Effects of Solvent Viscosity on Conformational Dynamics of Heme-pocket in Myoglobin and Hemoglobin

  • The influence of solvent viscosity on conformational dynamics of the heme-pocket, a small vacant site near the binding site of myoglobin (Mb) and hemoglobin (Hb), and playing a functionally important role by serving as a station in ligand binding and escape, was studied by probing time-resolved vibrational spectra of CO photodissociated from MbCO and HbCO in $D_2O$, 75 wt% glycerol/$D_2O$, and trehalose at 283 K. Two absorption bands ($B_1$ and $B_2$) of the sample in viscous solvents, arising from CO in the heme pocket, are very similar to those in $D_2O$. Two bands in Mb and Hb under all three solvents exhibit very similar nonexponential spectral evolution ($B_1$ band; blue shifting and broadening, $B_2$ band; narrowing with a negligible shifting), suggesting that in the present experimental time window of 100 ps, the extents of the spectral shift and narrowing is much influenced neither by the viscosity of solvent nor by the quaternary contact of Hb. Spectral evolution can be described by a biexponential function with a fast universal time constant of 0.52 ps and a slow time constant ranging from 13 to 32 ps. For both proteins in all three solvents majority of spectral evolution occurs with the fast universal time constant. The magnitude of the slow rate in the spectral shift of B1 band decreases with increasing solvent viscosity, indicating that it is influenced by global conformational change which is retarded in viscous solvent, thereby serve as a reporter of global conformational change of heme proteins after deligation.

스테레오 시간분해 입자영상유속계를 이용한 외부경계층 수직날 배열에 의한 마찰저항 저감 기구에 관한 실험적 조사 (Experimental Investigation on the Drag Reduction Mechanism of Outer-layer Vertical Blades Array using Stereoscopic Time-Resolved PIV)

  • A stereo PIV measurements in a circulating water channel has been performed to investigate the skin friction reduction mechanism of the outer-layer vertical blades first devised by Hutchins. In a recent PIV measurement study, considerable skin friction reduction was achieved as much as 2.73%~7.95% by outer-layer vertical blades array. In the present study, the influence of vertical blades array upon the characteristics of the turbulent coherent structures was analyzed by proper orthogonal decomposition method. It is observed that the vortical structures are cut and deformed by blades array and also the turbulent intensity and the Reynolds stress were weakened by the blades. These phenomena strongly associate the skin-friction drag reduction mechanism in the turbulent boundary layer flow.

Effect of Growth Temperature on the Luminescence Properties of InP/GaP Short-Period Superlattice Structures

  • The optical properties of InP/GaP short-period superlattice (SPS) structures grown at various temperatures from $400^{\circ}C$ to $490^{\circ}C$ have been investigated by using temperature-dependent photoluminescence (PL) and emission wavelength-dependent time-resolved PL measurements. The PL peak energy for SPS samples decreases as the growth temperature increases. The decreased PL energy of ~10 meV for the sample grown at $425^{\circ}C$ compared to that for $400^{\circ}C$-grown sample is due to the CuPt-B type ordering, while the SPS samples grown at $460^{\circ}C$ and $490^{\circ}C$ exhibit the significant reduction of the PL peak energies due to the combined effects of the formation of lateral composition modulation (LCM) and CuPt-B type ordering. The SPS samples with LCM structure show the enhanced carrier lifetime due to the spatial separation of carriers. This study represents that the bandgap energy of InP/GaP SPS structures can be controlled by varying growth temperature, leading to LCM formation and CuPt-B type ordering.

  • 펄스형 질소레이저를 사용한 time-resolved laser-induced fluorimetry를 이용하여 우라늄 작업자의 뇨속에 함유되어 있는 우라늄의 농도를 간단한 전처리만으로 정량분석하는 방법을 연구하였다. 형광분석할 때에 뇨에 함유된 chloride ion은 우라늄 형광을 심하게 quenching하는 것으로 알려져 있으며, 따라서 이를 제거하기 위한 전처리 과정에서 많은 시간 소모와 큰 실험 오차를 유발하고 있다. 본 방법에서는 10% Fluran 수용액을 뇨에 첨가하여 뇨함량이 약 1% 정도가 되었을 때 뇨속의 chloride에 의한 quenching 영향을 최소한으로 줄일 수 있었으며, 시간 0에서의 형광강도를 계산하여 형광강도의 농도에 대한 직선성을 측정한 결과 우라늄 농도 범위 10-500 ppb에서 우수한 직선성을 나타내었다.

Effect of Ganglioside $G_{M3}$ on the Erythrocyte Glucose Transporter (GLUT1): Conformational Changes Measured by Steady-State and Time-Resolved Fluorescence Spectroscopy

  • Interactions between ganglioside $G_{M3}$ and glucose transporter, GLUT1 were studied by measuring the effect of $G_{M3}$ on steady-state and time-resolved fluorescence of purified GLUT1 in synthetic lipids and on the 3-O-methylglucose uptake by human erythrocytes. The intrinsic tryptophan fluorescence showed a GLUT 1 emission maximum of 335 nm, and increased in the presence of $G_{M3}$ by 12% without shifting the emission maximum, The fluorescence lifetimes of intrinsic tryptophan on GLUT1 consisted of a long component of 7.8 ns and a short component of 2,3 ns and $G_{M3}$ increased both lifetime components. Lifetime components were quenched by acrylamide and KI. Acrylarnide-mduced quenching of long-lifetime components was partly recovered by $G_{M3}$ However. KI-induccd quenching of short- and long-lifetime components was not rescued by $G_{M3}$. The anisotropy of 1.6-diphenyl-1.3.5-hexatriene (DPH)-probed dimyristoylphosphatidylcholine (DMPC) model membrane was also increased with $G_{M3}$ incorporation, The transport rate of 3-O-methylglucose increased by 20% with $G_{M3}$ incorporation on the erythrocytes, Therefore, $G_{M3}$ altered the environment of lipid membrane and induced the conformational change of GLUT1.

