A STUDY OF THE ROOT SURFACE AREA, ANATOMIC STRUCTURE AND LINEAR VARIATION OF THE ROOT SURFACE AREA OF THE MANDIBULAR SECOND MOLAR (하악 제2대구치 치근의 형태학적 구조 및 표면적에 관한 연구)

  • The thirty six mandibular second molars, which were extracted because of hopeless tooth due to advanced periodontal disease, were measured the length of mesial and distal root and the distance from cementoenamel junction to root separation. The molars were cross-sectioned every 1.5 milimeter from cementoenamel junction to root apex perpendicular to long axis and each section was photographed, projected and measured with a calibrated Digital Curvi-Meter(Com Curvi-8. Japan). The root surface area (RSA), percentage of the RSA and the linear variation of the RSA were calibrated for each 1. 5 mm section. The results were as follows. 1. The mean length of the roots was 12. 98mm for mesial root, 11.84 mm for distal root. The mesial root was longer than distal root.(p<0.01) 2. The mean distance from the cementoenamel junction to the point at which the root separate from the root trunk was 3.82mm for the buccal furcation and 4.75mm for lingual furcation. The buccal root separation was coronal than the lingual root separation.(p<0.01) 3. The total root surface area was $317.78mm^2$. 4. The mean surface area of the root trunk was $150.06mm^2$ and averaged 42.54% of the total root surface area. 5. The mean root surface area was $88.79\;mm^2$ for the mesial root, $78.93mm^2$ for distal root, The mesial root surface area was wider than the distal root surface area.(p<0.05) 6. In comparision, the mean root trunk surface area of the mandibular 2nd molar was wider than that of mandibular 1st molar(p<0.01), but each root of 2nd molar was smaller than that of 1st molar(p<0.01).

Effect of Pot Depth on Root Development and Distribution during Seedling Growing Period in Tobacco. (육묘포트 깊이가 담배의 뿌리발달 및 분포에 미치는 영향)

  • The study was carried out to clarify reasonable production of healthy seedling, optimal Pot depth, and appropriate transplanting time, which can be deduced from understanding of seedling quality. Seedling quality results from growth of root and shoot, morphology and distribution of root system under influence of Pot depth during seedling growing period. Stem height, shoot dry weight, leaf area and leaf number were increased in proportion to depth of pots. Growth of shoot and root during seedling growing period showed the most dramatic development between 20th and 25th day after temporary planting. Root number increased as pot depth decrease and total root length and dry weight increased as pot depth increase. In 5cm pot, relative multiplication rate was higher and mean extension rate was lower than other depth of Pot. The limitation of pot volume in which rhizosphere was located enhance the development of roots of second and third order. At 20th days after temporary Planting root distribution was relatively uniform in length and development of adventitious root on stem base was poor as Pot depth decreased.

Effect of Trichokonins on the Growth Characteristics of Paeonia ostii 'Fengdan' Seedling Roots

  • The growth characteristics of Paeonia ostii 'Fengdan' seedlings roots in response to trichokonins-spray treatment were investigated in this study. One-year-old seedlings of P. ostii 'Fengdan' were potted in plastic cups containing garden mold and grown under field conditions. The results showed that application of trichokonins significantly promoted root growth in P. ostii 'Fengdan' seedlings. The total root projection area, total root surface area, total root volume, total root length, root number and root diameter of seedlings treated with 0.25 mg/L trichokonins were higher by 141.70, 116.59, 119.44, 55.97, 348.88 and 127.78%, respectively, than that of the control. Thus, the results supported the hypothesis that good growth condition for roots could directly improve their nutrient absorption and utilization efficiency, promoting plant growth and development.

Enhancing Effect of Shimizuomyces paradoxus on Seed Germination and Seedling Growth of Canola, Plant Growth of Cucumber, and Harvest of Tomato

  • Shimizuomyces paradoxus showed no inhibitory effect against plant pathogen fungi, such as Fusarium oxysporum f. sp. lycopersici and Alternaria solani. The S. paradoxus culture filtrate showed higher seed germination and seedling growth rates in canola than distilled water and potato-dextrose broth. A conidial suspension of $1.0{\times}10^4/mL$ resulted in the highest growth stimulating effects on total plant length, and fresh and dry weight of shoots and roots in cucumber, when compared to the highest suspension concentration. Total plant length and shoot weight increased with the foliar spray treatment, and root length and root weight increased by simultaneous treatments of soil drenching and foliar spray in cucumber. Lower concentrations of the S. paradoxus conidial suspension increased the harvest of tomato fruit.

