Effects of Psychrotrophic Bacteria, Serratia liquefaciens and Acinetobacter genomospecies 10 on Yogurt Quality

  • The aim of this study was to evaluate the effect of proteolytic (Serratia liquefaciens, match %: 99.39) or lipolytic (Acinetobacter genomospecies 10, match %: 99.90) psychrotrophic bacteria (bacterial counts, analysis of free fatty acids (FFA) and analysis of free amino acids) on the microbial and chemical properties (yogurt composition), and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) of yogurt during storage. Yogurts were prepared with raw milk preinoculated with each psychrotrophic bacteria. The total solid, fat, and protein content were not affected by preinoculation, but the pH of yogurt preinoculated with psychrotrophic bacteria was higher than in control. There was a dramatic increase in short chain free fatty acids among FFA in yogurt with Acinetobacter genomospecies 10. For 14 d of cold storage condition, SCFFA was 25.3 mg/kg to 34.4 mg/kg (1.36 times increased), MCFFA was 20.4 mg/kg to 25.7 mg/kg (1.26 times increased), and LCFFA was 240.2 mg/kg to 322.8 mg/kg (1.34 times increased). Serratia liquefaciens (match %: 99.39) in yogurt caused a greater accumulation of free amino acids (FAA), especially bitter peptides such as leucine, valine, arginine, and tyrosine, but SDS-PAGE showed that the inoculation of Serratia liquefaciens did not affect the degree of casein degradation during storage. Taken together, the excessive peptides and FFA in yogurt generated from psychrotrophic bacteria could develop off-flavors that degrade the quality of commercial yogurt products.

Evaluation of the Nutritional and Storage Quality of Meatballs Formulated with Bee Pollen

  • In this study, the nutritional and storage quality of meatballs formulated with different levels (0, 1.5, 3.0, 4.5 and 6.0%) of bee pollen were investigated during storage at $41^{\circ}C$ for 9 d. Protein content of meatballs increased, while moisture content decreased with increased pollen. The addition of pollen improved cooking loss but decreased the redness (Hunter a value) and sensory scores. Textural parameters (hardness, springsness, gumminess, and chewiness) were affected by pollen addition and the hardness and gumminess values of meatballs decreased as the pollen content increased. While C18:0 content of meatballs slightly decreased with pollen addition, C18:2n-6c, C18:3n-3, C20:5n-3, and PUFA contents increased. The PUFA/saturated fatty acids (P/S) ratio increased from 0.05 in the control to 0.09 in meatballs with 6.0% pollen. The n-6/n-3 ratio decreased from 11.84 in the control to 3.65 in the meatballs with 6.0% pollen. The addition of pollen retarded the lipid oxidation and inhibited the bacterial growth in meatballs. The pH, redness, TBA value and total aerobic mesophilic bacteria, coliform bacteria and S. aureus counts values changed significantly during storage. The results suggest that bee pollen could be added to enhance the nutritional and storage quality of meatballs with minimal changes in composition and/or sensory properties.

발효쪽 추출물의 생리적 기능과 염색특성(제1보) (A Study on the Physiological Effects and Dyeing Properties of the Extract of Fermented (Part I))

  • The purpose of this study was to investigate the antimicrobial activity, antimutagenic and anticancer effects and dyeing properties of the fermented indigo extract. The physiological effects of natural color extracts from colorant plants(gardenia, beet and indigo) were studied. The methanol extract of indigo showed an inhibitory effect on the growth of E. coli and Staph. aureus, and also showed a strong antimicrobial effect on Trich. mentagrophytes compared to others. The methanol extract of indigo showed antimutagenic activities against aflatoxin B1(AFB1) in the Ames test using Salmonella typhimurium TA 100. The proliferation of Clone M-3 mouse melanoma cells and A431 human epidermoid carcinoma cells was inhibited by the methanol extract of indigo. So we decided to use natural indigo for dyeing the fabrics because of those effects. Dried indigo leaves were fermented at variouss temperature and the fermented indigo was reduced by using alkaline(NaOH, Ca(OH)2) and glucose to dye the fabrics. The values of K/S fermented indigo showed the highest value when it was fermented at 3$0^{\circ}C$. The indigo fermented at 3$0^{\circ}C$ had the greatest number of total bacterial counts and we identified one of the main microorganisms as Aspergillus niger. This microorganism was responsible for the indigo fermentation and accelerated indigo fermentation. So it can be supposed to reduce the fermentation period of indigo by inoculating Aspergillus niger into the indigo leaves at 3$0^{\circ}C$.

