하이브리드 이산화티탄의 자기조직체 형성공법을 이용하여 제조된 하이브리드 이산화티탄의 자외선차단 상승효과

  • 화장료의 UV차단과 은폐효과를 갖는 이산화티탄을 사용하여 자기조직체 형성공법을 적용한 하이브리드 이산화티탄을 제조하고 형태, 성질, 공정의 최적조건과 자외선차단 개선을 확인하였다. 하이브리드 이산화티탄은 마이크로 이산화티탄(250~300nm)의 표면에 나노 이산화티탄(20~30nm)을 자기조직체 형성공법을 이용해 결합시킨, 이산화티탄 대 이산화티탄의 결합체를 말한다. 하이브리드 이산화티탄 제조의 최적조건을 알아내기 위해 (-)을 띄는 마이크로 이산화티탄의 표면에 양이온의 링크로 써 $AlCl_3$를 농도별로 조정하고, 그에 따른 마이크로와 나노 이산화티탄의 투입비율을 달리하여 각각의 조건에서 만들어진 시료를 광학분석, 입도분석, 전위차분석 등을 이용해 확인하고 최적의 제조 조건을 알 수 있었다. 최적의 제조 조건에서 만들어진 하이브리드 이산화티탄의 자외선차단 상승효과를 확인하기 위하여 하이브리드 이산화티탄이 첨가된 화장료와 사용된 하이브리드 이산화티탄과 같은 비율의 마이크로와 나노 이산화티탄을 첨가한 화장료의 SPF in-vitro를 측정하였고, 15%내지 30%의 자외선차단 상승 효과를 확인하였다.

Chitosan/hydroxyapatite composite coatings on porous Ti6Al4V titanium implants: in vitro and in vivo studies

  • Purpose: Titanium implants are widely used in the treatment of dentition defects; however, due to problems such as osseointegration failure, peri-implant bone resorption, and periimplant inflammation, their application is subject to certain restrictions. The surface modification of titanium implants can improve the implant success rate and meet the needs of clinical applications. The goal of this study was to evaluate the effect of the use of porous titanium with a chitosan/hydroxyapatite coating on osseointegration. Methods: Titanium implants with a dense core and a porous outer structure were prepared using a computer-aided design model and selective laser sintering technology, with a fabricated chitosan/hydroxyapatite composite coating on their surfaces. In vivo and in vitro experiments were used to assess osteogenesis. Results: The quasi-elastic gradient and compressive strength of porous titanium implants were observed to decrease as the porosity increased. The in vitro experiments demonstrated that, the porous titanium implants had no biological toxicity; additionally, the porous structure was shown to be superior to dense titanium with regard to facilitating the adhesion and proliferation of osteoblast-like MC3T3-E1 cells. The in vivo experimental results also showed that the porous structure was beneficial, as bone tissue could grow into the pores, thereby exhibiting good osseointegration. Conclusions: Porous titanium with a chitosan/hydroxyapatite coating promoted MC3T3-E1 cell proliferation and differentiation, and also improved osseointegration in vitro. This study has meaningful implications for research into ways of improving the surface structures of implants and promoting implant osseointegration.

인공 체액 조건에서 임플랜트용 티타늄 소재의 부식 특성 (Corrosion Behavior of Titanium for Implant in Simulated Body Fluids)

  • The corrosion of pure titanium (CP- Ti Grade 2) and titanium alloy (Ti6Al4V ELI) were studied under various conditions of simulated body fluids. The static immersion test and the electrochemical test were performed in accordance with ISO 10271 : 2001. For the electrochemical test, the open circuit potential was monitored as a function of time, and the cyclic polarization curve was recorded. The corrosion resistance was evaluated from the values of corrosion potential, passivation current density, breakdown potential, and the shape of hysteresis etc. The effects of alloy type, surface condition, temperature, oxygen, and constituents in the fluids such as acid, chloride were estimated. Both specimens had extremely low dissolution rate in the static immersion test. They showed strong passivation characteristics in the electrochemical test. They maintained negligible current density throughout the wide anodic potential range. The passive layer was not broken up to 2.0 V (vs. SCE). The hysteresis and the shift of passivation potential toward the anodic direction was observed during the reversed scan. The passivation process appeared to be accelerated by oxygen in air or that dissolved in the fluids. The passivation also proceeded without oxygen by the reaction of constituents in the fluids. Acid or chloride in the fluids, specially later weakened the passive layer, and then induced higher passivation current density and less shift of passivation potential in the reversed scan. CP-Ti Grade 2 was more reactive than Ti6Al4V ELI in the fluids containing acid or chloride, but thicker layer produced on its surface provided higher corrosion resistance.

