Comparative analysis of active components and antioxidant activities of Brussels sprouts (Brassica oleracea var. gemmifera) and cabbage (Brassica oleracea var. capitata)

  • The applicability of Brussels sprouts, which are widely cultivated in Jeju, as a functional biomaterial in the cosmetics and food industries is investigated. The active ingredients (total phenols and flavonoids, β-carotene, vitamin C, free sugar, minerals, glucosinolates, and isothiocyanates) and antioxidant activities of 70% ethanol extracts of Brussels sprouts and cabbage were analyzed. The total phenol, flavonoid, vitamin C, and β-carotene contents of Brussels sprouts were approximately 36%, 2.5 times, 2.3 times, and 65% higher than those of cabbage, respectively. The total free sugar content of Brussels sprouts was 58%-72% lower than that of cabbage. The K content was the highest among the other minerals, and there was little difference between the two samples. The Na content was approximately three times higher in cabbage than in Brussels sprouts. The total glucosinolate content of Brussels sprouts was 34.5 mol SE/g DW, twice that of cabbage. The sinigrin content of Brussels sprouts was thrice (10.06 μmol/g DW) that of cabbage. The isothiocyanate (sulforaphane, I3C) content in Brussels sprouts was double that of cabbage. The antioxidant activity of Brussels sprout extract was 2.5 to 2.8 times higher in 1,1-Diphenyl-2-picrylhydrazyl radical scavenging capacity and 3.3 to 3.6 times higher in 2,2'-Azino-bis(3-ethybenzothiazoline-6-sulfonic acid) radical scavenging capacity than those of cabbage extract. These are important basic data for the study of glucosinolates and isothiocyanates, which have anticancer activity, as well as antioxidant-related substances of Brussels sprout, which has high potential for use as a biomaterial in functional foods and cosmetics.

Impact of Transplanting on Tree Growth and Compartmentalization of Pruning Wounds in Acer palmatum Thunb

  • The objective of this study was to examine the impact of pruning (P treatment) and transplanting (T treatment) of Acer palmatum on cambial growth and compartmentalization of pruning wounds for one year after treatments. Changes of cambial electrical resistance (CER), sizes of pruning wounds, cambial growth of trunks and stems near the wounds, and total phenols at branch unions during the period were examined using a total of 49 trees. After harvesting, areas of discolored wood behind the wounds, relative proportions of extractives, holocellulose and lignin at branch unions were also determined. CER and the cambial growth of trunk at 30 cm above the ground (TGR) were inversely correlated, and differences of CER and TGR among three treatments were significant. TGRs of control, P treatment and P+T treatment after the treatments were 112.2%, 72.4% and 52.5% of the annual growth for the year before the treatments, respectively. The cambial growth rate of stem (SGR) at 1.5 cm above the branch bark ridge and the closure rate of pruning wound (WCR) for one year after treatments were positively correlated, and WCR of P treatment of 39.8% was significantly higher than that of P+T treatment of 31.8%. Wounds of P+T treatment formed greater discolored area per unit area of pruning wound (D/W Ratio) than those of P treatment significantly. Lower WCR and higher D/W Ratio of P+T treatment suggested less ability of compartmentalizing the wounds than P treatment. Total phenols at branch core of pruning wound for both treatments heightened a month after treatment, and then lowered. The contents at below core of the wound were higher than those at control ones continuously, while they became similar each other at above core. Relatively high phenol contents of the extractives at P+T treatment implied that trees with P+T treatment allocated more energy to compartmentalize their wounds. Holocellulose and lignin contents at the branch core of treated branch unions of both treatments were lower and higher, respectively, than at the same part of the union with living branch, as results of the tree reaction to protection from wounding and microbial invasion.

