Total polyphenols and antioxidant activties of the extract from leaves of Stachys sieboldii MIQ.

  • 초석잠(stachys sieboldii MIQ)의 용매 추출물에서 측정한 total polyphenols 함유량이 15${\sim}$21%으로 나타나 일반적으로 알려져 있는 녹차의 함유량인 20${\sim}$25%와 비슷하며 특히 에틸아세테이트 추출물에서 가장 높은 함유량을 나타내었다. DPPH법에 의한 항산화 활성 측성에서도 기존의 항산제인 ${\varpropto}-topophenol$, BHT 그리고 BHA에 비교해서 뛰어난 활성도를 나타내었고, total polyphenols 함유량이 많으면 높은 항산화 활성도를 보였다. 가장 좋은 활성도를 나타낸 에틸아세테이트 추출물을 sephedex LH-20 column을 통해 5개의 분획을 얻었고 각각의 분획물의 최대 흡광 범위는 일반적으로 알려져 있는 polyphenols 흡광범위인 200nm${\sim}$400nm와 유사함을 나타내었다.

반응표면분석법을 이용한 안젤리카로부터 폴리페놀 성분의 추출공정 최적화 (Optimization of Extraction Process for Total Polyphenols from Angelica Using Response Surface Methodology)

  • 본 연구에서는 항산화성분함량이 높다고 알려진 안젤리카를 이용하여 폴리페놀을 추출하고 반응표면분석법을 이용하여 추출과정을 최적화하였다. 반응표면분석법의 반응치로는 추출수율과 폴리페놀함량을 설정하였고, 추출공정의 계량인자로는 추출시간, 주정/초순수 부피비, 추출온도를 이용하였다. 추출수율과 폴리페놀함량 모두 계량인자의 주효과도와 교호효과도를 모두 고려하였을 때 가장 큰 영향을 미치는 인자는 추출시간이었다. 또한 반응표면분석 결과 안젤리카의 최적추출조건은 추출시간이 2.8 h, 주정/초순수 부피비 64.0 vol%, 추출온도 $56.6^{\circ}C$로 나타났다. 이 조건의 추출수율은 24.6%, 폴리페놀함량은 8.76 mg GAE/g으로 산출되었다. 추출수율과 폴리페놀함량에 대한 회귀방정식의 결정계수 $R^2$은 각각 81.4%와 75.4%이었으며, 종합 만족도는 D = 0.80, 유의성은 5% 이내의 수준에서 인정되었다.

자생식물과 생약자원 추출물의 폴리페놀, 플라보노이드 함량 및 항산화 활성 탐색 (Total Polyphenols, Total Flavonoid Contents, and Antioxidant Activity of Korean Natural and Medicinal Plants)

  • 본 연구에서는 전통적으로 약으로써 이용되었거나 민간요법으로 사용되던 식물을 문헌고찰을 통해 40여종(자생식물 26종과 생약식물 14종)을 선발하여, 폴리페놀과 플라보노이드 함량 측정 및 DPPH 라디칼 소거능을 측정하였다. 자생식물인 비수리의 폴리페놀(228.90 mg/g extract)과 플라보노이드 함량(90.15 mg/g extract)이 가장 높았으며, DPPH 라디칼 소거능(75.69%)이 모두 높게 나타났다. 감나무와 관중의 폴리페놀 함량(감나무 112.98 mg/g extract, 관중 121.46 mg/g extract)과 플라보노이드 함량(감나무 46.10 mg/g extract, 관중 58.69 mg/g extract)은 높은 편은 아니었으나, DPPH라디칼 소거능은 각각 72.87%와 79.50%로 가장 높게 나타났다. 생약식물에서는 삼백초의 총 폴리페놀함량이 194.60 mg/g extract, 애엽이 203.93 mg/g extract로 폴리페놀 함량이 높았으며, DPPH 라디칼 소거능(삼백초 66.27%, 애엽 66.22%) 또한 높게 나타났다. 그러나 삼백초와 애엽을 제외한 12종류의 생약식물의 DPPH 라디칼 소거능이 50% 미만인 것으로 조사되었다. 최근에 다양한 자생식물과 약용식물의 폴리페놀, 플라보노이드 함량 및 기능성 탐색에 대한 연구가 많이 진행되고 있지만, 아직까지도 연구되지 않은 무수히 많은 식물 소재들이 많아서 지속적인 연구가 필요하다. 본 연구 결과, 그 동안 대중화 되어있지 않았던 식물 소재들의 기능성 소재로의 가능성이 새롭게 조명 될 수 있을 것으로 사료되며 이와 관련된 의약품, 미용제품 및 건강보조식품 등의 개발에 기초자료로 이용될 수 있을 것으로 기대된다. 향후 본 연구에서 조사한 자생식물과 생약식물 중 총 폴리페놀함량과 플라보노이드 함량이 높고, DPPH 라디칼 소거능이 뛰어나게 나타난 몇몇 식물에 대한 심도 있는 연구가 필요할 것으로 판단된다.