Surface Treatment of a Titanium Implant using a low Temperature Atmospheric Pressure Plasma Jet

  • The surface treatment of a titanium implant is investigated with a non-thermal atmospheric pressure plasma jet. The plasma jet is generated by the injection of He and $O_2$ gas mixture with a sinusoidal driving voltage of 3 kV or more and with a driving frequency of 20 kHz. The generated plasma plume has a length up to 35 mm from the jet outlet. The wettability of 4 different titanium surfaces with plasma treatments was measured by the contact angle analysis. The water contact angles were significantly reduced especially for $O_2/He$ mixture plasma, which was explained with the optical emission spectroscopy. Consequently, plasma treatment enhances wettability of the titanium surface significantly within the operation time of tens of seconds, which is practically helpful for tooth implantation.

Magnesium vs. machined surfaced titanium - osteoblast and osteoclast differentiation

  • PURPOSE. This study focused on in vitro cell differentiation and surface characteristics in a magnesium coated titanium surface implanted on using a plasma ion source. MATERIALS AND METHODS. 40 commercially made pure titanium discs were prepared to produce Ti oxide machined surface (M) and Mg-incorporated Ti oxide machined surface (MM). Surface properties were analyzed using a scanning electron microscopy (SEM). On each surface, alkaline phosphatase (ALP) activity, alizarin red S staining for mineralization of MC3T3-E1 cells, and quantitative analysis of osteoblastic gene expression, were evaluated. Actin ring formation assay and gene expression analysis of TRAP and GAPDH performing RT-PCR were performed to characterize osteoclast differentiation on mouse bone marrow-derived macrophages (BMMs). RESULTS. MM showed similar surface morphology and surface roughness with M, but was slightly smoother after ion implantation at the micron scale. M was more hydrophobic than MM. No significant difference between surfaces on ALP activity at 7 and 14 days were observed. Real-time PCR analyses showed similar levels of mRNA expression of the osteoblast phenotype genes; osteopontin (OPN), osteocalcin (OCN), bone sialoprotein (BSP), and collagen 1 (Col 1) in cell grown on MM at 7, 14 and 21 days. Alizarin red S staining at 21 days showed no significant difference. BMMs differentiation increased in M and MM. Actin ring formation assay and gene expression analysis of TRAP showed osteoclast differentiation to be more active on MM. CONCLUSION. Both M and MM have a good effect on osteoblastic cell differentiation, but MM may speed the bone remodeling process by activating on osteoclast differentiation.

Physical Properties of Mercaptopyruvic-acid Layer Formed on Gold Surfaces

  • We studied the physical properties of the mercaptopyruvic-acid layer formed on gold surfaces, which has the interactions with the titanium dioxide surface for design of gold- titanium dioxide distribution. Surface force measurements were performed, using the atomic force microscope (AFM), between the surfaces as a function of the salt concentration and pH value. The forces were analyzed with the DLVO (Derjaguin-Landau-Verwey-Overbeek) theory, to evaluate the potential and charge density of the surfaces quantitatively for each salt concentration and each pH value. The difference in the properties reflected the effect of the isoelectric point on the surface forces. The forces were interpreted for the evaluation with the law of mass action and the ionizable groups on the surface. The salt concentration dependence of the surface properties, found from the measurement at pH 8.0, was consistent with the prediction from the law. It was found that the mercaptopyruvic-acid layer had higher values for the surface charge densities and potentials than the titanium dioxide surfaces at pH 8, which may be attributed to the ionized-functional-groups of the mercaptopyruvic-acid layer.

티타늄 표면조도가 조골세포의 부착 및 분화에 미치는 영향 (Effect of titanium surface roughness on adhesion and differentiation of osteoblasts)

  • The success of an implant is determined by its integration into the tissue surrounding the biomaterial. Surface roughness is considered to influence the behavior of adherent cells. The aim of this in vitro study was to determine the effect of surface roughness on Saos-2 osteoblast-like cells. Titanium disks blasted with 75 ${/mu}m$ aluminum oxide particles and machined titanium disks were prepared. Saos-2 were plated on the disks at a density of 50,000 cells per well in 48-well dishes. After 1 hour, 1 day, 6 days cell numbers were counted. One day, 6 days after plating, alkaline phosphatase(ALPase) activity was determined. Compared to experimental group, the number of cells was significantly higher on control group. The stimulatory effect of surface roughness on ALPase was more pronounced on the experimental group than on control group. These results demonstrate that surface roughness alters proliferation and differentiation of osteoblasts. The results also suggest that implant surface roughness may play a role in determining phenotypic expression of cells.

Analysis of microstructural characteristics and components of red and yellow ink pigments used in permanent makeup

  • Our purpose in this study is to analyze the microstructural characteristics and constituent elements of inorganic substances added to the yellow ink and red ink pigments used in permanent makeup. We observed the microstructural properties of inorganic pigments added to the ink using a scanning electron microscopy (SEM) and analyzed the constituent elements of the inorganic pigment particles using an energy dispersive X-ray spectroscopy (EDX). In red wine-colored ink, cubic titanium dioxide with a diameter of 110 to 200 nm was the major component, and rod-shaped iron oxide was rarely observed. Most of the ingredients of taupe yellow ink were rod-shaped yellow iron oxide, and a small amount of cubic titanium dioxide was observed. Red wine-colored ink and taupe yellow ink contained lumps composed of titanium dioxide particles. In red wine-colored ink, lumps were formed by agglomeration. However, we observed that the surface of the lump composed of titanium dioxide in the taupe yellow ink had a smooth surface caused by external physical compression. The titanium dioxide particle mass which found in taupe yellow ink in this study is an artificial product. When this mass accumulates in the dermis, it may cause a color mismatch. Therefore, permanent makeup using fine pigments should be free of foreign substances that may cause trouble in the skin. In addition, there is a need to improve the quality of the ink so that the required color can be safe and long lasting in the dermis.

