시호(柴胡)가 Alloxan 투여가토혈청중(投與家兎血淸中) Cholesterol 함량(含量) 및 Transaminase 활성도(活性度)에 미치는 영향(影響) (STUDIES ON THE EFFECT OF THE BUPLEURI RADIX ON THE SERUM TRANSAMINASE ACTIVITIES AND THE CONTENTS OF TOTAL CHOLESTEROL IN SERUM FROM THE ALLOXAN-DIABETIC RABBITS)

  • The effects of root of the Bupleurum falcatum L. on the serum transaminase (SGP-T, SGO-T) activities and the content of total cholesterol in serum from normal and alloxan-diabetic rabbits were observed. The preparations were extracted by methanol, and extracted samples were orally administrated to the normal and alloxandiabetic rabbits. The transaminase activities and the content of total cholesterol in serum of normal rabbits were significantly increased by administration of the alloxan. The increased serum transaminase activities of the alloxandiabetic rabbits were significantly decreased after 96 hours by administration of the extracted samples. The increased content of total cholesterol in serum of the alloxan-diabetic rabbits were gradually decreased by administration of the extracted samples and were recovered to the control levels after 12 days. According to the above results, it would be concluded that the extract of Bupleuri Radix influences for the recovery of the increased contents of total cholesterol and serum transaminase activities by alloxan.

한우(韓牛)의 혈청(血淸) Transaminase 활성도(活性度)에 대(對)하여 (Serum Transaminase Activity of Korean Cattle)

  • The effects of sex and age on the serum transaminase activity of 57 healthy Korean cattle (22 females and 35 males) were observed. Serum glutamic oxalacetic transaminase (SGOT) activity of the female revealed significantly higher value than that of the males. In the females, SGOT activity of 4 and 5 years group was a little decreased than 2 and 3 years, but difference was not recognized. On the other hand, SGOT activity of 4 and 5 years showed high significant increase as compared with that of 2 and 3 years in the males. The effects of sex and age on serum glutamic pyruvic transaminase (SGPT) activity and total protein levels were not found.

  • Transaminase represents the most important biocatalysts used for the synthesis of chiral amines due to their stereoselectivity. They allow asymmetric synthesis with high yields and enantioselectivity from their corresponding ketones. Due to their environmentally friendly access for the preparation of chiral amines, they have attracted growing attention in recent times. Thus, the production of chiral compounds by transaminase catalysed reactions is considered as an important application in synthetic organic chemistry. Therefore, transaminase is considered to be an important enzyme in the pharmaceutical and chemical industries. ω-Transaminase holds great potential because of its wide substrate specificity thus making it a suitable enzyme to be used at an industrial scale. This review highlights the reaction mechanism, classification, substrate specificity, and biochemical properties. The review also showcases the application of ω-transaminase in organic chemistry with a focus on the production of active pharmaceutical ingredients (APIs).

  • Eight cattle and ten sheep were administrated various degree of PMA-B and EPN, and S-GOT, S-GPT and serum cholinesterase activity were tested. Serum cholinesterase activity showed no typical tendency, however, S-GOT/S-GPT ratios were decreased by the administration of PMA-B. It was suggested some degree of liver damage by the administration of the chemicals was recognized.

사염화탄소에 의해 유발된 생쥐의 간독성에 미치는 누에 추출물의 영향 (Effects of Bombyx mori Larvae Extracts on Carbon Tetrachloride-Induced Hepatotoxicity in Mice)

  • To evaluate the protective effects of extracts of silkworm on carbon tetrachloride-induced hepatotoxicity, serum glutamic-oxaloacetic transaminase, glutamic-pyruvic transaminase and lactic dehydrogenase activities, malondialdehydes values and glutathione-S-transferase activity were measured in ICR mice. Extracts of silkworm was administered orally at 30min after the administration of CCl4 Mice were sacrificed at 24h after the administration of extracts of silkworm. The activities of serum aminotransferase and the hepatic content of lipid peroxide after carbon tetrachloride treatment were markedly increased than normal control but those levels were decreased by the treatment of butanol soluble fraction of silkworm methanol extract. Glutathione S-transferase activity was decreased by carbontetrachloride than control, but also inhibited by the treatment of butanol soluble fraction of silkworm methanol extract.

Transaminase 활성에 관한 연구(제 1보) (The Studies on the Transaminase Activities(1))

  • Transaminase has been a popular methods in the clinical laboratory for the diagnosis of particular diseases. The methods we are familiar with should be considered further on the serum states provided on the test to be done. (serum freshness, temperature and duration preserved) I have studied transaminase activities of the serum preserved under frozen (-10$\circ$C), refrigerated (4$\circ$C) and room temperature conditions. At a given interval those activities of serum are measured and then the results are compared with the optical density and calculated for the standard deviation and percent variation (Table 1. & Fig. 1.) G.O.T. activity is maintained decreased at a relatively constant rate from 2nd day to 14th day preservation under frozen and refrigerated but its activity variation at 25$\circ$C are far greater than the above conditions throughout the test periods. Elevated G.P.T. level of serum compared with G.O.T. are remained at any temperature and duration. Unless the determination of amino acid level study can not ruled out clearly.

