Assessment of Rhizosphere Microbial Community Structure in Tomato Plants after Inoculation of Bacillus Species for Inducing Tolerance to Salinity (토마토에 염류 내성을 유도하는 바실러스 균주 처리 후 근권 미생물 군집 구조 연구)

  • BACKGROUND: Soil salinity causes reduction of crop productivity. Rhizosphere microbes have metabolic capabilities and ability to adaptation of plants to biotic and abiotic stresses. Plant growth-promoting bacteria (PGPB) could play a role as elicitors for inducing tolerance to stresses in plants by affecting resident microorganism in soil. This study was conducted to demonstrate the effect of selected strains on rhizosphere microbial community under salinity stress. METHODS AND RESULTS: The experiments were conducted in tomato plants in pots containing field soil. Bacterial suspension was inoculated into three-week-old tomato plants, one week after inoculation, and -1,000 kPa-balanced salinity stress was imposed. The physiological and biochemical attributes of plant under salt stress were monitored by evaluating pigment, malondialdehyde (MDA), proline, soil pH, electrical conductivity (EC) and ion concentrations. To demonstrate the effect of selected Bacillus strains on rhizosphere microbial community, soil microbial diversity and abundance were evaluated with Illumina MiSeq sequencing, and primer sets of 341F/805R and ITS3/ITS4 were used for bacterial and fungal communities, respectively. As a result, when the bacterial strains were inoculated and then salinity stress was imposed, the inoculation decreases the stress susceptibility including reduction in lipid peroxidation, enhanced pigmentation and proline accumulation which subsequently resulted in better plant growth. However, bacterial inoculations did not affect diversity (observed OTUs, ACE, Chao1 and Shannon) and structure (principle coordinate analysis) of microbial communities under salinity stress. Furthermore, relative abundance in microbial communities had no significant difference between bacterial treated- and untreated-soils under salinity stress. CONCLUSION: Inoculation of Bacillus strains could affect plant responses and soil pH of tomato plants under salinity stress, whereas microbial diversity and abundance had no significant difference by the bacterial treatments. These findings demonstrated that Bacillus strains could alleviate plant's salinity damages by regulating pigments, proline, and MDA contents without significant changes of microbial community in tomato plants, and can be used as effective biostimulators against salinity stress for sustainable agriculture.

Effect of Fermented Soy Bean Extract Containing Herbal Medicines (Godjang) on Blood Glucose Levels and Histomorphology in Streptozotocin-induced Diabetic Rat (한약재 함유 대두발효 추출물이 streptozotocin으로 유발된 당뇨 흰쥐의 혈당 및 조직형태학적 변화에 미치는 영향)

  • Objective : This study was designed to investigate anti-diabetic effects of fermented soy bean extract with herbal medicines (Godjang) in diabetic rat models induced by streptozotocin (STZ) injection. Method : Changes in body weight, drinking water, and food intake were observed for 4 weeks before and after induction of diabetes mellitus in rats. The anti-diabetic capacity of Godjang was analyzed by fasting blood glucose (FBG) every week. Also, after 4 weeks of administration, the oral glucose tolerance test (OGTT) was performed, and then blood levels of insulin were checked. And serum levels of total cholesterol and triglycerides were determined. Histomorphological changes of liver, kidney and pancreatic tissues were also observed in STZ-induced diabetic rats and Godjang administered rats. Result : In Godjang administered group, body weight and water intake were more lower than that of STZ-induced diabetic rats. FBG was decreased in the Godjang administered group than STZ-induced diabetic group. According to OGTT, blood glucose levels at 30 minutes and 60 minutes significantly decreased in Godjang administered group than in STZ-induced diabetic control group. Administration of Godjang extract for 4W significantly decreased levels of serum glucose, total cholesterol (TC), triglycerides (TG) in diabetic rats. In histomorphological analysis of kidney, liver, Godjang administrated groups showed the inhibition of pathological damage. Conclusion : These results suggest that Godjang extract has an anti-diabetic action through decrease in serum glucose, TC, TG levels and recovery of the morphological changes in kidney and liver in STZ-induced diabetic rats.

