An experiment was conducted to study the effect of soluble protein supplements on concentration of soluble non-ammonia nitrogen (SNAN) in the liquid phase of ruminal (RD) and omasal digesta (OD) of Korean native steers, and to investigate diurnal pattern in SNAN concentration in RD and OD. Three ruminally cannulated Korean native steers in a $3{\times}3$ Latin square design consumed a basal diet of rice straw and corn-based concentrate (control), and that supplemented (kg/d DM basis) with intact casein (0.24; IC) or acid hydrolyzed casein (0.46; AHC). Ruminal digesta was sampled using a vacuum pump, whereas OD was collected using an omasal sampling system at 2.0 h intervals after a morning feeding. The SNAN fractions (free amino acid (AA), peptide and soluble protein) in RD and OD were assessed using the ninhydrin assay. Concentrations of free AA and total SNAN in RD were significantly (p<0.05) lower than those in OD. Although free AA concentration was relatively high, mean peptide was quantitatively the most important fraction of total SNAN in both RD and OD, indicating that degradation of peptide to AA rather than hydrolysis of soluble protein to peptide or deamination may be the most limiting step in rumen proteolysis of Korean native steers. Diurnal variation in peptide concentration in OD for the soluble protein supplemented diets during the feeding cycle peaked 2 h post-feeding and decreased thereafter whereas that for the control was relatively constant during the entire feeding cycle. Diurnal variation in peptide concentration was rather similar between RD and OD.
This study performed verification experiment for the removal of total nitrogen in sewage from a Town M village sewage treatment plant using YPNR processes. The total nitrogen discharged after the denitrification process was maintained at a level of 8-15 mg/L, which results in the total nitrogen removal efficiency above 68% on average. The total nitrogen components in discharged water consisted of 16% of ammonia nitrogen, 6% of nitrite nitrogen, and 77% of nitrate nitrogen, which reaches a 95% nitrification efficiency. Hence, the YPNR advanced treatment process used in this study can be successfully applied to sewage treatment.

It was investigated whether the removal of nitrogen ions included livestock wastewater were increased by increasing the reaction time of livestock wastewater and microbubbles with catalyst. For this study, the nitrogen reduction system using microbubbles with catalyst was used. The two reactors were consecutively arranged, and the second reactor (Step 2) was located to next the first reactor (Step 1). Each reactor was reacted for 2 hours and air or oxygen as oxidant was fed into the reactor during operation before microbubble device. When oxygen was used, ammonia nitrogen was removed each 18.3% and 52.8% during 2 (only step 1) and 4 (step 1 and step 2) hours reactions. This value was higher than that of when air was fed. When oxygen was used, the longer the reaction time, the ammonia nitrogen removal was higher. The longer the reaction time, the higher the nitrite and nitrate was also removed such as ammonia nitrogen. Also this system was examined whether organic matter removal is effective. The total chemical oxygen demand (TCOD) removal was higher than the soluble chemical oxygen demand (SCOD). Some materials among causing substances COD were difficult to decompose biologically. Therefore, it means that it will be easy to operate the biological processes following step and reduce the concentration of organic contaminants in effluent.
Principal aim of this study is to suggest the statistical equation model which can predict an amount of ammonia emission according to the degree of compost maturity during composting livestock manure. Composting process was classified with intial, midterm and final phase based on germination index of compost samples. Total Kjeldahl nitrogen(TKN) and organic matter(OM) were selected as the independent variables available to contribute to ammonia emission from composting pile. Ammonia concentration measured in the samples taken at the intial phase was about 10ppm, sharply increased to 50ppm at the midterm phase, and gradually decreased to about 10ppm. The contents of Total Kjeldahl nitrogen and organic matter through whole composting period were ranged from 0.6 to 1.2% and from 30 to 40%, respectively, were reduced slightly at the midterm phase, but generally showed no constant fluctuation pattern. In estimating ammonia emission with application of the statistical equation model, the coefficients of independent variables at the midterm phase when an average concentration of ammonia was highest showed a relatively high values whereas those at the initial phase when an that of ammonia was lowest indicated a relatively low values. However, no statistical significance was found in the coefficients of independent variables and the equation model. Additionally, the further research, which can include the considerable analysis data with more samples taken than this study, is needed in order to suggest the statistically significant equation model available to predict ammonia emission during composting process.
