Metallothionein, a cysteine-rich stress response protein that is naturally induced by a variety of immunologic stressors, has been shown to suppress autoimmune disorders through mechanisms not yet fully defined. In the present study, we examined the underlying mechanisms by which metallothionein might mediate such regulation of autoimmunity. $Na\ddot{i}ve\;CD4^+$ T cells from metallothionein-deficient mice differentiated to produce significantly less IL-10, $TGF-{\gamma}$, and repressor of GATA, but more $IFN-{\gamma}$ and T-bet, when compared with those from wild-type mice. The levels of IL-4 and GATA-3 production were not different between the two groups of mice. Conversely, treatment with exogenous metallothionein during the priming phase drove $na\ddot{i}ve$ wild-type $CD4^+\;T$ cells to differentiate into cells producing more IL-10 and $TGF-{\beta}$, but less $IFN-{\gamma}$ than untreated cells. Metallothionein-primed cells were hyporesponsive to restimulation, and suppressive to T cell proliferation in an IL-10-dependent manner. Lymphocytes from metallothionein-deficient mice displayed significantly elevated levels of AP-1 and JNK activities in response to stimulation compared with those from wild-type controls. Importantly, transgenic mice overexpressing metallothionein exhibited significantly reduced susceptibility to collagen-induced arthritis and enhanced IL-10 level in the serum, relative to their nontransgenic littermates. Taken together, these data suggest that metallothionein is able to promote the generation of IL-10-and $TGF-{\beta}$-producing type 1 regulatory T-like cells by downregulating JNK-dependent AP-1 activity. Thus, metallothionein may play an important role in the regulation of Th1-dependent autoimmune arthritis, and may represent both a potential target for therapeutic manipulation and a critical element in the diagnostic assessment of disease potential.
Transgenic animals are produced primarily by microinjecting exogenous DNA into the male pronuclei of a zygote. Microinjection method for gene transmitting is successful in mice but not efficient in farm animals, limiting it's general utility such as a large scale facility and labour. Based on our finding that sperm cells bind with exogenous DNA, sperm was used as a vector for producing transgenic animals to introduced green fluorescence protein(GFP) gene. (omitted)

Recently, production of transgenic animal by nuclear transfer has been known as a useful method. The production of cloned offspring derived from nuclear transfer depends upon a variety of factors such as species, donor cells type and cell cycle, and source of recipient ova. Therefore, we attempted a different transgenic methods using follicular granulosa cells (GCs). In general, ovulated GCs undergoes lutenization and transformation in vitro which might defective effects on developmental potential. In order to avoid the GCs transformation in vitro culture system, we introduced a direct injection of retrovirus into the follicles and then collected them mechanically from ovaries of 6-8 week-old ICR mice. Retrovirus vector constructed with pLN $\beta$ EGFP was injected into the follicles. The follicles are cultured in $\alpha$ -MEM supplemented with human FSH, LH and ITS in Costar Transwell dish for 4 days. Survival rate of virus injected follicles was 52.1% (12/23) and expression rate of EGPP gene was 33.3% (4/12). In this study, we found GCs performed transgenesis in our culture system. In addition, the GCs in follicle may be developed in vivo like environment rather than in vitro environment. Thus, the use of GCs as donor cells may be useful in the nuclear transfer for cloning of genetic modification. Therefore, these results suggest that follicular GCs can be transfected by viral vector during folliculogenesis in vitro.
