Research on the Development of Microneedle Arrays Based on Micromachining Technology and the Applicability of Parylene-C (미세가공 기술 기반의 마이크로니들 어레이 개발 및 패럴린 적용 가능성에 관한 연구)

  • In this research, we studied the development of a SUS304 microneedle array based on microfabrication technology and the applicability of Parylene-C thin film, a medical polymer material. First of all, four materials commonly used in the field of medical engineering (SUS304, Ti, PMMA, and PEEK) were selected and a 5 ㎛ Parylene-C thin film was deposited. The applicability of Parylene-C coating to each material was confirmed through SEM analysis, contact angle measurement, surface roughness(Ra) measurement, and adhesion test according to ASTM standards for each specimen. Parylene-C thin film was deposited based on chemical vapor deposition (CVD), and a 5 ㎛ Parylene-C deposition process was established through trial and error. Through characteristic experiments to confirm the applicability of Parylene-C, SUS304 material, which is the easiest to apply Parylene-C coating without pretreatment was selected to develop a microneedle array based on CNC micromachining technology. The CNC micromachining process was divided into a total of 5 steps, and a microneedle array consisting of 19 needles with an inner diameter of 200 ㎛, an outer diameter of 400 ㎛, and a height of 1.4 mm was designed and manufactured. Finally, a 5 ㎛ Parylene-C coated microneedle array was developed, which presented future research directions in the field of microneedle-based drug delivery systems.

Effects of Dietary Coconut Oil as a Medium-chain Fatty Acid Source on Performance, Carcass Composition and Serum Lipids in Male Broilers

  • This study was conducted to investigate the effects of dietary coconut oil as a medium-chain fatty acid (MCFA) source on performance, carcass composition and serum lipids in male broilers. A total of 540, one-day-old, male Arbor Acres broilers were randomly allotted to 1 of 5 treatments with each treatment being applied to 6 replicates of 18 chicks. The basal diet (i.e., R0) was based on corn and soybean meal and was supplemented with 1.5% soybean oil during the starter phase (d 0 to 21) and 3.0% soybean oil during the grower phase (d 22 to 42). Four experimental diets were formulated by replacing 25%, 50%, 75%, or 100% of the soybean oil with coconut oil (i.e., R25, R50, R75, and R100). Soybean oil and coconut oil were used as sources of long-chain fatty acid and MCFA, respectively. The feeding trial showed that dietary coconut oil had no effect on weight gain, feed intake or feed conversion. On d 42, serum levels of total cholesterol, low-density lipoprotein cholesterol, and low-density lipoprotein/high-density lipoprotein cholesterol were linearly decreased as the coconut oil level increased (p<0.01). Lipoprotein lipase, hepatic lipase, and total lipase activities were linearly increased as the coconut oil level increased (p<0.01). Abdominal fat weight/eviscerated weight (p = 0.05), intermuscular fat width (p<0.01) and subcutaneous fat thickness (p<0.01) showed a significant quadratic relationship, with the lowest value at R75. These results indicated that replacement of 75% of the soybean oil in diets with coconut oil is the optimum level to reduce fat deposition and favorably affect lipid profiles without impairing performance in broilers.

Anti-hyperlipidemic Effects of the Herbal Extract Mixture of Cnidium officinale, Petasites japonicus and Coptis chinensis on Mice with Tyloxapol (천궁(川芎), 머위, 황연(黃連) 추출물 조성이 생쥐의 지질대사에 미치는 영향)

  • This study was performed to evaluate the effect of the herbal extract mixture of Cnidium officinale, Petasites japonicus and Coptis chinensis (CPC) on the lipids metabolism in mice with tyloxapol. ICR mice weighing between 30-40 g were divided into four groups: normal group, 600mg/kg tyloxapol injected group, $50\;{\mu}g/g$ CPC treated group 6h after tyloxapol injection (SAM1), and tyloxapol and CPC treated group (SAM2), respectively. Tyloxapol or CPC was injected intraperitoneally. Tyloxapol caused an elevation of total cholesterol (TC), triglycerides(TG), and LDL-cholesterol and a decrease of HDL-cholesterol. In addition, tyloxapol induced accumulation of lipid including cholesterol in both the liver and kidney. Serum levels of TC, TG and LDL-cholesterol were decreased whereas HDL-cholesterol was increased by CPC. CPC increased in HDL-cholesterol /total cholesterol ratio and lowered atherogenic index. The levels of TC, TG and LDL-cholesterol by CPC were rather lower in SAM2 than SAM1. CPC also inhibited lowering HDL-cholesterol by tyloxapol. CPC reduced lipid blots and cholesterol particles in both the deposition and size in the liver and kidney with tyloxapol. These results suggest that CPC might be expected to be beneficial for protection and treatment of hyperlipidemia by the disturbance of lipid metabolism.

