비만아동의 지질과산화물 형성과 항산화 체계에 관한 연구 (LDL Oxidation, Total Radical Trapping Antioxidant Potential and Plasma Antioxidant Vitamin Systems in Obese School Children)

  • The purpose of this study was to examine the lipid peroxidation, plasma antioxidant status and insulin resistance in childhood obesity. To this end, we measured blood lipid profiles, glucose, insulin concentrations, plasma antioxidant vitamins, baseline conjugated diene formation as a measure of LDL oxidation in vivo and TRAP (total radical trapping antioxidant potential) of 93 school children (58 nonobese, 35 overweight-obese). Insulin resistance was estimated by homeostasis model assessment of insulin resistance (HOMA-IR). The overweight-obese children showed significantly higher levels of leptin (p < 0.0001) and triglyceride (p < 0.05) and significantly lower level of plasma Iycopene (p < 0.001) and $\gamma$-tocopherol (p < 0.05) compared with the normal weight children. Furthermore, the levels of TRAP were significantly lower in overweight-obese children (p < 0.05). Significant positive relationships between plasma leptin and conjugated dienes formation (p < 0.005) and inverse relationship between plasma leptin and lipid corrected levels of $\beta$-carotene (p < 0.05), Iycopene (p < 0.05) were observed. Our results showed an increased lipid peroxidation and decreased antioxidant capacity in childhood obesity which could be involved in the atherosclerotic process.

규칙적인 운동습관이 남자 성인의 적혈구내 항산화효소활성과 혈장 항산화능력(TRAP)에 미치는 영향 (Effects of Regular Physical exercise Habits on the Activities of Erythrocyte Antioxidant Enzyme and Plasma Total Radical-trapping Antioxidant Potential in Health Male Subjects)

  • In the present work we investigated the effect of regular physical exercise on the activities of erythrocyte antioxidant enzyme, plasma total radical-trapping antioxidant potential(TRAP) and plasma level of lipid peroxidation(malondialdehyde, MDA) in 64 healthy male, aged 34-67 years. The study population were divided in two groups: small amount of exerciser(exercise time less than 10min/d) and moderate amount of exerciser(exercise time more than 20min/d) according to their physical exercise habits measured by a questionnaire. Erythrocyte superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) and catalase(CAT), plasma TRAP, as well as plasma MDA were determined. Erythrocyte GSH-Px and plasma TRAP were higher in moderate amount of exercisers than those in small amount of exercisers by 17% and 26%, respectively. No significant differences were observed in erythrocyte SOD, CAT and plasma MDA between the two groups. Mean exercise time was positively correlated with the erythrocyte GSH-Px activity and plasma TRAP significantly. The results would sugest that regular moderate exercise enhances antioxidant defences against reactive oxygen species and may increase the likelihood of a healthier life span.

혈장 총 율기 포집 능력(TRAP) 수준 및 항산화 비타민 영양상태와 인체 임파구 DNA 손상정도와의 상호관련성 연구 (The Associations between Plasma Concentrations of Total Radical-Trapping Antioxidant Potential(TRAP), Antioxidant Vitamins and DNA Damage in Human Lymphocytes)

