A report of 31 unrecorded bacterial species in South Korea belonging to the class Gammaproteobacteria

  • During recent screening to discover indigenous prokaryotic species in South Korea, a total of 31 bacterial strains assigned to the class Gammaproteobacteria were isolated from a variety of environmental samples including soil, tidal flat, freshwater, seawater, and plant roots. From the high 16S rRNA gene sequence similarity (>98.7%) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that these 31 species have been described in South Korea; therefore 5 species of 3 genera in the order Alteromonadales, 11 species of 3 genera in the order Pseudomonadales, 8 species of 6 genera in the order Enterobacteriales, 2 species of 1 genera in the order Vibrionales, 1 species of 1 genera in the order Oceanospirillales, 3 species of 3 genera in the order Xanthomonadales, and 1 species in the order Spongiibacter_o within the Gammaproteobacteia are reported for proteobacterial species found in South Korea. Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs are also described in the species description section.

  • An outcome of the study to discover indigenous prokaryotic species in Korea, a total of 26 bacterial species assigned to the classes Bacteroidetes and Firmicutes were isolated from diverse environmental samples collected from soil, tidal flat, freshwater, seawater, wetland, plant roots, and fermented foods. From the high 16S rRNA gene sequence similarity (>99.0%) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that these 26 species have been described in Korea; therefore 14 strains for the order Flavobacteriales and two strains for the order Cytophagales were assigned to the class Bacteroidetes, and 8 strains for the order Bacillales and 4 strains for the order Lactobacillales were assigned to the class Firmicutes are reported for new bacterial species found in Korea. Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs are also described in the species description section.

  • As a subset study to discover indigenous prokaryotic species in Korea, a total of 39 bacterial strains assigned to the classes Betaproteobacteria and Gammaproteobacteria were isolated from diverse environmental samples collected from soil, tidal flat, freshwater, seawater, seaweed, wetland, plant roots, guts of insects, and fermented foods. From the high 16S rRNA gene sequence similarity (>99.1%) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that these 39 species have been described in Korea; therefore 4 species of 4 genera in the order Burkholderiales and 1 species in the order Neisseriales within the class Betaproteobacteria, and 10 species of 6 genera in the order Alteromonadales, 11 species of 3 genera in the order Pseudomonadales, 4 species of 4 genera in the order Enterobacteriales, 2 species of 2 genera in the order Vibrionales, 1 species in the order Aeromonadales, 3 species of 3 genera in the order Oceanospirillales, 2 species of 2 genera in the order Xanthomonadales, and 1 species in the order Chromatiales within the Gammaproteobacteia are reported for proteobacterial species found in Korea. Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs are also described in the species description section.

Report on 30 unrecorded bacterial species of the phylum Firmicutes isolated from Korea in 2016

  • During the course of investigation of indigenous prokaryotic species in Korea, a total of 30 bacterial strains belonging to the phylum Firmicutes were isolated from diverse environmental sites such as soil, avian feces, wastewater treatment plants, fermented vegetables, seawater, algae, sea cucumber, octopus and tidal flat sediment. Phylogenetic analysis based on 16S rRNA gene sequences revealed that each strain showed high sequence similarity (${\geq}98.7%$) to the closest type strain and formed a robust phylogenetic clade with the most closely related species in the phylum Firmicutes. To date, there is no official record of these 30 species in Korea. Therefore, we report 26 species of 12 genera in the order Bacillales and 4 species of 4 genera in the order Lactobacillales which have not been reported in Korea. Morphological and biochemical characteristics, isolation sources and NIBR deposit numbers are described in the species descriptions.

염전으로부터 농화배양된 호염 메틸영양미생물 군집의 특성 (Prokaryotic Communities of Halophilic Methylotrophs Enriched from a Solar Saltern)