A Study on Exploring Purification Management Indicators of Depositied Waste in Coastal Waters (연근해 침적폐기물 정화관리지표 개발에 관한 연구)

  • This study aims to explore a set of indicators to be used for purification management. by estimating the total quantity of depositied waste in coastal waters, Through 3 rounds of Delphi technique, this study sorts out the following 5 variables to be used as major indicators determining priorities for purification in coastal areas: the ratio of re-deposition after collection, the area where fishing gear are lost, the total quantity of depositied waste of each coastal water, the total quantity of deposited waste in the past, and the quantity of depositied waste by unit area. Although the fact that the delphi surveys were done only on 27 specialists limits its external validity, this study will serve as a base for in-depth studies with more expanded panels that will provide better indicators for purification management in coastal waters of Korea.

  • This work was intended to study the effect of a partial pressure ratio and a total pressure of reactive gases on the properties of TiC$_{x}$N$_{1-x}$ . coated layer. In this regard, various TiC$_{x}$N$_{1-x}$ coatings were synthesized with C2112 and N2 Mixture gas of different compositions by Arc Ion Plating process which has been highlighted for an industrial purpose. It was revealed from colors and X-ray diffraction patterns that the concentration of carbon of a TiC$_{x}$N$_{1-x}$ coating increases with a partial pressure ratio (PC$_{2}$H$_{2}$/PN$_{2}$) as well as a total pressure Of $C_{2}$H$_{2}$ and N$_{2}$ mixture gas. Accordingly, the hardness of TiC$_{x}$N$_{1-x}$ coated layer increased but the adhesion to the substrate of SKH 51 was degraded. On the other hand, the deposition rate was independent of a partial pressure ratio and a total pressure of mixture gas. It was found that a uniform gas distribution is critical for an industrial application since the composition of a coating depends strongly on the location of a substrate inside of the furnace. As a result of milling tests with different TiC$_{x}$N$_{1-x}$ coated end mills, the one which has a low carbon concentration was better than others studied in this work.d in this work.

Examining the Interrelation of Total, Soluble, and Bioavailable Metals in the Sediments of Urban Artificial Lakes (도심인공호 퇴적물의 총중금속, 용존중금속, 생물이용성 중금속의 연관성 규명)

  • Total metals, soluble metals, and bioavailable metals were monitored at the sediments of urban lakes located in Seoul, Korea during spring season 2006. The metals measured were zinc, arsenic, chromium, copper, nickel, and cadmium, which are known to be toxic to human health and ecosystems. The main sources of heavy metals in the lakes were urban runoff and atmospheric deposition associated with air pollution in urban areas. Extraction by using a weak electrolyte solution (0.1 M $Ca(NO_3)_2$) was used to predict bioavailability of the metals. Among the six heavy metals studied, copper was the most bioavailable, based the weak electrolyte extraction techniques. Since metal toxicity is related to metal bioavailability, the results were consistent with the high ecotoxicity of copper, compared to other heavy metals. Overall results suggest that there was no direct relationship between total and bioavailable metal concentration, although zinc, copper and cadmium show some relationships.

Effects of Boyangmaksung-bang (BYMSB) on the Membranous Nephropathy Induced by Cationic Bovine Serum Albumin in Mouse Model (보양막성방(補陽膜性方)이 Cationic Boivine Serum Albumin 투여로 유발된 Mouse의 Membranous Nephropathy에 미치는 영향)

  • Objective: The purpose of experimental study was to prove the effects of Boyangmakseong-bang (BYMSB) treatment on cBSA-induced in a MN mouse model. Methods: We divided mice into 4 groups. The Normal group had no treatment. We used cBSA and induced MN mouse model to the other 3 groups. The Control group was treated with cBSA (9mg/kg i.p) only. The second group, named 'BY-250', was treated with cBSA (9mg/kg i.p) and BYMSB extract (250mg/kg, p.o). The third group, named 'BY-500', was treated with cBSA (9mg/kg i.p) and BYMSB extract (500mg/kg, p.o). After cBSA and BYMSB extract treatment for 4 weeks, the increase in percentage of body weight, proteinuria, serum albumin, total cholesterol, creatinine and BUN of all groups were measured. The CD3+, CD19+, CD4+, CD8+ cell levels of spleen of all groups were analyzed. IgA, IgG, IgM, IL-$1{\beta}$, TNF-${\alpha}$, IL-6 and IFN-${\gamma}$ levels of all groups were gauged. H&E staining, immunofluorescence staining and electron microscopy of kidney were observed. Results: BYMSB showed significant decrease in the 24hrs proteinuria, serum total cholesterol, serum IgG levels and BUN levels, and showed significant increase in the serum albumin levels compared with the control group. BYMSB showed increase in the increasing percentage of body weight and IFN-${\gamma}$ levels compared with the control. BYMSB showed decrease in the CD3+ T cells, CD4+ Th cells, IL-$1{\beta}$, TNF-${\alpha}$ and IL-6 levels, but did not show significant change compared with the control. BYMSB showed considerable decrease in the thickening of the GBM on H&E staining, deposition of IgG on immunofluorescence staining and deposition of electron-density on electron microscopy of kidney compared with the control. Conclusions: According to the above results, it is suggested that BYMSB decreases the symptoms of MN induced by cBSA in a mouse model. Therefore BYMSB seems to be applicable to MN in clinical practice.