Assessment of Phytochemicals, Quality Attributes, and Antioxidant Activities in Commercial Tomato Cultivars

  • To assess South Korean commercial tomato cultivars, regular and cherry tomato cultivars were grown in the greenhouse and evaluated for color attributes, titratable acidity, pH, total soluble solids, carotenoids (lycopene and ${\beta}$-carotene), total phenols, flavonoids, vitamin C, and antioxidant activity. Significant differences (p < 0.05 using Duncan's multiple range test (DMRT)) were observed in the levels of most phytochemicals, quality parameters, and antioxidant activity among the twenty South Korean tomato cultivars tested. Lycopene and ${\beta}$-carotene contents varied significantly (p < 0.05 using DMRT), from $0.95mg{\cdot}100g^{-1}$ to $5.12mg{\cdot}100g^{-1}$ and $0.65mg{\cdot}100g^{-1}$ to $3.56mg{\cdot}100g^{-1}$ of fresh weight, respectively. ${\beta}$-carotene contents exhibited the highest genetic variation (59.2%), followed by naringenin (52.8%) and other phytochemicals. Most of the cherry tomato cultivars had statistically higher levels (p < 0.05 using DMRT) of carotenoids, phenols, flavonoids, vitamin C, and antioxidant activity compared to the regular tomato varieties, suggesting their higher nutritional value. Lycopene content was highest in the cultivars YoYo, Jicored, Titi-Chal, TY-Endorphin, and Rubyking. Cultivars Rubyking, TY-Endorphin, and Titi-Chal also showed relatively higher antioxidant activities in three assays: 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and ferric ion reducing antioxidant power (FRAP) assays. All the antioxidants, except luteolin, were positively correlated with antioxidant activities; the highest correlation was observed between total phenol and antioxidant activities, followed by the correlation between rutin and vitamin C. Cultivars identified to have superior nutritional status would be useful in tomato breeding programs to further improve quality and health benefits of tomatoes for the fresh and processed markets.

The Effects of Bioactive Compounds and Fatty Acid Compositions on the Oxidative Stability of Extra Virgin Olive Oil Varieties

  • The aim of this study was to determine the various bioactive components of five olive oil varieties, as well as to assess their contribution to the oxidative stability of the oils. Fatty acids, ${\alpha}$-tocopherol, ${\beta}$-carotene, total flavonoids, total phenols, and certain phenolic compounds of extra virgin olive oils (EVOO; blended, arbequina, hojiblanca, and picual) and pure olive oil (POO) were examined. Oxidation stability was evaluated by the peroxide value (POV). The total content of all the studied antioxidant compounds was significantly higher in the EVOOs than the POO (p<0.05). Among the EVOOs, picual had the highest levels of ${\alpha}$-tocopherol ($10.18{\pm}0.40\;mg/100\;g$), ${\beta}$-carotene ($557{\pm}8\;{\mu}g/100\;g$), and total phenols ($110.7{\pm}1.3\;mg/g$), which correlated strongly with antioxidative capacity. Furthermore, the lowest POV occurred in picual EVOO and correlated with the highest monounsaturated fatty acid (MUFA, C16:1 and C18:1) and lowest polyunsaturated fatty acid (PUFA, C18:2 and C18:3) compositions, suggesting the ratio of MUFA to PUFA is a critical parameter for the oxidative stability of olive oil. Our results indicate that the oxidative stability and antioxidant potential of EVOO depends not only on the antioxidant vitamins, but also on the amount of phenolic compounds and fatty acid profile of the oil.

Homobrassinolide and chemical composition of curcuma longa L. (turmeric) rhizome

  • The effect of homobrassinolide (HBL) on the nutrient value of Curcuma longa L. (turmeric) rhizome grown in Nizamabad, Telangana State, India was studied. Application of homobrassinolide (HBL) as foliar spray to turmeric plants on the 20th, 40th and 60th day from sowing resulted in enhanced chemical composition of turmeric rhizome. Application of homobrassinolide (HBL) resulted in enhanced total sugars, principal ingredient curcumin, total poly phenol content, total flavonoid content, total tannin content, crude fat, crude fibre and essential oils (turmerone, zingiberene, cineole and p-cymene) present in the turmeric rhizome indicating the ability of homobrassinolide (HBL) as a potential plant growth regulator (PGR).

