트랩식물로써 토마토에 대한 가지의 담배가루이 유인효과 (Effects of Egg-plant as a Trap Plant Attracting Bemisia tabaci Genn. (Hemiptera: Aleyrodidae) Adults Available on Tomato Greenhouses)

  • 본 연구는 토마토에 대하여 가지가 트랩식물로써 담배가루이를 효과적으로 유인할 수 있을지에 대한 잠재성을 평가하였다. 큰망초, 가지, 오이, 토마토에 대한 담배가루이의 선호성을 비교하기 위하여 선택성 조사가 수행되었다. 담배가루이 성충은 후각적인 자극보다는 시각적인 자극에 더 민감함을 보여주었고 큰망초보다는 오이와 가지를 더 선호하였다. 가지와 오이의 담배가루이 유인률은 각각 82.3%와 82.5% 였다. 가지는 오이보다 관리측면에서 더 쉽기 때문에 우리는 본 실험에서 오이를 제외하였다. 담배가루이 성충에 대한 가지의 유인률은 토마토의 초장과 같거나 더 클 때 90% 이상 이었다.

  • Gray leaf spot of tomato was observed on tomato plants growing in a middle of a pepper field where pepper plants were badly infected with gray leaf spot. Both Stemphylium solani and S. lycopersici were isolated from the lesion. Both species were highly pathogenic on tomato and pepper. This is the first report in Korea of occurrence of gray leaf spot of tomato caused by Stemphylium solani Weber.

  • Black dot root rot occurred severely in greenhouse tomatoes in jangseong area of Korea in April, 1996. The causal fungus of the disease was identified as Colletotrichum coccodes based on the morphological and cultural characteristics. Pathogenicity tests revealed that isolates of the fungus were responsible for the disease, and two cultivars of cherry tomato were less susceptible to the disease than other cultivars of tomato and cherry tomato tested.

Alternaria Spots in Tomato Leaves Differently Delayed by Four Plant Essential Oil Vapours

  • Alternaria leaf spot disease has been a concern during a tomato production in greenhouse. In vitro antifungal activities of vapours of four plant essential oils, cinnamon oil, fennel oil, origanum oil and thyme oil, were investigated during in vitro conidial germination and mycelial growth of Alternaria alternata causing the tomato leaf spots to find eco-friendly alternatives for chemical fungicides. The four plant essential oils showed different antifungal activities against in vitro conidial germination of A. alternata in dose-dependent manners, and cinnamon oil vapour was most effective to suppress the conidial germination. The four plant essential oils showed similar antifungal activities against the in vitro mycelial growth of A. alternata in dose-dependent manners, but low doses of thyme oil vapour slightly increased in vitro mycelial growth of A. alternata. Necrotic lesions on the A. alternata-inoculated tomato leaves were reduced differently depending on kinds and concentrations of plant essential oils. Delayed conidial germination and germ-tube elongation of A. alternata were found on the tomato leaves treated with cinnamon oil and origanum oil vapours at 6 hpi. These results suggest that volatiles from cinnamon oil and origanum oil can be provided as alternatives to manage Alternaria leaf spot during the tomato production eco-friendly.

Expression of Arabidiopsis CAX4 in tomato fruits increases calcium level with no accumulation of other metallic cations

  • We generated transgenic tomato plants with Arabidopsis thaliana $H^+$/cation exchanger gene (C4X4) by Agrobactrium-mediated transformation. We confirmed transgene copy number and transcription by Southern and Northern blot analyses. The intact CAX4-expressing tomato (Lycopersicon esculentum) fruits contained 63-71% more calcium ($Ca^{2+}$) than wild-type fruits. Moreover, ectopic expression of C4X4 in tomato fruits did not show any significant increase of the four kinds of metallic cations analyzed ($Mg^{2+}$, $Fe^{2+}$, $Mn^{2+}$, and $Cu^{2+})$. The C4X4-expressing tomato plants including their fruits did not show any morphological alternations during whole growth period. These results suggest the enhanced Ca-substrate specificity of CAX4 exchanger in tomato. Therefore, intact CAX4 exchanger can be a useful tool for $Ca^{2+}$ nutrient enrichment of tomato fruits with reduced accumulation of undesirable cations.