Inositol 1,4,5-trisphosphate ($IP_3$) plays an important role in the release of $Ca^{2+}$ from intracellular stores into the cytoplasm in a variety of cell types. $IP_3$ translocation dynamics have been studied in response to many types of cell signals. However, the dynamics of cytosolic $IP_3$ in salivary acinar cells are unclear. A green fluorescent protein (GFP)-tagged pleckstrin homology domain (PHD) was constructed and introduced into a phospholipase C ${\delta}1$ (PLC ${\delta}1$) transgenic mouse, and then the salivary acinar cells were isolated. GFP-PHD was heterogeneously localized at the plasma membrane and intracellular organelles in submandibular gland and parotid gland cells. Application of trypsin, a G protein-coupled receptor activator, to the two types of cells caused an increase in GFP fluorescence in the cell cytoplasm. The observed time course of trypsin-evoked $IP_3$ movement in acinar cells was independent of cell polarity, and the fluorescent label showed an immediate increase throughout the cells. These results suggest that GFP-PHD in many tissues of transgenic mice, including non-cultured primary cells, can be used as a model for examination of $IP_3$ intracellular dynamics.
유전적으로 결정되는 두개안면 기형의 발생 기전을 밝히기 위해 관련된 유전자의 기능 변화에 의한 효과를 이해하는 것이 필수적이다. 섬유아세포성장인자수용체-2 (FGFR2)의 활성형 돌연변이가 어퍼트 및 크루즌 증후군에서의 봉합의 조기유합의 원인이 된다고 알려져 있으나 인류에서는 다양한 개인차가 존재하므로 임상적으로 정의된 두 증후군에서의 유전형-표현형의 상관관계에 대해서는 의문이 제기되어 왔다. 본 연구의 목적은 어퍼트(Pro253Arg)및 크루즌(Cys278Phe) 돌연변이를 갖는 골특이성 FGFR2를 발현하도록 제작된 형질변환 쥐에서 결과적인 표현형의 차이를 분석하여 유전형에 의한 기형 형성의 인과관계를 추정하기 위한 것이다. 유전자 조작을 하지 않은 정상군과 정상 FGFR2 유전자를 가진 군을 대조군으로 하여 육안 관찰 및 micro-CT를 이용한 형태계측학적 방법으로 주로 전방두개 및 전두개저 부위의 이상을 분석하여 다음과 같은 결론을 얻었다. 첫째, 어퍼트 및 크루즌 돌연변이를 갖는 각각의 형질변환 쥐는 두개 봉합의 유합과 전후방적 두부 길이 감소를 공히 보였으나 어퍼트 형질변환 쥐에서만 전치부 반대교합이 나타났다. 또한 어퍼트 개체는 크루즌 개체 및 대조군에 비해 전방두개 및 전두개저 굴곡에 있어서 정상군과 비교해 차이를 나타냈으며 이는 유합을 보이는 봉합의 부위 및 순서에 있어서의 차이에 기인하는 것으로 사료된다. 둘째, 정상 FGFR2 유전자를 주입한 형질변환 쥐는 정상적인 두개안면 형태를 보였다. 이상의 결과를 토대로 어퍼트 및 크루즌 돌연변이는 각각의 유전형에 특이한 두개안면 기형을 유발할 것으로 보이며 정상 FGFR2의 발현 강도보다는 기능의 이상이 두개골 유합과 관련이 있는 것으로 사료된다. 변형된 FGFR2와 각 봉합에서의 기능과의 상관성은 추가 연구가 필요할 것으로 사료된다.EX> 와이어에서 열처리만 시행한 실험군이 열처리를 시행하지 않은 군보다 더 낮은 austenite finish ($A_f$) 온도를 보였다. $0.018"\;{\times}\;0.025"$ 및 $0.0215"0.028"$ 와이어에서 열처리를 시행하여 굴곡을 부여한 실험군은 열처리만 시행한 실험군과 열처리를 시행하지 않은 대조군에 비해 부하-변위 곡선이 상방 이동되었으며, 열처리 시간을 1초 증가시켜 굴곡을 부여한 실험군에서 가장 높은 부하-변위 곡선을 나타냈다. $0.018"\;{\times}\;0.025"$ 그리고 $0.0215"\;{\times}\;0.028"$ 와이어에서 $A_f$ 온도는 열처리 시간을 1초 증가시켜 굴곡을 부여한 실험군에서 가장 낮게 관찰되었고 열처리를 시행하여 굴곡을 부여한 실험군, 열처리만 시행한 실험군 그리고 열처리를 시행하지 않은 대조군 순으로 높게 관찰되었다. 이상의 결과를 종합할 때, 임상에서 니켈-티타늄 합금 와이어에 굴곡을 부여하기 위해 열처리하는 경우 초탄성 특성은 유지될 수 있으나, 부하-변위 곡선의 상방 증가가 나타나므로, 와이어에 의한 교정력이 증가될 수 있음에 유의하여야 한다. $day^{-1}$인 인공습지), scenario 2(면적 4.2ha인 저류지)가 각각 연평균 6.9%, 4.8%, 7.1%의 감소를 보였다. TN은 4.7%, 3.4%, 13.4%의 삭감율을 나타내었으며, TP는 5.6%, 3.9%, 7.3%의 삭감율을 나타내었다. 본 연구에서는 적용하지 못하였으나, 인공습지와 저류지의 적절한 연계시스템을 적용한다면 저감시설 설치 부지면적과 비용의 감소뿐만 아니라 보다 효과적인 수질개선효과를 가져올 수 있으리라 판단된다.

