Studies on Nutrio-physiological Response of Rice Plant to Root Environment (근부환경(根部環境)에 따른 수도(水稻)의 영양생리적(營養生理的) 반응(反應)에 관(關)한 연구(硏究))

  • The nutriophysiological response of rice plant to root environment was investigated with eye observation of root development and rhizosphere in situation. The results may be summarized as follows: 1) The quick decomposition of organic matter, added in low yield soil, caused that the origainal organic matter content was reached very quickly, in spite of it low value. In high yield soil the reverse was seen. 2) In low yield soil root development, root activity and T/R value were very low, whereas addition of organic matter lowered them still wore. This might be contributed to gas bubbles around the root by the decomposition of organic matter. 3) Varietal difference in the response to root environment was clear. Suwon 82 was more susceptible to growth-inhibitine conditions on low-yield soil than Norin 25. 4) Potassium uptake was mostly hindered by organic matter, while some factors in soil hindered mostly posphorus uptake. When the organic matter was added to such soil, the effect of them resulted in multiple interaction. 5) The root activity showed a correlation coeffieient of 0.839, 0.834 and 0.948 at 1% level with the number of root, yield of aerial part and root yield, respectively. At 5% level the root-activity showed correlation-coefficient of 0.751, 0.670 and 0.769 with the uptake of the aerial part of respectively. N, P and K and a correlation-coefficient of 0.729, 0.742 and 0.815 with the uptake of the root of respectively N.P. and K. So especially for K-uptake a high correlation with the root-activity was found. 6) The nitrogen content of the roots in low-yield soil was higher than in high-yield soil, while the content in the upper part showed the reverse. It may suggest ammonium toxicity in the root. In low-yield soil Potassium and Phosphorus content was low in both the root and aerial part, and in the latter particularly in the culm and leaf sheath. 7) The content of reducing sugar, non-recuding sugar, starh and eugar, total carbohydrates in the aerial part of plants in low yield soil was higher than in high yield soil. The content of them, especially of reducing sugar in the roots was lower. It may be caused by abnormal metabolic consumption of sugar in the root. 8) Sulfur content was very high in the aerial part, especially in leaf blade of plants on low yield soil and $P_2O_5/S$ value of the leaf blade was one fifth of that in high yield soil. It suggests a possible toxic effect of sulfate ion on photophosphorization. 9) The high value of $Fe/P_2O_5$ of the aerial part of plants in low yield soil suggests the possible formation of solid $Fe/PO_4$ as a mechanical hindrance for the translocation of nutrients. 10) Translocation of nutrients in the plant was very poor and most nutrients were accumulated in the root in low yield soil. That might contributed to the lack of energy sources and mechanical hindrance. 11) The amount of roots in high yield soil, was greater than that in low yield soil. The in high-yield soil was deep, distribution of the roots whereas in the low-yield soil the root-distribution was mainly in the top-layer. Without application of Nitrogen fertilizer the roots were mainly distributed in the upper 7cm. of topsoil. With 120 kg N/ha. root were more concentrated in the layer between 7cm. and 14cm. depth. The amount of roots increased with the amount of fertilizer applied.

  • Ethylene glycol (EG) is produced commercially in large amounts and is widely used as antifreeze or deicing solution for cars, boats, and aircraft. EG poisoning occurs in suicide attempts and infrequently, either intentionally through misuse or accidental as EG has a sweet taste. EG has in itself a low toxicity, but is in vivo broken down to higher toxic organic acids which are responsible for extensive cellular damage in various tissues caused principally by the metabolites glycolic acid and oxalic acid. The most conclusive analytical method of diagnosing EG poisoning is determination of EG concentration. However, victims are sometimes admitted at a late stage to hospitals or died during emergency treatment like a gastric lavage or found rotten dead, when blood EG concentrations are low or not detected. Therefore, in this study, the identification of EG was not only performed by gas chromatograpyc-mass spectrometry (GC-MS) following derivatization but also further toxicological analyses of metabolites, glycolic acid (GA) and oxalic acid (OA), were performed by ion chromatography in various biological specimens. A ranges of blood concentrations (3 cases) was $10\sim2,400\;{\mu}g/mL$ for EG, $224\sim1,164\;{\mu}g/mL$ for GA and ND $\sim40\;{\mu}g/mL$ for OA, respectively, In other biological specimens (liver, kidney, bile and pleural fluid), a range of concentrations (3 cases) was ND $\sim55,000\;{\mu}g/mL$ for EG, ND $\sim1,124\;{\mu}g/mL$ for GA and ND $\sim60\;{\mu}g/mL$ for OA, respectively. Liver and kidney tissues were recommended specimens including blood because OA, a final metabolite of EG, was identified large amounts in these despite no detectable EG caused by some therapy.

