Orotic acid 투여 흰쥐의 조직 과산화지질 농도에 미치는 capsaicin의 영향 (Effort of Capsaicin on the Lipid Peroxidation in Tissues of Rats Fed Orotic Acid.)

  • Orotic acid 및 capsaicin (0.02 and 0.04%)을 단독 또는 병합 투여한 상태에서 흰쥐의 각 조직 중에 과산화지질 농도를 thiobarbituric acid reactive substances (TBARS) 측정법으로 검토하였다. 체중 증가량, 식이섭취량, 식이효율 및 뇌, 신장, 심장, 비장, 고환의 상대무게는 각 실험군간에 유의적인 차이가 없었다. 간의 상대적 무게는 정상군과 비교해서 OA군에서는 유의적으로 증가하였고, 0.02% 및 0.04% Cap군에서는 차이를 보이지 않았으나 OA+0.02% Cap군 및 OA+0.04% Cap군에서는 감소하는 경향을 보였다. 간 조직의 과산화지질 농도는 정상군에 비해 OA군에서 유의적으로 증가하였고, 이러한 증가는 0.04% capsaicin 병합투여에 의해 더욱 증가하는 것으로 나타났다. 그러나, 간 조직의 과산화지 질 농도는 정상군보다 0.02% Cap군에서는 유의적인 차이가 없었으나, 0.04% Cap군에서는 유의적으로 증가하여 농도별 차이를 보여주었다. 신장 조직의 과산화지질 농도는 정상군에 비해 OA군에서 증가하였고, OA를 함유한 OA+0.02% Cap군 및 OA+0.04% Cap군에서도 증가하였다. 그러나, 0.02% Cap군 및 0.04% Cap군에서는 오히려 감소하였다. 간의 비헴철 함량은 정상군에 비해 OA군과 capsaicin을 병합 투여한 OA +0.02% Cap군 및 OA+0.04% Cap군에서 유의적으로 증가하였다. 한편, 뇌, 심장, 비장 및 고환 조직의 과산화지질 농도는 각 실험군간에 유의한 차이가 없는 것으로 나타났다. 이상의 결과에서 OA투여 흰쥐의 간 조직에서 과산화지질 농도 증가와 동시에 비헴철 함량이 증가하였고, 이러한 증가는 0.04% capsaicin 병합투여에 의해 더욱 증가하는 것으로 나타났다.

녹차 추출물의 첨가 수준이 김치 발효 소시지의 젖산균, 산화안정성 및 향기에 미치는 영향 (Effect of Addition Level of Green Tea Extract on the Lactic Acid Bacteria, Oxidative Stability, and Aroma in Kimchi-fermented Sausage)

  • 본 연구는 녹차 추출물의 첨가 수준(0 ppm, 400 ppm, 800 ppm 및 1,200 ppm)이 김치 발효 소시지의 젖산균, 산화안정성 및 향기에 미치는 영향을 구명하고자 실시하였다. 시료는 $24^{\circ}C/RH$ 89%에서 pH 4.9까지(17시간) 발효시킨 후 $10^{\circ}C/RH$ 75-80%에서 6일간 건조시켰다. 젖산균수와 pH는 최종 6일째에 각각 7.5-7.7 Log CFU/g sausage 및 4.30-4.33이었으며, 녹차 추출물의 첨가 수준에 따른 차이가 없었다. TBARS(2-thiobarbituric acid reactive substances) 함량은 첨가 수준이 증가함에 따라 현저하게 억제되었다. 명도($L^*$)와 적색도($a^*$)는 녹차 추출물의 첨가에 의해 숙성 중 감소되었으며, 황색도($b^*$)는 증가되었다. 전자코(electronic nose)에 의한 향기 패턴은 첨가구들과 대조구간에 뚜렷한 차이를 보였다. 따라서 녹차 추출물의 첨가 수준의 증가는 김치 발효 소시지의 지방산화 안정성을 향상시켰다. 또한 첨가 수준과 관계 없이 녹차 추출물은 색깔 안정성을 저하시키고, 젖산균의 생장에 영향을 미치지 못하였으나, 향기를 변화시켰다.

Effect of Replacing Beef Fat with Poppy Seed Oil on Quality of Turkish Sucuk

  • Sucuk is the most popular dry-fermented meat product. Sucuk has a relatively high fat. Poppy seed oil as animal fat replacer was used in Turkish sucuk and effects of its use on sucuk quality were investigated. There was a significant (p<0.5) treatment × ripening time interaction for moisture, pH (p<0.05) and 2-thiobarbituric acid reactive substances (TBARS) values (p<0.01). Increasing poppy seed oil level decreased (p<0.05) TBARS values. Addition of poppy seed oil to the sucuks had a significant effect (p<0.01) on hardness, cohesiveness, gumminess, chewiness and springiness values. Cholesterol content of sucuks decreased (p<0.05) with poppy seed oil addition. Using pre-emulsified poppy seed oil as partial fat replacer in Turkish sucuk decreased cholesterol and saturated fatty acid content, but increased polyunsaturated fatty acids. Poppy seed oil as partial animal fat replacer in Turkish sucuk may have significant health benefits.