Investigation of Root Morphological and Architectural Traits in Adzuki Bean (Vigna angularis) Cultivars Using Imagery Data

  • Roots play important roles in water and nutrient uptake and in response to various environmental stresses. Investigating diversification of cultivars through root phenotyping is important for crop improvement in adzuki beans. Therefore, we analyzed the morphological and architectural root traits of 22 adzuki bean cultivars using 2-dimensional (2D) root imaging. Plants were grown in plastic tubes [6 cm (diameter) × 40 cm (height)] in a greenhouse from July 25th to August 28th. When the plants reached the 2nd or 3rd trifoliate leaf stage, the roots were removed and washed with tap water to remove soil particles. Clean root samples were scanned, and the scanned images were analyzed using the WinRHIZO Pro software. The cultivars were analyzed based on six root phenotypes [total root length (TRL), surface area (SA), average diameter (AD), and number of tips (NT) were included as root morphological traits (RMT); and link average length (LAL) and link average diameter (LAD) were included as root architectural traits (RAT)]. According to the analysis of variance (ANOVA), a significant difference was observed between the cultivars for all root morphological traits. Distribution analysis demonstrated that all root traits except LAL followed a normally distributed curve. In the correlation test, the most important morphological trait, TRL, showed a strong positive correlation with SA (r = 0.97***) and NT (r = 0.94***). In comparison, between RMT and RAT, TRL showed a significantly negative correlation with LAL (r = -0.50***); however, TRL did not show a correlation with LAD. Based on RMT and RAT, we identified the cultivars that ranked 5% from the top and bottom. In particular, the cultivar "IT 236657" showed the highest TRL, SA, and NT, while the cultivar "IT 236169" showed the lowest values for TRL, SA, and NT. In addition, the coefficient of variance for the six tested root traits ranged from (14.26-40%) which suggested statistical variability in root phenotypes among the 22 adzuki bean varieties. Thus, this study will help to select target root traits for the adzuki bean breeding program in the future, generating climate-resilient adzuki beans, especially for drought stress, and may be useful for developing biotic and abiotic stress-tolerant cultivars based on better root trait attributes.

  • The present study was to evaluate the accuracy of the frequency dependent type apex locator, Root-ZX. The subjects included 505 root canals of 238 teeth treated by the Department of Conservative Dentistry, and 22 human premolars which were schduled to be extracted for the orthodontic reasons. The results were as follows ; 1. The working lengths determined by Root-ZX were compared with radiographic readings. Of the total 505 root canals, 66 % showed coincidence within ${\pm}0.5mm$ and the average readings of Root-ZX were $0.13mm{\pm}1.05$ longer than those of radiographic readings. 2. The length difference between the file tip determined by Root-ZX and the apical constriction in extracted teeth were measured. Of the total 24 root canals, 70.8 % showed coincidence within ${\pm}0.5mm$ and the average readings of Root-ZX were $0.12mm{\pm}0.50$beyond the apical constriction. 3. The vitality of the teeth did not show any statistical difference(p>0.05) in the accuracy of the Root-ZX readings. The presence of the periapical lesions, however, significantly lowered the percentage of ${\pm}0.5mm$ accuracy in Root-ZX measurements.(p<0.05). In the presence of periapical lesions, the percentage within ${\pm}0.5mm$ was significantly lower.

Effects of Water Stress on Leaf Water Potential, Photosynthesis and Root Development in Tobacco Plant (수분 스트레스가 담배의 잎 수분 포텐셜, 광합성 및 뿌리발달에 미치는 영향)

  • Development of shoot and root, leaf water potential and photosynthetic rate affected by water stress in early growing stage of tobacco were surveyed to interpret stress response in terms of plant physiological and agricultural aspects. The growth of shoot and root was highly suppressed by water stress and the difference in dry weight by rewatering was smaller in root than in shoot. The total root length was highly decreased by water stress and the lengths of root for water stress and non-stress were 74m and 84m, respectively, after rewatering. The root growth treated by water stress was increased between 2nd and 3rd day after treatment indicating that temporary water stress at early growing stage might have increased of root zone activity for early growth stage. The leaf water potentials were decreased to -7.63MPa, -9.47MPa, -11.89MPa, -13MPa at the 2nd, 3rd, 4th and 5th day by water stress. The relative water contents were 75%, 62% and 57% at the 3rd, 4th and 5th day after treatment. Photosynthesis was reduced largely by water stress. The photosynthetic rate after treatment at 2nd day and 3rd day was dropped to 18.15$\mu$mol. $CO_2$/$m^2$ㆍsec$^{-1}$ and 9.35$\mu$mol. $CO_2$/$m^2$ㆍsec$^{-1}$. It was never recovered to the normal, even after rewatering. Stomatal conductance had been reduced since 2nd day after treatment and increased after rewatering.