본 연구의 목적은 inbred 마우스 (C57BL/6J)의 수정란을 이용하여 형질전환마우스를 생산할 때, 수정란이식의 효율성을 증진시키기 위한 것이다. C57BL/6J 및 BCF1 마우스로부터 과배란처리 방법에 의해 수정란을 얻고, DNA를 1 세포기 수정란에 미세 주입한 다음, 1세포기 또는 2 세포기의 수정란을 가임신된 마우스의 한쪽 또는 양쪽 난관에 각각 이식하였다. 1세포기의 수정란을 0.75 d.p.c. 가임신된 마우스의 한쪽 난관에 이식했을 때, 임신율이 C57BL/6J는 68.8$\pm$7.83%, BCF1은 48.3$\pm$14.22% 이었고, 이식한 수정란 당 산자의 발달율은 C57BL/6J가 11.9$\pm$5.51%, BCF1은 10.5$\pm$8.03%로 성적이 저조하였다. 그러나, 2세포기의 수정란을 0.5 d.p.c. 가임신된 마우스의 양쪽 난관에 이식했을 때, 임신율이 C57BL/6J는 94.4$\pm$9.64%, 13CFl은 100$\pm$0% 이었고, 이식한 수정란 당 산자의 발달율은 C57BL/6J가 22.1 $\pm$0.4%, BCF1은 21.8$\pm$0.38%였다. 따라서 C57BL/6J 마우스의 2세포기 수정란을 0.5 d.p.c. 가임신된 마우스의 양쪽 난관에 이식하는 것이, BCF1마우스와 유사한 성적을 얻어 경쟁력이 있는 것으로 판단되었다. 이러한 결과에 영향을 미치는 인자가 여러 가지 있을 것으로 판단되지만, C57BL/6J 마우스의 2세포기 수정란을 0.5 d.p.c.가임신된 마우스의 양쪽 난관에 이식하는 방법이 다른 방법보다 형질전환마우스를 생산하는데 효율성이 더 높은 것으로 본 실험에서 확인되었다.
