This study was carried out to investigate the effects of pine needle extract on the color, hardness, springiness, chewiness, pH, thiobarbituric acid reactive substances (TBARS) value, and total bacterial number of seasoned pork meat stored at $4^{\circ}C$ for 9 days. The pH levels of sauce samples were not affected by the mixing rate of the extracts. Acidity, soluble solids, and salinity gradually increased as the amount of added extract increased. Total polyphenolic contents in the sauce ranged from $1.01{\pm}0.02$ mg GAE/mL to $1.41{\pm}0.04$ mg GAE/mL, DPPH radical scavenging activity ranged from $0.06{\pm}0.01$ AEAC to $0.12{\pm}0.01$ AEAC, and ABTS radical scavenging activity ranged from $0.11{\pm}0.01$ AEAC to $0.19{\pm}0.01$ AEAC. The pH levels significantly decreased as the amount of added extract increased. The lightness ($L^*$), redness ($a^*$) and yellowness ($b^*$) values of meat tended to decrease with longer storage period (p<0.05). Hardness and chewiness also increased with longer storage period (p<0.05). The TBARS values decreased as the amount of added extract increased after 6 days (p<0.05). Total bacterial numbers of P5, P10, and P15 decreased compared to the control (p<0.05). In the sensory evaluation, taste and palatability were not significantly different among C, P5, and P10 (p<0.05). Further, flavor, color, tenderness, and juiciness were not different among the seasoned pork meats. These results suggest that pine needle extract can inhibit protein degradation, lipid oxidation, and bacterial growth when used as an additive to seasoned pork meat.
The effect of irradiation at dose of 3~10 kGy on the extension of the shelf-life of sliced dried squid and dried file and on its quality were investigated during nine months of storage at room temperature. The number of microorganisms, TBA value and TMA-N content were used as objective indices concerned to the sensory evaluation. In the number of initial microbial loading, total bacterial count of sliced dried squid and dried file fish were $2.7{\times}10^{8}\;and\;2.0{\times}10^{5}$, yeast and mold were $5.7{\times}10^{5}\;and\;6.2{\times}10^{5}$ and coliform group were $2.1{\times}10^{5}$ and above 10 per gram of the samples, respectively. The number of total bacteria was sterilized by 99% with irradiation of 3~10 kGy and irradiation of above 7 kGy was shown to be effective for the radurization of yeast, mold and coliform group. TBA value of irradiated groups were higher than storage time. The TMA-N content of nonirradiated group was markedly increased by microbial spoilage during storage, however above 8 kGy irradiated group were indicated about 10 mg% after nine months storage. In the sensory evaluation, the nonirradiated group was off favor after three months storage and was changed in color of dried fishes but 7~10 kGy irradiated group were maintained good qualit compared with those of fresh samples after nine months storage at room temperature.
Endophytic bacterial communities of tidal flat plants antagonistic to oomycete plant pathogens were studied by the isolation of 256 root colonizing endophytic bacteria from surface-disinfected root tissues of six plants ($Rosa$$rugosa$, $Suaeda$$maritima$, $Vitex$$rotundifolia$, $Carex$$scabrifolia$, $Glehnia$$littoralis$ and $Elymus$$mollis$) growing in a tidal flat area of Namhae Island, Korea. To understand the antagonistic potential, an $in$$vitro$ antagonistic assay was performed to characterize and identify strains that were antagonistic to the oomycete plant pathogens $Phytophthora$$capsici$ and $Pythium$$ultimum$ from the total population. Nine percent of the total number of isolated bacteria exhibited in vitro inhibitory activity against target plant pathogenic oomycetes. Taxonomic and phylogenetic placement of the antagonistic bacteria was investigated by analysis of the 16S rRNA gene sequences. The sequence analysis classified the antagonistic strains into four major classes of the domain bacteria ($Firmicutes$, ${\alpha}-Proteobacteria$, ${\gamma}-Proteobacteria$ and $Actinomycetes$) and 10 different genera. Further production of secondary metabolites, hydrolytic enzymes and plant growth promoting traits were determined for the putative new species of antagonistic endophytic bacteria. These new strains could not be identified as known species of ${\alpha}-Proteobacteria$, and so may represent novel bacterial taxa. The unexpected high antagonistic bacterial diversity associated with the tidal flat plants may be indicative of their importance in tidal flat plants as a promising source of novel antimicrobial compounds and biocontrol agents.

