Studies on the Growth of Bidens L. Along the Environmental Gradient (환경구배에 따른 Bidens L.속 식물의 성장에 관한 연구)

  • The growth and niche breath of four species, i.e., native plants such as Bidens tripauita and B. bipinnata and naturalized plants such as B. frondosa and B. pilosa var. minor, were studied as related with environmental gradients including light intensity, soil moisture and soil nutrient. There were no significant differences in the phonology within each environmental gradients while considerable differences were found between species; the two naturalized species bloomed and produced fruit later than the two native species. Two naturalized species exhibited relatively higher total dry weight than the two native counterparts within all environmental gradients. Total dry weight showed positive responses to light intensity and soil moisture, and negatively to soil nutrient. The relative contribution of the environmental factors to total dry weight decreased in the order of soil nutrient, soil moisture and light intensity. Both B. bipinnata and B. pilosa var. minor showed significantly higher net assimilation rate (NAR) than other species. Also, NARs of B. bipinnata and B. pilosa var. minor decreased with increasing soil nutrient. Relative growth rate (RGR) decreased in order of B. tripartita, B. bipinnata, B. frondosa and B. pilosa var. minor in response to light intensity. In addition, RGRs of B. bipinnata and B. piEosa yay. minor increased in response to soil moisture, while those of B. pilosa var. minor and B. tripartita decreased with increment of soil nutrient. No significant fluctuations of shoot/root ratio were not observed in three species, but a native species, B. tripartita showed n decreased shoot/root ratio in response to soil nutrient. Comparing the growth characteristics of the species, B. pitosa var. minor and 3. tripartita revealed vigorous growth on barren soil. On the other hand, B. frondosa exhibited vigorous growth on fertile soil. Morphologically, B. tripartita adapted to light and required considerable moisture. On the contrary, the leaves of B. bipinnata did not change considerably in their area, although it preferred habitat with abundant light. In terms of niche breadth, B. bipinnata showed the widest ranges of 0.875 and 0.845 for light intensity and soil moisture gradients, respectively. B. pilosa var. minor showed a value of 0.933 for soil nutrient gradient. B. tripartita showed narrow ranges for the three environmental factors, whereas B. frondosa showed wide values for light intensity and soil nutrient, but relatively narrow value for soil moisture.

Changes of Components in Salt-fermented Northern Sand Lance, Ammodytes personatus Sauce during Fermentation (숙성기간에 따른 까나리액젓의 성분변화)

  • To investigate changes of components in salt-fermented northern sand lance, Ammodytes personatus sauce during fermentation, various chemical properties were examined at 1$\~$3 months intervals during 18 months fermentation. The moisture content decreased slightly, but the content of VBN and crude protein, total nitrogen, amino nitrogen, degree of hydrolysis, and absorbance at 453 nm increased gradually during fermentation. On the other hand, ash content, pH, and salinity showed almost no change. The contents of total nitrogen, amino nitrogen, and degree of hydrolysis increased sharply until 6$\~$8 months fermentation and showed the gentle increment after that, The Hx and uric acid were the most abundant in ATP related compounds, ranging from $83.1\%$ to $92.9\%$, After 18 month of fermentation, sauce was rich in free amino acids, such as glutamic acid, alanine, Iysine, leucine, isoleucine, valine, aspartic acid in that order.

Effects of Dietary Supplementation of Betaine on Performance, Lipid Metabolic Parameters, and Blood and Ileal Osmolality in Laying Hens (비태인의 급여가 산란계의 생산성과 지질대사 관련인자, 소화물의 삼퉁성에 미치는 영향)

