성장인자/키토산이 담지된 온도감응성 하이드로젤의 유착방지제로서의 응용 (Thermosensitive Chitosan-based Hydrogel with Growth Factor as Adhesion Barrier)

  • 조직과 장기 유착은 외과적 수술 후 부작용으로 자주 발생된다. 이러한 조직 유착을 방지하기 위하여 유착부위에 장벽을 형성하여 장기 유착을 방지할 수 있는 온도감응성 하이드로젤 유착방지제를 제조하고자 하였다. 폴록사머, 키토산과 표피성장인자를 가지고 온도감응성 키토산 하이드로젤을 제조하였다. 제조한 키토산 하이드로젤은 인체 내 온도와 유사한 $35^{\circ}C$에서 졸-젤 전이현상을 보였고, 1분 이내에 빠르게 젤화되었다. 키토산 하이드로젤은 표피성장인자를 7일 동안 천천히 방출하였고, 마우스모델에서 평가한 결과 조직 유착 방지를 효과적으로 하는 것을 확인하였다. 온도 감응성과 항균성을 갖는 키토산 하이드로젤이 장기 부착의 증가와 표피성장인자를 천천히 방출하는 유착방지제로서 유용하게 사용될 수 있다.

  • Studies on some biodegradable polymers and other materials such as hydrogels have shown the promising potential for a variety of surgical applications. Postoperative adhesion caused by the natural consequence of surgical wound healing results in problems of the repeated surgery. Recently, scientists have developed absorbable anti-adhesion barriers that can protect a tissue from adhesion in case they are in use; however, they are dissolved when no longer needed. Although these approaches have been attempted to fulfill the criteria for adhesion prevention, none can perfectly prevent adhesions in all situations. Overall of this work, a new method to fabricate an anti-adhesion membrane using biodegradable polymer and hydrogel has been developed. The ideal barrier for preventing postoperative adhesion would have the following properties; it should be (i) resorbable (ii) non-reactive (iii) easy to apply (iv) capable of being fixed in position. In order to fulfill these properties, we adopted solid freeform fabrication method combined with surface modification which includes the hydrogel coating, therefore, inner or outer structure can be controlled and the property of anti adhesion can be improved.

성견 1면 골내낭에서 탈회 냉동 건조골과 calcium sulfate 혼합 이식 및 calcium sulfate 차단막 사용이 치주조직 치유에 미치는 영향 (The Effect of composite Graft of allogenic DFDB and Calcium Sulfate with and without Calcium Sulfate barrier in Periodontal 1 wall intrabony defects in Dogs)

  • Numerous bone graft materials have been used in Periodontics, in an attempt to reach the main goal of periodontal therapy, i.e. the regeneration of periodontal tissue lost due to destructive periodontal diseases. The present study investigates the effect of composite graft of DFDB and Calcium sulfate with and without Calcium sulfate barrier in Periodontal 1-wall intrabony defects in dogs. Following the initiation of general anesthesia by I.V. administration of 40mg/Kg of Pentobabital, second premolar was extracted and full thickness flap elevated. The crown portion of premolars was removed. Exposed root canals were sealed with Caviton and covered completely with flap. After the healing period of 8 weeks, the surgical sites were re-opened and 1-wall intrabony defects were created, and treated with flap operation alone(control group), with composit graft of 80% DFDB and 20% Calcium sulfate(Experimental group 1), with composite graft of DFDB and calcium sulfate with calcium sulfate membrane( Experimental group 2). Healing response was histologically observed after 8 weeks and the results were as follows : 1. New bone formation was 70 % in the control group, 93 % in the Experimental group I, 89 % in the Experimental group II. There was a no differences between Experimental groups. 2. New cementum formation was not significantly different between control and two Experimental groups. 3. The length of connective tissue adhesion was 30 % in the control, 7% in the Experimental group I and 11 % in the Experimental group II. 4. After 8weeks, calcium sulfate was completely resorbed, while DFDB particle remained. These results suggest that the use of composite graft of allogenic DFDB and Calcium sulfate with and without Calcium sulfate barrier in periodontal 1 wall intrabony defects have little effect on connective tissue adhesion, but has beneficial effect on new alveolar bone and new cementum formation, and prevent downgrowth of epithelium and connective tissue effectively.

Recently, scientists have developed absorbable anti-adhesion barriers that can protect a tissue from adhesion in case they are in use; however, they are dissolved when no longer needed. Although these approaches have been attempted to fulfill the criteria for adhesion prevention, none can perfectly prevent adhesions in all situations. Overall, we developed a new method to fabricate an anti-adhesion membrane using biodegradable polymer and hydrogel. It employed a highly accurate three-dimensional positioning system with pressure-controlled syringe to deposit biopolymer solution. The pressure-activated microsyringe was equipped with fine-bore nozzles of various inner-diameters. This process allowed that inner and outer shapes could be controlled arbitrarily when it was applied to a surgical region with arbitrary shapes. In order to fulfill the properties of the ideal barriers f3r preventing postoperative adhesion, we adopted the pre-mentioned method combined with surface modification with the hydrogel coating by which anti-adhesion property was improved.