Effect of horseradish, mushroom waste and almond hull on the concentrations of odorous compounds in swine manure for spreading on grassland in spring

  • The objective of the current study was to investigate the effect of odor reducing agents on the levels of pH, total carbon, total nitrogen, and odorous compounds [phenols, indoles, short chain fatty acid (SCFA), branched-chain fatty acid (BCFA), and ammonium nitrogen] of swine manure during the spring season (temperature around $20^{\circ}C$). Odor reducing agents included horseradish powder, mushroom waste powder, and almond hull powder. A manure sample (15 L) was taken from the pit under the pens of a swine feeding operation and incubated with 0.03% horseradish powder, 1% mushroom waste powder, and 1% almond hull powder, respectively, in acryl chambers for 14 days. Addition of almond hull powder showed the lowest pH (p < 0.05) and the highest level of total carbon (p < 0.05) among treatments of odor reducing agents. Although addition of odor reducing agents increased the level of phenols (p < 0.05), addition of almond hull powder decreased the level of indoles (p < 0.05). Levels of SCFA and BCFA were higher in almond hull powder than those in control (p < 0.05). Taken together, the results from our current study showed that odor reducing agents can be used for reducing the odor of swine manure by providing fermentable carbohydrates. At $20^{\circ}C$, however, the function of odor reducing agents might be decreased due to lower microbial activity.

Pathogenicity of Bacillus Strains to Cotton Seedlings and Their Effects on Some Biochemical Components of the Infected Seedlings

  • Pathogenicity of eight Bacillus strains to seedlings of four cotton cultivars was evaluated under greenhouse conditions. Each of the tested cultivars was individually treated with powdered inoculum of each bacterial strain. Untreated seeds were planted as control treatments in autoclaved soil. Effects of the tested strains on levels and activities of some biochemical components of the infected seedlings were also assayed. The biochemical components included total soluble sugars, total soluble proteins, total free amino acids, peroxidase, polyphenol oxidase, phenols, and lipid peroxidation. ANOVA showed that Bacillus strain (B) was a very highly significant source of variation in damping-off and dry weight. Cotton cultivar (V) was a nonsignificant source of variation in damping-off while it was a significant source of variation in dry weight. B × V interaction was a significant source of variation in damping-off and a nonsignificant source of variation in dry weight. Bacillus strain was the most important source of variation as it accounted for 59.36 and 64.99% of the explained (model) variation in damping-off and dry weight, respectively. The lack of significant correlation between levels and activities of the assayed biochemical components and incidence of damping-off clearly demonstrated that these biochemical components were not involved in the pathogenicity of the tested strains. Therefore, it was hypothesized that the pathogenicity of the tested strains could be due to the effect of cell wall degrading enzymes of pathogenic toxins. Based on the results of the present study, Bacillus strains should be considered in studying the etiology of cotton seedling damping-off.

반응표면분석법에 의한 노루궁뎅이 버섯 에탄올 추출조건의 최적화 (Optimization of Extraction Conditions from Hericium erinaceus by Response Surface Methodology)

  • 노루궁뎅이 버섯의 기능성 식품소재 개발과 가능성을 검토하기 위하여, 반응표면분석에 의해 추출조건의 최적화 및 추출특성을 모니터링하였다. 중심합성계획에 따라 에탄올 농도$(X_1)$, 시료의 용매비$(X_2)$를 요인변수로 하고 추출물의 특성 즉, 가용성 고형분$(Y_1)$, 총페놀 함량$(Y_2)$, 조단백$(Y_3)$, 전자공여능$(Y_4)$, 갈색도$(Y_5)$를 종속변수로 하여 추출을 실시하였다. 추출물의 가용성 고형분은 에탄올 농도에 의해 거의 영향을 받지 않는 것으로 나타났으며 시료의 용매비가 증가할수록 증가하는 경향을 나타내었다. 에탄올 농도가 낮을수록, 시료의 용매비가 높을수록 페놀성 성분의 함량이 증가하는 반응표면을 나타내었으며, 조단백 함량의 경우 에탄올 농도가 감소할수록 증가하였다가 일정시점 이후로 약간의 감소하는 경향을 나타내었으며, 시료의 용매비가 증가할수록 증가하는 경향을 나타내었다. 추출물의 전자공여작용을 제외하고는 에탄올 농도보다는 시료의 용매비에 더 큰 영향을 미치는 것으로 나타났다. 이들 추출물의 특성을 모두 만족시키는 최적 추출조건은 에탄올 농도 $36{\sim}52%$. 시료의 용매비 $3.9{\sim}5.0\;g/100\;mL$로 나타났으며 예측된 최적 추출조건의 임의의 점에서 실험한 결과, 각 반응변수들의 예측값과 실제값이 유사하였다.