  • Current military command information system uses electronic equipment a lot on which semiconductor chip is attached. It seems its' importance will increase more with latest information communication technology developing. Electronic equipment which uses electricity contains regular tolerance to high output electric signal. And EMC specification is the standardized of this electronic equipment's tolerance. On the other hand, the Institute of Atomic Energy Research has ever declared that high output electromagnetic pulse(EMP) will be broken out within the radius of 170Km when 10kt nuclear explosion occurs at an altitude of 40Km above Seoul. Then, the region suffer from the damage of most electronic equipments. Therefore, the norm to protect the influences in that case is defined by EMP protection specification. Most common electronic equipments meet the EMC norm, but there is no way to check whether they meet the EMP norm or not. That is because it is difficult to check whether they meet EMP protection norm and is on the matter of cost. Except inevitable cases, there is no review of checking whether they meet the norm or not. Considering the above, in this research, we speculate about the measures to improve military EMP protection ability by analyzing the EMC-EMP correlation and checking the EMP protection ability of general electronic equipment through the analysis.

The Effects of Cadmium on Seed Germination and Growth of Sunflower and Rape (카드뮴이 해바라기와 유채 발아 및 성장에 미치는 영향)

  • Sunflower (Sunking4505) and Rape (Sunmang) are oil-seeds containing high oleic acid, and these are used for the production materials of bio-diesel and applying for phytoremediation. The effect of cadmium on germination rate and the growth of the plants is evaluated. Object seeds were placed in Cd (0, 1.5, 4, 12, 30, 60, 100, 150, 300, 500 mg/L) solutions for seven days, and germination rate, root length, shoot length, seedling length, and dry weight were observed. $IC_{50}$, seedling vigor index, and tolerance indices were computed, and data were statistically analyzed by Analysis of Variance (ANOVA). Germination rate as well as root, shoot, and seedling length decreased as the cadmium concentration increased except dry weight. The $IC_{50}$ of sunflower and rape are 112 and 10 mg-Cd/L, respectively. Only one of the sunflower seeds is germinated at 500 mg-Cd/L whereas rape seeds are not germinated more than 150 mg-Cd/L solution. Root has higher cadmium sensitivity than shoot, and sunflower has higher germination rate, growth, and seedling vigor index than rape. In case of tolerance indices, sunflower has lower value than rape at relatively low concentration, but has higher value at high concentration.

  • After signing the WHO FCTC in 2003, South Korea ratified the FCTC in 2005. This study was conducted to provide data on toxic constituents that can be used as useful information for the level of exposure to Korean smokers. Emissions data from five brands of cigarettes were tested under the ISO and "Canadian Intense (HCI)" smoking regimes, respectively. We conducted an analysis of 25 compounds containing nicotine, tar, carbonyls, phenolics, volatile organic compounds (VOCs), and semi-VOC cigarette smoke. Tar and nicotine showed levels of 4.3 to 5.8 mg/cig and 0.4 to 0.5 mg/cig, respectively, which are within the range of tolerance presented in ISO 8243. In the case of carbonyls, formaldehyde was detected within a range of 8.2 to $14.3{\mu}g/cig$, and acetaldehyde was present within a range of 224.7 to $327.2{\mu}g/cig$ under the ISO smoke regime. Crotonaldehyde was not detected under the ISO regime, and all of the carbonyls showed values 2.3 to 4.5 times higher under the HCI regime than those under the ISO regime. Catechol, which showed a level of 47.0 to $80.5{\mu}g/cig$ under the ISO regime and 117.5 to $184.7{\mu}g/cig$ under the HCI regime, was the highest constituent among the phenols. The amount of isoprene was 91.7 to $158.3{\mu}g/cig$ under the ISO regime and 221.0 to 377.0 under the HCI regime. To summarize, most of the constituents showed a tendency to be detected at levels 2 to 4 times higher under the HCI regime than under the ISO regime. Above all, these results represent the first analysis in Korea from an independent institute of tobacco companies under accreditation of ISO 17025.

