추출용매에 따른 다릅나무 수피 추출물의 항산화 활성 (Stem bark of Maackia amurensis Extract according to extraction Solvent)

  • Objectives : The objective of this research was to investigate the antioxidant activities of stem bark of Maackia amurensis extract.Methods : Stem bark of Maackia amurensis extract were prepared using 70% methanol. Methanol extracts were fractionated to hexane, chloroform, ethyl acetate, butyl alcohol, water fractions and investigated. The antioxidant activities of fractions was evaluated by four different assays as total polyphenol contents, total flavonoid contents, DPPH(1,1-diphenyl-2-picrylhydrazyl) free-radical scavenging activity and ABTS(2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) scavenging ability.Results : The yield of methanolic extracts from stem bark of Maackia amurensis was 10.16%, whereas those of its solvent fractions (hexane, chloroform, ethyl acetate, butyl alcohol, and water) were 5.45, 11.39, 13.88, 26.07, and 40.80%, respectively. The total polyphenol contents and electron donating ability of 70% methanol extracts from stem bark of Maackia amurensis were 15.44 mg/g and 194.15 μg/mL of its IC50, respectively. The 70% methanol extracts showed the highest antioxidant activity. The total polyphenol content and total flavonoid content of chloroform fractions were higher in each of 201.98 mg/g and 13.55 mg/g. The chloroform fraction showed the lowest levels of DPPH(IC50, 183.95 μg/mL) and ABTS scavenging activity(IC50, 10.0 μg/mL). The antioxidant activity was detected in methanol extract, chloroform fractions.Conclusions : These results indicate that 70% methanol extract and its fractions of stem bark of Maackia amurensis, especially chloroform fraction, have the properties of anti-oxidant suggesting stem bark of Maackia amurensis may be a candidate for natural and functional materials.

수수 및 옥수수 첨가 비율별 발효주의 품질 특성 (Quality Characteristics of Korean Traditional Wines with Different Amounts of Sorghum and Corn)

  • In this study, we attempted to compare the quality characteristics of korean traditional wines with different amounts of corn and sorghum. These samples were analyzed for pH, total acidity, ethanol contents, total polyphenol, total flavonoid and tannin contents, ABTS and DPPH radical scavenging activities. The pH of fermented wines ranged from 3.83 to 4.64 and the total acidity of samples ranged from 0.30~0.63% on the 12th day of fermentation period. After 12 days of fermentation, the alcohol content of sorghum and corn fermented wines ranged between 11.6~15.5%. The a value(Redness) of fermented wines was the highest at 8.13 in B treatment and the a value decreased as the rate of corn addition increased. Total polyphenol and flavonoid contents had the highest values(162.64 and 18.44 mg/100 mL, respectively) in the E treatment which is the fermented wines of 25% Sorghum and 75% Corn. The ABTS and DPPH radical scavenging activity of the samples were 71.06~74.57% and 15.46~36.20% respectively. Antioxidant activity was higher in fermented wines with sorghum and corn than in control. As a result, this study provides useful scientific information that quality characteristics of fermented wines containing sorghum and corn and forms a basis in the food and wine industry.

참나무 수종별 톱밥재배에 따른 표고의 항산화 특성 (Antioxidant Properties of Lentinula edodes after Sawdust Bag Cultivation with Different Oak Substrates)

  • 본 연구에서는 참나무 수종을 달리하여 톱밥재배한 표고의 항산화 활성을 알아보기 위해 건조된 표고를 70% methanol로 추출하여 총 폴리페놀 함량, 총 플라보노이드 함량, 2,2-Diphenyl-1-picrylhydrazyl (DPPH) 라디칼 소거활성, ferric reducing antioxidant capacity(FRAP), reducing power를 측정하였다. 총 폴리페놀 함량과 총 플라보노이드 함량을 측정한 결과, 갈참나무로 톱밥재배 한 표고가 3.12 mg GAE/g과 0.84 mg QE/g으로 다른 수종에 비해 가장 높게 나타났다. DPPH 라디칼 소거활성 측정 결과, 10 mg/mL일 때 다른 수종에서 재배한 표고에 비해 갈참나무(89.4%)와 상수리나무(87.9%)에서 가장 높았다. FRAP은 갈참나무일 때 0.425로 가장 높게 나타났고, reducing power도 역시 갈참나무에서 0.359로 가장 높게 나타내었다. 참나무 수종에 따른 표고의 항산화 활성은 갈참나무로 톱밥재배한 것이 가장 우수한 것으로 나타났다. 본 연구의 결과로 동일한 균주를 이용하더라도 톱밥재료에 따라 표고의 항산화 활성이 차이가 있다는 것을 확인할 수 있었다.