구개편도 및 인두편도에서 혈관내피성 접착분자의 발현에 관한 연구 (The Study on the Expression of Vascular Endothelial Adhesion Molecule in Palatine and Pharyngeal Tonsil)

  • The palatine tonsils(tonsils) and pharyngeal tonsils(adenoids) are situated at the entrance of the respiratory and alimentary tracts and represent the first site of contact with a variety of microorganisms and other antigens present in food and inhaled air. They are known as lymphoid organs carrying out the function of cellular and humoral immunity, and so they form a local protective barrier. And the expression of the vascular endothelial adhesion molecules is known to play an important role for the inflammatory reaction in tonsils and adenoids as well as in other inflammatory tissues, by binding with the receptors on the surface of leukocytes. But although several scientific hypotheses on the role of these lympoid tissues have been suggested, their complete functions have remained unknown. The purpose of this study is to present an basic data of the knowledge on the immunologic physiology of the tonsils and adenoids and their role as active immunologic organs that reinforce the mucosal immunity of the entire upper aerodigestive tract. We examined 16 human tonsils and adenoids and the expression of three endothelial adhesion molecules, vascular endothelial adhesion molecule-1(VCAM-1), intracellular adhesion molecule-1(ICAM-1), and E-selection, in tissue sections using immunohistochemistry. We used the inferior turbinate mucosa obtained from 9 patients getting septal surgery as a control group. The expressions of vascular endothelial adhesion molecule-1(VCAM-1) and intracellular adhesion molecule-1 (ICAM-1) were significantly higher in the tonsils and adenoids. But respectively, there were no significant differences between the tonsils and adenoids. The expression of E-selection was significant higher in the tonsils, but not in the adenoids. We observed that tonsils and adenoids showed significantly higher expressions of vascular endothelial adhesion molecule-1(VCAM-1), intracellular adhesion molecule-1(ICAM-1), and E-selection (in the case of E-selection, only in the tonsils). We propose that these adhesion molecules play an important role for the immunologic reaction by the transendothelial migration of lymphocytes and binding with the receptors on the surface of leukocytes.

Effect of laser-dimpled titanium surfaces on attachment of epithelial-like cells and fibroblasts

  • PURPOSE. The objective of this study was to conduct an in vitro comparative evaluation of polished and laser-dimpled titanium (Ti) surfaces to determine whether either surface has an advantage in promoting the attachment of epithelial-like cells and fibroblast to Ti. MATERIALS AND METHODS. Forty-eight coin-shaped samples of commercially pure, grade 4 Ti plates were used in this study. These discs were cleaned to a surface roughness (Ra: roughness centerline average) of 180 nm by polishing and were divided into three groups: SM (n=16) had no dimples and served as the control, SM15 (n=16) had $5-{\mu}m$ dimples at $10-{\mu}m$ intervals, and SM30 (n=16) had $5-{\mu}m$ dimples at $25-{\mu}m$ intervals in a $2{\times}4mm^2$ area at the center of the disc. Human gingival squamous cell carcinoma cells (YD-38) and human lung fibroblasts (MRC-5) were cultured and used in cell proliferation assays, adhesion assays, immunofluorescent staining of adhesion proteins, and morphological analysis by SEM. The data were analyzed statistically to determine the significance of differences. RESULTS. The adhesion strength of epithelial cells was higher on Ti surfaces with $5-{\mu}m$ laser dimples than on polished Ti surfaces, while the adhesion of fibroblasts was not significantly changed by laser treatment of implant surfaces. However, epithelial cells and fibroblasts around the laser dimples appeared larger and showed increased expression of adhesion proteins. CONCLUSION. These findings demonstrate that laser dimpling may contribute to improving the peri-implant soft tissue barrier. This study provided helpful information for developing the transmucosal surface of the abutment.

콩 잎 엽모에 의한 불마름병균 부착 저해 (Hairs as Physical Barrier against Adhesion of Xanthomonas axonopodis pv. glycines on Soybean Leaf)

  • 콩에 발생하는 불마름병은 주로 엽맥을 따라 병징이 형성되며, 이러한 병징의 형성에 대한 원인을 밝히고자 광학현미경 및 주사전자현미경으로 표면을 관찰하였다. 광학현미경 관찰에 의한 조직의 차이는 볼 수 없었으나 주사전자현미경으로 표면 관찰시, 잎 뒷면 미세한 엽모가 잎표면에 존재하였으며 엽맥 주위로는 엽모가 관찰되지 않았다. 세균은 엽맥 주위 엽모가 없는 부분에 주로 부착되어 있었고, 엽모가 있는 부분에서 세균은 관찰되지 않았다. 또한 잎에 불마름병균을 접종 후 표면 미생물을 배양시 엽맥주위에서 주로 세균이 검출되어 엽모의 존재가 불마름병균의 부착을 방해하여 엽맥주변으로 병징이 발현된 것으로 판단된다.