Ruminal protozoa, especially entodiniomorphs, engulf other members of the rumen microbiome in large numbers; and they release oligopeptides and amino acids, which can be fermented to ammonia and volatile fatty acids (VFAs) by amino acid-fermenting bacteria (AAFB). Studies using defaunated (protozoa-free) sheep have demonstrated that ruminal protozoa considerably increase intraruminal nitrogen recycling but decrease nitrogen utilization efficiency in ruminants. However, direct interactions between ruminal protozoa and AAFB have not been demonstrated because of their inability to establish axenic cultures of any ruminal protozoan. Thus, this study was performed to evaluate the interaction between Entodinium caudatum, which is the most predominant rumen ciliate species, and an AAFB consortium in terms of feed degradation and ammonia production along with the microbial population shift of select bacterial species (Prevotella ruminicola, Clostridium aminophilum, and Peptostreptococcus anaerobius). From an Ent. caudatum culture that had been maintained by daily feeding and transfers every 3 or 4 days, the bacteria and methanogens loosely associated with Ent. caudatum cells were removed by filtration and washing. An AAFB consortium was established by repeated transfers and enrichment with casamino acids as the sole substrate. The cultures of Ent. caudatum alone (Ec) and AAFB alone (AAFB) and the co-culture of Ent. caudatum and AAFB (Ec + AAFB) were set up in three replicates and incubated at 39℃ for 72 h. The digestibility of dry matter (DM) and fiber (NDF), VFA profiles, ammonia concentrations, pH, and microscopic counts of Ent. caudatum were compared among the three cultures. The co-culture of AAFB and Ent. caudatum enhanced DM degradation, VFA production, and Ent. caudatum cell counts; conversely, it decreased acetate: propionate ratio although the total bacterial abundance was similar between Ec and the Ec + AAFB co-culture after 24 h incubation. The ammonia production and relative abundance of C. aminophilum and P. anaerobius did not differ between AAFB alone and the Ec + AAFB co-culture. Our results indicate that Ent. caudatum and AAFB could have a mutualistic interaction that benefited each other, but their interactions were complex and might not increase ammoniagenesis. Further research should examine how such interactions affect the population dynamics of AAFB.
Rumen bacterial amino acids in sheep on urea diet were monitored to assess a possible change in amino acid synthesis as a long term response to high rumen ammonia environment. A sheep was fed a semipurified diet with soybean meal, followed by a diet with urea as a main nitrogen source. Mixed rumen bacteria were harvested from ruminal fluid taken 3 h after feeding (twice in soybean meal feeding and 6 times in urea feeding) and fractionated as cell wall, proteins and protein-free cell supernatant of monitor amino acids in each fraction. Ruminal ammonia concentration at the sampling ranged from 5.7 to 39.5 mgN/dl. Cell wall and protein fractions of mixed rumen bacteria were stable in their amino acid composition regardless of nitrogen sources of diet and the feeding duration. However, protein-free cell supernatant fraction showed a higher alanine proportion with urea feeding (18.6 and 28.2 molar % of alanine for samples from sheep fed soybean meal and urea, respectively) and its duration (20.6 and 32.9 molar % for samples from sheep on urea diet for 1 and 65 days, respectively). Total free amino acid level of bacteria was depressed in the initial period of urea feeding but restored on 65th day of the feeding. These results suggest that an alanine synthesizing system may develop in rumen bacteria as urea feeding becomes longer.
