. Recently, vanadium oxide nanostructures have been drawn attentions due to their superior electronic, catalytic and ion insertion properties. However, anodization of vanadium metal to form oxide layers is relatively difficult due to ease formation of highly soluble complex in water contained electrolyte during anodization. Yang et al. reported $[TiF_6]^{2-}$ or $[BF_4]^-$ in electrolyte helps to formation of stable oxide layer [1, 2]. However, the reported approaches are very sensitive in other parameters. In this presentation, we deal with the other important key parameters to form ordered anodic vanadium oxide such as pH, temperatures and applied potential.