  • We investigated antioxidant activity and lignan contents by harvesting times to expand use of Schisandra chinensis (Turcz.) Baillon. Total polyphenol and flavonoid contents of seed was higher than those of flesh but there is not much difference in harvesting times. As $RC_{50}$ value, that was, the concentration of sample required for 50% reduction of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical, was 13.7~24.2 ${\mu}g/m{\ell}$ in seed thus it showed a high antioxidant activity. Among lignan components, schizandrin content was the highest and followed by gomisin N and gomisin A in all of flesh and seed. Also these components in seed were 4~9 times more contained than those of flesh. All of them were decreased by harvesting times in flesh. But the contents of schizandrin and gomisin N were high in August 3rd and September 15th in seed, respectively. As the results, the seed of S. chinensis had high antioxidant activity and lignan contents so it could be potentially developed as a resource.

Antioxidant Potentials and Quantification of Flavonoids in Mung Bean (Vigna radiata L.) Seeds

  • Mung bean (Vigna radiata L.) is an increasingly important human food source, as well as a new functional agent, mainly due to its potent antioxidant activity. This study was conducted to determine antioxidant activity of fractions from mung bean seeds by measuring DPPH radical scavenging activity and to quantify the flavonoids by means of HPLC analysis. Vitexin and isovitexin were present in both ethanol and water extracts in highest amount. Flavonoids, vitexin and isovitexin were quantified from 195 germplasms of mung beans and their concentrations varied by 4.7 fold. Especially, the breeding line KM99004-4B-2 (Suwon28/KM94004), which has grown in Jeollanamdo Agricultural Research and Extension Services, showed the highest amount (15.88 mg/g) of total flavonoids. The vitexin portion was averaged $70.73{\pm}1.38\%$. High positive correlation $(r=0.96^{***})$ between vitexin and isovitexin contents showed. However, the flavonoid content showed very low correlation with the 24 growth and ecological characteristics. Seed coats of mung beans had the highest flavonoid amount, showing $50\~70$ times more than cotyledons. Flavonoid contents in the seed, the cotyledon, and the seed coat were decreased as the seed imbibition time increased.

Antibacterial Effect of Allium ampeloprasum and Allium porrum Extracts on Staphylococcus aureus and Pseudomonas aeruginosa

  • Objectives: The most prominent microorganisms that cause hospital infections and acquire antibiotic resistance are Staphylococcus aureus and Pseudomonas aeruginosa. The present study aimed to compare the phenolic and flavonoid compounds of various Allium ampeloprasum and Allium porrum extracts and evaluate the antibacterial effects of these extracts against these two microorganisms. Methods: The total phenolic and flavonoid contents of the acetone, methanol, aqueous, and hexane leeks extracts from A. ampeloprasum and A. porrum were measured. The antibacterial activity of these extracts against S. aureus and P. aeruginosa was tested using the disk diffusion method for 24, 48, and 72 hours. Further, the minimum inhibitory concentrations and the minimum bactericidal concentrations of these extracts for these two bacteria were evaluated and compared with those of common antibiotics. Results: The aqueous extracts showed the highest phenolic and flavonoid contents and at concentrations of 35 and 40 mg per disk, showed the most antibacterial activity against S. aureus and P. aeruginosa; P. aeruginosa showed more sensitivity to the aqueous extracts than S. aureus. Conclusion: Aqueous A. ampeloprasum and A. porrum extracts may prevent the growth of hospital pathogens, especially P. aeruginosa; our findings will aid the discovery of new antimicrobial substances against antibiotic-resistant bacteria.

Antioxidant and Anti-Inflammatory Effects of Various Cultivars of Kiwi Berry (Actinidia arguta) on Lipopolysaccharide-Stimulated RAW 264.7 Cells

  • The present study evaluated the total phenolic and flavonoid contents as well as total antioxidant capacity (TAC) of three cultivars of Actinidia arguta Planch. kiwi berries; cv. Mansoo (Mansoo), cv. Chiak (Chiak), and cv. Haeyeon (Haeyeon). In addition, the anti-inflammatory effects of the three cultivars of kiwi berries were investigated using a lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophage cell line. Mansoo had the highest total phenolic content and TAC among the three cultivars, whereas Chiak had the highest total flavonoid content. The total antioxidant capacities of the kiwi berry extracts were more strongly correlated with total phenolic content than with total flavonoid content. The kiwi berry extracts suppressed the secretion of pro-inflammatory cytokines, including interleukin-6 and tumor necrosis factor-α, from LPS-stimulated RAW 264.7 cells. The release of nitrite, an indirect indicator of nitric oxide, was also ameliorated by pre-treatment with the kiwi berry extracts in a dose-dependent manner. Cellular-based measurements of antioxidant capacity exhibited that the kiwi berry extracts had cellular antioxidant capacities. Such cellular antioxidant effects are possibly attributed to their direct antioxidant capacity or to the inhibition of reactive oxygen species generation via anti-inflammatory effects. Our findings suggest that kiwi berries are potential antioxidant and anti-inflammatory agents.