Sudax fodder (Sorghum sudanense ${\times}$ Sorhum vulgare) was ensiled in laboratory silos with or without 20, 30, or 40 percent broiler litter and 6 percent molasses with or without Candida yeast. The samples were analyzed for pH, lactic acid and nitrogen fractions at the start of the experiment and at 5 days interval, thereafter till 40 days. A sharp decline in pH and increase in lactic acid content was observed on fifth day of ensiling. Thereafter, the rate of pH decline decreased till 20 days and that of lactic acid increase till 25 days and the remained constant. Increasing levels of broiler litter had adverse effect on pH drop and lactic acid increase of silages. Total-N content of the silages had little variation throughout the ensiling period. A sharp decline in protein-N and increase in ammonia-N content was observed on day 5 of ensiling. Thereafter, the content of protein-N increased till 20 days and that of ammonia-N decreased till 15 days, but these changes were very small compared to that occurred during the first 5 days of ensiling. The level of broiler litter had inverse relationship with protein degradation and direct relationship with ammonia production. The yeast inoculum failed to produce any significant effect.

Ammonia microdiffusion method and colorimetric measurement are described for the nitrogen determination. The diffusion of ammonia could be successfully induced by using a microdiffusion cell. It is a simple and rapid technique, which is suitable for transforming the nitrogen in digests into $NH_4CI$ for the colorimetric N determination with ammonia color reagent. Above 99% of N recovery were obtained with microdiffusion up to 15 hours. The coloration method of collected $NH_4CI$ for the colorimetric N determination was also estabilshed with a scanning in U.V. spectrophotometer. Under the proposed coloration method (0.5 mL of sample digest, 4.0 mL of $H_2O$ and 0.5 mL of ammonia color reagent), a maximal absorbance was observed at 410 nm. The kinetic measurement of absorbance showed a high stability from 5 to 45 minutes after color development. Absorbance was directly proportional to the amount of $NH_4^+-N$ present. The microdiffusion-ammonia coloration method was successfully applied to the nitrogen determination in the forms of protein-N or total -N in plant tissue. Comparing with Kjeldahl distillation method, the values obtained with described method were slightly higher and more reliable.
The effect of cecal ligation on the utilization of dietary [15N]urea in chickens fed 5 % protein diet plus urea were investigated. Nitrogen balance and utilization tended (P<0.01) to increase by cecal ligation. Total uric acid excretion was significantly decrease by (P<0.01) cecal ligation in chickens from origin of diet and urea (P<0.01). Excretion of ammonia was increased in chickens from origin of diet, where as it decreased in chickens an urea diet (P<0.01). Amount of urea nitrogen excretion from origin of urea was significantly decrease (P<0.01) by cecal ligation, but cecal ligated chicken fed 5% protein diet with urea showed 51.6% urea utilization. Result obtained in present study indicates that ceca is having beneficial role for urea utilization in chicken fed protein deficient diets, but ceca do not always positive role for nitrogen utilization.

The objective of this study was to prove the effect of pig slurry application with charcoal on nitrogen use efficiency (NUE), feed value and ammonia (NH3) emission from maize forage. The four treatments were applied: 1) non-pig slurry (only water as a control), 2) only pig slurry application (PS), 3) pig slurry application with large particle charcoal (LC), 4) pig slurry application with small particle charcoal (SC). The pig slurry was applied at a rate of 150 kg N ha-1, and the charcoal was applied at a rate of 300 kg ha-1 regardless of the size. To determine the feed value of maize, crude protein, dry matter intake, digestible dry matter, total digestible nutrient, and relative feed value were investigated. All feed value was increased by charcoal treatment compared to water and PS treatment. Also, the NUE for plant N was significantly higher in charcoal treatments (LC and SC) compared to PS treatment. On the other hand, there is no significant difference for feed value and NUE between LC and SC. The NH3 emission was significantly reduced 15.2% and 27.9% by LC and SC, respectively, compared to PS. Especially, SC significantly decreased NH3 emission by 15% compared to LC. The present study clearly showed that charcoal application exhibited positive potential in nitrogen use efficiency, feed value and reducing N losses through NH3 emission.
