This experiment was carried out to study on the safety assessment of Bas tures . Ursus thibetanus extract solution(BU) for Herbal acupuncture. SD rats and ICR mice were used for acute toxicity test the results were summerized as follows; 1. In rats and mice, LD50 value could not be measured. 2. There were no abnormal finding in acute toxicity test treated BU for Herbal-acupuncture.
This study was aimed to evaluate the effect of oxygen on motor activity and toxicity in male mice. The modified Porsolt forced swim test (FST) was used and the distance and time of movement by mice were analyzed in 15。C water bath for 20 minutes using the automatic Ethovision videotracking system. Analyses were carried out before and after 20 minutes of exposure to 10%-70% concentration of normobaric oxygen. The effects of inspired oxygen tension on the distance and time of movement showed the similar trends, but changes in distance were more prominent. Both the distance and time of movement increased after exposure to 30% and 40% oxygen concentration. The distance and time of movement also increased upon exposure to 50% and 60% oxygen. In contrast, increases En movement and time under exposure to 21% oxygen concentration were suppressed when exposed to over 50% oxygen concentration. With exposure to 10% oxygen, there was a significant decrease in the distance of movement and a slight suppression of movement time. During the swim test, 12.5%, 37.5%, and 87.5% of the mice drowned after exposure to 10%, 60%, and 70% oxygen concentration, respectively. These results suggest that motor activity can be enhanced by inspired oxygen up to 40% concentration. When hypoxic and hyperoxic oxygen exposure over 50%, motor activity is reduced and toxicity may be induced.
This research investigates how using algae as an ecotoxicological test species is easier than using daphnia for identifying toxic causative substances. From the results of the ecotoxicity measurements on the Shihwa lake and its tributaries, heavy metals were considered as one of major factors in causing toxicity. The algae ecotoxicity value was 9.6 while the daphnia ecotoxicity value was 0.8 in the Jeongwang stream. By using algae as the test species, we could identify the toxicity that causes heavy metals which might otherwise have been missed with only daphnia. The results from the EDTA addition test showed that zinc and copper were the main toxic causative substances in the Jeongwang stream and Gunja stream.
Ecotoxicity assessments with the physiochemical water quality items and the bioassay test using Daphnia magna were conducted for 18 selected effluents of 6 industrial types (metal processing, petroleum refining, synthetic textile manufacturing, plating, alcohol beverage manufacturing, inorganic compound manufacturing) being detected toxicity from industrial effluent in Ulsan city, and the interrelationship between total toxic unit (${\Sigma}TU$) and concentrations of Water Quality Conservation Act in Korea were investigated. The average toxic unit(TU) of effluents for 6 industrial types displayed the following ascending order: petroleum refining (0.2) < synthetic textile manufacturing (0.6) < alcohol beverage manufacturing (0.9) < metal processing (1.3) ${\leq}$ inorganic compound manufacturing (1.3) < plating (3.0). These values were less than effluent permission standard. Based on the result of substances causing ecotoxicity, the correlation analysis was not easy because most of heavy metals were not detected or were less than effluent permission standard. Toxicological assessment of industrial effluent was suitable for the evaluation of the mixture toxicity for pollutant. The whole effluent toxicity test using a variety of species was needed for the evaluation of industrial wastewater.
Benthic amphipod, Grandidierella japonica widely inhabits the Korean coastal waters and is developed as a standard test species for sediment toxicity tests. We exposed G. japonica to various pollutants including 4 kinds of inorganic metals (Ag, Cd, Cu and Hg), tributyltin [TBT], ammonia and 7 polycyclic aromatic hydrocarbon (PAH) compounds (acenaphthene, chrysene, fluoranthene, fluorene, naphthalene, phenanthrene and pyrene) to estimate the no observed effect concentration (NOEC) and the median lethal concentration (LC50) of each pollutant during the 96-hour acute exposure. Among all tested pollutants, TBT was most toxic to G. japonica, and Rg was most toxic among inorganic metals. The toxicity of pyrene to G. japonica was greatest among PAH compounds, followed by fluoranthene, phenanathrene, acenaphthene, fluorene and naphthalene. The toxicity of PAH compounds was closely related to their physico-chemical characteristics such as $K_ow$ and water solubility. G. japonica responded adequately to pollutant concentrations and exposure durations, and the sensitivity of G. japonica to various inorganic and organic pollutants was generally comparable to other amphipods used as standard test species in ecotoxicological studies, indicating this species can be applied in the assessment of environments polluted by various harmful substances.
To date, various techniques have been utilized to assess the contractility of cardiomyocytes and their response to drug-induced toxicity. However, these techniques are either invasive or involve complex fabrication methods and expertise. Here, we introduce the use of video-based analysis software to track the motion of cardiomyocytes and assess their contractility. The software, called "Tracker", is freely available and this is the first attempt at using it for cardiac contractility measurement. We used the software to measure the contractile properties of cells cultured on a rigid substrate and two flexible polydimethylsiloxane (PDMS) substrates having different elastic moduli day-wise up to eight days. Contractility was found to be highest in the most flexible substrate. Subsequently, the cardiotoxicity response of the cells on three different substrates was analyzed with verapamil. It was observed that the cells on rigid substrate were primarily affected by drug-induced toxicity, while the drug had a lesser impact on cells on the more flexible PDMS substrate. Evidently, the flexible substrate aided the maturation of cells and had lower drug toxicity, while the cells on PS could not fully mature. The assessment of cardiomyocytes using "Tracker" proved to be simple and reliable.
