Bangpungtongseong-san(BPTSS, Fangfengtongsheng-san in Chineses) is a traditional herbal formula comprising 18 medicinal herbs. In the present study, we performed the simultaneous analysis for four compounds of BPTSS and examined anti-allergic effects in human keratinocytes and mouse splenocytes. The column for separation of four compounds was used Luna C18 column and maintained at 40℃. The mobile phase for gradient elution consisted of two solvent systems. The analysis was carried out at a flow rate of 1.0 mL/min with PDA detection at 254 and 280 nm. To evaluate production and expression of Th2 chemokines, ELISA and RT-PCR were conducted in tumor necrosis factor (TNF)-α and interferon (IFN)-γ-stimulated HaCaT cells with or without BPTSS or silymarin, a positive control for skin inflammation. To measure Th2 cytokines, primary mouse splenocytes were treated with BPTSS and performed ELISA for interleukin (IL)-4, 5, 13. Calibration curves were acquired with r2>0.9999. The contents of geniposide, liquiritin, baicalin, and glycyrrhizin in BPTSS were 5.06 ㎎/g, 7.33 ㎎/g, 27.56 ㎎/g, and 7.81 ㎎/g, respectively. BPTSS reduced TARC and RANTES production and mRNA expression in TNF-α and IFN-γ-treated HaCaT cells. BPTSS inhibited IL-4, 5, and 13 production in mouse splenocytes. Our data will be a helpful information to upgrade quality control and anti-allergic effects of BPTSS.
최근 가상 현실 및 증강 현실 기술의 발달로 다양한 형태의 기술이 제안되고 발전되고 있다. 특히 가상의 학습공간이나 가상의 건축 공간들을 위한 기술로서 원하는 형태의 그림을 그릴 수 있는 가상 드로잉 툴은 필수적인 요소 중 하나로 볼 수 있다. 기존의 래스터 기반 드로잉 알고리즘은 공간 이동이 잦은 가상 현실에서는 부적합 할 수 있는데, 가상 현실의 특성상 근접하거나 멀어지는 경우가 많기 때문이다. 본 논문에서는 연필 및 에어브러시의 물리적인 특성을 고려하여 실제 결과와 흡사하면서도 확대 및 축소에 강건한 가상의 드로잉 툴 렌더링 알고리즘을 제안한다. 캔버스로부터의 거리, 분사되는 잉크의 양, 움직이는 속도, 마찰력, 압력 등을 고려한다. 드로잉의 기본 단위는 정사각형 패치이며 각 패치는 슈도 랜덤값을 기반으로 하는 입자 형태의 이미지를 가진다. 이러한 입자 패치는 슈도 랜덤 식을 사용하고 매 프레임 마다 재 생성되기 때문에 확대 축소 시 깨짐 현상이 발생하지 않는다. 제안된 알고리즘은 2차원 환경 및 가상 현실 환경에 구현되었다. 2차원 환경은 안드로이드 기반 스마트폰에서 구현하였으며 가상 현실 환경은 언리얼 엔진 4를 사용하여 구현하였다. 구현 결과 확대 축소 시에도 이질감 없이 원래의 질감을 유지하는 렌더링 결과를 얻을 수 있었다.
The development of Korean glaze originated from the development of ash glazes. Ash not only has strong solubility but also can shows the glaze a variety of different colors according to what the glaze has in it as the main component. In addition, it gives a feeling of lucidness and softness. For these reasons, there are a lot of needs for ash and many potters want to take advantage of ashes as glaze. But natural ashes have not been widely used as glaze primarily because it is relatively hard to find or manufacture. Considering the difficulty of finding or manufacturing natural ashes, this study aims to formulate synthetic ash which not only is available to the potters in general but also has the sam ε characteristics as the natural ashes. To achieve this aim, this study examines the characteristics of the pine tree ash, the main component of the glaze of celadon porcelains, and the red pepper stem ash, the main component of the brown glaze, both of which have been used by the Korean traditional potters. In this study, the alkaline component of the glaze. A important ingredient when the ashes are synthesized, was supplied by mixing of $Na_2CO_3$, Chungju limestone and rice straw ash. Furthermore, the synthetic ash, when it shows no change of pH in its composition rate of 6:2:2, was found to be usable as the most stable material. In conclusion, the formula which frits some raw $materials-Na_2CO_3$, Chungju limestone and rice straw ash-can duplicate the synthetic ash which is simila to the natural ash.