  • Various bonegraft materials and the technique of guided tissue regeneration have been used to regenerate lost periodontal tissue. Calcium sulfate has been known as a bone graft material because of good biocompatibility, rapid resorption and effective osteoinduction. It has been known that calcium sulfate works as a binder to stabilize the defect when it is used with synthetic graft materials. The effects on the regeneration of pericxiontal tissue were studied in dogs after grafting 3-wall intrabony defects with calcium carbonate and calcium sulfate and covering with calcium sulfate barrier. The 3-wall intrabony defectstdmm width, 4mm depth, 4mm length) were created in anterior area and treated with flap operation alone(contol group), with porous resorbable calcium carbonate graft alonetexperirnental group 1), with calcium sulfate graft alonetexperimental group 2) and with composite graft of 80% calcium carbonate and 20% calcium sulfate with calcium sulfate barriertexperimental group 3). Healing responses were histologically observed after 8 weeks and the results were as follows: 1. The alveolar bone formation was $0.59{\pm}0.19mm$ in the control group, $1.80{\pm}0.25mm$ in experimental group 1, $1.61{\pm}0.21mm$ in experimental group 2 and $1.94{\pm}0.11mm$ in experimental group 3 with statistically significant differences between control group and all experimental groups(P<0.05). There were statistically significant differences between experimental group 1 and group 2 (P<0.05). 2. The new cementum formation was $0.48{\pm}0.19mm$ in the control group. $1.72{\pm}0.26mm$ in experimental group 1, $1.43{\pm}0.17mm$ in experimental group 2, $1.89{\pm}0.15mm$ in experimental group 3 with statiscally significant differences between control group and all experimental groups (p<0.05). There were statistically significant differences between experimental group 1 and group 2, and between experimental group 2 and group 3(P<0.05). 3. The length of junctional epithelium was $1.61{\pm}0.20mm$ in the contol group, $0.95{\pm}0.06mm$ in experimental group 1, $1.34{\pm}0.16mm$ in experimental group 2, $1.08{\pm}0.11mm$ in experimental group 3 with statiscally significant differences between control group and experimental group 1. and btween control group and experimental group 3(p<0.05). There were statistically significant differences between experimental group 1 ,and group 2, and between experimental group 2 and group 3(P<0.05). 4. The connective tissue adhesion was $1.67{\pm}O.20mm$ in the control group, $1.33{\pm}0.24mm$ in experimental group 1. $1.23{\pm}0.16mm$ in experimental group 2, $1.08{\pm}0.14mm$ in experimental group 3 with statistically significant differences between control group and all experimental groups(p<0.05). There were nostatistically significant differences between all experimental groups. As a result, epithelial migration was not prevented when calcium sulfate was used alone, but new bone and cementum formation were enhanced. Epithelial migration was prevented and new bone and cementum formation were also enhanced when calcium carbonate was used alone and when both calcium carbonate and calcium sulfate were used.

참깨 내탈립성의 원인과 검정 방법 (Grain Shattering Resistance and Its Screening Method of Sesame)

  • 기존의 내탈립성 참깨들은 우리나라 환경에서의 적응성이 떨어지며 수량성도 낮았으나, 계속적인 개량으로 인해 기존품종보다 내탈립성이 뛰어나면서도 수량성 등 제반 형질이 뒤지지 않는 새로운 내탈립성 참깨의 개발을 목전에 두고 있다. 그러나 앞으로도 계속적인 개량이 필요한 이들 내탈립성 참깨의 효율적인 육성 기술은 아직 부족한 실정이다. 따라서 본 시험에서는 이와 관련된 기술로 참깨 종자의 내탈립성을 나타내는 구체적인 기작 및 효율적인 탈립성 검정 방법 등을 밝히고자 수행하였던 결과를 요약하면 다음과 같다. 참깨 종자의 내탈립성의 원인을 검토한 결과 PA형의 내탈립성은 꼬투리(capsule)내 태좌의 종자 보유력이 크기 때문에 나타난 것으로 추정되었으며, ID형은 꼬투리의 중과피(mesocarp) 두께가 일반 참깨보다 두껍기 때문이었으며, SL형은 꼬투리에 seam이 존재하지 않기 때문이었다. 또한 내탈립성 참깨들은 일부의 개체에서 탈립성이 큰 변이가 나타났는데, 이는 이들 참깨들의 진화방향이 탈립성이 큰 방향임을 의미하는 것으로 판단되었다. 탈립성 측정을 위한 적합한 건조 조건은 상온에서는 참깨 수확 후 최소 20일, $40^{\circ}C$의 건조기 내에서는 10일 이상의 건조기간이 필요한 것으로 나타났다.
