인삼이 적혈구세포의 해당과정 및 막 투과도에 미치는 영향 (Effect of Panax Ginseng Saponin on Metabolism and Ion Transport in Human Erythrocytes)

  • Red cell glycolytic intermediates, metabolites and metabolic ratios were studied. Glycolytic intermediates were measured in neutralized perchloric acid extracts of red cell suspensions after 3 hr incubation at $37^{\circ}C$ in the presence and absence of saponin. Adenosine triphosphate(ATP), adenosine diphosphate(ADP), pyruvate and lactate were measured by enzymatic procedures involving stoichiometric oxidation or reduction of a pyridine nucleotide. Glucose was determined using glucose oxidase after zinc hydroxide extraction. The redox state was calculated from the lactate dehydrogenase equilibrium. Adenosine triphosphatase activity(ATPase) was measured by determining the amount of phosphate released from ATP by washed erythrocyte membranes(ghost) during 20 min. incubation. Both total hydrolysis and the amount of hydrolysis that occured in the presence of ouabain were measured. The second measurement yields Mg-ATPase and represents nonspecific ATPase activity of the membranes. The difference between total and Mg-ATPase activity can be attributed to Na-K-ATPase. For the measurement of sodium fluxes, human erythrocytes were preincubated in $^{22}Na$ for 3 hr at $37^{\circ}C$, washed and suspended in a tracer-free medium. The amount of $^{22}Na$ transported out of cells at any time was determined by analysis of supernatant samples taken at various time after addition of the labeled cells to isotope-free medium. The cells and medium were separated and the radioactivity appearing in the medium was measured. From the total radioactivity in the suspension and the radioactivity appearing in the medium at known time, the rate constant for sodium release was computed. The results are summarized as follows: 1) ATP and ATP/ADP were found to increase at every concentration of saponin tested whereas ADP declined at every cone. of saponin. The increase in pyruvate and lactate were observed at every cone, of saponin and thus $NAD^+/NADH$ computed from pyruvate/lactate also increased. Glucose utilization was stimulated by saponin. 2) $Na^+-K^+-ATPase$ activities showed a biphasic response to saponin, first increasing in lower concentration and then decreasing in higher concentration of saponin. 3) The efflux of sodium was significantly increased by saponin in the range of 5 to 10 mg%. The stimulatory effect of saponin on the rate constants for active(ouabain-sensitive) sodium efflux was inhibited by addition of ouabain.

국내 인삼시장에서 유통되고 있는 홍삼 파우치 제품의 사포닌 함량 및 이화학적 특성 (Saponin Contents and Physicochemical Properties of Red Ginseng Extract Pouch Products Collected from Ginseng Markets in Korea)

  • 홍삼 파우치 제품의 성분 및 제조방법의 표준화를 위한 일환으로 국내 유통 중인 홍삼 파우치의 사포닌 함량 및 이화학적 특성을 분석하였다. 총 ginsenoside 함량은 5.5~185.7 mg/100 mL의 함량을 나타내었고 항암효과로 알려진 ginsenoside $Rg_3$, $Rg_2$, $Rh_1$$Rh_2$의 함량 분포는 $Rg_3$는 1.6~46.3 mg/100 mL, $Rg_2$는 미검출~22.0 mg/100 mL, $Rh_1$은 미검출~4.3 mg/100 mL, $Rh_2$는 미검출~20.4 mg/100 mL의 분포를 나타내었다. 항 당뇨 효과가 있는 ginsenoside $Rb_2$와 Re의 함량분포는 $Rb_2$는 미검출~10.8 mg/100 mL, Re는 미검출~7.0 mg/100 mL의 함량을 나타내었으며 그 밖의 사포닌 중 ginsenoside $Rb_1$는 미검출~25.2 mg/100 mL, Rc는 미검출~12.5 mg/100 mL, Rd는 미검출~11.3 mg/100 mL, Rf는 미검출~5.9 mg/100 mL, $Rg_1$는 미검출~4.4 mg/100 mL까지의 함량 분포를 보였다. 이화학적 특성 분석결과 총당함량은 226.6~3,102.9 mg/100 mL, 당도는 $1.4\sim9.5^{\circ}Bx$, 탁도는 82.2~100.0%, pH는 4.1~5.0의 범위를 나타내었다. 국내에서 수집한 홍삼 파우치 제품의 약 50%(21~24개 제품)가 ginsenoside $Rb_1$, $Rb_2$, Rc, Rd, Re, $Rg_1$이 미검출 되었으며, 각 제품별 사포닌 함량 차이가 매우 크게 나타나, 국내유통 중인 홍삼 제품에 대한 기준 설정 및 생산방법의 표준화가 필요할 것으로 사료된다.