  • The spontaneous frequency of genetic damage and the possible relationship of this damage to total radical-trapping antioxidant potential(TRAP) and antioxidant vitamins, including plasma levels of $\alpha$-carotene, $\beta$-carotene, cryptoxanthin, retinol, $\alpha$-tocopherol and ${\gamma}$-tocopherol in humans were investigated in 57 subjects using two indices of genetic damage, SCE & HFC frequency. The mean of SCE and HFC frequencies were weakly correlated with plasma TRAP(r=-0.305, p<0.1 for SCEs: r=-0.297, p<0.1 for HFCs, respectively), but those were strongly negatively correlated with plasma $\beta$-carotence(r=-0.385, p<0.01 for SCEs : r=-0.392, p<0.01 for HFCs) and cryptoxanthin(r=-0.312, p<0.05 for SCEs : r=0.335, p<0.05 for HFCs, respectively) levels in the subjects. However, those DNA damage markers including SCE and HFC did not correlate with either plasma $\alpha$-carotene, $\alpha$-tocopherol or retinol concentrations. The mean of SCE and HFC frequencies were positively correlated with plasma ${\gamma}$-tocopherol level(r=0.421, p<0.01 for SCEs : r=0.399, p<0.01 for HFCs, respectively). These findings indicate that increased cytogenetic DNA changes, as determined by SCE and HFC frequencies are possibly associated with generation of free radicals in lymphocytes and decreased plasma antioxidant vitamin($\beta$-carotene and cryptoxanthin) status in the subjects.   • Recently developed TRAP assay(Total Radical-trapping Antioxidant Potential) is a new technique which enable effectively the measurement of total antioxidant status in the plasma. Researches using TRAP method have rarely been reported in Korea. Therefore, we investigated effects of various life-style and dietary factors on plasma TRAP level in Korean adult population. The subjects consisted of 161 health adult male(n=105) and female (n=56) aged 19-57. A self-administered questionnaire ascertained life-style factors including smoking habits, alcohol use, physical activity. Dietary factors were analyzed by 24 hour recall method. TRAP was determined spectrophotometrically in plasma at 734nm. Mean plasma TRAP concentrations were slightly higher in males (1.35$\pm$0.02mM) and females(1.30$\pm$0.02mM) without significance. There were significant correlations between plasma TRAP levels and life-style factors including exercise and smoking Plasma TRAP levels were affected more by exercise than by smoking. Moderate amount exerciser(exercise time $\geq$30min/day) had significantly higher level of plasma TRAP levels in male smokers(1.33$\pm$0.03mM) were significantly lower than those in male nonsmokers(1.39$\pm$0.01mM, p<0.05), but the difference was not seen in female subjects. Intakes of vitamin C(r=0.288, p=0.007) or folate (r=0.258, p=0.016) were positively correlated with plasma TRAP levels in nonsmokers, but those relations were not seen in smokers. Dietary factors such as vegetables (r=0.288, p=0.007)or folate (r=0.258, p=0.016) were positively correlated with plasma TRAP levels in nonsmokers, but those relations were not seen in smokers. Dietary factors such as vegetables (r=0.277, p=0.009) and legumes(r=0.263, p=0.013) consumption were positively related to plasma TRAP concentrations also in nonsmokers only. Regular green tea drinkers showed significantly greater level of plasma TRAP than the non green tea drinkers(1.36$\pm$0.01mM vs 1.30$\pm$0.03mM, p<0.05). These results suggest that the regular exercise($\geq$30min daily), nonsmoking, and consumption of vegetables, legumes and green tea would contribute to improving plasma TRAP levels in Korean adults.

Antioxidant Activity of Orange Flesh and Peel Extracted with Various Solvents

  • The aim of this study was to investigate the antioxidant activity of orange (Citrus auranthium) flesh (OF) and peel (OP) extracted with acetone, ethanol, and methanol. Antioxidant potential was examined by measuring total phenolic content (TPC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity (RSA), total radical-trapping antioxidant potential (TRAP), oxygen radical absorbance capacity (ORAC), and cellular antioxidant activity (CAA). The comet assay was used to determine the protective effects of OF and OP against $H_2O_2$-induced DNA damage. TPC was highest in the acetone extracts of OF and OP. DPPH RSA was also higher in the acetone extracts than in the ethanol extracts. The DPPH RSA was highest in the acetone extracts of OF. The TRAP and ORAC values of the all extracts increased in a dose-dependent manner. In the TRAP assay, the acetone extracts of OF and OP had the lowest $IC_{50}$ values. In the CAA assay, the methanol and acetone extracts of OP had the lowest $IC_{50}$ values. All of the samples protected against $H_2O_2$-induced DNA damage in human leukocytes, as measured by the comet assay, but the acetone extracts of OP had the strongest effect. These results suggest that acetone is the best solvent for the extraction of antioxidant compounds from OF and OP. Furthermore, the high antioxidant activity of OP, which is a by-product of orange processing, suggests that it can be used in nutraceutical and functional foods.