  • This study examined the antioxidant activity of methanol and water extracts from Orostachys japonicus A. Berger according to harvest times, by measuring electron donating ability(EDA), reducing power, superoxide dismutase(SOD)-like activity, thiobarbituric acid reactive substances(TBARS) and antioxidant activity within a linoleic acid emulsion. EDA increased proportionally with the extract concentration and the methanol extract had slightly stronger effects than the water extract. And reducing power and SOD-like activity were highest in the methanol extract. Overall, based on the data, the methanol extract of O. japonicus A. Berger harvested during $August{\sim}October$ presented the highest level of antioxidative activity and may be a good candidate as a natural antioxidant source.

근육식품에서 지방산화에 대한 피틴산, 저장기간 및 온도의 영향 (Effects of Phytic Acid Content, Storage Time and Temperature on Lipid Peroxidation in Muscle Foods)

  • Phytic acid, making up 1~5% of the composition of many plant seeds and cereals, is known to form iron-chelates and inhibit lipid peroxidation. Thiobarbituric acid reactive substances (TBARS), as an indication of lipid peroxidation, were measured in beef round, chicken breast, pork loin, and halibut muscle after the meats were stored for 0, 1, 3, 5, and 7 days at various temperatures [frozen (~2$0^{\circ}C$), refrigerator (4$^{\circ}C$), and room temperature ($25^{\circ}C$)]. Phytic acid effectively inhibited lipid peroxidation in beef round, chicken breast, halibut, and pork loin muscle (p<0.05). The inhibitory effect of phytic acid was dependent on concentration, storage time, and temperature. At frozen temperature, the inhibitory effect of phytic acid was minimal, whereas at room temperature, the inhibitory effect of phytic acid was maximal, probably due to the variation of the control TBARS values. At the concentration of 10 mM, phytic acid completely inhibited lipid peroxidation in all the muscle foods by maintaining TBARS values close to the level of the controls, regardless of storage time or temperature (p<0.05). The rate of lipid peroxidation was the highest in beef round muscle, although they had a close TBARS value at 0 day. Addition of phytic acid to lipid-containing foods such as meats, fish meal pastes, and canned seafoods may prevent lipid peroxidation, resulting in improvement of the sensory quality of many foods and prolonged shelf-life.

감나 폴리페놀이 흰쥐의 생체내 지질과산화에 미치는 영향 (Effect of Potato Polyphenolics on Lipid Peroxidation in Rats)

  • The total contents of polyphenolics in potatoes measured by Folin Denis method were 42~76mg/100g in fresh weight. A major phenolic component contained in potato polyphenolics was identified as chlorogenic acid(3.6~15mg/100g in fresh weight). The antioxidative effects of potato polyphenolics and chlorogenic acid on the lipid peroxidation of liver microsome were studied in vivo and in vitro systems by measuring the formation of thiobarbituric acid reactive substances(TBARS) and the content of urine 8 hydroxy deoxyguanosine(OHdG). The TBARS contents of liver showed an increase in the cholesterol diet compared to those in the normal diet. This trend, however, was minimized when potato polyphenolics and chlorogenic acid were supplemented in the cholesterol diet. On the other hands, urinary 8 OHdG contents showed a marked increase with the supplementation of potato polyphenolics in the cholesterol diet. However, there was a trend of marked decrease by the supplementation of chlorogenic acid. In vitro study, potato poly phenolics and chlorogenic acid effectively inhibited the formation of TBARS in liver microsomal system in a dose dependent manners. These results suggest that potato polyphenolics exerts an antioxidative activity in cholesterol fed rat liver.

Oxidative Stress in the Heart of Rats Infected with Trypanosoma evansi

  • This study was conducted to investigate the occurrence of oxidative stress in the heart tissue of rats infected with Trypanosoma evansi. Rats were divided into 2 groups (A and B) with 12 animals each, and further subdivided into 4 subgroups (A1 and A2, 6 animals/each; and B1 and B2, 6 animals/each). Animals in the groups B1 and B2 were subcutaneously inoculated with T. evansi. Thiobarbituric acid reactive substances (TBARS), superoxide dismutase activity (SOD), glutathione S-transferase activity (GST), reduced glutathione activity (GSH), and non-protein thiols (NPSH) in the heart tissue were evaluated. At day 5 and 15 post-infection (PI), an increase in the TBARS levels and a decrease in the SOD activity (P<0.05) were observed. GSH and GST activities were decreased in infected animals at day 15 PI (P<0.05). Considering the proper functioning of the heart, it is possible that the changes in the activity of these enzymes involved in the oxidative stress may be related, at least in part, in the pathophysiology of rats infected with T. evansi.