The Removal of Organics and Nitrogen with Step Feed Ratio Change into the Anoxic and Anaerobic reactor in Advanced Sewage Treatment process Using Nonsurface-modified and Surface-modified Media Biofilm (비개질/개질 생물막을 이용한 오수고도처리공정에서 혐기조와 무산소조의 원수 분배율에 따른 유기물 및 질소 제거)

  • This study was accomplished using attached $A^2/O$ process that contains nonsurface-modified and surface-modified polyethylene media inside the Anaerobic/Anoxic, Oxic tank, respectively. We could make the hydrophobic polyethylene media have hydrophilic characteristics by radiating ion beam on the surface of the media. The objectives of this study is to investigate the removal efficiencies of the organics and nitrogen when the step feed ratio of raw wastewater into anaerobic and anoxic tank is changed. In this case, we assumed that the denitrification rate can be improved because the nitrifiers in anoxic tank can perform denitrification using RBDCOD instead of artificial carbon sources (for example, methanol, etc.). The wastewater injection rate into anaerobic/anoxic tank was set up by the ratio of 10 : 0, 9 : 1, 8 : 2, 6 : 4, and the results of BOD removal efficiency showed similar trends with $93.3\%,\;92.6\%,\;92.4\%\;and\;91.6\%$, respectively. But the BOD removal efficiency (utilization of the organics) in the anoxic tank was in the order of 9 : 1 $(84.8\%)$, 10 : 0 $(77.0\%)$, 8 : 2 $(75.3\%)$, and 6 : 4 $(61.1\%)$. The T-N removal efficiency was most high when the ratio is 9 : 1 $(67.4\%)$, and other conditions, 10 : 0, 8 : 2, 6 : 4, showed $61.3(\%),\;60.7\%,\;55.5\%$, respectively; the ratio 6 : 4 was found to be lowest T-N removal efficiency, lower than the ratio 9 : 1 by $12\%$. Though the nitrification rate of the ratio 10 : 0, 9 : 1, and 8 : 2 showed similar levels, the ratio 6 : 4 showed considerable inhibition of nitrification, ammonia was the great portion of the effluent T-N. The advantages of this process is that this process is cost-saving, and non-toxic methods than injecting the artificial carbon source.

Growth inhibition of hydrotrope-combined copper against Microcystis aeruginosa and evaluation of its toxicity (Microcystis aeruginosa에 대한 hydrotrope-combined copper의 생장억제 및 독성 평가)

  • Hydrotrope-combined copper (HCC) is a copper ($Cu^{2+}$)-based algicide, which is combined with a hydrotrope that keeps copper ion in solution to improve performance. This study assessed the growth inhibition effect of HCC against Microcystis aeruginosa which is one of the most common toxic cyanobacterium in eutrophic freshwater environment. Various HCC doses, ranging from 5.5 to $550{\mu}g/L$ as $Cu^{2+}$, were applied to either BG-11 or 1/4 diluted medium with low- or high-inoculum density of M. aeruginosa. Growth inhibition was monitored based on a decrease in chlorophyll-a content in culture medium during the incubation. Results showed that HCC significantly inhibited the growth of M. aeruginosa in a dose-dependent manner. In case of 1/4 diluted BG-11 medium, HCC dose as low as $5.5{\mu}g$ $Cu^{2+}/L$ completely inhibited the production of chlorophyll-a by M. aeruginosa. It was found that HCC did not induce any significant release of microcystin-LR from M. aeruginosa. Acute toxicity of HCC was tested using Daphnia magna, and the 24-h $EC_{50}$ value was 0.30 mg/L as $Cu^{2+}$ which was much higher than the actual inhibition dose. Ames test was performed using Salmonella enterica serovar Typhimurium TA100, and HCC showed no increase in the number of revertant colonies. The result suggested that HCC does not have any mutagenic potential in the aquatic environment. In addition, no genotoxic effect of HCC was also confirmed based on the SOS ChromoTest using Escherichia coli PQ37. Therefore, HCC could be used as a relatively safe and effective pre- and post-treatment agent to control hazardous algal blooming in aquatic environments.

