Benthic amphipod, Grandidierella japonica widely inhabits the Korean coastal waters and is developed as a standard test species for sediment toxicity tests. We exposed G. japonica to various pollutants including 4 kinds of inorganic metals (Ag, Cd, Cu and Hg), tributyltin [TBT], ammonia and 7 polycyclic aromatic hydrocarbon (PAH) compounds (acenaphthene, chrysene, fluoranthene, fluorene, naphthalene, phenanthrene and pyrene) to estimate the no observed effect concentration (NOEC) and the median lethal concentration (LC50) of each pollutant during the 96-hour acute exposure. Among all tested pollutants, TBT was most toxic to G. japonica, and Rg was most toxic among inorganic metals. The toxicity of pyrene to G. japonica was greatest among PAH compounds, followed by fluoranthene, phenanathrene, acenaphthene, fluorene and naphthalene. The toxicity of PAH compounds was closely related to their physico-chemical characteristics such as $K_ow$ and water solubility. G. japonica responded adequately to pollutant concentrations and exposure durations, and the sensitivity of G. japonica to various inorganic and organic pollutants was generally comparable to other amphipods used as standard test species in ecotoxicological studies, indicating this species can be applied in the assessment of environments polluted by various harmful substances.
오늘날 전세계적으로 수환경에서의 잔류 의약물질들의 오염에 대한 연구결과들이 많이 보고되고 있다. 이들 잔류 의약물질들은 다양한 종류와 그들이 가지는 물리 화학적인 특성들로 인해 수환경에서의 거동, 오염현황, 영향 및 독성 등도 매우 다양하여 수환경 및 수처리 공정에서의 거동을 예측 평가하기가 어렵다. 선진 외국의 경우 환경 중에서의 오염 현황 및 사용량을 정량화하기 위한 조사를 이미 시작하였고, 잔류 의약물질들의 인체에 대한 잠재적 위험성뿐만 아니라 상수나 하수처리 공정에서의 제거기술에 관한 다양한 연구도 활발히 진행하고 있다. 그러나 국내의 경우 잔류 의약물질들에 대해 전국 주요하천과 상수원에 대한 모니터링 결과도 매우 부족한 실정이다. 우선 국내의 경우는 상수원으로 이용되는 전국 주요 하천과 호소에 대해 주기적이고 체계적인 오염현황 평가가 선행되어야 하며, 잔류 의약물질들이 비교적 고농도로 검출되는 상수원에 대해서는 주오염원에 대한 조사도 병행되어야 한다. 또한, 의약물질 사용량은 지속적으로 증가할 것으로 전망되기 때문에 하수처리 시설에 대한 부하도 지속적으로 증가할 것이다. 따라서 수환경에서의 유해도를 저감시키기 위해 효과적인 하수처리 공법의 도입이 절실하며, 상수에 대한 안전성 확보를 위해서도 잔류 의약물질들의 수환경중에서의 물리 화학적 거동에 대한 면밀한 연구가 요구된다.
The acute oral LD5O toxicity values of isazofos, pyraclofos, diazinon and methomyl were determined for Japanese quail based on OECD guideline. The $LD_{50}$ of isazofos, pyraclofos and diazinon was 16.26 mg/kg, and 7.11mg/kg body weight In female respectively. And the $LD_{50}$ of each chemical in male was 21.44, 35.64, 8.28 mg/kg body weight respectively. Diazinon was the most susceptible compounds to Japanese quail in both sexes. The $LD_{50}$ of methomyl was 21.24 mg/kg body weights in female, and 28.28 mg/kg body weight in male respectively. Diazinon, isazofos and methomyl were more toxic In the female than male. The symptoms of poisoning were similar in quails administrated with each chemicals. The clinical sign in Japanese quail were ataxia, salivation, diarrhea, ruffled feather and convulsion at dead point. There were severe hemorrhage and catarrhal inflammation from duodenum to ileum In all compounds. In Japanese quail treated with organophosphorus and carbamate compounds, brain acetylcholinesterase was inhibited by 88-96. The recovery was not observed after 5 h in sublethal dose.
Butyltin compounds in seawater, sediments and organisms inside Jeju Harbor of Jeju Island, Korea, were quantitatively determined to assess the extent of contamination and to evaluate the bioaccumulation in Thais clavigera (gastropod), Liolophura japonica and Cellana nigrolineata. In addition, imposex in T. clavigera was used to assess the sublethal effect of tributyltin compounds and its usage as a specific biomarker for these compounds was also studied. The main species in seawater was tributyltin (TBT) in March and June, monobutyltin (MBT) in August, but in sediments and organisms it was dibutyltin (DBT) and TBT, respectively, irrespective of survey time. TBT, the most toxic to aquatic organisms among BTs, was found at concentrations which were sufficient to have a serous effect on the sensitive organisms upon chronic exposure. The high correlations (r>0.7) between BTs indicated that DBT and MBT were mainly degraded from TBT based on antifouling paints and their sources were negligible. The sedimentary organic matters have little influence on the distribution of BTs in marine environment and the lipid content in T. clavigera showed a significant correlation with TBT concentrations. Measurement of imposex T. clavigera was expected to be a very helpful tool for preliminary survey of BTs prior to trace analysis of those.