초음파 처리에 의한 인삼 잎과 줄기의 진세노사이드 성분 변화 (Changes in Ginsenosides Composition of Ginseng Leaf and Stem after an Ultrasonication Process)

  • The purpose of this study is to develop a new preparation process of ginseng leaf and stem extracts having high concentrations of ginsenoside Rg2, Rg3, Rg5, Rh1, a special component of red and black ginseng. Chemical transformation from ginseng saponin glycosides to prosapogenin was analyzed by the HPLC. Extracts of ginseng (Panax ginseng) leaf and stem were processed under several treatment conditions including ultrasonication treatments. The content of total saponin reached their heights at 17 hr (UGL-17) of ultrasonication treatment, followed by 16 hr (UGL-16) and 7 hr (UGL-7) of ultrasonication treatment at $100^{\circ}C$. UGL-17 findings show that the ginseng leaf and stem that had been processed with ultrasonication for 17 hours peaked in the level of Rg2, Rg3 and Rh1. In addition, UGL-16 contained ginsenoside Rg5 at high concentrations. It is thought that such results provide basic information in preparing ginseng leaf and stem extracts with functionality enhanced.

팽화처리가 홍미삼의 품질특성에 미치는 영향 (Effect of Puffing on Quality Characteristics of Red Ginseng Tail Root)

  • 팽화처리가 홍미삼의 사포닌 성분, 당류, 페놀성분, 미세구조 및 소화율 등에 미치는 영향을 조사하였다. 홍미삼의 추출된 조사포닌 함량은 팽화처리후 약 26.5% 증가하였다. 사포닌 조성에서 팽화홍미삼은 홍미삼에 비해 $Rb_1$, $Rb_2$, Rc, Re, $Rg_1$함량이 다소 감소하였으나 Rd와 $Rg_3$함량은 증가하였고, 특히 $Rg_3$함량은 0.49 mg/g에서 0.72 mg/g로 증가하였다. 가용성 총당은 팽화전후 비슷하였고, 산성다당체 함량은 각각 7.15, 6.44%로 팽화후 다소 감소하였다. 총페놀함량은 7.86%에서 9.94%로 증가하였다. 개별 페놀산의 경우 gentisic acid는 홍미삼에서만, salicylic acid는 팽화후 홍미삼에서만 검출되었고, gallic acid, p-coumaric acid, caffeic acid 및 ferulic acid 등은 팽화로 크게 감소한 반면 syringic acid는 약 6배 증가하였다. 팽화후 홍미삼의 절단면 미세구조는 팽화전보다 훨씬 균일화한 입자모양을 나타내었으며, 팽화전후 펩신소화율은 각각 22.4, 46.2%로 통계적으로 유의한 차이가 있었다 (p<0.05). 본 결과는 팽화처리에 의해 홍미삼의 생리활성성분의 증가와 기호성 및 소화율을 높일 수 있음을 시사하는 것으로 판단되었다.

미삼의 추출 조건에 따른 인삼 프로사포게닌 성분 변화 (Changes in the Contents of Prosapogenin in Ginseng Radix Palva (Panax ginseng) Depending on the Extracting Conditions)

  • This study compared the contents of ginseng prosapogenin depending on the extracting conditions of Ginseng Radix palva(Panax ginseng) to provide basic information for developing Ginseng Radix palva-based functional foods. Our findings show that the content of crude saponin peaked at 18 hours of extraction and when extracted twice at $100^{\circ}C$ (GRP-18). However, the content of total saponin reached its height at 6 hours of extraction at $100^{\circ}C$ (GRP-6) and when extracted twice. On the other hand, the content of ginsenoside $Rg_3$, $Rg_5$ and $Rk_1$ from Red and Black ginseng reached their heights at 18 hours of extraction, followed by 72 hours and 15 hours of extraction at $100^{\circ}C$. And at $100^{\circ}C$ the main prosapogenin of the content of Black ginseng ginsenoside $Rg_5$ and $Rk_1$ reached their heights at 18 hours of extraction, followed by 72 hours and 15 hours of extraction.