  • C-1화합물은 고염분성 환경의 혐기적인 퇴적층에서 관찰되며, 이 퇴적층의 표면과 수면에는 호기성 메틸영양미생물의 잠재적인 서식지가 된다. 염전과 갯벌에서 채취한 토양 시료를 접종원으로 하여 메탄올을 탄소원과 에너지원으로 공급하고 염분농도에 따라 계대배양한 후 메탄올 산화 세균 성장 조건을 살펴 본 결과, 메탄올 산화 세균이 성장 할 수 있는 염분의 최대 농도는 20% 조건이었다. 변성 구배 젤 전기영동 (Denaturing gel gradient electrophoresis)을 이용하여 농화배양액 내 미생물 군집구조를 분석한 결과, 메탄올 산화 미생물인 Methylophaga 관련 세균이 우점하는 것으로 나타났다. 정량 PCR결과 고세균이 세균의 1-10%로 존재하는 것으로 나타났다. 세균용 항생제 실험결과, 메탄올 산화가 억제되어 고세균이 메탄올 산화에 관여하지 않는다는 것을 추정할 수 있었다. 이번 연구를 통해, 메틸영양세균이 고염분환경(염분 농도 20%까지)에서도 C-1 화합물을 산화할 수 있음을 확인 할 수 있었다.

A report of 17 unrecorded bacterial species of Korea belonging to the phylum Bacteroidetes

  • While screening indigenous prokaryotic species in Republic of Korea in 2017, a total of 17 bacterial strains assigned to the phylum Bacteroidetes were isolated from a variety of environmental habitats including water of fountain, tidal flat, plant root, soil, the gut of Russian grayling butterfly, ginseng field, seawater, lagoon and seashore sand. From the 16S rRNA gene sequence similarity of more than 98.7% and the formation of a robust phylogenetic clade with the closest species, it was found that the 17 strains belong to independent and recognized bacterial species. There has been no official report that the identified 17 species have been previously isolated in the Republic of Korea. Thus, 15 species in 10 genera of one family in the order Flavobacteriales, one species in one genus of one family in the order Cytophagales, and one species in one genus of one family in the order Sphingobacteriales are proposed as unrecorded species of the phylum Bacteroidetes found in the Republic of Korea. Gram reaction, colony and cell morphology, basic phenotypic characteristics, isolation source, taxonomic status, strain ID and other information are described in the species descriptions.

A report of 38 unrecorded bacterial species in Korea within the classes Bacilli and Deinococci isolated from various sources

  • A total of 38 bacterial strains within the classes Bacilli and Deinococci were isolated from various sources in Korea. Samples were collected from animal intestine, urine, soil, tidal flat mud, and kimchi. In the sequence comparison and phylogenetic analysis of 16S rRNA sequences, the 38 isolates were assigned to the classes Bacilli and Deinococci with sequence similarities more than 98.7%. Twenty-four strains and 13 strains were classified the order Bacillales and Lactobacillales in the class Bacilli, respectively. In the order Bacillales, there were nine species in the genus Bacillus, seven species in the genus Paenibacillus, and the remaining eight species in the genera Domibacillus, Halobacillus, Virgibacillus, Lysinibacillus, Paenisporosarcina, Planococcus, Savagea, and Staphylococcus. In the order Lactobacillales, there were four species in the genus Lactobacillus, three species in the genus Leuconostoc, three species in the genus Lactococcus, and the remaining three species in the genera Aerococcus, Enterococcus, and Streptococcus. One species was related to the genus Deinococcus of the order Deinococcales. Most of the isolated strains were Gram-stain-positive, but some were Gram-stain-variable or Gram-stain-negative. Cells were rod or cocci-shaped. Based on the results of 16S rRNA analysis, we report 38 strains as previously unrecorded species to Korea, and the basic characteristics of strains are described herein.

A report of 24 unrecorded bacterial species in Korea belonging to the Phyla Proteobacteria and Bacteroidetes isolated in 2020

  • In 2020, 24 bacterial strains were isolated from algae, kudzu leaf, mud, pine cone, seashore sand, sea water, soil, tidal flat, and wetland from the Republic of Korea. Isolated bacterial strains were identified based on 16S rRNA gene sequences, and those exhibiting at least 98.7% sequence similarity with known bacterial species, but not reported in Korea, were highlighted as unrecorded species. These isolates were allocated to the phyla Bacteroidetes and Proteobacteria as unrecorded species in Korea. The four Bacteroidetes strains were classified into the families Chitinophagaceae, Flavobacteriaceae, and Sphingobacteriaceae (of the orders Chitinophagales, Flavobacteriales, and Sphingobacteriales, respectively). The 20 Proteobacteria strains belonged to the Aeromonadaceae, Marinobacter, Microbulbiferaceae, Enterobacteriaceae, Erwiniaceae, Morganellaceae, Yersiniaceae, Lysobacteraceae, Halomonadaceae, Moraxellaceae, Pseudomonadaceae, Steroidobacteraceae, Xanthomonadaceae, and Myxococcaceae (of the orders Aeromonadales, Alteromonadales, Cellvibrionales, Enterobacterales, Lysobacterales, Oceanospirillales, Pseudomonadales, Steroidobacter, Xanthomonadales, and Myxococcales). This study focused on the description of 24 unreported bacterial species in Korea in the phyla Bacteroidetes and Proteobacteria belonging to six classes.