Physicochemical and microbial characteristics of domestic commercial semi solid type yogurt

  • Yogurt is a food produced by bacterial fermentation of milk and the bacteria used to make it are known as "yogurt cultures". Most of them belong to probiotics such as Lactobacillus delbrueckii subsp. bulgaricus and Streptococcus thermophilus bacteria. Domestic fermented milk market is increasing and about 30 companies are producing yogurt. The purpose of this study was to analyze the quality characteristics of domestic commercial semisolid type yogurt. We collected 20 types of commercial yogurt at local markets. Physicochemical properties including pH, sugar content, acidity, viscosity and microbial characteristics of lactic acid bacteria counts were measured. The yogurt showed pH 4.5, 7.4~18.1% of sugar contents, 0.6~1.3% of total acids and 282~748 cP of viscosities. In the microorganism populations, lactic acid bacteria count were 6.5~11.5 Log CFU/mL and anaerobic lactic acid bacteria count were 7.2 ~ 11.1 Log CFU/mL. The quality characteristics were different depending on the constituents of the sample and the microorganisms used. These results are related to the quality characteristics of yogurts which are useful information about identifying new trends in domestic fermented milk industry.

성남시 무료 노인 급식에서 제공되는 고구마줄기무침의 미생물학적 위해 분석 (Microbiological Hazard Analysis for Seasoned Sweet Potato Stems in a Free Meal service Operation for the Elderly in Sungnam)

  • In this study, we investigated the application of HACCP for a prepared side dish of sweet potato stems, within a free meal service system for the elderly in the Sungnam area. Total bacterial counts (TBC) and levels of coliforms and Esherichia coli (E.coli) were analyzed through an eight step cooking process. The TBCs of the raw samples ranged between 3.30 and 1.37${\times}10^4$ CFU/g per 100 cm$^2$ The trimmed, blanched, and drained sweet potato stems showed a mean TBC value of 1.37${\times}10^4$ CFU/g, and the level of coliforms was 1.48${\times}10^3$ CFU/g. Among the eight samples, however, after stir-frying and serving, the TBC decreased to a standard satisfactory level, and a coliforms and E. coli were not found. A five step process was used and samples were taken to check the microbial quality of the cook and cooking equipment. Here we tested for TBC, coliforms, Staphylococcus aureus (S. aureus), and Salmonella. Specifically, the TBC and number of coliforms were examined on the cook's hand's, cutting board, and knife, as they represented hazards for cross-contamination. The three inspection steps of preparation of the trimmed, rinsed, blanched, and drained sweet potato stems, cook's' hands, cutting board and knife were all considered CCPs, and a manual of cooking process management was established to improve the risk factors in this study. In conclusion, this study reinforces that microbiological analysis is as a valuable tool for checking what points and stages of the cooking process must be controlled.

Effect of Modified Atmosphere Packaging and Vacuum Packaging on Quality Characteristics of Low Grade Beef during Cold Storage

  • Many studies have been carried out with respect to packaging methods and temperature conditions of beef. However, the effects of packaging methods and temperature conditions on the quality characteristics have not been extensively studied in low-grade beef. Low-grade beef samples were divided into 3 groups (C: ziplock bag packaging, T1: vacuum packaging, and T2: modified atmosphere packaging (MAP), $CO_2/N_2$ = 3:7) and samples were stored at $4^{\circ}C$ for 21 days. The water-holding capacity (WHC) was significantly lower in T1 than in the other samples up to 14 days of storage. The thiobarbituric acid reactive substances and volatile basic nitrogen values were significantly lower in T1 and T2 than in C after 7 to 14 days of storage. The total bacterial counts were significantly lower in T1 and T2 than in C after 14 days of storage. In a sensory evaluation, tenderness and overall acceptability were significantly higher in T1 and T2 than in C at the end of the storage period (21 days). We propose that the MAP method can improve beef quality characteristics of low-grade beef during cold storage. However, the beneficial effects did not outweigh the cost increase to implement MAP.

Quality of Irradiated Plain Yogurt during Storage at Different Temperatures

  • To develop a safer yogurt for immuno-compromised or allergy patients and to extend shelf-life, a plain yogurt was irradiated with doses of 0, 1, 3, 5, and 10 kGy using a gamma ray and the chemical and microbiological quality and allergenicity change were investigated. There was no difference in the content of protein, total solid, and amino acids of the plain yogurt by irradiation treatment and different storage temperatures (4, 20, and $35^{\circ}C$). The lactic acid bacterial counts of irradiated plain yogurt had approximately 3-decimal reduction at 3 kGy, and no viable cell at 10 kGy regardless of storage time and temperature. The binding ability of rabbit antiserum to milk proteins in irradiated plain yogurt showed that 10 kGy of irradiation produced significantly higher binding ability than other treatments. Sensory evaluation indicated that only appearance of the plain yogurt irradiated at 3 kGy or higher had a lower value than the non-irradiated control when stored at $20^{\circ}C$. Results suggest that irradiation of plain yogurt does not significantly affect the chemical and sensory quality of plain yogurt, but can extend the shelf-life, possibly reduce allergenicity, and provide a safer product.