홍삼 사포닌의 항불안 효과 (Anxiolytic Effect of Ginseng Total Saponin)

  • 홍삼사포닌분획의 항불안작용의 양상을 검색하고자 사포닌분획을 사용하여 실험 동물에서 항불안작용을 검색하고 이들의 작용을 benzodiazepine의 대표적 약물인 diazepam의 효과와 비교하여 보았다. 실험동물에서 항불안효과의 검정을 위하여 생쥐에게 각각 상이 한 단일성분 함량을 지닌 여러분획들을 투여 후 elevated plus maze법을 사용하여 항불안효과를 비교, 관찰하였다. 실험결과 홍삼 ginsenoside Rg1, Rf 및 Re 등의 함량이 보다 높은 TSI 분획에서 항불안 효과를 관찰하였다. 중추신경계에 작용하여 진정효과를 나타내는 약물 중 특히 항불안제의 작용에 관여하는 신경전달물질 수용체로 현재까지 가장 주목을 받고 있는 것은 GABA A-benzodiazepine 수용체-chloride통로 복합체에 있는 benzodiazepine 수용체이다. 본 실험결과 관찰한 홍삼 사포닌의 항불안작용의 기전 규명을 위한 접근방법으로 홍삼 사포닌의 benzodiazepine 수용체에 대한 수용체 결합력을 관찰하고자, 흰쥐의 대뇌 피질을 검체로 하여 benzodiazepine 수용체에 대한 [$^3H$] Ro15-1788 결합 실험을 실시하였다. 홍삼 사포닌 분획들은 benzodiazpine 수용체에 결합하는 반응성을 보였으며, ginsenoside Rb, Re 및 Rd등의 함량이 높은 TSIV 분획이 가장 높은 결합력을 나타내었는데, 이 분획에서는 항불안 작용을 관찰할 수 없었다. 이상의 결과에서 볼 때 홍삼사포닌은 항불안작용을 나타내었으며, 이 항불안작용과 benzodiazepine 수용체에 대한 결합력과의 연관성은 관찰되지 않았다.

Effects of Panax ginseng on Morphine-induced Immune Suppression

  • To investigate the possibility of Panax ginseng as a therapeutic agent for the immune suppression, ginseng total saponin (GTS) extracted from korean red ginseng was tested on immune functions from morphine-induced immune suppressed mice. To study how immune functions are affected by morphine and also to test whether GTS can be an useful therapeutic agent for morphine toxicity, several parameters were employed, body weight, immune organ weight, B cell functions, and T cell function. Morphine impaired the development of body weight and immune organ weight such as spleen and thymus. Morphine also depressed a B-cell function, antibody production. T-cell functions studied by type IV hypersensitivity test were most markedly affected by morphine treatment. GTS restored most of morphine-induced immune suppression. GTS restored the morphine-induced decrease in spleen weight to body weight ratio in a dose dependent manner, but not the body weight decrease. Also all of the morphine-induced impairments of B cell functions and cellmediated immunity were fully recovered by GTS. These results suggest that ginseng product could be very helpful for the treatment of immune suppression occurring in morphine abusers.

고온 열처리가 홍삼물추출물의 이화학적 특성에 미치는 영향 (Effects of High Temperature Heating on the Some Physicochemical Properties of Korean Red Ginseng (Panax ginseng C.A. Meyer) Water Extract)

  • This study was carried out to investigate the some physicochemical properties of Korean red ginseng (Panax ginseng C.A. Meyer) water extract (RGWE) after heated with high temperatures above $100^{\circ}C$ for 2 hours. RGWEs were heated at 100, 110 and $120^{\circ}C$ for 2 hours by using autoclave. After RGWEs were heated at high temperature for 2 hours without not adjustment of pH, the changes of saponin, free sugars, mineral and color in the RGWEs were investigated. Total ginsenoside content in control was 1.99%, while those of RGWE were 1.65, 1.49 and 1.29% when treated at 100, 110 and $120^{\circ}C$, respectively. The contents of total ginsenoside showed decreased tendency as heating temperatures were increased. The ginsenoside-$Rh_{2}$ and $-Rg_{3}$, which have been reported as very stable red ginseng ginsenosides, showed relatively strong spots on TLC when RGWEs were heated at 110 and $120^{\circ}C$. In case of free sugars in RGWEs, fructose, glucose and maltose showed high contents when compared with control, while Fe, Ca and Mg ions showed very low contents. Value of L in RGWE treated with high temperature was almost the same with control, while values of a and b were increased. Values of a were increased from -0.86 of control to +0.04, +0.05 and +1.14 when treated with 100, 110 and $120^{\circ}C$, respectively. Values of b also were increased from 27.68 of control to 33.61, 33.61 and 37.42 when treated with 100, 110 and $120^{\circ}C$, respectively. Values of total color in RGWEs treated with high temperatures, E, were finally increased by values of a and b.