Growth Characteristics, Main Constituents and Antioxidant Activities in Local Accessions of Sophora flavescens AIT. (고삼 수집종의 생육특성, 주요성분 및 항산화 활성)

  • Background: Sophora flavescens AIT root is used as a herbal medicine in Asian culture. This study was aimed at selecting the best cultivars, by comparing growth characteristics, active ingredients, and antioxidant activities. Methods and Results: A total twenty nine accessions of S. flavescens were collected from five different regions in Korea. Plant height, leaf length, peduncle length, fresh root weight, and seed weight were the growth characteristics selected. Antioxidant activity was determined by measuring the total polyphenol (TPC) and total flavonoid contents (TFC) as well as the radical scavenging activity of DPPH, ABTS and FRAP. Oxymatrine and matrine, active ingredients in S. flavescens were analyzed by HPLC. Results indicated that accession YS-11 had the highest plant height (152.5 cm) and peduncle length (54.0 cm). The fresh root weight was highest in JA-01 at 4.9 kg, while the highest total seed weight was recorded in accession BH-04 at 77.7 g. The YS-03 accession contained the highest oxymatrine and matrine total contents (0.9616% and 0.9638%, respectively). Accessions BH-02, YS-11, YS-05, and MJ-03 had the highest levels of antioxidant activity (p < 0.05). Oxymatrine showed a high correlation with TPC and FRAP. TPC showed high correlations with TFC, DPPH, ABTS and FRAP. TFC showed a high correlation with DPPH. Conclusions: The superior lines can be selected for use as standard variety breeding material by comparing the growth characteristics of the accessions.

Difference in Growth Characteristics of 5-Year-Old Ginseng Grown by Direct Seeding and Transplanting (품종 증식을 위한 매년 채종시 직파와 이식에 따른 5년생 인삼의 품종별 지하부 생육 특성)

  • Background : In order to determine the effects of planting methods on root growth of ginseng varieties, two different methods, direct seed sowing and transplanting were compared in terms of their effects on different root growth characteristics. Methods and Results : Higher fresh root weight was observed in ginseng grown by direct seed sowing. Direct seed sowing of three cultivars (Sunhyang, Chungsun and K-1) resulted in higher yield, whereas no difference was observed in the yield of one cultivar (Chungsun). Gumpoong was highly tolerant to physiological stress, as it showed fewer symptoms of rusty and rough skin root diseases in both direct seed sowing and transplanting. The average main root length per total root length of ginseng grown by direct seed sowing was 33.6%, whereas that of ginseng grown by the average of those by transplanting was 22.4%. Other root growth characteristics, including root length, main root diameter, and number of side roots, improved when the direct seed sowing method was used. Conclusions : To our knowledge, this is the first study reporting the differences in root growth parameters of ginseng varieties grown by direct seed sowing or transplanting at the same planting density. Because of the advantages of direct sowing during ginseng planting, developing new varieties and improving cultivation methods are imperative.

Symbiotic Effectiveness of Bradyrhizobium japonicum USDA 110 in Supernodulating Soybean Mutant SS2-2

  • In the absence of exogeneous nitrogen supply, evaluation of a symbiosis effectiveness of Bradyrhizobium japonicum USDA 110 in a supernodulating soybean mutant, SS2-2, its wild type, Sinpaldalkong 2, and control genotype, Jangyeobkong, was conducted in this study. Nodules in SS2-2 were initially white and similar to its wild type, Sinpaldalkong 2. At the late stage, the wild type nodules became dark pinkish by maturation, by contrast, mature nodules in SS2-2 remained light green to pinkish, indicating a lack of leghemoglobin. Tap root length was short in nodulated symbiotic SS2-2 than that of its wild type and the control genotype. Nodulated root length and nodule density on root length were significantly increased by B. japonicum inoculation, but no significant increase was observed on root length and percentage of nodulation to total root length. Regardless of Bradyrhizobium inoculation, SS2-2 showed higher nodule dry weight and higher acetylene reduction activity (ARA) when compared with its wild type and the control genotype. Inoculation of B. japonicum leaded the increase of ARA in 47 days after planting (DAP), in part because of nodule development. Supernodulating mutant, SS2-2, less responded to B. japonicum induction in terms of nitrogen fixation and nodulation characteristics than its wild type. Thus, interaction of supernodulating soybean mutant with Bradyrhizobium had less symbiotically associated response than normal nodulating soybean.