  • The gas hydrate exploration using seismic reflection data, the detection of BSR(Bottom Simulating Reflector) on the seismic section is the most important work flow because the BSR have been interpreted as being formed at the base of a gas hydrate zone. Usually, BSR has some dominant qualitative characteristics on seismic section i.e. Wavelet phase reversal compare to sea bottom signal, Parallel layer with sea bottom, Strong amplitude, Masking phenomenon above the BSR, Cross bedding with other geological layer. Even though a BSR can be selected on seismic section with these guidance, it is not enough to conform as being true BSR. Some other available methods for verifying the BSR with reliable analysis quantitatively i.e. Interval velocity analysis, AVO(Amplitude Variation with Offset)analysis etc. Usually, AVO analysis can be divided by three main parts. The first part is AVO analysis, the second is AVO modeling and the last is AVO inversion. AVO analysis is unique method for detecting the free gas zone on seismic section directly. Therefore it can be a kind of useful analysis method for discriminating true BSR, which might arise from an Possion ratio contrast between high velocity layer, partially hydrated sediment and low velocity layer, water saturated gas sediment. During the AVO interpretation, as the AVO response can be changed depend upon the water saturation ratio, it is confused to discriminate the AVO response of gas layer from dry layer. In that case, the AVO modeling is necessary to generate synthetic seismogram comparing with real data. It can be available to make conclusions from correspondence or lack of correspondence between the two seismograms. AVO inversion process is the method for driving a geological model by iterative operation that the result ing synthetic seismogram matches to real data seismogram wi thin some tolerance level. AVO inversion is a topic of current research and for now there is no general consensus on how the process should be done or even whether is valid for standard seismic data. Unfortunately, there are no well log data acquired from gas hydrate exploration area in Korea. Instead of that data, well log data and seismic data acquired from gas sand area located nearby the gas hydrate exploration area is used to AVO analysis, As the results of AVO modeling, type III AVO anomaly confirmed on the gas sand layer. The Castagna's equation constant value for estimating the S-wave velocity are evaluated as A=0.86190, B=-3845.14431 respectively and water saturation ratio is $50\%$. To calculate the reflection coefficient of synthetic seismogram, the Zoeppritz equation is used. For AVO inversion process, the dataset provided by Hampson-Rushell CO. is used.

The Fish Community and Habitat Analysis in the Bongseonsa Stream within Gwangneung Forest, South Korea (광릉숲 내 봉선사천의 어류군집 및 서식처 분석)

  • The present study is to understand the Bongseonsa stream in the National Arboretum fish fauna variation through comparison with historical data and to evaluate the stream health situated. We performed investigations over three times from April to September 2015. In the survey, 2,960 individuals which belonging to 22 species, 8 families were collected. Dominant species by number was Zacco platypus and subdominant species was Zacco koreanus. Seven Korean endemic species (Squalidus gracilis majimae, Microphysogobio yaluensis, Zacco koreanus, Koreocobitis rotundicausata, etc) were observed and showed a ratio of 36.4%. The community analysis revealed that the structure of fish community in the study sampling sites was instability in having dominance 0.79 (${\pm}0.15$), diverse 1.21 (${\pm}0.60$), evenness 0.58 (${\pm}0.15$) and species richness 1.49 (${\pm}0.83$). The values in the Qualitative Habitat Evaluation Index (QHEI) was averagely 122.9 (${\pm}44.8$) in the Bongseonsa stream and this was showed to have favorable habitat surroundings. As a result of tolerance guild analysis, the total number of sensitive species and intermediate species were higher than tolerant species. Analysis was divided into A and B two groups of fiducial 12.25% in Cluster analysis degree of similarity between study sampling sites. Fish Assessment Index (FAI) was rated A and B grade in Bongseonsa stream that stream health showed favorable. However Wangsuk stream as a urban stream rated C grade and analysed the lowest grade in the whole study sampling sites. There was high correlation beteween FAI and various indexes, dominance, diverse, evenness and sensitive species and intermediate species.