  • Virus disease occurred up to 62% in average in the greenhouse production of table tomato Seokwang in Suwon, Korea. From symptomatic transition of the labeled tomatoes, two different symptoms, mosaic and bud necrosis, were developed independently. Cucumber mosaic virus necrosis strain (CMV-N) was isolated from table tomato showing bud necrosis symptoms. The isolate caused the bud necrosis on four tomato cultivars and locally infected Chenopodium spp. and Vicia faba by mechanical inculation. The 5th RNA segment, satellite RNA, was identified from CMV-N-infected plants by dsRNA analysis. Crystals of virus particles were observed in cytosols and vacuoles. The virus particles of CMV-N presented abundantly in xylem vessel.

기계시각장치에 의한 토마토 작물의 병해엽 검출 (Machine Vision Based Detection of Disease Damaged Leave of Tomato Plants in a Greenhouse)

  • Machine vision system was used for analyzing leaf color disorders of tomato plants in a greenhouse. From the day when a few leave of tomato plants had started to wither, a series of images were captured by 4 times during 14 days. Among several color image spaces, Saturation frame in HSI color space was adequate to eliminate a background and Hue frame was good to detect infected disease area and tomato fruits. The processed image ($G{\sqcup}b^*$ image) by OR operation between G frame in RGB color space and $b^*$ frame in $La^*b^*$ color space was useful for image segmentation of a plant canopy area. This study calculated a ratio of the infected area to the plant canopy and manually analyzed leaf color disorders through an image segmentation for Hue frame of a tomato plant image. For automatically analyzing plant leave disease, this study selected twenty-seven color patches on the calibration bars as the corresponding to leaf color disorders. These selected color patches could represent 97% of the infected area analyzed by the manual method. Using only ten color patches among twenty-seven ones could represent over 85% of the infected area. This paper showed a proposed machine vision system may be effective for evaluating various leaf color disorders of plants growing in a greenhouse.

Aspergillus terreus JF27 Promotes the Growth of Tomato Plants and Induces Resistance against Pseudomonas syringae pv. tomato

  • Certain beneficial microorganisms isolated from rhizosphere soil promote plant growth and induce resistance to a wide variety of plant pathogens. We obtained 49 fungal isolates from the rhizosphere soil of paprika plants, and selected 18 of these isolates that did not inhibit tomato seed germination for further investigation. Based on a seed germination assay, we selected four isolates for further plant tests. Treatment of seeds with isolate JF27 promoted plant growth in pot tests, and suppressed bacterial speck disease caused by Pseudomonas syringae pathovar (pv.) tomato DC3000. Furthermore, expression of the pathogenesis-related 1 (PR1) gene was higher in the leaves of tomato plants grown from seeds treated with JF27; expression remained at a consistently higher level than in the control plants for 12 h after pathogen infection. The phylogenetic analysis of a partial internal transcribed spacer sequence and the b-tubulin gene identified isolate JF27 as Aspergillus terreus. Taken together, these results suggest that A. terreus JF27 has potential as a growth promoter and could be used to control bacterial speck disease by inducing resistance in tomato plants.

Expression of dehydration responsive element-binding protein-3 (DREB3) under different abiotic stresses in tomato

  • We investigated the expression pattern of dehydration responsive element-binding protein-3 in tomato under different abiotic stresses. Full length LeDREB3 cDNA was isolated from tomato plant, followed by phylogenetic analysis based on deduced amino acid sequences that revealed significant sequence similarity to DREB proteins belonging to diverse families of plant species. Southern blot analysis showed duplicate copies of LeDREB3 in the tomato genome while organ-specific expression profiling indicated constitutive expression of LeDREB3 in all tested organs, which was particularly strong in flower. LeDREB3 expression was significantly induced by Nacl, drought, low temperature and $H_2O_2$. Moreover, LeDREB3 was slightly regulated by treatment with ABA and MV. These observations suggest that the LeDREB3 gene may be involved in the response of the tomato plant to stress.

  • Tomato aspermy virus(TAV-B)는 두 개의 다른 CMV균주 D 및 K에서 분리한 satellite RNA의 증식 및 encapsidation에 대하여 helper virus의 역할을 하였다. 이 두 satellite RNA는 역시 ??? system에서 보이는 바와 같이 TAV가 감염된 담배에서 TAV 병징을 크게 감소시켰으며, 이는 감염된 식물내의 Virus 함량의 감소와 관계가 있었다. 또한 CMV가 감염된 토마토에서 lethal necrosis를 유발시킴을 볼 수 있었다.
