Ginseng seedlings were transplanted in the perlite and grown with various $CO_2$ concentration for 14 days. The days for full emergence was decreased by 3 days at 6 to 8% $CO_2$ but increased above 13%. Stem length showed significant negative linear correlation with $CO_2$ concentration at 14 days but little difference at 64 days. Relative growth (%) to seedling at 64 days tended to increased till 8% $CO_2$ and decrease thereafter without significance. Seedlings showed 65% emergence with short stem (60%) under 22% $CO_2$ for 64 days. Seedlings were all sleeping under 35% $CO_2$ for 64 days. Above results indicates that $CO_2$ may be useful for growth control or post harvest storage of ginseng.
Transgenic tobacco plants expressing either a sweet potato anionic peroxidase (POD) (swpal) or neutral POD (swpnl) were irradiated by gamma radiation, and the gamma radiation-induced biochemical changes in antioxidant enzymes and plant growth inhibition were investigated at 30 days after treatment. Gamma radiation significantly inhibited the growth of all plants regardless of transgenic or nontransformed plants, showing a dose-dependent inhibition. In high dosage of 50 and 70 Gy, plant heights were severely retarded and new leaves does not emerged. No significant changes in antioxidant enzymes such as POD, superoxide dismutase and catalase were observed in all plants regardless of irradiation dosages ranging from 10 to 50 Gy. These results suggest that sweet potato PODs may be not involved in the protection against the oxidative stress induced by gamma radiation.
Among the 34 strains of Nicotinophiles selected in the previous experiments, strain NCT27 identified with Pseudomonas putida and strain NCT30 identified with Arthrobacter oxydans biotype nan thus were Investigated for optimization of growth conditions for nicotine degradation and other cultural characteristics. The compositions of optimized medium were to be following: $KH_2PO_4$ 2.Ogr, KCI 5.Ogr, $MgSO_4$.$7H_2O$ 20mg, $MnSO_4$.$6H_2O$ 0.2mg, $FeSO_4$.$7H_2O$ 1.Omg, Col$^{++}$ (Cobalt Acetate),2.O$\gamma$, N1$^{++}$ (NiSO4,6H2O) 0.5$\gamma$, and yeast extract 80mg per liter. The optimum initial concentrations of nicotine for growth were 0.4% for Pseudomonas and 0.1% for Arthrobacter, respectively. The optimum temperature and pH were 3$0^{\circ}C$ and 7.0 for both of strains. The pH of culture medium of Pseudomonas was changed from acidic condition to basic one in going from the logarithmic growth phase to the stationary growth phase. In contrast with Pseudomonas, it remained constant in case of Arthrobacter. The growth of Arthrobacter was completely inhibited in the nicotine concentration of 0.7&. However, Pseudomonas could grow even in the nicotine concentration of 1.0%. Moreover, it could grow successfully in the tobacco extract media as well as media containing carbon and nitrogen sources other than nicotine. The maximum rates of nicotine degradation were to be 1.22 gr./hr./liter for Pseudomonas and 0.186 gr./hr./liter for Arthrobacter, respectively.
Microbial antagonism between virulent and avirulent isolates of Pseudomonas solanacearum was studied in relation to the control of bacterial wilt of tobacco. In nutrient broth media or in soil, the avirulent isolate of P. solanacearum grew faster than did the virulent one. Inhibitory effect of avirulent isolate against growth of virulent one was negligible in mixed culture of the two isolates. The disease severity of bacterial wilt was significantly reduced when the roots of cultivar BY 4 susceptible to bacterial wilt was dipped in suspension of an avirulent isolate for 6 hours prior to transplanting to the soil infested with virulent bacteria. When the seedlings of tobacco were poured with the suspension of an avirulent isolate onto the soil in pre-planting pots 24 hours before ransplanting, there was a significant reduction in disease severity in the field. However, the reduction was noticed until early July, but after middle of July, no difference between the avirulent isolate-treated and non-treated plants was found in severity of the bacterial wilt.

This study was carried out to obtain basic information for gene manipulation in potent edible tobacco (Nicotiana tabacum cv. TI 516). N. tabacum cv. TI 516 is a plant for a possible candidate to use as an edible vaccine, since it contains a low level of nicotine. The effective plant regeneration system through leaf disc culture was achieved using a MS basal medium supplemented with 0.1 mg $1^{-1}$ NAA and 0.5 mg $1^{-1}$ BA. In order to transform the N. tabacum cv. TI 516 with the green fluorescent protein (GFP) gene, Agrobacterium tumefaciens LBA 4404 containing the GFP gene was used. Genomic PCR confirmed the integration of the GFP gene into nuclear genome of transgenic plants. Expression of the GFP gene was identified in callus, apical meristem and root tissue of transgenic N. tabacum cv. TI 516 plants using fluorescence microscopy. Western blot analysis revealed the expression of GFP protein in the transgenic edible tobacco plants. The amount of GFP protein detected in the transgenic tobacco plants was approximately 0.16% of the total soluble plant protein (TSP), which was determined by ELISA.