본 연구에서는 건물 실내에서 주로 사용되고 있는 합판, 실내 마감용 목재(루바; 소나무), 압축 스티로폼 단열재, 강화마루 및 PVC 장판 5 종류의 연소가스 유해성을 분석하였다. 연소가스의 유해성 분석은 Naval Engineering Standard(NES) 713 규격 및 미국국방성 규격(MIL-DTL)을 적용하였다. $CO_2$의 방출량은 5종의 내장재 모두 NES 713 규격 한계치인 100,000 ppm을 초과하지 않았다. CO 방출량의 경우 압축 스티로폼 단열재가 6,098 ppm으로 규격 한계치인 4,000 ppm을 초과하는 값을 나타내었다. 포름알데히드는 PVC 장판과 압축 스티로폼 단열재에서 각각 25 ppm, 49 ppm 방출되었다. $NO_X$는 합판에서 955 ppm으로 가장 높은 양이 방출되었으며, 규정 한계치 250 ppm에 비하여 높은 값으로 나타났다. 독성 지수는 합판 5.19, PVC 장판 4.13, 압축 스티로폼 단열재가 2.35, 강화마루 2.34, 그리고 실내 마감용 목재 1.22로 산출되었다. 본 연구에서는 화재 시 발생되는 연소가스 측정 및 유해가스 농도를 확인하여, 향후 건축 내장재의 화재 안전성을 평가하기 위한 기초 데이터로 활용하고자 한다.
A series of experiments were conducted to develop standard test organisms and test protocols for measuring sediment toxicity using candidate amphipods such as Mandibulophoxus mai, Monocorophium acherusicum, Haustorioides indivisus, and Haustorioides koreanus, which are indigenous to Korea. The relevant association of test species with sediment substrates was one of the important factors in sediment bioassay. The indigenous amphipods M mai and M. acherusicum were well associated with test sediments when they were exposed to various sediment substrates from sand to mud. The tolerant limits to various physico-chemical factors affecting bioassay results such as temperature, salinity and ammonia, as well as sensitivities to reference toxicant and contaminated sediments, were investigated using M. mai and M. acherusicum in the present study. These amphipods were tolerant to relatively wide ranges of salinity $(10{\sim}30\;psu)$ and ammonia (<50 ppm), and displayed relevant sensitivity to temperature as well. They are more sensitive to Cd, the reference toxicant, when compared to the standard test species used in other countries. Field-sediment toxicity tests revealed that M. mai would be more sensitive to sediment-associated pollutants than M. acherusicum, while the sensitivity of M. acherusicum was comparable to that of Leptocheirus plumulosus, which has been used as a standard test species in the United States of America. Overall results of this first attempt to develop an amphipod sediment toxicity test protocol in Korea indicated that M. mai and M. acherusicum would be applicable in the toxicity assessment of contaminated sediments, following the further evaluation encompassing various ecological and toxicological studies in addition to test method standardization.
Objectives: The purpose of this study was to examine the single dose toxicity with oral administration and genotoxicities of Ssanghwa-tang fermented with Lactobacillus acidophyllus. Materials and Methods: Clinical signs, weight changes, lethal doses$(LD_{50})$, and postmortem evaluation were determined by Globally Harmonized Classification System(GHCS) in a single-dose oral toxicity study. In vitro mammalian chromosomal aberration test was conducted with Ames test by cell proliferation suppression assessment using the cultivated CHO-K1(Chinese hamster ovary fibroblast) origins. Bacterial reversion assay was performed using Salmonella typhimurium (TA98, TA100, TA1535, and TA1537) and Escherichia coli (WP2uvrA). In vivo micronucleus test was performed using ICR mouse bone marrow. Results: No clinical sign was observed and none of the groups with doses up to 2000 mg/kg showed significant acute oral toxicity in the single dose oral administration. None of the sample doses taken during the 6 to 18 hour groups showed significant aberrant metaphases comparing to the negative control group in the in vitro mammalian chromosomal aberration test. No evidence of mutagenicity was seen for Escherichia coli (WP2uvrA) or Salmonella typhimurium (TA98, TA100, TA1535, and TA1537). No significant increase in the frequency of micronuclei was seen in the micronucleus test. Conclusion: These results indicate that the $LD_{50}$ value of Ssanghwa-Tang fermented with Lactobacillus acidophyllus may be over 2000 mg/kg and it have no acute oral toxicity and genotoxicity.

This research is to estimate the toxicity of Atractylodis Rhizoma Alba (ARA) in F344 rats and to find a dose level for the 13 weeks toxicity study. A hot water extract of ARA (ARWE) was administered orally to F344 rats at dose levels of 0 (vehicle control), 500, 1000, 2000, 3500, and 5000 mg/kg/day for 2 weeks. Each group was composed to five male and five female F344 rats. According to the result, there were no ARWE-related adverse changes in mortality, body weights, food consumption, urinalysis, hematology, clinical chemistry, gross finding at necropsy, and organ weight examination. Salivation was observed in 3500 and 5000 mg/kg/day in male and female rats but it could not have found any relationship with ARWE administration. Based on our findings, ARWE may not cause toxicity in rats under the experimental conditions. Therefore, dose level of 5000 mg/kg/day as a highest treatment group in 13-week exposure study is recommended for further toxicity assessment.