Objectives: Naesohwangryeon-tang (NHT) is a type of traditional herbal formula, however, little is known about its antitumor activity. In this study, the antitumor properties of NHT was evaluated in human lung adenocarcinoma cells. Methods: To check the inhibitory effect of NHT, MTT assay was performed. Cell cycle analysis and detection of ROS production were conducted by flow cytometry. To evaluate the signaling pathway, Western blotting was conducted. Results: Our results showed that the decrease of cell proliferation by NHT stimulation occurred more significantly in A549 cells than in NCI-H460 cells. In addition, NHT-induced apoptosis was associated with the activation of caspases and production of reactive oxygen species (ROS). NHT-induced apoptosis was attenuated after pretreatments with z-VAD-fmk or N-acetylcysteine, suggesting that NHT-induced apoptosis was caspaseand ROS-dependent. Interestingly, NHT treatment led to the development of autophagic vesicular organelles and upregulation of several autophagy-related genes. The pretreatment of bafilomycin A1 decreased apoptosis slightly but increased cell viability in the presence of NHT. Conclusion: These findings indicated that NHT induces both apoptosis and cell-protective autophagy in human lung cancer cells. This data suggests that NHT might be a novel herbal drug for lung cancer.
Gagamhwanglyeonhaedog-tang(GHH), a Korean genuine medicine, is a newly designed herbal drug formula based on the traditional oriental pharmacological knowledge for the purpose of treating tumorous diseases. Apoptosis is an evolutionarily conserved suicide program residing in cells. It leads to cell death through a tightly regulated process resulting in the removal of damaged or unwanted tissue. In the present study, the apoptosis inducing activities of the decocted water extract of GHH were studied. Results of the 3- [4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay showed that GHH had a strong cytotoxic effect on HL-60 cells. The number of live cells was less than 20% after exposure to 1㎎/㎖ GHH for 48 hr. GHH increased cytotoxicity of HL-60 cells in a dose- and timedependent manner. Cell apoptosis by GHH was confirmed by flow cytometric analysis of the DNA-stained cells. The percentage of apoptotic cells increased to 28%, 31% and 37% 24 hr and 37%, 44% and 81% 48 hr after treatment with 0.01, 0.1 and 1㎎/㎖ GHH, respectively. Flow cytometric analysis of GHH treated HL-60 cells showed increase of hypodiploid apoptotic cells in a dose- and time- dependent manner. DNA fragmentation also occurred in apoptosis and was characterized by a ladder pattern on agarose gel. In addition, GHH (0.01 and 0.1㎎/㎖) increased the secretion of tumor necrosis factor-alpha in 24 and 48 hr. The author showed that GHH-induced apoptosis was accompanied by activation of caspase-3. These results suggest that GHH induces activation of caspase-3 and eventually leads to apoptosis.

Dangguisoo-san (DGSS), a traditional Korean herbal formula, has been prescribed to patients who suffer from various symptoms related with blood stagnation. Since inflammation can cause blood stagnation, we hypothesized that DGSS suppresses inflammation, relieving the symptoms associated with blood stagnation. In this study, given that DGSS is prepared in a mix of alcohol and water, we prepared the methanol (mDGSS) and water extract (wDGSS) of DGSS and compared their activities in suppressing inflammatory reaction. Western blot analyses show that mDGSS was more effective than wDGSS in activating Nrf2, a key factor that suppresses inflammation. Semi-quantitative RT-PCR shows that mDGSS activating Nrf2 resulted in the induction of Nrf2-dependent genes expression. However, mDGSS was not effective in suppressing the nuclear translocation of $NF-{\kappa}B$, a key factor that promotes inflammation, and the expression of $NF-{\kappa}B$-dependent genes such as TNF-a and IL-1b. When comparing with wDGSS, mDGSS was less effective in suppressing luciferase activity driven by $NF-{\kappa}B$. Therefore, our results show that mDGSS has the anti-inflammatory function by mainly activating Nrf2, while wDGSS does by both activating Nrf2 and suppressing $NF-{\kappa}B$. Our results suggest that preparing DGSS in a mix of water and methanol is a better way to achieve a strong anti-inflammatory efficacy of DGSS.