Purpose: The aim of this study was to compare the prevalence and bacterial load of 6 main periodontal pathogens between pairs of periodontal patients with and without type 2 diabetes mellitus. Porphyromonas gingivalis and Aggregatibacter actinomycetemcomitans genotypes were also investigated. Methods: Twenty patients affected by chronic periodontitis and type 2 diabetes were retrospectively selected and matched to 20 patients without diabetes on the basis of the degree and severity of periodontal disease. Microbiological data of subgingival biofilms were analysed and compared for the examined pathogens: A. actinomycetemcomitans, P. gingivalis, Prevotella intermedia, Treponema denticola, Fusobacterium nucleatum, and Tannerella forsythia. Results: The pairs were balanced in terms of demographic and clinical parameters, except for bleeding on probing and suppuration. In the microbiological test sites (4 for each patient), the mean probing pocket depth was 6.34±1.63 mm in patients with diabetes and 6.41±1.78 mm in patients without diabetes. No significant difference between pairs in the prevalence of P. gingivalis or the distribution of its genotypes was recorded. Patients with diabetes had a significantly greater amount of total bacterial load, P. gingivalis, T. denticola, T. forsythia, and F. nucleatum (P<0.05). Moreover, patients with diabetes had a higher number of sites with a greater cell count than patients without diabetes. When compared to the total bacterial load, only T. forsythia maintained its relative load in patients with diabetes (P=0.001). Conclusions: This retrospective matched study supports the hypothesis that microbiological differences exist among periodontal patients with and without diabetes mellitus.
This study was conducted to survey the relationship between the somatic cell count (SCC), and California mastitis test (CMT) & mastitis. A total of 328 quarter milk samples were collected from 211 cows suspected to have mastitis; Both SCC and CMT were performed on the samples. Milk smear was stained with Broadhurst and Paley stain and the cells were classified into either epithelial or blood cells. Bacterial isolation was made and antimicrobial susceptibility was tested. Of the 328 quarters, 78 ($23.8{\%}$) were CMT negative with SCC <750,000/ml. As expected, CMT score increased with the increase of SCC. The number of epithelial cells decreased with the increasing number of somatic cells, while the opposite was the case with the number of blood cells. The critical point was when the SCC reached 1,000,000/ml. Up to 1,000,000 cells/ml, the number of epithelial cells was greater than that of blood cells. The results indicate that when epithelia:blood cell ratio is 58.1:41.9, the milking line should be checked and bacterial isolation be performed on the samples in order to identity mastitis.

Presence and distribution of bacteria and fungi in inner compartment of PCs(Personal Computers) were investigated. Samples for the analysis were collected from inside of PCs which had been used in various facilities including public computer facilities, laboratories and computer training rooms of a university. Total number of PC examined in this study was 51 each. When the total CFU(colony forming unit) of the inner compartment of the PCs was measured, the bacterial count was found to be dependent on the operation time(total running time) of PCs. When the distribution of bacteria in the inner compartment of PCs was estimated, CPU(Central Processing Unit) cooling fan area showed the highest bacterial concentration(average 605 $CFU/cm^2$). In the case of the fungi, various opportunistic pathogens including Aspergillus sp. and Penicillium sp. were isolated and identified in the inner compartment of PCs. And the average of bacterial number in the dust collected from the PCs was 212 CFU/mg. These results indicated that handling of PC might have a risk of infection by the microorganism.
The composition of the bacterial populations in Gorosoe(Acer mono Max.) sap was characterized during storage with different heat treatments($63^{\circ}C$ for 30 min and $73^{\circ}C$ for 15 sec). The saps were aseptically collected at 0, 15 and 30 days of storage and analyzed by dilution plating and 16S rDNA PCR-DGGE analysis. There were significant differences in the total number of colony forming units(CFUs) of bacteria between heated and nonheated saps. Bacteria of nonheated sap were present at a level of $3.4{\times}10^7CFU/m{\ell}^{-1}$, whereas living bacteria were not detected in the heated sap. According to the 16S rDNA sequence and DGGE analysis, Pseudomonas sp. was the most abundant bacterial strain in the samlpes, and the bacterial community structures become more simplified with time and were composed of the Chryseobacterium sp. with time. These results allowed us to characterize the dominant bacteria involved in Gorosoe sap and to better understand their dynamics throughout storage.