  • Two experiments were conducted to investigate the effect of betaine on egg production, lipid metabolism, and osmoregulation in 18-to 42-week-old ISA Brown laying hens. In experiment 1, three hundred and sixty one hens were fed a com-soy basal diet contailing 16% crude protein (CP), 2800 kcal/kg metabolizable energy (ME), 0.33% methionine, and 0, 300, 600, or 1200 mg betaine per kg diet. Egg production, egg weight, feed consumption, feed conversion, and egg quality were measured every eight weeks. Betaine concentration in live and egg were determined along with serum cholesterol, abdominal fat, total serum protein and albumin levels. In experiment 2, twenty thirty-three-week-old laying hens were fed the same diets as those used in experiment 1 in individual cages and the amount of feed and water consumption were measured for two weeks. At the end of experiment 2, all birds were killed to determine blood plasma and ileal osmopressure, arginine vasotocin (AVT), and liver moisture content. In experiment 1, egg production between the treatments during the first eight weeks were not different, whereas the significant increment of egg production were noticed in the birds fed more than 600 ppm betaine after reaching the peak egg production stage (p<0.05). The egg weight was reduced significantly by the betaine supplementation for the first 8 weeks (p<0.05). Feed conversion tended to improve by betaine supplement. Egg quality was not enhanced by betaine supplementation. Liver betaine level increased with betaine feeding compared to the control but betaine concentration in eggs decreased with betaine supplementation. Betaine supplementation elevated the level of serum total cholesterol and triglyeerides compared to the control. Abdominal fat content was increased by betaine supplementation, whereas liver fat content decreased. In experiment 2, water consumption significantly increased in hens fed diets containing 300 and 600 mg betaine/kg (p<0.05) and osmotic pressure of ileal digesta increased with betaine supplement. Liver moisture content was not affected by betaine, but AVT increased in hens fed betaine. The overal results suggested the possibility of using betaine as a feed additives in the laying hens beacuse of its positive contribution to improving egg production and other metabolic parameters related to lipid metabolism.

Mass Culture and Ginsenoside Production of Ginseng Hairy Root by Two-Step Culture Process (2계단 배양방법을 이용한 인삼 모상근의 대량배양과 Ginsenoside 생산)

  • A hairy root clone of Panax ginseng C.A. Meyer, HRB-15 was cultured iu various conditions with 3 L bubble type bioreactor to enhance both growth and ginsenoside production. The hairy roots were more rapidly grown under the dark condition than under the light condition. However, total amount of ginsenoside of hairy roots cultured under the light for 30 days increased 2 folds as compared with the dark condition and was 1.10% based on 6 ginsenosides. Especially, ginsenoside-Re was significantly increased and some ginsenosides except for ginsenoside-Re was slightly reduced. Also, the growth of hairy roots decreased about 30% as compared with the dark condition. In contrast, addition of sodium acetate led to decreased production of ginsenoside and growth of hairy roots under light condition. The influence of potassium dihydrogenphosphate concentration was examined in MS medium and a 1.25 mM concentration was found to be the most appropriate for growth and ginsenoside production under light condition. Two-step process of hairy roots culture with yeast elicitation or without ammonia in culture medium was developed to enhance growth and giusenoside synthesis. $50\;\mu\textrm{g}$ of yeast elicitor per g of fresh weight showed a synergistic effect on the ginsenoside synthesis of hairy roots on 20 days after culture. At that time, the content of total ginsenoside was 1.15%, while the growth of hairy roots decreased 21 % as compared with the dark condition. In addition, when elimination of ammonia on 20 days after culture, the content of total ginsenoside was 1.26% with significant increment of ginsenoside-Rd (0.27%) in addition to ginsenoside-Re and the growth of hairy roots decreased 10% as compared with the dark condition. In this system, we have demonstrated a unique two-step process of hairy root cultures to maximize biomass and secondary metabolites. It has found possibility to enhance ginsenosides production by growing hairy roots in this method.

Changes of Haematological Constituents in the Nile tilapia, Oreochromis niloticus Exposed to HCB and PCBs (HCB와 PCBs에 노출된 나일틸라피아, Oreochromis niloticus의 혈액성분의 변화)

  • The study was carried out to investigate the changes of haematological constituents induced by dietary exposure of Hexachlorobenzene (HCB) and Polychlorinated biphenyls (PCBs) in nile tilapia, Oreochhromis niloticus. The nile tilapia were fed pellet with 0.05, 0.25 and 0.50 mg/kg body weight/day of HCB and PCBs for 30 days, respectively. HCB ($\ge$ 0.25 mg/kg) induced a significantly decreased the red blood cell count at 30 days, and the hemoglobin concentration and hematocrit value were significantly decreased after 20 days to 0.50 mg/kg. The diminution of red blood cell count, hemoglobin concentration and hamatocrit value by PCBs was found after 20 days at $\ge$0.25 mg/kg. The concentrations of calcium decreased significantly at 30 days at $\ge$0,25 mg/kg of PCBs-diet. However, magnesium and inorganic phosphorus did not show any noticeable changes in the range of 0.05$\~$o.50 mg/kg of HCB and PCBs compared to control group during the experimental period, The low concentration of total protein was observed at the last day of experiment to 0.50 mg/kg of HCB-diet, while total protein initiated to increase from 20 days at the 0.25 mg/kg of PCBs-diet. The glucose was significantly increased after 20 days at $\ge$0,25 mg/kg of HCB and PCBs, A significant increment GOT and GPT activities was noticed after 20 days at 0.50 mg/kg of HCB and PCBs, These results indicate that hematological and serum biochemical changes in the nile tilapia by dietary exposure to HCB and PCBs are affected at more than 0,25 mg/kg after 20 days.