고들빼기 김치가 단백질 소화율에 미치는 영향 (Effect of Godulbaegi(Korean Lettuce, Ixeris sonchifolia H.) Kimchi on the in vitro Digestibility of Proteins)

  • Extent of reduction in protein digestibility by Godulbaegi(lxeris sonchifolia H.) kimchi powder appeared to be related to the kimchi weight-to-protein weight ratio, the kind of protein sources, part of plants and fementation period. In vitro digestibilities of protein were significantly(p<0.05) reduced as the freeze dried kimchi powder weight-to-protein ration increased from 0.5 : 1.0 to 1.0 : 1.0 for all Godulbaegi samples and protein sources. Overall digestibility of protein sources in the presence of kimchi powder, the reduction ranked in the following order : soybean>casein>beef>squid. Lower(p<0.005) reduction occurred for each protein source when raw plant was exchanged for kimchi products. Some greater reduction of digestibility was noted in young plants and leaf samples than ripe or root samples. Trypsin inhibitor, which expressed as soybena trypsin inhibitor, was inversely related(r=0.8437) to in vitro protein digestibility of casein in the presence of Godulbaegi kimchi powder. More than three times of total polyphenols contained in leaves than in roots. Young leaves had 30% more total polyphenols(37.64mg/g sample) than that in ripe ones. Soaking in 5% NaCl solution for 24 hrs was markedly reduced in total polyphenols as 73% for leaves and 33% for roots. Remarkable reduction in total polyphenols was not checked during fermentation followed after soaking. Trypsin inhibitor content correlated well(r=0.8873) with total polyphenols in all of Godulbaegi samples.

Polyphenolic Contents and Antioxidant Activities of Underutilized Grape (Vitis vinifera L.) Pomace Extracts

  • Grape pomace is an abundant source of underutilized winery by-products. Polyphenols were extracted from grape pomace using cellulase and gluco-amylase enzymes. 2,2-diphenyl-1-picrylhydrazyl (DPPH) and Folin-Ciocalteu's assays were used to measure antioxidant activity and total polyphenolic contents. Both cellulase, and gluco-amylase digested grape pomace showed efficient radical scavenging activity. In addition, the total polyphenolic contents of cellulase digested grape pomace showed lower concentrations were effective compared to higher concentrations, whereas glucoamylase enzyme did not show remarkable variations. The DPPH radical scavenging activity and total polyphenolic contents were significantly higher in the cellulase digested grape pomace compared to the gluco-amylase digested and the not digested grape pomace. It is notable that enzymatic digestions were efficient for extracting polyphenols from grape pomace. The underutilized grape pomace polyphenols can be further used for food safety as a natural antioxidant.

Development of an Analytical Approach for the Utilization of Edible Tree Sprouts

  • This study evaluated the general nutritional ingredients such as crude fats, crude ashes, crude proteins, total polyphenols, and total flavonoids in 18 kinds of edible tree sprouts. The tree sprouts of Philadelphus schrenckii, Lycium chinense, and Morus alba had the highest crude fat, crude ash, and crude protein content, respectively. The tree sprouts of Cedrela sinensis (CSS) with high ABTS+ radical scavenging activities had a high content of total polyphenols (175.65 mg/g ext.) and total flavonoids (75.18 mg/g ext.). The simultaneous determination of flavonoids such as rutin, isoquercitrin, quercitrin, afzelin, and quercetin in CSS was conducted using high-performance liquid chromatography with a wavelength of 270 nm. Among the flavonoids, the content of quercitrin in CSS was the highest at 59.28 mg/g ext. This study also aids the quality control of many edible tree sprouts by analyzing the general components, total polyphenols, and total flavonoids.