The Uptake of $Sr^{90}$ by Paddy Rice from Soil and its Distribution in the Plant (답토양(畓土壤)에서 수도(水稻)의 Strontium-90 흡수(吸收)와 수도체내(水稻體內) 분포(分布))

  • Because of the drastic development of nuclear industries, the contamination of natural environments by the disposal of radioactive materials which are released from nuclear facilities have aroused a considerable concern in relation to agricultural practices. Therefore the present investigation, through pot experiment, was performed to find out the aspect of the uptake of $Sr^{90}$ by rice plants and its distribution in them in five different types(physicochemical and minerallogical properties) of paddy soils. The results obtained were as follows; 1) Visual toxic symptoms on the growth of rice plant due to treatment of $Sr^{90}$ up to $40{\mu}Ci/10㎏$ in a pot were not observed even though uptake of $Sr^{90}$ by rice plant was proportionally increased with the $Sr^{90}$ treatment. 2) Distribution of $Sr^{90}$ in the rice plant was the highest in the leaves (84.5%) followed in the order by stems (13.5%) and rough grain (2.0%). The ratio of $Sr^{90}$ to Ca was higher in the leaves (872) and stems (667) than in the rice grain (89). 3) $Sr^{90}$ absorption in the rice plant ranged $0.15{\sim}0.30%$ at harvesting time. Uptake of $Sr^{90}$ by rice plants decreased by the increase of soil pH and exchangeable canons in the soils, but $Sr^{90}$ uptake increased when nitrogen, organic matter and clay content in soil was high, and uptake of this nuclide in the rice plant was higher with low Illite and Vermiculite content in the soils.

Risk Analysis of Inorganic Arsenic in Foods (식품 중 무기비소의 위해 분석)

  • Arsenic and its compounds vary in their toxicity according to the chemical forms. Inorganic arsenic is more toxic and known as carcinogen. The provisional tolerable weekly intake (PTWI) of $15{\mu}g/kg$ b.w./week established by the Joint FAO/WHO Expert Committee on Food Additives (JECFA) has been withdrawn, while the EFSA panel suggested $BMDL_{0.1}$ $0.3{\sim}8{\mu}g/kg\;b.w./day$ for cancers of the lung, skin and bladder, as well as skin lesions. Rice, seaweed and beverages are known as food being rich in inorganic arsenic. As(III) is the major form of inorganic arsenic in rice and anaerobic paddy soils, while most of inorganic arsenic in seaweed is present as As(V). The inorganic arsenic in food was extracted with solvent such as distilled water, methanol, nitric acid and so on in heat-assisted condition or at room temperature. Arsenic speciation analysis was based on ion-exchange chromatography and high-performance liquid chromatography equipped with atomic absorption spectrometry and inductively coupled plasma mass spectrometry. However, there has been no harmonized and standardized method for inorganic arsenic analysis internationally. The inorganic arsenic exposure from food has been estimated to range of $0.13{\sim}0.7{\mu}g/kg$ bw/day for European, American and Australian, and $0.22{\sim}5{\mu}g/kg$ bw/day for Asian. The maximum level (ML) for inorganic arsenic in food has established by EU, China, Australia and New Zealand, but are under review in Korea. Until now, several studies have conducted for reduction of inorganic arsenic in food. Inorganic arsenic levels in rice and seaweed were reduced by more polishing and washing, boiling and washing, respectively. Further research for international harmonization of analytical method, monitoring and risk assessment will be needed to strengthen safety management of inorganic arsenic of foods in Korea.