유통 중인 패류 단순가공품의 세균학적 위해요소 분석 (Bacteriological Hazard Analysis in Minimally Processed Shellfish Products Purchased from Korean Seafood Retail Outlets)

  • The purpose of this study was to assess bacteriological hazards in the following 7 kinds of minimally processed shellfish products purchased from Korean seafood retail outlets: raw oysters stored with seawater in polyethylene bags, frozen oysters, raw mussels, frozen mussels, boiled mussels, raw short-necked clam, and frozen short-necked clams, obtained from Korean seafood retail outlets. The total coliform and Escherichia coli counts determined in all samples were detected below regulation limit of the Korean government guidelines (Food Code). In addition, the high-risk bacterial pathogens, Listeria monocytogenes, Salmonella spp., and E. coli O157:H7 were not detected in any samples. Low-risk pathogens such as Staphylococcus aureus and Vibrio parahaemolyticus were detected at levels above $1.0{\times}10^2$ colony forming unit (CFU)/g in some minimally processed shellfish products. Notably, S. aureus was detected in all samples. Raw oysters stored with seawater in polyethylene bags, frozen oysters, and boiled blue mussels are commonly ingested without heating, and therefore these minimally processed shellfish products pose bacteriological hazards. The detection of S. aureus in all shellfish products, an organism indicative of poor personal hygiene, which can grow and multiply during distribution, indicates the necessity of improving the food safety of minimally processed shellfish products.

Effect of direct-fed microbials on culturable gut microbiotas in broiler chickens: a meta-analysis of controlled trials

  • Objective: This meta-analysis was conducted to evaluate the overall effect of direct-fed microbial (DFM) or probiotic supplementation on the log concentrations of culturable gut microbiota in broiler chickens. Methods: Relevant studies were collected from PubMed, SCOPUS, Poultry Science Journal, and Google Scholar. The studies included controlled trials using DFM supplementation in broiler chickens and reporting log concentrations of the culturable gut microbiota. The overall effect of DFM supplementation was determined using standardized mean difference (SMD) with a random-effects model. Subgroups were analyzed to identify pre-specified characteristics possibly associated with the heterogeneity of the results. Risk of bias and publication bias were assessed. Results: Eighteen taxa of the culturable gut microbiota were identified from 42 studies. The overall effect of DFM supplementation on the log concentrations of all 18 taxa did not differ significantly from the controls (SMD = -0.06, 95% confidence interval [-0.16, 0.04], p = 0.228, $I^2=85%$, n = 699 comparisons), but the 18 taxa could be further classified into three categories by the direction of the effect size: taxa whose log concentrations did not differ significantly from the controls (category 1), taxa whose log concentrations increased significantly with DFM supplementation (category 2), and taxa whose log concentrations decreased significantly with DFM supplementation (category 3). Category 1 comprised nine taxa, including total bacterial counts. Category 2 comprised four taxa: Bacillus, Bifidobacterium, Clostridium butyricum, and Lactobacillus. Category 3 comprised five taxa: Clostridium perfringens, coliforms, Escherichia coli, Enterococcus, and Salmonella. Some characteristics identified by the subgroup analysis were associated with result heterogeneity. Most studies, however, were present with unclear risk of bias. Publication bias was also identified. Conclusion: DFM supplementation increased the concentrations of some beneficial bacteria (e.g. Bifidobacterium and Lactobacillus) and decreased those of some detrimental bacteria (e.g. Clostridium perfringens and Salmonella) in the guts of broiler chickens.

빵 부패미생물에 대한 녹차의 항균작용 (Antimicrobial Activity of Green Tea against Putrefactive Microorganism in Steamed Bread)

  • 빵 부패미 생물에 대한 녹차추출물의 항균활성을 조사하고 녹차가루를 첨가한 찐빵의 저장 중 항균효과를 알아보고자 하였다. 사용된 세균 3종류인 Bacillus subtilis ARCC 6633, Bacillus pumilus KCTC 3348, Bacillus cereus IFO 12113과 곰팡이 Aspergillus niger KCCM l1239 모두 GTE 1% 이상에서 우수한 항균력을 가지고 있으며, 생육이 억제됨을 확인하였다. GTE의 열 안정성은 넓은 온도 범위(50~20$0^{\circ}C$)에서 열을 가하지 않았을 때와 동일하게 항균력이 유지되었고, pH 사용범위 (4~9)에 관계없이 동일한 환으로 항균력을 나타내었다. 녹차를 첨가한 찐빵을 $25^{\circ}C$에 저장하면서 총 곰팡이 수와 총 세균 수를 측정한 결과 대조군, GTP 1%, 3% 첨가군 모두 저장 3일째에 곰팡이가 나타났으나 GTP 5% 첨가군에서는 저장 3일째에 나타났다. 총 세균 수는 저장 3일 이후 $10^{7}$ CFU/g을 넘어 부패가 진행 중이었으나 GTP 3%는 5일째에, GTP 5% 첨가군은 7일째에 $10^{7}$ CFU/g을 나타났다. 그러므로 찐빵에서의 녹차의 항균 효과에 따른 저장기간 연장은 녹차가루 첨가량 5% 이상의 수준에서 가능하리라고 추측된다.