Granular activated carbon(GAC) is highly effective in removing organic compounds which are resistant to biological disintegration in wastewater treatment. However, GAC has reached its full adsorptive capacity, GAC needs to be regenerated before it can be used for a further adsorption cycle. Countercurrent oxidative reaction (COR) technique has been developed and evaluated for the regeneration of spent GAC. Various parameters such as flame temperature, the loss of carbon, destruction and removal efficiency (DRE) of organic compounds, surface area, surface structure, adsorptive capacity, etc. were examined to determine the performance of COR. The results of these tests showed that adosorptive capacity of regenerated GAC was completely recovered, the loss of carbon was controllable, flame temperature was high enough to insure complete destruction and removal $(\geq99.9999%)$ of specific organics of interest, polychlorinated biphenyls (PCBs), that are thermally stable, and on formation of toxic byproducts such as polychlorinated dibenzo-p-dioxins (PCDDs) or polychlorinated dibenzofurans (PCDFs) were detected during the regeneration process. The COR technique is environmentally benign, easy to use and less copital intensive than other available regeneration technologies.
최근 몇 년간 유기인 계열 독성물질이 민간인을 대상으로 사용되어 전 세계적으로 큰 위협이 되고 있다. 독성물질에 대한 예방이 불가능한 현 치료대책 대신, 보다 개선된 치료 대책으로서의 바이오스캐빈저에 대한 연구가 활발히 진행됐다. 바이오스캐빈저는 유기인 계열 독성물질이 인체 내 표적 기관에 도달하기 전, 독성물질 자체를 비활성 상태로 전환하거나 독성물질과 기질 간의 결합을 차단함으로써 중독을 예방하는 단백질 및 효소를 일컫는다. 특히 독성물질을 분해하는 과정에서 활성 상태를 유지함으로써 적은 양의 단백질로도 독성물질의 중독을 빠르게 치료하는 촉매성 바이오스캐빈저 개발에 많은 노력이 투여되어 왔다. 본 리뷰에서는 촉매성 바이오스캐빈저 개발을 위해 분자진화 및 단백질 공학 기술을 적용한 최신 연구들에 대해 소개하고, 끝으로 이러한 효소들을 임상적으로 승인된 약으로 개발하기 위해 남은 몇 가지 과제들을 간단히 제시할 것이다.
본 논문에서는 해마다 수 십만 개씩 발생되는 폐타이어의 열적 처리 방법인 건류 소각 처리 방법에서 발생되는 이차 오염 물질을 정량하여 폐타이어의 안전한 최종 처리 방법까지 고찰하였다. 건류-소각처리 방법에서 발생되는 배기 가스에서 이차 독성 오염 물질을 화학 평형 모델을 응용하여 예측하였고 실질적인 잔류 물질인 건류재와 소각재에서 유기성 독성 물질과 중금속을 정량하였다. 건류재에서는 폐타이어의 불완전 연소에 따른 유기성 이차 오염 물질이 다량 포함되어 있는 반면에 소각재에서는 납과 카드뮴과 같은 중금속 화합물의 함량이 높았다. 건류재 및 소각재의 안전한 최종 매립을 위하여는 서로 다른 전처리 방법이 요구된다.
Hymenoxys brachyactis지상부의 디클로로메탄 추출액으로부터 3개의 sesquiterpene lactone들과 새로운 lactone화합물인 isohymenograndin을 확인하였다. 그리고 2개의 sesquiterpene lactone축합물인 biennine C와 독성이 있는 것으로 알려진 flavone화합물인 hispidulin을 분리하여 각각을 고자장 NMR, Mass등의 기기를 이용하여 구조결정 하였다. Biennine C는 gas chromatography와 Mass분석을 통하여 독성화합물 hymenoxon과 화합물 8과의 Diels-Alder축합물임을 확인하였다. biennine C를 포함한 이상의 lactone화합물들은 ${\alpha},{\beta}$-unsaturated lactone으로서 독성화합물군으로서 알려진 hymenovin과 공통적인 구조를 갖고 있어서 생체독성을 나타낼 가능성이 높은 화합물들로 사료된다.

A relatively high homology between Escherichia coli prolidase and Alteromonas organophosphorous acid anhydrolase suggests that E. coli prolidase may have an activity to d egrade toxic organophosphorous compounds. To confirm this suggestion, we cloned and expressed a prolidase gene (pepQ) of E. coliBL2l. The recombinant E. coli prolidase that consisted of 443 amino acid residues exhibited activity and stereochemical selectivity against organopho sphorous compounds, although its activity was two to three orders of magnitude less than that of the other organophosphorous acid hydrolase isolated from Pseudomonas diminuta.
헤테로고리형 PPO 저해제 화합물들은 강한 제초활성을 갖고 있으면서도 동물과 인간, 환경에 대한 뛰어난 안전성 때문에 최근에 매우 활발하게 연구되고 있는 화합물이다. 우리는 이러한 여러 가지 헤테로고리형 PPO 저해제 화합물 중에서 4,5,6,7-tetrahyoindazole, maleimide 및 tetrahydrophthalimide type 화합물의 페닐 치환기의 5- 위치에 서로 다른 epoxy group이 치환된 6종의 유도체들을 합성하였고, 이들의 제초활성을 논조건에서 시험하였다. 4,5,6,7-tetrahydroindazole 화합물들은 매우 강한 제초활성을 나타냈는데 피와 물달개비, 너도방동사니 등에 대하여 16 g/ha 이하의 낮은 농도에서도 제초효과가 좋았으며, 벼에 대한 안전성은 뛰어난 결과를 나타냈다.
