Precise ultrasonic coating and controlled release of sirolimus with biodegradable polymers for drug-eluting stent

  • In the current study, a drug-eluting stent coated with biodegradable polymers and sirolimus was developed by using an ultrasonic nanocoater and characterized in aspects of surface smoothness and coating thickness. In addition, in vitro release profiles of sirolimus by changing top coating layer with different biodegradable polymers were investigated. Smooth surfaces with variable thickness could be fabricated by optimizing polymer concentration, flow rate, nozzle-tip distance, gas pressure, various solvents and ultrasonic power. Smooth surface could be generated by using volatile solvents (acetone, chloroform, and methylene chloride) or post-treating with solvent vapor. Coating thickness could be controlled by varying injection volume or polymer concentration, and higher concentration could reduce the coating time while obtaining the same thickness. The thickness measurement was the most effectively performed by a conventional cutting method among three different methods that were investigated in this study. Release profiles of sirolimus were effectively controlled by changing polymers for top layer. PLGA made the release rate 3 times faster than PDLLA and PLLA and all top layers prevented burst release at the initial phase of profiles. Our results will provide useful and informative knowledge for developing drug-eluting stents, especially coated with biodegradable polymers.

Fabrication and Microstructure/Properties of Bulk-type Tantalum Material by a Kinetic Spray Process (Kinetic Spray 공정을 이용한 벌크형 탄탈륨 소재의 제조 및 미세조직/물성)

  • A bulk-type Ta material is fabricated using the kinetic spray process and its microstructure and physical properties are investigated. Ta powder with an angular size in the range $9-37{\mu}m$ (purity 99.95%) is sprayed on a Cu plate to form a coating layer. As a result, ~7 mm-sized bulk-type high-density material capable of being used as a sputter material is fabricated. In order to assess the physical properties of the thick coating layer at different locations, the coating material is observed at three different locations (surface, center, and interface). Furthermore, a vacuum heat treatment is applied to the coating material to reduce the variation of physical properties at different locations of the coating material and improve the density. OM, Vickers hardness test, SEM, XRD, and EBSD are implemented for analyzing the microstructure and physical properties. The fabricated Ta coating material produces porosity of 0.11~0.12%, hardness of 311~327 Hv, and minor variations at different locations. In addition, a decrease in the porosity and hardness is observed at different locations upon heat treatment.

  • Atomic layer deposition (ALD) is widely used as a tool for the formation of near-atomically flat and uniform thin films in the semiconductor and display industries because of its excellent uniformity. Nowadays, ALD is being extensively used in diverse fields, such as energy and biology. By controlling the reactivity of the surface, either homogeneous or inhomogeneous coating on the shell of nanostructured powder can be accomplished by the ALD process. However, the ALD process on the powder largely depends on the displacement of powder in the reactor. Therefore, the technology for the fluidization of the powder is very important to redistribute its position during the ALD process. Herein, an overview of the three types of ALD reactors to agitate or fluidize the powder to improve the conformality of coating is presented. The principle of fluidization its advantages, examples, and limitations are addressed.

Characteristics of sub-80 nm three-layered film manufactured by continuous roll-to-roll processes (연속 롤투롤 공정을 이용한 80 나노 이하의 3층 구조 필름 제조 및 특성)

  • Three-layer nano-coated films in applications for the back cover of mobile cellular phones were prepared utilizing a roll-to-roll continuous process. By introducing a coating layer with a ceramic/metal/ceramic three-layer structure, the inherent reflective properties of the metals were maintained while electrically insulating properties were maintained. The thickness of the composite coating layer on a large area PET film with a length of 1,500 nm and width of 500 nm was less than 60 nm, and a uniform thickness was maintained in all areas. The transmittance according to the wavelength range (240~1600 nm) of the nanocoating film gradually increases as the wavelength increases, and is about 48 % at 1,000 nm, which is within the infrared region, and about 35.5 % at 550 nm, which is within the visible region. These results meet the required level of coated backcover (< 40 %).

An Investigation on the Mechanical Behaviors of Lubricant and Coating to Improve the Drawability of Non-heat Treated Steels (열처리 생략강의 인발특성 향상을 위한 윤활제와 피막제의 기계적 거동 고찰)

  • In this research, we developed a pilot wire-drawing machine as well as wire end-pointing roller. Using these machines, we performed a pilot wire-drawing test at different coating material and lubricant when the reduction ratio is 10 %. To inversely compute the friction coefficient between the coating layer of wire and the surface of die for a specific lubricant, we carried out a series of three dimensional finite element analysis. Results show that the drawing force is varied with the coating material of wire at the same reduction ratio and lubricant. It is noted that the frictional coefficient in drawing is dependent on the coupled property of coating material and lubricant, indicating the best coating material for a given lubricant.