뽕나무가루 첨가 배지에서 배양한 버섯균사체 배양물의 자유라디칼 유도 산화 억제 (Inhibition of Free Radical-Induced Lipid Oxidation by the Extract from Submerged-Liquid Culture of Mushrooms in the Medium Containing Mulberry Tree Powders)

  • 뽕나무가루 첨가 배지에서 배양한 느타리버섯균사체 배양물의 조추출물이 자유라다칼로 유도한 linloeic acid의 산화를 대조구에 비해 75.9% 감소시켰고, mouse liver microsome산화에서도 NADPH/Fe$^{++}$ system에서 64.3% 감소시켰다. 이와 같은 효과는 조추출물로부터 용매분획한 분획물의 단독 효과보다 우수하였다. 상황 및 동충하초버섯균사체 배양물의 조추출물도 느타리 버섯균사체 배양물의 조추출물과 효과가 유사하였지만 다소 낮았다.

상엽(桑葉)추출물의 항산화효과에 대한 연구 (Studies on the Antioxidant Effects of Mori Folium Extract)

  • Objective : The objective of this study was to investigate the antioxidant effects of Mori Folium extract. Methods Total antioxidant status was examined by total antioxidant capacity(TAC) and total antioxidant response(TAR) against potent free radical reactions. The effect of Mori Folium extract was examined by measuring total phenolic content, concentration at which 1,1-dipheny1-2-picrylhydrazyl(DPPH) radical scavenging activity was inhibited, inhibitory effect on lipid peroxidation, and the effect on reactive oxygen species(ROS) generation. Results : 1. TAC and TAR of Mori Folium extract at the concentration of 5 mg/ml were 1.61 and 1.24 mM Trolox equivalents, respectively. 2. Total phenolic content of Mori Folium extract at the concentration of 5 mg/Ml was 1.70 mM gallic acid equivalent. 3. Concentration of Mori Folium extract at which DPPH radical scavenging activity was inhibited by 50% was 2.29 m9/m4 as compared to 100% by Pyrogallol solution as a reference. 4. The inhibitory effect of the extract on lipid peroxidation was examined using rat liver mitochondria induced by FeSO$_4$/ascorbic acid. Mori Folium extract at the concentration of 10 mg/ml significantly decreased thiobarbituric acid reactive substances(TBARS) concentration. The extract prevented lipid peroxidation in a dose-dependent 5. The effect of Mori Folium extract on reactive oxygen species(ROS) generation was examined using a celt-free system induced by hydrogen peroxide FeSO$_4$. Addition of 1 mg/ml of Mori Folium extract significantly reduced dichlorofluorescein(DCf) fluorescence. The extract caused concentration-dependent attenuation of the increase in DCF fluorescence, indicating that the extract significantly prevented ROS generation in vitro. Conclusion ; The antioxidant effects of Mori Folium extract seem to be due, at least in part, to the prevention offree radical-induced oxidation, fllowed by inhibition of lipid peroxidation.

층꽃나무(Caryopteris incana Miq.) 추출물의 생리활성 (Biological activities of extracts from Caryopteris incana Miq.)

  • 본 연구에서 층꽃나무(Caryopteris incana Miq.)는 항산화, 피부미백 및 주름개선 활성을 측정하는 데에 사용되었다. 층꽃나무의 생리활성은 고형분 함량보다 첨가된 total phenolic compounds의 농도에 의해 영향을 받는 것으로 확인되어 이후 모든 실험은 층꽃나무로부터 추출한 물과 80 % ethanol 추출물의 total phenolic compounds 함량을 조절하여 실시하였다. 층꽃나무 추출물의 항산화 활성은 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azinobis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS), antioxidant protection factor (PF), Thiobarbituric acid reactive substances (TBARs)를 측정하였다. $200{\mu}g/mL$의 농도에서 물과 ethanol 추출물로 DPPH free radical 소거능을 측정한 결과 각각 84, 92 %의 활성을 나타내었고, ABTS radical 소거능은 물과 ethanol 추출물 둘 다 약 99 %의 활성을 나타내었다. PF를 측정한 결과 물과 ethanol 추출물이 각각 1.56, 1.67 PF 값을 나타내었고, TBARs는 각각 62, 82 %의 활성을 나타내었다. 주름개선 및 피부미백 활성을 측정하였을 때, $200{\mu}g/mL$의 농도에서 80 % ethanol 추출물이 물 추출물보다 눈에 띄게 높은 효과를 나타내었다. 주름개선과 연관된 elastase와 collagenase 저해활성은 ethanol 추출물에서 각각 58, 89 %로 측정되었다. 피부미백과 연관된 tyrosinase의 저해활성은 ethanol 추출물에서 13 %로 측정되었고, 수렴효과는 50 %로 측정되었다. 이러한 결과로 보아, 층꽃나무 추출물은 항산화 활성, 주름개선 및 미백효과가 우수하므로 기능성 화장품 원료의 새로운 자원이 될 것으로 기대되었다.