본 연구는 사람의 조혈촉진 유전자(hEPO)가 도입된 형질전환 돼지를 생산하기 위해 사계절동안 수행하였다. 약 8∼15개월령의 순종의 랜드레이스 경산돈 및 미경산돈 42두는 유전자 미세주입을 위한 1세포기 단계의 수정란 채란 및 이식을 위해 사용하였으며, 발정동기화 및 과배란 방법은 PG 600 주입 후 9일간 매일 20mg의 altrenogest를 사료에 첨가하여 급여하였다. Altrenogest를 9일간 급여 후 1,500IU의 PMSG와 500IU의 hCG를 주입하므로서 과배란을 유도하였다. 미세주입을 위한 유전자는 mouse whey acidic protein(mWAP) 프로모터에 hEPO 유전자를 연결하여 준비하였으며, 호르몬 처리후 23두의 공란돈으로 부터 650개의 난자를 회수하였으며, 이 중 83.1%(540/650)는 DNA 미세주입을 위해 전핵을 관찰할 수 있는 1-세포기의 수정란이었다. 이중 유전자가 미세주입 된 543개의 난자를 19두의 수란돈에 이식하였으며 7두의 임신돈으로부터 47두의 자돈을 생산하였다. 생산된 자돈 47두로부터 꼬리조직으로부터 분리된 DNA의 PCR 검정 결과 수컷 1두가 형질전환 양성반응을 나타내어 2.13%의 형질전환율을 나타내었으며, 이러한 연구의 결과는 생체반응기(bioreactor)연구에 있어서 형질전환 돼지생산의 성공적이며 유용한 정보를 제공할 것으로 사료된다.
The 2-year rodent carcinogenicity test involves long-term, repetitive dosing of animals that is both time consuming and expensive. Alternative approaches have been attempted using specific transgenic or knockout mice or toxicogenomics to predict carcinogenicity without conducting a 2-year rodent test. In addition, toxicogenomic analysis of carcinogen-treated animals could also enhance our understanding of molecular mechanisms and aid in the diagnosis of acute toxicity induced by carcinogens. Therefore, we investigated transcription profiles after administering the carcinogens 4,4-dimethylformamide (DMF) and 4-biphenylamine (ABP). BALB/c male mice were treated once with DMF (650 mg/kg i.p.) or ABP (120 mg/kg p.o.). Standard blood biochemistry and histological changes were observed. Gene expression profiles in the livers of mice treated with either vehicle or the carcinogens were analyzed using the Affymetrix $GeneChip^{(R)}$ assay. In all, 1,474 differentially expressed genes in DMF- or ABP-treated mice were identified as being either up- or down-regulated over 1.5-fold (P< 0.01), and these genes were analyzed using hierarchical clustering and Ingenuity Pathways Analysis. Of these, 107 genes were consistently regulated in both carcinogen-treated groups. Genes associated with cancer were upregulated (Por, S100a10, Tes, Ctcf, Ddx21, Eapp, Nel, and Pa2g4) or downregulated (Cbs and Gch1). Toxicological function analysis also identified genes involved in organ toxicity, including hepatotoxicity. These data may help to identify molecular markers for acute hepatotoxicity induced by carcinogens.
Thioredoxin (TRX) protein is an antioxidant responsible for reducing other proteins by exchanging cysteine thiol-disulfide and is also known for its anti-allergic and anti-aging properties. On the other hand, epidermal growth factor (EGF) is an important material used in the cosmetics industry and an essential protein necessary for dermal wound healing facilitated by the proliferation and migration of keratinocytes. EGF also assists in the formation of granulation tissues and stimulates the motility of fibroblasts. Hence, genetically modified soybeans were developed to overexpress these industrially important proteins for mass production. A single-dose oral-toxicity-based study was conducted to evaluate the potential toxic effects of TRX and EGF proteins, as safety assessments are necessary for the commercial use of seed-specific protein-expressing transgenic soybeans. To achieve this rationale, TRX and EGF proteins were mass purified from recombinant E. coli. The single-dose oral-toxicity tests of the TRX and EGF proteins were carried out in six-week old male and female Institute of Cancer Research (ICR) mice. The initial evaluation of the single-dose TRF and EGF treatments was based on monitoring the toxicity signatures and mortality rates among the mice, and the resultant mortality rates did not show any specific clinical symptoms related to the proteins. Furthermore, no significant differences were observed in the weights between the treatment and control groups of male and female ICR mice. After 14 days of treatment, no differences were observed in the autopsy reports between the various treatment and control groups. These results suggest that the minimum lethal dose of TRX and EGF proteins is higher than the allowed 2,000 mg·kg-1 limit.