Electrochemical Properties of Cathode according to the Type of Sulfide Electrolyte and the Application of Surface Coating

  • The electrochemical performance of all-solid-state cells (ASSCs) based on sulfide electrolytes is critically affected by the undesirable interfacial reactions between oxide cathodes and sulfide electrolytes because of the high reactivity of sulfide electrolytes. Based on the concept that the interfacial reactions are highly dependent on the type of sulfide electrolyte, the electrochemical properties of the ASSCs prepared using three types of sulfide electrolytes were observed and compared. The Li2MoO4-LiI coating layer was also introduced to suppress the interfacial reactions. The cells using argyrodite electrolyte exhibited a higher capacity and Coulombic efficiency than the cells using 75Li2S-22P2S5-3Li2SO4 and Li7P3S11 electrolytes, indicating that the argyrodite electrolyte is less reactive with cathodes than other electrolytes. Moreover, the introduction of Li2MoO4-LiI coating on the cathode surface significantly enhanced the electrochemical performance of ASSCs because of the protection of coating layer. Pulverization of argyrodite electrolyte is also effective in increasing the capacity of cells because the smaller size of electrolyte particles improved the contact stability between the cathode and the sulfide electrolyte. The cyclic performance of cells was also enhanced by pulverized electrolyte, which is also associated with improved contact stability at the cathode/electrolyte. These results show that the introduction of Li2MoO4-LiI coating and the use of pulverized sulfide electrolyte can exhibit a synergic effect of suppressed interfacial reaction by the coating layer and improved contact stability owing to the small particle size of electrolyte.

Al2O3 Coating and Filling of Carbon Nanotubes

  • Aluminum oxide ($Al_2O_3$) nanotubes and nanorods were fabricated by coating and filling of multiwalled carbon nanotubes (MWNTs) with atomic-layer deposition (ALD). $Al_2O_3$ material was deposited on the MWNTs at a substrate temperature of $300^{\circ}C$ using trimethylaluminum and distilled water. Transmission electron microscopy, high resolution transmission electron microscopy, energy-dispersive X-ray spectroscopy, and selected area electron diffraction of the deposited MWNTs revealed that amorphous $Al_2O_3$ material coats the MWNTs conformally and that this material fills the inside of the MWNTs. These illustrate that ALD has an excellent capability to coat and fill any three-dimensional shapes of MWNTs conformally without producing any crystallites.

Preparation and Dissolution Characteristics of the Compression-Coated Controlled Release Tablet Exhibiting Three-step Release (압축코팅법에 의한 3단계 약물방출형 지속성제제의 제조 및 용출특성)

  • A novel oral controlled release tablet which may offer more uniform drug level in the body than simple zero-order was developed. The tablet is composed of three layers; outer film layer, middle part compression-coated hydroxypropylmethylcellulose (HPMC) matrix layer, and inner core layer. Each layer contains nicardipine HCl as a model drug. In vitro dissolution test showed that the tablet released the drug in clear three steps; a rapid initial release, followed by a constant rate of release, and then a second phase of fast release of drug. The dissolution characteristics could be modified easily by changing the grade of HPMC, thickness of matrix layer, content of methylcellulose in matrix layer, content of active ingredient in each layer. The pH of dissolution medium did not affect the release profile. This three-step release system is expected to raise the blood concentration rapidly to effective level and to maintain effective blood level longer than simple slow-release systems.

Hydroxyapatite+TiO2 Composite Sol Coating on Cp-Ti (Cp-Ti 표면의 Hydroxyapatite+TiO2 복합 Sol 코팅에 관한 연구)

  • In this study, $Hydroxyapatite+TiO_2(HAp+TiO_2)$ composite sol coatings on Cp-Ti substrates were deposited by using a sol-gel derived precursor. Prior to hydroxyapatite coating, the samples were micropolished and divided into three sets. The first set was coated with hydroxyapatite (HAp) directly on Cp-Ti. The second set was first coated with intermediate titania layer and then coated with HAp. The third set samples were coated with $HAp+TiO_2$ (50:50) composite sol. Each samples were predried at $200^{\circ}C$, and heat treated at $600^{\circ}C$. The formation of hydroxyapatite has been confirmed by XRD analyses and the substrate material was found to be oxidized with negligible amount of CaO in the coating. The NaOH treated samples showed the presence of rutile crystal. The SEM studies revealed surface morphologies of each samples. $HAp+TiO_2$ composite sol coating layer was found to be smooth. The bonding strength of each samples were calculated using pull out tests. The bonding strength of the $HAp+TiO_2$ composite sol coating on substrate was 29.35MPa.

A Study on the High Speed Characteristics of Plastic Mould Steel using Ball End Mill AlTiN Coated Layers (볼 엔드밀 AlTiN코팅 층수에 따른 플라스틱금형강의 고속가공에 관한 연구)

  • This paper investigated into process characteristics of AlTiN coated layers for machining to the direction of upper and lower in plastic mold material (KP-4) with the cemented carbide ball endmill with the diameter of 8mm coated AlTiN layers (1~4) step by step using machining center. The material used in experiments was KP-4 that was machined by three types of inclined angles; $15^{\circ}$, $30^{\circ}$ and $45^{\circ}$ As estimated mechanical properties of AlTiN coated layers, it was shown the most result in the condition of three layered coating that the coating that the coating depth, the hardness of the coated layer and the surface roughness of the coated layer were $13{\mu}m$, Hv 3027.3 and $0.042{\mu}m$, respectively. The cutting component was better at the condition of upper direction than that of lower direction in all experimental conditions and indicated to be less which the bigger angle of the material was increased the effective diameter of the tool.