상수리 나무중 Polyphenol 성분들의 분포에 관한 연구 (Studies on the Distribution of Polyphenols in the Parts of Quercus acutissima)

  • 상수리나무(Quercus acutissima, 3년생)의 잎, 줄기, 뿌리, 껍질, 및 열매에 각각 존재하고 있는 polyphenol 성분의 분포를 thin layer chromatography와 column chromatography를 이용하여 조사하였다. 총 25종류의 polyphenol 성분을 분리하였는데, 이들 중 잎에서 15개, 줄기에서 11개, 뿌리와 껍질에서 각각 7개를 분리하였으며, 열매에서는 4개의 성분을 분리하였다. 분리한 polyphenol 성분들 중에는 catechin 성분이 모든 부위에서 비교적 많이 분포하고 있었으며 일부 flavonoid 성분들도 확인되었다. 잎을 제외한 모든 부위에서 3종류의 polyphenol isomer의 존재를 확인했으며, 잎에서는 monomer와 oligomer성 polyphenol의 존재를 확인했으나 polymeric polyphenol이 확인되지 않았다. Quercus acutissima에서 polyphenol의 총함량은 껍질에서 상대적으로 가장 많았고, 다음으로는 뿌리, 잎, 열매의 순으로 많이 분포하였으며, 줄기에서 가장 적은 양이 확인되었다.

미숙사과의 페놀계 화합물의 특성 (Properties of Phenolic Compounds in Unripened Apples)

  • Abundant amounts of unripe apples, which were picked out before maturation, were abolished every year fer wastes. Since unripen apples are known to be rich in functional components, the ultimate objective of this research was to utilize them for food resources. The concentrations of polyphenols in hot water extract were the lowest, those in 75% acetone extract the highest. Concentrations of polyphenols in 5/30 sample were 3.5 times higher than those in 6/30 sample regardless of solvents used for extraction. The degree of condensation of flavanolic tannins in the peel was lower than those of the other parts, and the hot water extract was more condensed than the acetone extract. The amount of total flavonoids in the peel and 5/30 samples were 2.7 and 5.0 times higher than those of the flesh and 6/30 samples, respectively.

  • Dendropanax morbifera H.Lev is an evergreen tree that lives in subtropical climates. About 75 species of D. morbifera live in East Asia, but only one species live in Korea(Korean dendropanax). D. morbifera belonging to Aralicacae family 98% of D. morbifera are distributed in Jeollanam-do in the Korea and the grow wild in Gyeongsangnam-do and Jeju Island. The harvest time and usage of D. morbifera were recorded in traditional medicinal books. The roots and stems of D. morbifera had been used for traditional medicine to treat migraine, menstrual irregularity and skin disease. And D. morbifera leaves are contain flavonoids and polyacetylene compounds. In this study, we were investigated the physiological activity of D. morbifera by different areas collected at the same time, and compared to characteristics of plants. D. morbifera collected from Jeollanam-do (Goheung-gun), Gyeongsangnam-do (Namhae-gun) and Jeju Island, and dried at $50^{\circ}C$ for three days. We used dried D. morbifera powder for antioxidant tests. Each sample was extracted with hot water under the same conditions. The contents of total polyphenols and total flavonoids from D. morbifera were identified. Also, we performed to DPPH radical scavenging activity, ABTS cation radical scavenging activity and Superoxide anion scavenging activity efficacy for antioxidant activity determination. The contents of total polyphenols and total flavonoids in hot water extract of D. morbifera harvested from Gyeongsangnam-do and Jeollanam-do were higher than Jeju. However, D. morbifera harvested from Gyeongsangnam-do and Jeollanam-do showed no significant difference those content of total polyphenols and total flavonoids. And the antioxidant capacity was showed the similar patterns in antioxidant activity.

Effects of polyphenols of Cocos nucifera husk fibreon selected indices of cardiovascular diseases in mice

  • Cocos nucifera (C. nucifera) oil is indigenously used to treat cardiovascular diseases. However, coconut husk fibre (which is rich in polyphenols) has not been screened for this property. Based on the ethnomedicinal use of polyphenols in treating cardiovascular diseases, this study was carried out to evaluate the effects of polyphenols of C. nucifera husk fibre on selected cardiovascular disease indices in mice. Fifty adult male Swiss albino mice were assigned randomly into five groups (A-E). Mice in groups B, C, D and E were administered 31.25, 62.5, 125, and 250 mg/kg body weight polyphenols of ethyl acetate extract of C. nucifera husk fibre respectively while the control group (A) mice received 5% DMSO for seven days. The mice were sacrificed twenty four hours after the last administration of polyphenols. Heart and plasma lactate dehydrogenase (LDH), alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) activities and plasma lipid profile were determined. Results revealed significant reduction (*p< 0.05) in plasma levels of total cholesterol and LDL-cholesterol with no significant change (*p> 0.05) in HDL-cholesterol, triglyceride and VLDL levels in the plasma at all doses of polyphenols administered compared to controls. There was significant reduction (*p< 0.05) in the activities of heart AST and LDH while plasma ALT, AST, and ALP activities were not significantly altered (*p> 0.05) at all doses of polyphenols administered compared to controls. These results suggest that the polyphenols of C. nucifera husk fibre possess cardio-protective properties and also indicate their possible use in the treatment of cardiovascular diseases.
