Cancer stem cell surface markers on normal stem cells

  • The cancer stem cell (CSC) hypothesis has captured the attention of many scientists. It is believed that elimination of CSCs could possibly eradicate the whole cancer. CSC surface markers provide molecular targeted therapies for various cancers, using therapeutic antibodies specific for the CSC surface markers. Various CSC surface markers have been identified and published. Interestingly, most of the markers used to identify CSCs are derived from surface markers present on human embryonic stem cells (hESCs) or adult stem cells. In this review, we classify the currently known 40 CSC surface markers into 3 different categories, in terms of their expression in hESCs, adult stem cells, and normal tissue cells. Approximately 73% of current CSC surface markers appear to be present on embryonic or adult stem cells, and they are rarely expressed on normal tissue cells. The remaining CSC surface markers are considerably expressed even in normal tissue cells, and some of them have been extensively validated as CSC surface markers by various research groups. We discuss the significance of the categorized CSC surface markers, and provide insight into why surface markers on hESCs are an attractive source to find novel surface markers on CSCs.

Numerical Analysis of Free Surface Flows Using Adaptable Surface Particle Method based on Grid System (격자기반 적합 표면입자법을 이용한 자유표면유동 수치해석)

  • In this study, the surface marker method, one of the particle tracking methods, used to track the free surface is extended to cover the more general cases easily including the collision and separation of the free surface. In surface particle method to redistribute particles effectively using the grid system, the free surface is composed of the sum of quadrilaterals having four curves where fixed markers are placed at ends of each curve. Fixed markers are used to know how curves are connected to each other. The position of fixed markers can move as the free surface deforms but all fixed markers cannot be deleted during all time of simulation to keep informations of curve connection. In the case of the collision or separtion of the free surface where several curves can be intersected disorderly, severe difficulties can occur to define newly states of curve connection. In this study, the adaptable surface parTicle method without fixed markers is introduced. Intersection markers instead of the fixed markers are used to define quadrilaterals. The position of the intersection markers is defined to be the intersection point between the free surface and the edge of the grid and it can be added or deleted during the time of simulation to allow more flexibilities. To verify numerical schemes, two flow cases are simulated and the numerical results are compared with other's one and shown to be valid.

Effects of Age and Gender on the Viability and Stem Cell Markers, mRNA, and Protein Expression of Bone Marrow-Derived Stem Cells Cultured in Growth Media

  • Purpose: Bone marrow has long been a source of primary cells. This study was performed to evaluate the effects of age and sex on the cellular viability and expression of stem cell markers of mRNA and on the protein expression of bone marrow stem cells (BMSCs) derived from healthy donors. Materials and Methods: Stem cells were isolated from human bone marrow and plated in culture plates. The shape of the BMSCs was observed under inverted microscope. Quantitative cellular viability was evaluated using a Cell-Counting Kit-8 assay. The expression of stem cell surface markers was tested and a series of quantitative real-time polymerase chain reaction (qRT-PCR) and Western blot was performed to evaluate the expression in each group. Result: The shapes of the cells at 20s, 30s, and 50s were similar to each other. No significant changes in cellular viability were noted among different age groups or sex groups. The BMSCs expressed CD44, CD73, and CD90 surface markers but did not express CD14 and CD34. There were no noticeable differences in CD surface markers among the different age groups. The expressions of CD surface markers were similar between men and women. No significant differences in the secretion of vascular endothelial growth factors (VEGFs) were noted at Day 3 between different age groups. qRT-PCR regarding the expression showed differences between the age groups. However, Western blot analysis showed a decrease in expression but did not reach statistical significance (P>0.05). Conclusion: This study clearly showed no significant differences in shape, cell viability, expression of stem cell surface markers, or secretion of human VEGF among different age groups. However, western blot analysis showed a tendency of age-related decrease which did not reach statistical significance. Collectively, autologous or allogeneic BMSCs should be meticulously applied to obtain optimal results regarding age and sex.

  • Tubular markers for road safety facilities are used to lead the driver's sight line and separate the lanes on the road. Such tubular markers are usually installed on the road and frequently hit by vehicles, they are accordingly requested to assure the product durability. The traditional evaluation method of tubular markers include only quality tests of the material properties. However, most of consuming agencies in charge of road management at fields have proposed problems on long-term performance of the products hit by vehicles under various weather conditions. Therefore, the objectives of this study are to develop the reliability test methods and equipments to simulate the product failures of tubular markers due to vehicle collision and wheel compression and the delamination and discoloration of reflection sheets attached on the surface of the products under high and low temperatures.