Environmental Damage to Nearby Crops by Hydrogen Fluoride Accident (불화수소 누출사고 사례를 통한 주변 농작물의 환경피해)

  • BACKGROUND: Hydrogen fluoride is one of the 97 accident preparedness substances regulated by the Ministry of Environment (Republic of Korea) and chemical accidents should be managed centrally due to continual occurrence. Especially, hydrogen fluoride has a characteristic of rapid diffusion and very toxic when leaking into the environment. Therefore, it is important to predict the impact range quickly and to evaluate the residual contamination immediately to minimize the human and environmental damages. METHODS AND RESULTS: In order to estimate the accident impact range, the off-site consequence analysis (OCA) was performed to the worst and alternative scenarios. Also, in order to evaluate the residual contamination of hydrogen fluoride in crop, the samples in accident site were collected from 15-divided regions (East direction from accident sites based on the main wind direction), and the concentration was measured by fluoride ($F^-$) ion-selective electrode potentiometer (ISE). As a result of the OCA, the affected distance by the worst scenario was estimated to be >10 km from the accident site and the range by the alternative scenario was estimated to be about 1.9 km. The residual contamination of hydrogen fluoride was highest in the samples near the site of the accident (E-1, 276.82 mg/kg) and tended to decrease as it moved eastward. Meanwhile, the concentrations from SE and NE (4.96~28.98 mg/kg) tended to be lower than the samples near the accident site. As a result, the concentration of hydrogen fluoride was reduced to a low concentration within 2 km from the accident site (<5 mg/kg), and the actual damage range was estimated to be around 2.2 km. Therefore, it is suggested that the results are similar to those of alternative accident scenarios calculated by OCA (about 1.9 km). CONCLUSION: It is difficult to estimate the chemical accident-affecting range/region by the OCA evaluation, because it is not possible to input all physicochemical parameters. However simultaneous measurement of the residual contamination in the environment will be very helpful in determining the diffusion range of actual chemical accident.

Study on Skin pH Improvement Effect through Regulation of Na+/H+ Exchanger 1 (NHE1) Expression of Prunella vulgaris Extract and Its Active Compound, Caffeic Acid (꿀풀 추출물과 그 활성 화합물인 카페인산의 Na+/H+ exchanger 1 (NHE1) 발현 조절을 통한 피부 pH 개선 효과에 대한 연구)

  • This study was conducted to discover substances that regulate skin surface acidification using human epidermal keratinocyte cell lines, and to investigate their effects on the moisturizing ability and skin barrier function of the stratum corneum. Prunella vulgaris (P. vulgaris) is an herb widely distributed in Northwest Africa and North America that has been studied for its anti-apoptotic, antioxidant, and anti-inflammatory effects. However, research on the regulation of NHE1 expression and the restoration of skin barrier function has not been conducted. Analysis of P. vulgaris revealed the presence of rosmarinic acid and caffeic acid as active ingredients, which were tested for toxicity in human epidermal keratinocyte cell lines (HaCaT), and showed no toxic effects were observed at high concentarion (100 ㎍/mL or 100 µM). It is known that sodium-hydrogen ion exchange pumps (NHE1) decrease in expression in aging skin to maintain the acidic pH of the stratum corneum, and it is hypothesized that this decrease plays an important role in the impaired restoration of skin barrier function in aging skin. P. vulgaris extract and caffeic acid increased the expression of NHE1 in keratinocytes, increased the expression of natural moisturizing factor (NMF) precursor filaggrin and ceramide synthesis enzyme serine palmitoyl transferase (SPT). In addition, P. vulgaris and caffeic acid decreased the extracellular pH of keratinocytes, indicating a direct effect on skin pH regulation. Taken together, these results suggest that P. vulgaris and caffeic acid can regulate skin pH through NHE1 modulation, and may help to restore skin barrier function by increasing NMF and ceramide synthesis. These results show the possibility that honeysuckle and caffeic acid can have a positive effect on skin health, and can be the basis for the development of new skin protection products using them.