Effect of Ethanol Addition on Efficacy of Dental Adhesive (에탄올의 첨가가 치과용 접착제의 효율에 미치는 영향)

  • The purpose of this study was to evaluate the effect of ethanol addition on efficacy of two-step total-etch adhesive under over-wet condition by measurement of remaining volatile part (RVP), microtensile bond strength (${\mu}TBS$), and degree of conversion (DC). Two-step total-etch adhesive, Optibond Solo Plus (Kerr, Orange, USA), was used. Experimental groups were divided into 8 groups: Group 1 (only 10 ${\mu}l$ adhesive), Group 2 (mixture of 3 ${\mu}l$ distilled water and 10 ${\mu}l$ adhesive), From Group 3 to Group 8 (mixture of 3 ${\mu}l$ distilled water, 10 ${\mu}l$ adhesive, and ethanol added in 1 ${\mu}l$ increment from 1 ${\mu}l$ to 6 ${\mu}l$). The mixtures were placed on slide glass and evaporated for 10 s, 30 s, and 60 s by air-drying. The weight of RVP was measured by precision weight. Same procedures were performed for ${\mu}TBS$ test and measurement of DC. The condition of mixed solution was observed under light microscope. For RVP weight, the weights of experimental groups except for group 1 decreased with the increase of air-drying time (p<0.05). The DC increased with the increase of air-drying time in only group 5 and 6 (p<0.05). The ${\mu}TBS$ increased with the increase of air-drying time in group only 5, 6, and 7 (p<0.05). The phase separation was examined and water blisters were diminished with the increase of air-drying time in group 5, 6, 7, and 8. Within the limits of this study, ethanol additionally applied to adhesive decreased RVP and increased DC and ${\mu}TBS$ under over-wet condition. It was shown that the addition of ethanol to two-step total-etch adhesive under over-wet condition would remove water and increase the efficacy of adhesive.

An Appreciation of Functional Role of Macrophage in the Acute Lung Injury in the Neutropenic Rat. (호중구 감소증을 보이는 백서의 급성폐손상에서 대식세포의 기능적 역할)

  • Background : It has long been suggested that neutrophils and their products are implicated as the central mediators of the acute lung injuries. Contrary to the dominant role of neutrophils in ARDS, many cases of ARDS has occurred in the setting of severe neutropenia without pulmonary neutrophil infiltration. Therefore it is certain that effector cell(s) other than neutrophil play an important role in the pathogenesis of ARDS. This experiment was performed to define the mechanism of ARDS in the setting of neutropenia, 1) by comparing the severity of endotoxin-induced lung injury, 2) by measurement of hydrogen peroxide production and cytokine concentration in the bronchoalveolar lavage cells and fluids obtained from different rats with and without cyclophosphamide-pretreatment. Method : The male Sprague-Dawleys were divided into the normal control (NC)-, endotoxin (ETX)-, and cyclophosphamide (CPA)-group in which neutropenia was induced by injecting cyclophosphamide intraperitoneally. Acute lung injury was evoked by injecting lipopolysaccharide (LPS) into a tail vein. The bronchoalveolar lavage (BAL) was performed at 3 and 6 hour after administration of LPS to measure the change of cell counts and concentrations of protein and cytokines, tumor necrosis factor-alpha (TNF-alpha) and interleukin-6 (IL-6). Hydrogen peroxide (HPO) production from BAL cells was measured at 6 hour after LPS administration by phenol red microassay with and without zymosan stimulation. Results : The results were as follows. A change of leukocyte counts in the peripheral blood after treatment with CPA : More than 95% of total leukocytes and neutrophils were reduced after CPA administration, resulting in severe neutropenia. A change of BAL cells : In the ETX-group, the number of total cells (p < 0.01) and of macrophage and neutrophil (p < 0.05) were increased at 3 and 6 hour after LPS administration compared to those of NC-group. In the CPA-group, the number of total leukocyte and macrophage were not changed after LPS administration, but neutrophil counts were significantly reduced and it took part in less than 0.1% of total BAL cells (p < 0.01 vs NC-group). BAL cells in this group were almost all macrophages (99.7%). A change of protein concentration in the BALF : In the ETX-group, protein concentration was increased at 3 hour and was more increased at 6 hour after LPS administration (p < 0.05 and < 0.01 vs NC-group, respectively). In the CPA-group, it was also significantly elevated at 3 hour after LPS administration (p < 0.05 vs NC-group), but the value was statistically not different from that of ETX-group. The value measured at 6 hour after LPS administration in the CPA-group became lower than that of ETX-group (p < 0.05), but showed still a higher value compared to that of NC-group (p < 0.05). A change of cytokine concentration in the BALF : TNF -alpha and IL-6 were elevated in the ETX - and CPA-group compared to those of NC-group at both time intervals. There was no statistical difference in the values of both cytokines between the ETX- and CPA-groups. Measurement of hydrogen peroxide production from BAL cells : There was no intergroup difference of HPO production from resting cells. HPO production after incubation with opsonized zymosan was significantly elevated in all groups. The percent increment of HPO production was highest in the ETX-group (89.0%, p < 0.0008 vs NC-group), and was 42.85 in the CPA-group (p = 0.003 vs NC-group ). Conclusion : Acute lung injury in the setting of neutropenia might be caused by functional activation of resident alveolar macrophages.