Transportation and Deposition of Modern Sediments in the Southern Yellow Sea

  • Based on the data obtained under the China-Korea joint project (1997-2001) and historic observations, the distribution, transportation and sedimentation of sediment in the southern Yellow Sea (SYS) are discussed, and the controversial formation mechanism of muddy sediments is also explored. The sediment transport trend analysis indicates that the net transport direction of sediment in the central SYS (a fine-grained sediment deposited area) points to $123.4^{\circ}E,\;35.1^{\circ}N$, which is a possible sedimentation center in the central SYS. The sediment transport pattern is verified by the distribution of total suspended matter (TSM) concentration and ${\delta}^{13}C$ values of particulate organic carbon (POC), the latter indicates that the bottom water plays a more important role than the surface water in transporting the terrigenous material to the central deep-water area of the SYS, and the Yellow Sea circulation is an important control factor for the sediment transport pattern in the SYS. The carbon isotope signals of organic matter in sediments indicate that the Shandong subaqueous delta has high sedimentation rate and the deposited sediments originate mainly from the modern Yellow River. The terrigenous sediments in deep-water area of the SYS originate mainly from the old Yellow River and the modern Yellow River, and only a small portion originates from the modern Yangtze River. The analytical results of TSM and stable carbon isotopes are further confirmed by another independent tracer of sediment source, polycyclic aromatic hydrocarbons (PAHs). Five light mineral provinces in the SYS can be identified and they indicate inhomogeneity in sources and sedimentary environment. The modern shelf sedimentary processes in the SYS are controlled by shelf dynamic factors. The muddy depositional systems are produced in the shelf low-energy environments, which are controlled by some meso-scale cyclonic eddies (cold eddies) in the central SYS and the area southwest of the Cheju Island. On the contrary, an anticyclonic muddy depositional system (warm eddy sediment) appears in the southeast of the SYS (the area northwest of the Cheju Island). In this study, we give the cyclonic and anticyclonic eddy sedimentation patterns.

Deposition Characteristics of Atmospheric PCBs Depending on Exposure Periods Using Pine Needles (소나무 잎을 이용한 대기 중 PCBs의 노출기간별 침착특성)

  • Pine needle samples(1 year, 2 year, 3 year old) were collected in Seoul and Ansung to investigate the deposition characteristics of atmospheric PCBs depending on exposure periods. Total PCBs concentration of pine needle samples in Seoul and Ansung showed 2,429, 1,198 pg/g DW(dry weight), respectively. PCB homologs concentration of pine needle samples decreased with increasing chlorine-substituted compounds. It showed that higher chlorine- substituted compounds in existing particle phase compared to lower chlorine-substituted compounds existing in gaseous phase were difficult to accumulate lipids of the pine needles samples. The results of regression analysis between exposure periods in the atmosphere and concentration of total PCBs measured in pine needles at two sites showed significant levels($R^2>0.94$, p<0.01), which implies that atmospheric PCBs accumulated on pine needles with positive linearity depending on exposure periods in this study. Therefore pine needle samples can be used as passive air sampler(PAS) for monitoring air contamination for a long time in different sites.

Effects of Artificially Acidified Soils on the Growth and Nutrient Status of Pinus densiflora and Quercus acutissima Seedlings (토양산성화가 소나무, 상수리나무 묘목의 생장 및 영양상태에 미치는 영향)

  • The effects of soil acidification on the seedling growth and nutrition of Pinus densiflora and Quercus acutissima were investigated. The relationship between the seedling growth and molar (Ca+Mg+K)/Al ratio of in soil solution was examined. The results suggested that growth inhibition of seedling Pinus densiflora and Quercus acutissima was due to the low pH of soil solution, which was followed by leach of Al into soil solution, and decrease of essential elements, such as Ca in aerial pant of the seeding caused by the increase of Al concentration in subterranean pant of the seedlings. The level of growth inhibition was determined not only by Al concentration, but also by the balance of inorganic elements, Al, Ca, Mg and K. The growths of two species in total dry weight were clearly inhibited when molar (Ca+Mg+K)/Al ratio of the soil was lower than 6.0. The growth in dry weight, in the condition of the molar ratio was 0.8, was decreased 60% or 50% for the seedling of Pinus densiflora or Quercus acutissima respectively. It was concluded that the molar (Ca+Mg+K)/ Al ratio could be an important index for evaluation of the effects of soil acidification, due to acid deposition such as acid rain, on growth of trees and nutrition. And it might be a more useful indicator for evaluation of critical load of acid deposition on forest ecosystems.