Lettuce (Lactuca sativa L.) is a major ingredient used in many food recipes in South Korea. Lettuce samples were collected during their maximum production period between April and July in order to investigate the microbiota of lettuce during different seasons. 16S rRNA gene-based sequencing was conducted using Illumina MiSeq, and real-time PCR was performed for quantification. The number of total bacterial was greater in lettuce collected in July than in that collected in April, albeit with reduced diversity. The bacterial compositions varied according to the site and season of sample collection. Potential pathogenic species such as Bacillus spp., Enterococcus casseliflavus, Klebsiella pneumoniae, and Pseudomonas aeruginosa showed season-specific differences. Results of the network co-occurrence analysis with core genera correlations showed characteristics of bacterial species in lettuce, and provided clues regarding the role of different microbes, including potential pathogens, in this microbiota. Although further studies are needed to determine the specific effects of regional and seasonal characteristics on the lettuce microbiota, our results imply that the 16S rRNA gene-based sequencing approach can be used to detect pathogenic bacteria in lettuce.
BACKGROUND: Although antibiotics have contributed to treatment of bacterial infection, the antibiotic abuse can lead to antibiotic resistant bacteria. Impact of human activities on distribution of antibiotic resistance has been intensively issued and occurrence of antibiotic resistant bacteria in contaminated environments would not be a surprise. Nonetheless, anthropogenic contamination with the dissemination of antibiotic resistance along uncontaminated environments has been less considered. The aim of this study is to investigate antibiotic resistant bacteria across Ulleungdo, known as antibiotic resistance free and anthropogenic pollution free environment in Rep. of Korea. METHODS AND RESULTS: Antibiotic resistant bacteria in coastal seawater of Ulleungdo were investigated in July 2021. Antibiotic susceptibility test using the disk diffusion method was applied with six drugs according to the Clinical and Laboratory Standards Institute (CLSI) guideline. Total 43 bacterial isolates were tested and 20 isolates among of them showed multidrug resistance. Particularly, the number and ratio of resistant bacteria were relatively high in a densely populated area of Ulleungdo. The bacterial communities were investigated using 16S rRNA gene metabarcoding approach in the coastal seawater and soils of Ulleungdo. In the bacterial communities, Firmicutes were selectively distributed only in seawater, suggesting the possibility of anthropogenic contamination in coastal seawater of Ulleungdo. CONCLUSION(S): We found antibiotic resistant bacteria in a populated area of Ulleungdo. The occurrence of antibiotic resistant bacteria in Ulleungdo seems to result from the recent anthropogenic impact. Consistent monitoring of antibiotic resistant bacteria in the uncontaminated environment needs to considered for future risk assessment of antibiotics.
Seasonal and spatial variations in propagule numbers of Trichoderma species were investigated every other month for one year in deciduous and coniferous forest soils and evaluated the relationships of Trichoderma spp. populations to soil environmental factors. The total population of Trichoderma spp. increased until summer and then declined until winter. The yearly mean frequency of Trichoderma spp. exceeded 1.4% of total fungal propagules in two sites. Decreases of absolute an relative propagule numbers of Trichoderma spp. with increasing soil depth were found and variation in Trichoderma spp. propagules caused by differences in soil depth ($0{\sim}50cm$) was greater than that caused by differences in sampling time. The most common species occurring in two sites was T. viride, followed by T. polysporum, T. koningii, and T. hamatum. Individual species of Trichoderma showed diferent abundance trend in accordance with sampling time. T. viride was dorminant from spring to autumn, while T. polysporum dominated over the other speicies in winter. Variations in propagule number of Trichoderma sppp. were principally mediated by the actions of biotic environmental factors rather than by the direct effects of abiotic factors. In multiple-regression analyses, 48% of the total vaiation in Trichoderma spp. propagules in deciduous site could be accounted for by total fungal propagules and soil CMCase actvity. In coniferous site, 65% of total variation could be accounted for by total fungal and bacterial propagules, moisture content and organic carbon content.