Effects of Soil Hardness on the Root Distribution of Pinus rigida Mill. Planted in Association with Sodding Works on the Denuded Land (사방시공지(砂防施工地)에 있어서 리기다소나무의 수근(樹根)의 분포(分布)에 미치는 토양견밀도(土壤堅密度)의 영향(影響))

  • Soil harness represents such physical properties as porosity, amount of water, bulk density and soil texture. It is very important to know the mechanical properties of soil as well as the chemical in order to research the fundamental phenomena in the growth and the distribution of tree roots. The writer intended to grip soil hardness by soil layer and also to grasp the root distribution and the correlation between soil hardness and the root distribution of Pinus riguda Mill. planted on the denuded hillside with sooding works by soil layer on soil profile. The site investigated is situated at Peongchang-ri 13, Kocksung county, Chon-nam Province. The area is consisted of 3.63 ha having on elevation of 167.5-207.5 m. Soil texture is sandy loam and parant rock in granite. Average slope of the area is $17^{\circ}-30^{\circ}$. Soil moisture condition is dry. Main exposure of the area is NW or SW. The total number of plots investigated was 24 plots. It divided into two groups by direction each 12 plots in NW and SW and divided into three groups by the position of mountain plots in foot of mountain, in hillside, and in summit of mountain, respectively. Each sampling tree was selected as specimen by purposive sampling and soil profile was made at the downward distance of 50cm form the sampling tree at each plot. Soil hardness, soil layer surveying, root distribution of the tree and vegetation were measured and investigated at the each plot. The soil hardness measured by the Yamanaka Soil Hardness Tester in mm unit. the results are as follows: 1) Soil hardness increases gradually in conformity with the increment of soil depth. The average soil indicator hardness by soil layer are as follows: 14.6mm in I - soil layer (0-10cm in depth from soil surface), 16.2mm in II - soil layer (10-20cm), 17.2 in III - soil layer (20-30cm), 18.3mm in IV - soil layer(30-40cm), 19.8mm in V - soil layer (4.50mm). 2) The tree roots (less than 20mm in diameter) distribute more in the surface layer than in the subsoil layer and decrease gradually according to the increment of soil depth. The ratio of the root distribution can be illustrated by comparing with each of five soil layers from surface to subsoil layer as follows: I - soil layer; 31%, II - soil layer; 26%, III - soil layer; 18%, IV - soil layer; 12%, V - soil layer; 13%, 3) Soil hardness and tree root distribution (less than 20mm in diameter) of Pinus rigida Mill. correlate negatively each other; the more soil hardness increases, the most root distribution decreases. The correlation coefficients between soil hardness and distribution of tree roots by soil layer are as follows: I - soil layer; -0.3675 (at the 10% significance level), II - soil layer; -0.5299 (at the 1% significance level), III - soil layer; -0.5573 (at the 2% significance level), IV - soil layer; -0.6922 (at the 5% significance level), V - soil layer; -0.7325 (at the 2% significance level). 4) the most suitable range of soil hardness for the growth of Pinus rigida Mill is the range of 12-14.9mm in soil indicator hardness. In this range of soil indicator hardness, the root distribution of this tree amounts to 41.8% in spite of 33% in soil harness and under the 20.9mm of soil indicator hardness, the distribution amounts to 93.2% in spite of 82% in soil hardness. Judging from above facts, the roots of Pinus rigida can easily grow within the soil condition of 20.9mm in soil indicator hardness. 5) The soil layers are classified by their depths from the surface soil.