Comparative Studies on the Analysis of Pesticide Residues. in Rice, Barley and Wheat (주곡중의 잔류농약분석에 관한 연구)

  • Recenty, the pesticides Pollutions in connection with the maintenance of dietary life are poised as serious sanitary problems. Author have established an appropriate analysis method in this connection and, at the same time, analysed the levels of pesticides residues, such as organochloride pesticides and organic mercurials, contained in main cereals (rice, barley, and wheat) collected throughout the country. Using gas liquid chromatography method, comparative analyses were made of organochloride Pesticides with electron capture detector, and organic mercurials with electron capture detector and the Dithizone Method. As a result, the organic mercurials analysis using gas liquid chromatography is believed to hold out an especially good method for both in terms of its sensitivity and its Practical applications. The summary obtained from these results is as follows; The detectable limit of organochloride pesticides is $5\times10^{-10}$ grams and that of organic mecurials is $5\times10^{-9}$ grams. The detections using the Dithizone Method are difficult. The gas liquid chromatographic analysis of organic mercurials is very simple in its operation and high in its sensitivity, compare with the analysis using the Dithizone Method. Therefore, this analysis is expected to be a good method applicable to the pesticide residues analysis. The levels of pesticide residues contained in samples are very little for tolerance and, therefore, no problem i.: foreseen for eating. The appropriate conditions of gas liquid chromatographic analysis obtained from these results are expected very useful for the date establishing an officially authorized analysis method.

Study for Residue Analysis of Pinoxaden in Agricultural Commodities

  • BACKGROUND: Pinoxaden is the phenylpyrazoline herbicide developed by Syngenta Crop Protection, Inc. and marketed on 2006. The maximum residue levels for wheat and barley were set by import tolerance. Thus, Ministry of Food and Drug Safety (MFDS) official analytical method determining Pinoxaden residue was necessary in various food matrixes. Satisfaction of international guideline of CODEX (Codex Alimentarius Commission CAC/GL 40) and National Institute of Food and Drug Safety Evaluation-MFDS (2017) are additional pre-requirements for analytical method. In this study, liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was investigated to analyze residue of Pinoxaden (M4), which is defined as pesticide residue in Korea, in foods. METHODS AND RESULTS: Pinoxaden (M4) was extracted followed by acid digestion (2hr reflux with 1N HCl) and pH adjusting (pH 4-5 with 3% ammonium solution). To remove oil, additional clean-up step with hexane saturated with acetonitrile was required to high oil contained sample before purification. HLB cartridge and nylon syringe filter were used for purification. Then, samples were analyzed by LC-MS/MS using reserve phase column C18. Five agricultural group representative commodities (mandarin, potato, soybean, hulled rice, and red pepper) were used to verify the method in this study. The liner matrix-matched calibration curves were confirmed with coefficient of determination (r2) > 0.99 at calibration range 0.002-0.2 mg/kg. The limits of detection and quantitation were 0.004 and 0.01 mg/kg, respectively, which were suitable to apply Positive List System (PLS). Mean average accuracies of pinoxaden (M4) were shown to be 74.0-105.7%. The precision of pinoxaden and its metabolites were also shown less than 14.5% for all five samples. CONCLUSION: The method investigated in this study was suitable to CODEX (CAC/GL 40) and National Institute of Food and Drug Safety Evaluation-MFDS (2017) guideline for residue analysis. Thus, this method can be useful for determining the residue in various food matrixes in routine analysis.

A Development of 3D Modeling-based Survivability Analysis System for Armored Fighting Vehicle using Importance of Components (부품의 중요도를 활용한 3차원 전차 모델 기반 생존성 분석 시스템 개발)

  • The mission capability of tank depends on its survivability. The survivability is ability for protection and tolerance by damage from threats. To improve the survivability of tank, we need an effectiveness analysis for loss of components, and accomplish performance enhancement using the result of analysis. In this paper, we develop a survivability analysis system for tank based on the importance. The importance numerically represents weight of each component which consisting of whole tank, also the importance is basic method of quantitative survivability analysis. To do this, we assign weight values to each component of tank, compose a weight tree, apply the importance calculation equation, and analyze the survivability of tank. Also we develop the system that consists of component structuralization and weight value setting program and survivability analysis and visualization program, and evaluate the system using implemented 3D CAD models of components of tank. The developed system apply to arrangement components.