Titanium Phosphide 표면에 대한 세포독성 및 골친화성의 평가 (Evaluation of cytotoxicity and bone affinity on the surface of a titanium phosphide)

  • Dental implants have been developed for enhancement of osseointegration. Biocompatibility, bone affinity and surface characteristics of dental implants are very important factors for osseointegration. The aim of the present study was to determine the cytotoxicity and the bone affinity of titanium phosphide(Ti-P) implant material. The Ti-P surface was obtained by vacuum sintering of titanium within compacted hydroxyapatite powder. The composition and the chemical change of the surface were determined by Auger electron spectroscopy. The in vitro cytotoxicity was evaluated by the viability of the bone cells and macrophages obtained from chicken embryo and rat,s peritonium, respectively. For the comparative evaluation, 316L stainless steel, commercially pure titanium and Ti-P materials, prepared in size of 1O.0mm in diameter and 5.0mm in height, were immersed separately in bone cells and macrophages for 10 days. For the evaluation of the in vivo bone affinity, 316L stainless steel, commercially pure titanium and Ti-P materials, prepared in size of 5.0mm in diameter and 10.0mm in length, were implanted after drilling in diameter 5.5mm in femurs of 2 dogs weighing 10Kg more or less. Six weeks after implantation the specimens were prepared for histopathological examination and were observed under light microscope. In comparison of in vitro bone cell viability, Ti-P and commercially pure titanium groups were not significantly different from control group (p>O.1), but 316L stainless steel group was significantly lower than control group(p<0.05). There was no statistical difference in the viability of macrophages between 3 different groups and control group(p>O.l). In comparison of in vivo study, 316L stainless steel and commercially pure titanium showed fibrous encapsulation, but Ti-P showed remarkable new bone formation without any fibrous tissue. The results demonstrate that Ti-P has favorable biocompatibility and bone affinity, and suggest that dental implants with Ti-P surface may enhance osseointegration.

산-처리 조건이 RBM처리한 티타늄 임플란트의 표면 특성에 주는 영향 (Effects of acid-treatment conditions on the surface properties of the RBM treated titanium implants)

  • 본 논문에서는 순수 티타늄(cp-Ti) 임플란트를 SLA (Sandblasting with Large grit and Acid) 처리할 때 산-처리 용액의 유형, 산-처리 온도 및 산-처리 시간 등이 티타늄 표면에 주는 영향을 평가하고자 하였다. 원판형의 cp-Ti 시편을 준비하여 표면을 인산칼슘계 세라믹 분말로 RBM (Resorbable Blast Media) 처리하였다. 산-처리 용액으로 염산을 30 vol%로 고정하고 황산의 농도를 10, 20, 30, 35 vol%로 증가시키며 혼합한 용액에 증류수를 추가하여 4종의 산-처리 용액을 준비하였다. 실험군은 4종의 산-처리 용액, 3 종의 처리온도 및 3 종의 처리시간 등 36 가지로 분류하여 실험군당 4개의 시편을 산-처리하였다. 산-처리 전 후 시편 무게를 전자저울로 측정하여 무게 감소비율을 계산하였고, 공초점주사전자현미경으로 표면거칠기를 측정하였다. X-선 회절분석기(XRD)로 XRD 패턴을 측정하였고, 주사전자현미경으로 표면 형상을 관찰하였으며, 에너지 분산형 분석기(EDX)와 광전자분광법(XPS)로 표면성분을 분석하였다. 무게 감소비율과 표면거칠기 측정값은 Tukey-multiple comparison test (p = 0.05)로 통계 분석하여 다음의 결과를 얻었다. 산-처리에 따른 티타늄 시편의 무게 감소는 황산의 농도 및 산-처리 용액의 온도가 높을수록 유의하게 증가하였다. 산-처리한 티타늄의 표면 거칠기는 산-처리 조건(황산 농도, 온도, 시간)에 일정한 영향을 받지 않았다. XRD 분석에서 산-처리한 모든 시편에서 티타늄(${\alpha}-Ti$)과 수소화 티타늄($TiH_2$) 결정상이 관찰되었고, XPS 분석으로 티타늄 표면에 얇은 n산화 티타늄 층이 형성된 것을 알 수 있었다. $90^{\circ}C$ 산-용액에서 처리할 경우 티타늄 표면이 과도하게 용해될 수 있으므로 주의하여야 한다.