복숭아 품종별 성숙정도에 따른 항산화 및 항염증 효과간의 상관관계 (In vitro correlation and analysis of anti-oxidant and anti-inflammatory activities by fruit ripening of peach cultivars)

  • 본 연구에서는 복숭아의 품종인 미황, 가납암백도 및 천홍을 성숙정도에 따라 미숙과와 성숙과로 분류하여 항산화 및 항염증 효과간의 상관관계 등을 분석함으로서 향후 염증 및 산화스트레스와 관련된 질병의 예방 또는 치료할 수 있는 기능성 소재를 발굴하고자 수행하였다. 총 페놀 및 플라보노이드 함량은 3품종 모두 성숙과에 비해 미숙과에서 함량이 많았고, 미숙과 중에서는 천홍이, 성숙과 중에서는 미황이 유의성 있게 높은 함량을 나타내었다. 항산화 활성은 미숙과 천홍에서 가장 높았고, 성숙과에서는 DPPH 라디칼 소거능 측정에 의해서는 미황이 유의성 있게 높았으나 FRAP과 CUPRAC 측정에서는 3품종간 유의적인 차이가 없었다. RAW 264.7 세포에서는 미황 미숙과 추출물이 약 30%의 NO 생성 억제 효과가 관찰되었고, 가납암백도 미숙과는 약 29%, 천홍 미숙과는 약 20%의 저해활성을 보이지만, 복숭아 성숙과 3품종에서는 NO 생성 억제 효과가 나타나지 않았다. 3품종 모두 성숙과에 비해 미숙과에서 저해 활성이 높게 나타났다. 총 페놀 함량과 플라보노이드 함량에 따른 항산화 활성들 간의 상관관계에서 고도의 정의 상관성을 나타내었으며, 항염증 활성과 항산화 활성, 항염증 활성과 총 페놀 및 플라보노이드 함량은 낮은 상관관계를 나타내었다.

태종대산 5종 돌미역의 화학성분 및 항산화활성 비교 (A Comparative Study on the Chemical Characteristics and Antioxidant Effects of Sea Mustards Sourced from Different Areas in Taejongdae)

  • 태종대 구역별 바람맥이, 굴통머리, 참물개, 조홍택, 고래등 구역에서 채취한 자연산 돌미역의 총 플라보노이드 함량을 비교한 결과 굴통머리 구역에서 채취한 돌미역의A+M 및 MeOH 추출물들의 총 플라보노이드 함량이 각각 1.44±0.04 mg/g 및 0.66±0.13 mg/g으로 5종 돌미역들 중 총 플라보노이드 함량이 가장 높았다. 지방산 조성분석 결과 굴통머리 돌미역은 30.1%의 n-6 지방산 및 10.0%의 총 n-3 지방산을 함유하였고 5종 돌미역 중 가장 높은 다가불포화지방산(polyunsaturated fatty acids, PUFA)을 함유하였다. 5종 돌미역 추출물들의 DPPH 및 ABTS 라디칼 소거능을 비교했을 때 모든 돌미역 A+M 추출물에 의한 소거활성이 MeOH 추출물보다 높았고 그 중 굴통머리A+M 추출물에 의한 소거능 활성이 가장 높았다(p<0.05). 5종 돌미역A+M 및 MeOH 추출물을 0.05 및 0.025 mg/ml의 농도로 인체 섬유육종세포(HT-1080)에 처리하여 세포 내 활성산소종을 측정한 결과 모든 추출물들은 측정시간 120분이 지남에 따라 높은 세포 내 활성산소종 생성 억제효과를 나타내었다. 바람맥이 및 굴통머리 구역에서 채취한 돌미역A+M 추출물에 의한 활성산소종 생성 억제효과가 높았다(p<0.05). 이상의 결과로부터 굴통머리 구역의 돌미역이 높은 함량의 플라보노이드 및 페놀 함량을 나타내었고 이러한 높은 함량은 세포내 활성산소종 생성 억제효과 및 DPPH 소거능과 상관성이 있다고 여겨지며 또한 굴통머리 구역 돌미역은 높은 함량의 EPA와 DHA를 함유하고 있어 향후 해조류유래 n-3 지방산 활용 기능성 식품 혹은 지역 대표 수산식품으로 개발 가능할 것으로 기대된다.