  • Effect of 4-nonylphenol (4-NP), endocrine disrupting compounds (EDCs), on glutamate oxaloacetate transaminase (GOT) and glutamate pyruvate transaminase (GPT) were investigated in the plasma of juvenile rockfish, Sebastes schlegeli. Fish were injected with 4­NP (10, 50, 100 and 200 mg/kg body weight) in $70\%$ ethanol twice at 3-day intervals and plasma sampling were extracted at 7 days after the last injection. Controls received solvent only. 4-NP significant increased GOT in a dose-dependent manner. GPT was markedly elevated to $61\%$ (P<0.05) and $82\%$ (P<0.01) than that of the control at the 4-NP doses of 100 and 200mg, respectively. These results suggest that the estrogenic activity of 4-NP increase plasma GOT and GPT by toxic effect on hepatocyte.

Dimethyl-esculetin이 Carbon-tetrachloride 부하가토혈청 Transaminase 활성도에 미치는 영향 (The Effect of Dimethyl-esculetin on the Serum Transaminase Activity of Rabbit Pretreated with Carbon-tetrachloride)

  • Antihepatotoxic effect of dimethyl-esculetin, the major constituent of herbal drug Artemisia capillaris $T_{HUNBERG}$ (Compositae) which has long been used as folk medicine for gall stone and hepatitis in Asian country (China and Japan, except Korea) was investigated on the rabbit intoxicated with carbon-tetrachloride. Ten rabbits were divided into two parts, group A and B in five each. The group A was injected with carbon-tetrachloride, 0.1ml per kg at begining of the experiments for control. The group B was injected with carbon-tetrachloride soon after 7 days treatment of dimethyl-esculetin, 30mg per day. The results obtained in this experiment were as follows.: 1) Antihepatotoxic activity of dimethyl esuletin on the rabbits intoxicated with carbon tetrachloride has shown the serum glutamic oxaloacetic transaminase activity and serum glutamic pyruvic transaminase activity decreased respectively. 2) The component of Artemisia capillaris, dimethyl-esculetin, has shown remarkable antihepatotoxic effect.

  • In order to evaluate the protective effects of extracts of Mori cortex radicis on carbon tetrachloride-induced hepatotoxicity, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, lactic dehydrogenase, malondiadehyde values and glutathione S-transferase activity were measured in ICR mice. Glutathione S-transferase activity was decreased by carbon tetrachloride than control, but that also inhibited by the treatment of butanol fraction of Mori cortex radicis methanol extract. These results demonstrate a possible hepatoprotective role of extract role of extract of Mori cortex radicis against ${CCl_4}-induced$ hepatoxicity in vivo.

바이러스성 하기도 감염 환자에서 간기능 이상과 중증도와의 연관성에 관한 연구 (A study about the relation between elev ated transaminase lev el and severity of viral lower respiratory tract infection in children)

  • 목 적 : 소아의 바이러스에 의한 급성 하기도 감염시간기능 이상을 동반하는 경우가 많다. 저자들은 바이러스성 하기도 감염시 간기능 이상이 질병의 중증도에 미치는 영향과 원인 바이러스, 연령별 분포에 대하여 알아보고자 본 연구를 시행하였다. 방 법 : 2003년 1월부터 2005년 6월까지 급성 하기도 감염으로 한림대학교 강남성심병원 소아과에 입원한 환아들의 비인두 분비물을 Hep-2세포를 이용하여 배양하였으며 의무기록지를 후향적으로 검토하였다. 비말 흡인 검사를 통한 바이러스 배양검사에서 양성결과가 나온 환아를 환자군(AST 혹은 ALT >45)과 대조군(AST 혹은 ALT ${\leq}45$)으로 나누어 연령, 성별, 진단명, 발열 기간, RR(Respiratory Rate)점수, RDAI(Respiratory Distress Assessment Instrument)점수, 산소흡입 여부, 입원 기간 등과 원인바이러스를 비교 분석하였다. 결 과 : 바이러스가 검출된 181례 중에서 기저 질환이 동반되거나 간독성 약제를 복용한 16례는 제외되었고, 환자군이 28례(17.0%), 대조군이 137례(83.0%)였다. 환자군과 대조군에서 연령, 성별 간에 유의한 차이는 없었으며 발열 기간, RR score, RDAI score, 산소흡입 여부, 입원 기간 등의 임상양상에서 유의한 차이점은 없는 것으로 나타났다(P>0.05). 바이러스 배양 결과 환자군에서는 RSV 17례(60.7%), parainfluenza virus 4례(14.3%), influenza B virus 4례(14.3%), adenovirus 3례(10.7%), influenza A virus 1례(3.6%)였고 대조군에서는 RSV 78례(56.9%) parainfluenza virus 28례(20.4%), influenza A virus 13례(9.5%), influenza B virus 9례(6.6%), adenovirus 6례(4.4%), coxsackie virus 3례(2.2%)로 양 군 간에 의미있는 차이는 없었으며 연령별 분포에서도 양 군 간에 큰 차이는 없었다. 결 론 : 본 연구에서 호흡기 바이러스 감염으로 진단된 환아 165례 중 환자군이 28례(17.0%)였고 대조군과의 비교 분석 결과 간기능 이상이 병의 중증도와 임상양상에 큰 영향을 미치지 않으며 원인 바이러스와 연령별분포 경향에서도 큰 차이는 없는 것으로 나타났다.