포도주스의 보충섭취가 흡연성인의 혈장 항산화 영양상태 및 DNA 손상 개선에 미치는 영향 (The Effects of Purple Grape Juice Supplementation on Improvement of Antioxidant Status and Lymphocyte DNA Damage in Korean Smokers)

  • The purpose of this project was to evaluate whether daily fruit juice consumption could reduce the DNA damage in healthy subjects. The study was performed using 67 healthy volunteers (29 smokers, 38 nonsmokers) who were supple-mented with 480 m1 of grape juice for 8 weeks. Eight weeks of grape juice consumption did not change any anthropometric parameters. Lymphocyte DNA damage before the study was significantly greater (p<0.05) in smoker than nonsmoker, but, grape juice consumption significantly reduced DNA damage in both smoker (26%) and nonsmoker (I7%) to the level where there was no difference remained between the two groups after the intervention trial. This preventive effect of grape juice against DNA damage was not affected by sex of the subjects in non-smokers. Plasma $\alpha$-carotene, Iyco-pene and ${\gamma}$-totopherol was significantly increased after the trial in smokers, while erythrocyte catalase was significan-tly increased in both smokers and nonsmokers. Total radical-trapping antioxidant potential (TRAP) level in all subjects was significantly reduced after the intervention, while GSH-Px activity was increased only in nonsmokers. These results suggests that daily consumption of grape juice may protect DNA damage in peripheral lymphocytes, and supports the hypothesis that grape juice might exert their effect partially via a decrease in oxidative damage to DNA in humans partly by improving their antioxidative defense system.

흰점박이꽃무지(Protaetia brevitarsis) 함유 음료의 in vitro 항산화 관련 생리활성효능 및 안전성 검증 (Antioxidant Activity and Safety Evaluation of Juice Containing Protaetia brevitarsis)

  • 본 연구에서는 흰점박이꽃무지 함유 음료의 총폴리페놀함량과 항산화활성(DPPH 라디칼 소거능, TRAP, ORAC assay, comet assay) 및 사용에 대한 안전성을 검증하기 위하여 ICR mouse를 사용하여 흰점박이꽃무지 무첨가군과 첨가군 단회 투여(2 g/kg) 시 나타나는 치사율, 체중, 생리적 증상, 장기무게, 생화학적 분석(triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol, aspartate transaminase, alanine transaminase)을 통해 급성독성시험을 실시하였다. 흰점박이꽃무지를 함유한 호박, 알로에, 돼지감자음료 1 pack의 총 페놀함량은 66.35~100.98 mg/pack, DPPH 라디칼 소거능은 67.1~90.0%/1 pack, TRAP 값은 $1.46\sim1.54{\mu}M$/trolox equivalent(TE)를 보였으며, 모든 음료의 ORAC 값은 1500보다 높았다. 모든 음료는 인체 임파구에서 세포독성을 보이지 않았으며, $H_2O_2$에 대한 DNA 손상에 대한 보호효과는 유의적이었다. 흰점박이꽃무지 첨가/무첨가 음료의 ICR mouse 암수 각각에 경구투여(2 g/kg) 후 14일까지 치사된 실험동물은 관찰되지 않았으며, 체중, 생리적, 병리적 및 생화학적 분석에서 대조군과 비교 시 그룹들 간의 유의한 변화는 관찰되지 않았다. 그러므로 2 g/kg의 흰점박이꽃무지 첨가/무첨가 음료를 ICR mouse에 단일 경구투여시 독성이 없는 것으로 판명되었다. 본 연구에서 규명된 흰점박이꽃무지 함유 음료의 항산화활성과 안전성은 산업적측면에서 약용 및 식용 자원으로 적극적인 활용을 기대할 수 있을 것으로 사료된다.

솔잎착즙액을 첨가한 설기떡의 가열조리에 따른 항산화 및 항유전독성 효능 검증 (Changes in the Antioxidative and Antigenotoxic Effects After the Cooking Process of Sulgidduk Containing Pine Needle Juice)