An ultra-performance liquid chromatography-electrospray ionization-mass spectrometer (UPLC-ESI-MS) method was established for the simultaneous quantification of eighteen marker compounds in traditional Korean formula, Cheonwangbosimdan (CWBSD). In addition, we evaluated the antioxidant effects of CWBSD. Eighteen marker components were separated on a UPLC BEH $C_{18}$ analytical column ($2.1{\times}100mm$, $1.7{\mu}m$) and kept at $45^{\circ}C$ by gradient elution with 0.1% (v/v) formic acid in water and acetonitrile as mobile phase. The flow rate was 0.3 mL/min and the injection volume was $2.0{\mu}L$. The antioxidant activities of CWBSD were assessed by measuring free radical scavenging activities on 2,2'-azinobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and 1-1-diphenyl-2-picrylhydrazyl (DPPH). The calibration curves of all analytes showed good linearity (correlation coefficient ${\geq}0.9937$) within the test ranges. The limits of detection and quantification for the 18 marker compounds were 0.01-4.71 ng/mL and 0.03-14.13 ng/mL, respectively. The contents of the 18 compounds in CWBSD extract ranged from none to $1701.00{\mu}g/g$. The CWBSD showed the radical scavenging activity in a dose-dependent manner. The concentration required for 50% reduction ($RC_{50}$) against ABTS and DPPH radicals were $149.42{\mu}g/mL$ and $339.24{\mu}g/mL$.
Galgeun-tang (GGT) is a traditional medicinal formula that is widely prescribed to treat cold, asthma, and hives in Korea. Fermented herbal medicines can be made more effective than normal herbal medicines by increasing the absorption and bioavailability of the active compounds. In this study, we fermented Galgeun-tang to produce bioconversion constituents using Lactobacillus plantarum (GGT144), and found that four peaks were decreased, three peaks were increased and two new peaks appeared in the HPLC-DAD chromatogram. After HPLC-DAD-guided fractionation of the newly-appearing compounds (1 and 5) and the increased (6, 7, and 9) compounds, the structure of the compounds was determined using NMR and MS. Using this approach the compounds were identified to be pyrogallol (1), daidzein (5), liquiritigenin (6), cinnamyl alcohol (7), and formononetin (9), respectively. In addition, the decreased compounds were identified to be daidzin (2), liquiritin (3), ononin (4), and cinnam aldehyde (8) using HPLC-DAD analysis with standard compounds. The high performance liquid chromatography method was used to quantify the nine constituents in GGT and GGT144. All calibration curves of the standard compounds displayed excellent linearity with a $R^2$ > 0.9968.
Objectives & Methods : In order to evaluate the in vitro synergic effects or Eunkyo-san which is a traditional poly-herbal formula that has been used in the treatment of respiratory diseases in oriental medicine, and quinolone antibiotics, rufloxacin (RUFX) and iprofloxacin(CPFX), experimentation was designed to determine minimal inhibitory concentration(MIC), $MIC_{50}\;and\;MIC_{90}$ of single use of quinolones and concomitant treatment with Eunkyo-san against 5 strains of aerobic gram negative bacteria, Escherichia coli, Klebsiella peumoniae, Hemophilus influenzae, Citrobacter freundii and Pseudomonas aeruginosae. Result : In the case of aerobic gram negative bacteria, the MIC, $MIC_{50}\;and\;MIC_{90}$ against Klebsiella peumoniae and Citrobacter freundii significantly decreased in concomitant-treated groups with Eunkyo-san compared to those of single-treated groups of RUFX and CPFX, respectively. However, no significant changes were demonstrated against Echerichia coli, Hemophilus influenzae and Pseudomonas aeruginosae. Conclusion : According to these results, concomitant use of Eunkyo-san against some strains of aerobic gram-negative bacteria dramatically increases in vitro antibacterial activity of RUFX and CPFX, and the increase and selectivity of antibacterial activities against these strains is attributable to Eunkyo-san, and not RUFX or CPFX activity.
웹은 하이퍼링크 및 야후와 같이 수동으로 분류된 웹 디렉토리 처럼 문서의 콘텐츠를 넘어선 다양한 정보의 소스가 풍부하다. 이 연구는 웹문서 내용을 활용한 텍스트기반의 검색 방식, 하이퍼 링크를 활용한 링크 기반의 검색 방식, 그리고 야후의 카테고리를 활용한 분류 기반의 검색 방식을 융합하므로서 여러 정보소스를 결합하면 검색 성능을 향상시킬 수 있다는 기존 융합검색연구들을 확장시켰다. 텍스트, 링크, 분류 기반 검색 결과를 여러가지 선형조합식으로 생성한 융합결과를 기존의 검색 평가 지표를 사용하여 각각의 검색 결과와 비교 한 후, 검색결과 오버랩의 중요성 또한 조사 하였다. 본 연구는 텍스트, 링크, 분류 기반 검색의 솔루션 스패이스들의 다양성이 융합검색의 적합성을 제시한다는 결론과 더불어 시스템 파라미터의 영향, 그리고 오버랩, 문서순위, 관련성들의 상호 관계 같은 융합 환경의 중요한 특성들을 분석하였다.