  • Eeffect of temperature (3-day or 7-day treatment under $15^{\circ}C$, $20^{\circ}C$ and $25^{\circ}C$) on chlorophyll, nitrogen and potassium content in the second leaf blade from top of IR667 line (Suwon 213 and 214) and leading local varieties (Jinheung and Paldal) was investigated at the end of nursery, maximum tillering and flowering stage using phytotron. 1. Chlorophyll content was higher in IR667 line than in the local at $25^{\circ}C$ but reversed at $20^{\circ}C$ or $15^{\circ}C$ and chlorophyll a/b value was always high in the local. 2. Chlorophyll content per fresh weight decreased with growth and low temperature effect was greater at the end of nursery and on chlorophyll a than on chlorophyll b. 3. Chlorophyll a/b value increases with the increase of chlorophyll content and the increasing rate of chl. a/b value per chlorophyll increment tends to decrease under unfavorable condition. This decrease is greater in low temperature sensitive IR667 than local varieties. 4. According to chlorophyll retention value IR667 line was weaker at low temperature. 5. The content of total nitrogen or soluble nitrogen (methanol soluble) was decreased by low temperature. 6. Chl. (a+b)/S-N value decreased with growth and seemed not to be greatly affected by temperature and always higher in the local. 7. Potassium content (total or methanol soluble) tends to decrease at low temperature and soluble K increased with chlorophyll content. 8. High yielding ability of IR667 seems to be attributable to its high chlorophyll content at high temperature and easy leaf discoloration by low temperature or by nitrogen depression of IR667 seems to be attributable to the low Chl./S-N value.

Relationship between Clozapine-Induced Therapeutic Responses and Plasma Homovanillic Acid and 5-Hydroxyindoleacetic Acid Levels in Patients with Chronic Schizophrenia (만성 정신분열증 환자에서 Clozapine의 치료반응과 혈장 Homovanillic Acid 및 5-Hydroxyindoleacetic Acid 농도와의 관계)

  • This study was done to examine changes of plasma homovanillic acid(HVA), 5-hydroxyindoleacetic acid(5-HIAA), and HVA/5-HIAA ratio during an 8-week clozapine trial and to investigate the relationship between the plasma monoamine metabolites and treatment responses. Twenty-seven chronic schizophrenic patiens were treated for 8 weeks with clozapine. The psychopathology was assessed at baseline just clozapine trial and then every 2 weeks until the end of 8-week clozapine treatment using the Positive and Negative Syndrome Scale(PANSS) and the Clinical Global Impression scale(CGI). The plasma HVA and 5-HIAA levels were measured also biweekly using high preformance liquid chromatography with electrochemical detection method. Plasma HVA and 5-HIAA levels were significantly decreased during a 8-week clozapine treatment, although plasma HVA/5-HIAA ratio showed no significant change. The changes of plasma HVA levels were in significant correlations with the changes of PANSS positive scores, of general psychophathology scores, and changes of total socres. The changes of plasma 5-HIAA levels were in signfificant correlations with the changes of PANSS negative scores. But the changes of plasma HVA/5-HIAA ratio had no significant correlation with any PANSS subscale score changes. 48% of the patients treated with clozapine was categorized as responders, who showed at least a 20% decrease in PANSS total socre and a CGI severity score of mildly ill or less(${\leq}3$) at the end pint of the study. The baseline plasma HVA levels and HVA/5-HIAA ratio were significantly higher in responders(N=13) than in nonresponders (N=14). But no significant difference in baseline levels of plasma 5-HIAA was found between responders and nonresponders. At the end point of the study, there was significant difference in the change of plasma HVA between responders(40.3% decrement) and nonresponders(3.1% increment). But no signficant differences in the change of plasma 5-HIAA and the change of plasma HVA/5-HIAA ratio between responders and nonresponders were observed. These results suggest that the antipsychotic effect of clozapine on positive symptoms may be associated with dopaminergic blocking activity, and that on negative symptoms may be associated with serotonergic blocking activity. The baseline plasma HVA levels and the change of HVA levels from baseline may be useful predictors of treatment response with clozapine.