  • Recently, two formulas of sulgidduk added to pine needle juice (PNJ) with various physiological activities were developed for metabolic syndrome patients in our lab. According to previous studies, cooking may alter antioxidant properties by initiating destruction, release or transformation of antioxidant compounds contained in food. Therefore, the aim of this study was to compare the antioxidant activities and antigenotixic effects of sulgidduk with/without PNJ and to note changes in these activities after cooking. The ingredients of sulgidduk was added on the basis of 100% rice flour as follows: conventional sulgidduk (S): 1.5% salt, 30.0% sugar; PNJ added to sulgidduk A (PS-A): 1.4% salt, 30.0% sugar, and 1.0% PNJ; PNJ added to sulgidduk B (PS-B): 1.5% salt, 21.4% sugar, and 1.4% PNJ. Ethanol and water extracts of sulgidduk were analyzed for the total phenolic content (TPC), DPPH radical scavenging activity (DPPH RSA), total radical-trapping antioxidant potential (TRAP), oxygen radical absorbance capacity (ORAC), and antigenotoxic effect by comet assay. The ethanol extracts PS-A and PS-B showed higher TPC and antioxidant activities (DPPH RSA, TRAP, and ORAC) than did the S ethanol extract before cooking. The more PNJ was added, the higher TPC and anitoxidant activities were observed in sulgidduk (PS-A$200{\mu}M$ of $H_2O_2$. Taken together, this study suggests that sulgidduk added to 1.44% of pine needle juice may be a good option antioxidant and antigenotoxic source.

Influence of Functional Food Containing Bacillus polyfermenticus SCD on Lipid and Antioxidant Metabolisms in Rats Fed a High-Fat and High-Cholesterol Diet

  • We evaluated the effects of newly developed functional food containing Bacillus polyfermenticus SCD as the main material on the lipid and antioxidant metabolisms of hypercholesterolemic rats. Thirty male SD rats were divided into three groups after a 1-week adaptation period and were fed with a high fat-cholesterol diet (control), or with a high fat-cholesterol diet supplemented with low or high doses ($3.1\;{\times}\;10^6\;cfu/day$ or $3.1\;{\times}\;10^8\;cfu/day$) of B. polyfermenticus SCD and other physiological active materials for 6 weeks. Both doses of B. polyfermenticus SCD significantly reduced hepatic total cholesterol and triglycerides, while increasing the fecal excretion rates of total lipid, total cholesterol and triglycerides. B. polyfermenticus SCD increased the total radical trapping antioxidant potential (TRAP). The erythrocytic glutathione peroxidase activity in the B. polyfermenticus groups was significantly lower than that in the control group. Plasma TRAP levels exhibited a highly significant negative correlation with hepatic total cholesterol while a significant positive correlation was detected between fecal total cholesterol and plasma TRAP. This hypolipidemic and antioxidative effect of B. polyfermenticus SCD seemed to be unrelated to its dosage. These results suggest that functional food containing B. polyfermenticus SCD can improve oxidative stress and hepatic lipid profiles by enhancing the excretion of cholesterol and triglycerides in feces of rats fed with high fat-high cholesterol diet.

사리장의 항산화 효과 (In vitro Antioxidant Effects of Sarijang)

  • 본 연구에서는 서목태를 기본으로 하여 제조된 전통 발효물인 사리장의 항산화 활성 분석 및 comet assay를 이용한 DNA 손상 억제 효과를 분석하고자 하였다. 사리장의 총 폴리페놀 함량은 $1.04{\pm}0.01$ mg GAE/mL로 나타났다. 항산화 활성을 분석한 DPPH 라디칼 소거능 및 TRAP는 농도 의존적으로 활성이 증가하였으며, 각각의 $IC_{50}$은 11.2 mg/mL와 1.2 mM로 나타났다. ORAC 활성 역시 농도 의존적 증가 활성을 나타내었다. 세포의 ROS 소거능(CAC)은 사리장 처리구의 모든 농도(10~100 ${\mu}g/mL$)에서 NC와 동일한 수준의 ROS 억제 활성을 나타내었다. Comet assay를 이용한 DNA 손상 보호 효과는 $H_2O_2$, Fe-NTA 그리고 HNE에 의한 산화적 스트레스에 의한 DNA 손상을 농도 의존적으로 보호하는 것으로 나타났으며, $IC_{50}$$H_2O_2$ 처리군이 13.4 ${\mu}g/mL$, Fe-NTA 처리군이 32.2 ${\mu}g/mL$, HNE 처리군이 59.9 ${\mu}g/mL$로 나타났다. 이상의 결과들은 사리장이 항산화 관련 생리활성을 가지는 것으로 판단되며, 향후 사리장에 포함된 생리활성 성분의 탐색과 in vivo 모델을 통한 생리활성 연구가 이루어져야 할 것으로 보인다.