식물성 당단백질의 제조 및 유효성 분석 (Preparation and Availability Analysis of Vegetable Glycoprotein)

  • This study is on the feasibility of use of glycoprotein in various areas such as cosmetics and food etc. by extracting, isolating and refining glycoprotein from carrots, red ginseng extract residue, sesame and pine needles using protease(pepsin) and by analyzing general characteristics and measuring various bioactivities. The results of analysis of nutritional composition showed protein contents of glycoprotein. In the analysis of constitutive amino acids, the ratio of contents of hydroxy proline and glycine, the characteristics of glycoproteins appeared similar and the contents of glutamic acid and aspartic acid appeared higher. As a result of measurement contents of total polyphenol and flavonoid, it showed that glycoprotein had more contents generally, and the effect of bioactivity of glycoprotein appeared higher although different kinds of glycoprotein showed a little DPPH radical and nitrite scavenging ability, total antioxidant capacity by ABTS, ACE inhibitory.

수확시기별 오렌지 5품종 부산물의 플라보노이드 함량과 항산화 활성 (Flavonoids composition and antioxidant activity of by-products of five orange cultivars during maturation)

  • 오렌지 생과 및 가공용으로 주로 사용되는 주요 5가지 오렌지 품종의 과피 및 착즙을 하고 난 부산물의 플라보노이드 성분과 유리 라디칼 소거활성을 성숙시기별로 분석하여 오렌지 부산물의 천연물 유래 기능성 소재로써의 활용 가능성을 검토하고자 본 연구를 수행하였다. 성숙시기에 따른 총 플라보노이드 함량은 모든 시료에서 성숙이 진행된 과일 보다 미성숙 상태의 과일인 9월에 수확한 과일에 함량이 가장 높은 것으로 나타났으며, 성숙이 진행될수록 감소하는 경향을 보였다. 품종간에는 과피 추출물의 경우 Hamlin(21.66 mg/g) > Sanguinello blood(20.39 mg/g) > Shamouti(18.49 mg/g), Tarocco blood(18.46 mg/g) > Olinda valencia(17.07 mg/g) 순으로 조사되었다. 플라보노이드 성분 변화에서는 모든 시료에 narirutin과 hesperidind의 함량이 높았으며, naringin과 neohesperidin은 검출되지 않았다. PMF 성분 중에는 9월에 수확한 미성숙과의 과피 추출물에 nobiletin이 $23{\sim}40{\mu}g/mL$ 함유되어 있음을 확인할 수 있었다. 유리 라디칼 소거활성에서는 모든 품종에서 과피 추출물이 과육 추출물에 비해 우수하였고, 과피 추출물의 DPPH 라디칼 소거활성은 40~58%, ABTS 라디칼 소거활성에서는 90~94%의 소거능을 보였다.

증숙처리에 따른 재래생강 및 중국종자생강의 항산화 및 기능성물질 비교 (Comparison of Antioxidant and Functional Compounds in Korean Conventional and Chinese Seed Ginger (Zingiber officinale Roscoe) Following Steam Treatment)

  • 본 연구에서는 재래생강과 중국종자생강 간의 증숙 시간별 총 페놀화합물, 총 플라보노이드, ABTS 및 DPPH 라디칼 소거능, gingerol, shogaol 함량을 비교 분석하였다. 총 플라보노이드와 총 페놀 화합물, ABTS radical 소거능의 경우 대체적으로 증숙 6시간에서 가장 높은 함량을 나타내다가 그 이상으로 증숙할수록 오히려 감소하는 경향을 보였으며, 생강 중에서는 재래생강인 WO-2가 가장 높게 나타났다. DPPH 라디칼 소거능은 증숙 시간별로 유의미한 차이가 없었다. 생강의 gingerol 함량은 대조군에서 WO-2가 1,351 mg/kg으로 가장 높은 함량을 나타냈다. 증숙 시간에 비례하여 gingerol의 함량은 감소하고, shogaol의 함량은 증가하였는데 증숙 8시간에서 WO-2가 생강 중에서도 가장 높았으며, 또한 종자별로도 재래생강이 중국 종자생강보다 shogaol 함량이 높은 것으로 나타났다. 본 연구결과 재래생강이 중국종자생강보다 기능성 함량이 높음을 확인하였으며 증숙 처리된 재래생강을 활용하여 다양한 식품을 개발할 수 있을 것으로 판단된다.