진동 유동장에서 원형 실린더의 lock-on 해석 (Lock-on states of a circular cylinder in the oscillatory flow)

  • Vortex lock-on or resonance in the flow behind a circular cylinder is visualized by a time-resolved PIV when a single frequency oscillation is superimposed on the mean incident velocity. Measurements are made of the $K{\'{a}}rm{\'{a}}n$ vortices in the wake-transition regime at the Reynolds number 360. Basically, natural shedding state is observed to compare with lock-on state. Wake motion by the change of the shedding frequency of lock-on state is investigated. When lock-on occurs, the vortex shedding frequency is found to be half the oscillation frequency as expected from previous experiments. The physical flow phenomena of natural shedding and lock-on states are analyzed with physical parameters of recirculation and vortex formation region. Consequently, it is found that the change of wake bubble plays an important role in the flow at the lock-on state. Vortex formation region is also actively changed like recirculation region as the lock-on occurs. Therefore, it is deduced that the recirculation region is closely related with the vortex formation region.

경계층 외부 수직날의 마찰저항 저감 기구에 대한 PIV 관측 (PIV Investigation on the Skin Friction Reduction Mechanism of Outer-layer Vertical Blades)

  • An experimental assessment has been made of the drag reducing efficiency of the outer-layer vertical blades, which were first devised by Hutchins. The drag reduction efficiency of the blades was reported to reach as much as 30%. However, the drag reduction efficiency was quantified only in terms of the reduction in the local skin-friction coefficient. In the present study, a series of drag force measurements in towing tank has been performed toward the assessments of the total drag reduction efficiency of the outer-layer vertical blades. A maximum 9.6% of reduction of total drag was achieved. The scale of blade geometry is found to be weakly correlated with outer variable of boundary layer. In addition, detailed flow field measurements have been performed using 2-D time resolved PIV with a view to enabling the identification of drag reduction mechanism.

펄스형 방전플라스마 장치에서 반경방향 Current Sheath의 속력 (Radial Speed of Current Sheath in Pulsed Discharge Plasma Device)

  • 목적: 펄스형 방전플라스마 포커스 장치에서 반경방향 플라스마 current sheath 의 속력을 측정하였다. 방법: 측정에는 시간분해 분광분석법과 로고프스키 코일이 사용되었다. 결과: 15 kV의 방전전압과 수십 torr의 He과 Ar의 기체기압에서 $10^5$ cm/s의 속력이 측정되었으며, 기체기압이 증가할수록 current sheath의 속력은 감소되었다. 결론: 최적조건인 수 torr의 기압에서는 $10^7$ cm/s의 속력이 나올 것으로 예상된다.

  • This paper describes the process of nanoparticle synthesis by laser ablation of microparticles and consolidated sample. We have generated nanoparticles by high-power pulsed laser ablation of AI, Cu and Ag microparticles using a Q-switched Nd:YAG laser (wavelength 355nm, FWHM 6ns, fluence $0.8{\sim}2.0J/cm^2$). Microparticles of mean diameter $18{\sim}80{\mu}m$ are ablated in the ambient air. The generated nanoparticles are collected on a glass substrate and the size distribution and morphology are examined using a scanning electron microscope and a transmission electron microscope. The effect of laser fluence, collector position and compacting pressure on the distribution of particle size is investigated. To better understand the process of laser ablation of microparticle(LAM), we investigated the Nd: YAG laser-induced breakdown of Cu microparticle using time-resolved optical shadow images. Nanosecond time-resolved images of the ablation process are also obtained by laser flash shadowgraphy. Based on the experimental results, discussions are made on the dynamics of ablation plume.

Photoluminescence Characteristics of p-Phenylene Vinylene and Its Derivatives in Solution and in Nanoaggregates

  • Oligomers of p-phenylene vinylene and its derivatives have drawn much attention due to their unusual emission characteristics of showing increased emission when they form into nanoparticles. We have investigated the optical properties of the oligo-(p-phenylene vinylene) and its cyano-substituted derivatives in solution and in nanoaggregate media by femtosecond and picosecond time resolved fluorescence as well as stationary spectroscopies. All the spectroscopic data are consistent with the conclusion that the cyano substitution on the ${\beta}$-position of oligo-(p-phenylene vinylene) leads to breakage of the otherwise planar structure of cyano-unsubstituted molecules, which opens up an extremely efficient, as fast as 100 fs, non-radiative relaxation channel of the excited state. Formation of the nanoaggregates reverts the effect to make the molecules planar and to block the non-radiative relaxation channel. Therefore, concerning the applications in organic electroluminescent devices and organic light emitting diodes, substitution by the cyano group is not advantageous, although such modification should be useful in respect of controlling fluorescence intensity in different media.
