Uptake and Phytotoxicity of TNT in Onion Plant

  • The uptake of $^{14}C$-2, 4, 6-trinitrotoluene (TNT) in hydroponics was studied using onion plants. Of the total TNT mass (5 $\mu\textrm{M}$ concentration), 75% was in the roots, 4.4% in the leaves, and 21% in the external solution at 2 days, The percent distribution in roots was lower with higher concentration in the external solution, but in leaves it was comparable at all concentrations (5-500 $\mu\textrm{M}$). Root concentration factor (RCF) in hydroponics was more than 85 in constant hydroponic experiment (CHE) at 5 $\mu\textrm{M}$ and 150 in non-constant hydroponic experiment (NHE) at 5 $\mu\textrm{M}$. The maximum RCF values in the hydroponic system were greater with lower solution concentration. Transpiration stream concentration factor (TSCF) values in the present study (NHE only: 0.31-0.56) were relatively similar to the values with predicted values (0.43-0.78), increasing with higher external TNT concentration. For phytotoxicity tested in hydroponics and wet paper method, 500 $\mu\textrm{M}$ was toxic to onion plant, 50 $\mu\textrm{M}$ was non-toxic for plant growth but limited the transpiration rate, and 5 $\mu\textrm{M}$ was non-toxic as control.

  • Four crop plant species were tested to assess an ecotoxicity in arsenic-amended soils. Test plants were Sorghum bicolor, Cucumis sativus, Triticum aestivum, and Phaseolus radiatus. The presence of arsenic decreased the root and shoot growths. Arsenite was more toxic than arsenate to all test plants. Root growths of Phaseolus radiatus and Cucumis sativus seem to be a good protocol to assess ecotoxicity of soils contaminted by arsenic.

  • As grouting materials for ground improvement, silicate materials such as water glass, silica-sol and so forth are used worldwide. However, they may pollute underground water. In this study, fish poison test(Korean standard industrial code KS M 0111) which estimates toxicity for fish is applied to evaluate the toxic effect by grouting materials. From the test result of the sample made of LW, LC50(Median Lethal Concentration) reaches within 24 hours. In case of the silica-sol, it does not even in 96 hours. Therefore, we can conclude that the sililca-sol grouting method is more free from the danger of underground water pollution. From the result that the mortality of fish under the condition of pH 8.6 is 0% within 96 hours, the groundwater can be saved by controlling pH below 8.6 for the grouting in the field.

Insecticidal Characterization of Thirteen Bacillus thuringiensis Isolates from Soil (III)

  • Thirteen strains of Bacillus thuringiensis were isolated from soil in Korea and characterized. The all strains produced parasporal crystals and spores in their cells. Two strains had bipyramidal crystals, seven strains contianed round ones and four strains had unregular ones. Only minor biochemical characteristics of the thirteen isolates were different and distinctive, however general characteristics were similar to the known serotypes of B. thuringiensis. Two strains were resistant to ampicilin. Three strains were resistant to bacitracin, six strains were resistant to cephalothin, two strains were resistant to colistin, HL-68 strain was resistant to gentamycin, HL-67 strain was resistant to kanamycin and HL-71 was resistant to tetracycline. Two strains were resistant to penicillin G. Four strains were toxic to Bombyx mori larvae and eleven strains were toxic to Culex pipiens larvae.

  • 토양오염의 독성을 탐지하기 위해 재조합 발광 박테리아의 고정화를 이용하여 바이오 센서를 제작하였으며 이를 이용하여 대표적 토양오염물질인 PAHs의 독성을 측정할 수 있었다. 또한, 이 바이오 센서를 이용하여 토양오염처리 전후의 처리 효율을 신속히 탐지할 수 있음을 확인하였다.

  • Bioremediation techniques have proved to be effective for restoring petroleum-contaminated soils. however some limitations still exist, especially biodegradation of hydrophobic organic compounds(HOCs) in soil is limited by their low solubility and sorption to solid surfaces. The principal objective of this study was to evaluate the effectiveness of biosurfactant sophorolipid on the biodegradation of hydrocarbons in soil. Experimental results showed that sophorolipid was not toxic to the HOC-degrading bacteria and enhanced biodegradation of HOCs in soil better than synthetic surfactants. when these models were treated with 1000mg/soil kg sophorolipid.

A Comparison of Electrical Stimulation for Electrodic and EDTA-Enhanced Phytoremediation of Lead using Indian Mustard (Brassica juncea)

  • The use of plants to remove toxic metals from soil (phytoremediation) is emerging as a cost-effective alternative to conventional methods for the removal of heavy metals from contaminated soil. Indian mustard (Brassica juncea) was used as the plant to accumulate high tissue concentrations of lead when grown in contaminated soil. For this study, the application of an electric field combined effectively with EDTA-enhanced phytoremediation. A stimulation of direct and alternating electric potential was compared and EDTA-enhanced phytoremediation of lead using Indian mustard has been performed. The effects of experimental parameters such as operating voltage with different concentration of EDTA, the number of graphite electrodes, and cultivation period on the removal of toxic metal were studied. Shoot lead accumulations in Indian mustard increased as the concentration of EDTA and dc electric potential was increased. Two to four folds was increased when EDTA plus a dc electric potential was applied, compared to an ac electric potential. The maximum lead accumulation in the shoots was achieved by applying EDTA plus dc electric potential with 6 graphite electrodes.

Assessment of toxic metals in vegetables with the health implications in Bangladesh

  • This study was conducted to investigate the levels of heavy metals in twelve species of vegetables and assessment of health risk. Samples were analyzed using inductively coupled plasma mass spectrometer (ICP-MS). The ranges of Cr, Ni, Cu, As, Cd and Pb in vegetables species were 0.37-5.4, 0.03-17, 0.35-45, 0.01-2.6, 0.001-2.2, and 0.04-8.8 [mg/kg, fresh weight (fw)], respectively. The concentrations of As, Cd and Pb in most vegetable species exceeded the maximum permissible levels, indicating unsafe for human consumption. Health risks associated with the intake of these metals were evaluated in terms of estimated daily intake (EDI), and carcinogenic and non-carcinogenic risks by target hazard quotient (THQ). Total THQ of the studied metals from most of the vegetables species were higher than 1, indicated that these types of vegetables might pose health risk due to metal exposure. The target carcinogenic risk (TR) for As ranged from 0.03 to 0.48 and 0.0004 to 0.025 for Pb which were higher than the USEPA acceptable risk limit (0.000001) indicating that the inhabitants consuming these vegetables are exposed to As and Pb with a lifetime cancer risk. The findings of this study reveal the health risks associated with the consumption of heavy metals through the intake of selected vegetables in adult population of Bangladesh.

  • The importance of a proper sample preparation and analysis technique has getting attention due to the complicity of toxic elements of interest from environmental samples depend on analytical purposes. It is critical to use proper analytical method to evaluate trace elements concentration in many environmental samples especially for making remediation decisions. Therefore, it is critical to apply a proper sampling and analytical method such as EPA publication SW-846 (Test Methods for Evaluating Solid Waste, Physical/chemical Methods). The objective of this study was to compare the USEPA Methods 3050B, modified 3050B, 305 la, and KBSI method (modified EPA 3052 Method) in term of recovery rate of metals. The NIST SRM (Montana soil) was used to compare the extraction and digestion efficiency. After sample has been collected the analysis were achieved by ICP-MS (Elan 6100, Perkin Elmer) as well as ICP-AES (Ultima 2C, JY) for trace elements and major elements.