수삼의 저장조건에 따른 홍삼의 이화학적 특성 (Physicochemical Properties of Red Ginseng on Storage Condition of the Fresh Ginseng)

  • 본 연구는 수삼의 저장기간 (무저장, 2일, 6일, 8일, 10일) 및 저장온도 $(20^{\circ}C,\;34^{\circ}C,\;-10^{\circ}C)$로 구분하여 제조된 홍삼의 품질 및 이화학적 특성변화를 비교 조사하였다. 수삼의 수분함량은 저장일이 길어질수록 감소하는 경향이었다. 수삼을 10일동안의 단기간 저장온도는 $34^{\circ}C$의 냉장저장이 적합하다고 생각한다. 전질소 함량은 저장일 10일로 갈수록 무저장구에 비해 다소 증가하는 경향이었다. 저장온도별로는 실온, 냉동, 냉장저장 순으로 무저장구보다 전질소 함량이 증가함을 보였다. 각각의 저장구별 페놀성 화합물의 함량은 무저장구에 비하여 저장기간이 길수록 증가하는 경향이었다. 저장 온도별로는 냉장 저장구가 실온, 냉동 저장구에 비하여 다소 페놀성 화합물의 함량이 높았다. 저장조건별 홍삼의 사포닌 Rf 에 대한 8종의 $ginsenoside\;Rb_1,\;Rb_2,\;Rc,\;Rd,\;Re,\;$Rg_3,\;Rg_1,\;Rg_2$의 상대적 함량 (%)비 변화를 분석한 결과, 각각 저장조건에서 Rf 대한 $Rb_1,\;Rg_1$의 상대적 함량비가 저장기간이 늘어날수록 감소하는 경향이었다. 기타 ginsenoside 함량변화는 시료간의 차이에서 나타난 것으로 판단되며 저장조건에 따른 변화 경향은 뚜렷하지 않았다. 관능적 성질에서 각각 저장구별로 홍삼의 품질, 맛과 향을 종합적으로 평가하여 볼 때, $34^{\circ}C$에서 냉장저장 운반하는 것이 홍삼 향미의 안정적 유지에 가장 적합한 것으로 평가되었다.

Inhibitory effects of total saponin from Korean red ginseng via vasodilator-stimulated phosphoprotein-Ser157 phosphorylation on thrombin-induced platelet aggregation

  • In this study, we have investigated the effects of total saponin from Korean red ginseng (TSKRG) on thrombin-induced platelet aggregation. TSKRG dose-dependently inhibited thrombin-induced platelet aggregation with $IC_{50}$ value of about 81.1 ${\mu}g/mL$. In addition, TSKRG dose-dependently decreased thrombin-elevated the level of cytosolic-free $Ca^{2+}$ ($[Ca^{2+}]_i$), one of aggregation-inducing molecules. Of two $Ca^{2+}$-antagonistic cyclic nucleotides as aggregation-inhibiting molecules, cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), TSKRG significantly dose-dependently elevated intracellular level of cAMP, but not cGMP. In addition, TSKRG dose-dependently inhibited thrombin-elevated adenosine triphosphate (ATP) release from platelets. These results suggest that the suppression of $[Ca^{2+}]_i$ elevation, and of ATP release by TSKRG are associated with upregulation of cAMP. TSKRG elevated the phosphorylation of vasodilator-stimulated phosphoprotein (VASP)-$Ser^{157}$, a cAMP-dependent protein kinase (A-kinase) substrate, but not the phosphorylation of VASP-$Ser^{239}$, a cGMP-dependent protein kinase substrate, in thrombin-activated platelets. We demonstrate that TSKRG involves in increase of cAMP level and subsequent elevation of VASP-$Ser^{157}$ phosphorylation through A-kinase activation to inhibit $[Ca^{2+}]_i$ mobilization and ATP release in thrombin-induced platelet aggregation. These results strongly indicate that TSKRG is a beneficial herbal substance elevating cAMP level in thrombin-platelet interaction, which may result in preventing of platelet aggregation-mediated thrombotic diseases.