A total of about 300 fungal isolates from forest havitats were screened for inhibitors of tobacco mosaic virus (TMV) infection using its local lesion host, Nicotiana tabacum cv. Xanthi nc. Ine of the isolates, 14T, showed a strong activity against TMV infection, and was identified as an Apiocrea sp. based on its morphological characterstics. Rice was an optimum culture medium for its fermentation, and two antiviral compounds, KGT 141 and KGT 142, were resolved from the rice culture through column chromatography, TLC, and HPLC. By NMR and FAB-MS, the two compounds were identified as chrysospermins B (KGT 141) and D (KGT 142), both of which are peptaibols with 19-mer amino acids possessing an acetylated N-terminus and a hydroxy-amino acid (tryptophanol) at the C-terminus. Both compounds showed inhibitory activities against TMV infection, but chrysospermin D showed the stronger activity than chrysospermin B. The former of $100{\;}\mu\textrm{g}/ml$ and 54.7% at $10{\;}\mu\textrm{g}/ml$, respectively. Furthermore, the chrysospermins were highly cytotoxic toward cancer cell lines of PC-3 (prostrate) and K562 (leukemia), and inhibited growth of the Gram-positive bacteria tested, especially the plant pathogenic bacterium Corynebacterium lilium. To the best of our knowledge, this is the first report on the inhibition of plant virus infection by antimicrobial peptaibols.
Ground-based remote sensing can be used as one of the non-destructive, fast, and real-time diagnostic tools for predicting yield, biomass, and nitrogen stress during growing season. The objectives of this study were: 1) to assess biomass and nitrogen (N) status of tobacco (Nicotiana tabacum L.) plants under N stress using ground-based remote sensors; and 2) to evaluate the feasibility of spectral reflectance indices for estimating an application rate of N and predicting yield of tobacco. Dry weight (DW), N content, and N uptake at the 40th and 50th day after transplanting (DAT) were positively correlated with chlorophyll content and normalized difference vegetation indexes (NDVIs) from all sensors (P<0.01). Especially, Green NDVI (GNDVI) by spectroradiometer and Crop Circle-passive sensors were highly correlated with DW, N content and N uptake. The yield of tobacco was positively correlated with canopy reflectance indices measured at each growth stage (P<0.01). The regression of GNDVI by spectroradiometer on yield showed positively quadratic curve and explained about 90% for the variability of measured yield. The sufficiency index (SI) calculated from data/maximum value of GNDVI at the $40^{th}$ DAT ranged from 0.72 to 1.0 and showed the same positively quadratic regression with N application rate explaining 84% for the variability of N rate. These results suggest that use of reflectance indices measured with ground-based remote sensors may assist in determining application rate of fertilizer N at the critical season and estimating yield in mid-season.

Streptomyces spp. were isolated from rhizosphere in fallow lands. The Streptomyces spp. were identified as Streptomyces griseus (MSS181), Streptomyces griseoaurantiacus (MSS269), Streptomyces microflavus (MSS275), Streptomyces herbaricolor (MSS276) based on 16S rRNA gene sequences. Afterwards, cucumber, pepper, tobacco and tomato were drenched with the isolates at early growth stages and plant growth such as height and dry weight of plants was measured. By treatment of Streptomyce spp., plant height of cucumber was increased by 16-29% compared to the control, But there were no statistically significant differences in dry weight. When the same isolates were treated on chili-pepper, plant height and dry weight of chili-pepper were increased respectively by 10-19% and 19-25% compared to the control. The dry weight of tobacco and tomato were increased by 44-73% and 65-165%, respectively compared to the control. When antifungal activities of the isolates were tested against plant pathogenic fungi, Streptomyces microflavus (MSS275) effectively inhibited the mycelial growth of Phytophthora capsici, Fusarium oxysporum, Rhizoctonia solani and Sclerotinia sclerotiorum.
This study was carried out to establish simple and easy methods to judge the survival, senescence and death of the protoplasts in culture system by identifying the marker substance related to metabolic status of the cells. When rice and tobacco protoplasts were cultured in MS and KM-8P media containing 2,4-D or coconut milk ABA decreased especially in the media containing coconut milk, but GA$_3$, IAA and zeatin increased as the cultures progressed. The decrease of ABA and increase of zeatin was especially remarkable. When the supraoptimal amount of osmoticum (mannitol) was added to the culture media ABA decreased after a momentary increase, but other growth hormones slowly increased as the concentration of the osmoticum increased. Contents of individual hormones were contrasted when protoplasts rice and tobacco were cultured on the same medium containing 10mM super mine or NaCl. Tobacco protoplasts were more sensitive to NaCl stress and stopped protoplast division at the late stage of culture. Protoplast viability decreased greatly in 48 hours when the protoplast were at 32$^{\circ}C$ on a medium lacking several components. ABA content increased up to 10 days from incubation in negative proportion to the protoplast viability. On the other hand contents of other growth hormones, especially zeatin, decreased. The present results clearly showed that the contents of individual growth hormones in the plant protoplasts in culture varied sensitively in response to environmental factors that they are faced with. This indicates that the physiological states of the protoplast, such as survival, senescence or death can be simply judged based on the quantitative analysis of those hormones by ELISA.
Cotton Glutathione S-Transferase (GST: EC 2.5.1.18) was cloned and overexpressed in tobacco (Nicotiana tabacum) plants. Northern blot analysis confirmed the successful transformation of cotton gst gene in tobacco plant. Type I and Type ll transcript patterns were identified in transgenic tobacco plants and only Type I transcripts were discussed in this paper, The activity of GST in the type II transgenic plants was about 1.5-fold higher than those of the wild type and non-expresser by using 1-chloro-2,4-dinitrobenzene (CDNB) and reduced glutathione as the substrate. The expression of cotton GST in tobacco plants proved that Gh-5 could be translated into functional protein. Type II transgenic plants produced functional GST in the cells. The effects of cotton GST in the seedlings was evaluated by growing the control and transgenic seedlings at $15^{\circ}C$ in the growth chamber in the light. Overexpressors were grown well compared to the control plants (non-expressors). lo test far tolerance to salinity, seeds of Gh-5 overexpressors and the wild type Xanthi seedlings were grown at 0, 50, 100, 150, and 200 mM NaCl solution. Gh-5 transgenic seedlings showed higher growth rate over control seedlings on 50 and 100 mM NaCl solution. There was no difference in growth rate at 150 and 200mM NaCl concentration.