경기도 화성 고정리 공룡알 화석지 공룡생태박물관 부지선정을 위한 지구물리 및 지질조사 (Geophysical and Geological Investigation for Selecting a Dinosaur Museum Site in the Dinosaur Egg Fossil Area, Gojeong-ri, Hwasung, Gyeonggi Province)

  • 이 연구에서는 많은 양의 공룡알 화석이 발견된 경기도 송산면 고정리의 시화호 남측 간척지내에 위치한 공룡생태박물관 건립부지의 $350{\times}750\;m^2$ 구역에서 탄성파 굴절법 탐사, 시추, 그리고 시추공에서 하향식 탄성파 탐사로 이루어지는 지질조사를 통해 부지내 기반암과 그 위에 쌓인 갯벌의 분포와 특성을 파악하였다. 전체 6,950 m 길이의 11개 측선에서 해머를 파원으로 이용하여 탄성파 굴절법 자료를 획득하여 역산으로 지질구조를 구하였다. 5개의 지점에서 지표에서 기반암까지 시추를 하였으며 이때 시료를 채취하여 입도분석 등을 통해 퇴적물을 분류하였으며 일부 시료에서 갯벌의 진화과정을 밝히기 위해 연대측정을 하였다. 2개 시추공에서 지층시료를 회수한 후에 해머를 음원으로 이용하여 깊이에 따라 탄성파의 속도를 측정하였다. 조사지역의 지질은 지표에서 아래로 갯벌, 풍화토, 그리고 기반암의 구조로 나타난다. 갯벌퇴적층과 풍화토층의 두께는 각각 5 ~ 12 m와 2 ~ 8 m이다. 기반암은 중생대 백악기 퇴적암층으로 해석되며 조사지역내에서 서쪽에서 동쪽으로 갈수록 얕게 분포하는 경향이 있다. 조사구역을 피복하고 있는 갯벌퇴적층과 풍화층의 부피는 약 140만 $m^3$, 무게는 350만 ton으로 계산된다. 조사지역에서 해수면 상승속도는 0.1 ~ 0.15 cm/yr로 추정되는데 이 값은 8,000년전 이후의 느린 해수면 상승속도를 반영한다고 볼 수 있다.

습지토양 및 연안퇴적물의 유기물질 및 영양물질 보유 특성 (Retention properties of organic matters and nutrients in wetland soils and coastal sediments)

  • 최근에 기후변화와 수질관리의 중요성이 증가하면서 습지토양의 탄소와 영양물질 보유 특성에 대한 관심이 증가되고 있다. 본 연구에서는 연안퇴적물과 인공습지, 자연습지(내륙, 하구, 갯벌)와 같이 연안퇴적물 및 다양한 유형의 습지토양에서의 유기물과 영양물질의 보유특성을 알아 보고자하였다. 또한 유기물, 영양물질, 입도에 따른 상관관계를 분석하여 이들 항목 간의 연관성을 살펴보았다. 연구 결과 유기물이나 영양물질의 보유정도가 습지의 유형에 따라 달라지는 것으로 나타났는데 내륙습지가 하구나 연안습지보다 질소의 보유 능력이 큰 것으로 나타났으며, 같은 육상생태계에서도 더 오랜 기간 동안 오염물질이 유입되고 자체적으로 유기물을 생산하여 축적하고 있는 자연습지에서 유기물과 질소의 농도가 인공습지보다 더 높은 것을 조사되었다. 연안지역의 경우 해수의 흐름이 원활한 하구나 갯벌 보다 해수의 유통이 느린 만 지역에서 유기물과 질소 농도가 높게 나타났다. 또한 화학적산소요구량은 총유기탄소 및 총질소농도와 높은 상관관계를 보여주어 화학적산소요구량 자료를 이용하여 해당 습지의 총유기탄소나 총질소 농도를 효과적으로 추정할 수 있음을 보여주었다.