  • This paper describes a novel experiment that measures skin movement with respect to the flexional motion of a hand. The study was based on MR images in conjunction with CAD techniques. The MR images of the hand were captured in 3 different postures with surface markers. The surface markers attached to the skin where employed to trace skin movement during the flexional motion of the hand. After reconstructing 3D isosurfaces from the segmented MR images, the global registration was applied to the 3D models based on the particular bone shape of different postures. Skin movement was interpreted by measuring the centers of the surface markers in the registered models.

Effects of Serial Passage on the Characteristics and Chondrogenic Differentiation of Canine Umbilical Cord Matrix Derived Mesenchymal Stem Cells

  • Mesenchymal stem cells (MSCs) are often known to have a therapeutic potential in the cell-mediated repair for fatal or incurable diseases. In this study, canine umbilical cord MSCs (cUC-MSCs) were isolated from umbilical cord matrix (n = 3) and subjected to proliferative culture for 5 consecutive passages. The cells at each passage were characterized for multipotent MSC properties such as proliferation kinetics, expression patterns of MSC surface markers and self-renewal associated markers, and chondrogenic differentiation. In results, the proliferation of the cells as determined by the cumulative population doubling level was observed at its peak on passage 3 and stopped after passage 5, whereas cell doubling time dramatically increased after passage 4. Expression of MSC surface markers (CD44, CD54, CD61, CD80, CD90 and Flk-1), molecule (HMGA2) and pluripotent markers (sox2, nanog) associated with self-renewal was negatively correlated with the number of passages. However, MSC surface marker (CD105) and pluripotent marker (Oct3/4) decreased with increasing the number of subpassage. cUC-MSCs at passage 1 to 5 underwent chondrogenesis under specific culture conditions, but percentage of chondrogenic differentiation decreased with increasing the number of subpassage. Collectively, the present study suggested that sequential subpassage could affect multipotent properties of cUC-MSCs and needs to be addressed before clinical applications.

Familial Clustering and Its Associated Factors in Carriers of Hepatitis B Surface Antigen (B형 간염의 가족집적성과 전파경로에 관한 연구)

  • To evaluate the possible route of intrafamilial transmission among carriers of hepatitis B surface antigen ($HB_sAg$), epidemiologic and serologic data were obtained on 107 household contacts of 35 carriers of HBsAg and on 71 household contacts of 25 controls who were negative for serologic markers of hepatitis B virus. The HBsAg prevalence was 26.5% among the contacts of carriers compared to 0.0% among the contacts of controls. And the combined prevalence for all hepatitis markers was 48.5% among the contacts of carriers compared to 26.0% among the contacts of controls(p<0.05). Especially the offspring of carriers showed significantly higher risk in the combined prevalence for all hepatitis markers (p<0.05). There were no significant relationship between HBV infection and past history like acupuncture, transfusion, operation and tattooed. Factors associated with the risk of intrafamilial transmission of HBV were not found in the sharing of household articles such as razor, towel, drinking glass, nail clippers, toothbrush and tableware.

  • This paper proposes a real-time geometric correction system based on a projector to project digital images onto deformable surface. Markers use to trace lots of corresponding points would spoil the projected image when the projector projects a digital image onto the surface because they leave marks on the surface. In addition, it is difficult to build a real-time geometric correction system since bottlenecks occur through the process of the geometric correction for projecting images. In this paper, we use invisible infrared markers and a vertex shader of GPU using Cg TookKit of NVIDIA in order to eliminate disadvantage and bottlenecks in the process of markers recognition so that it is possible to project natural correction images in real-time. As a result, this system overlays an interactive virtual texture onto the real paper by using the geometric transformation. Therefore, it is possible to develop variation of AR(Augmented Reality) based on digital contents systems.

In Vitro Selection of Cancer-Specific RNA Aptamers

  • In this study, nuclease-resistant RNA aptamers that are specific for Jurkat T leukemia cells were selected by a subtractive systemic evolution of ligands by exponential enrichment (SELEX) method. A randomized nuclease-resistant RNA library was incubated with normal peripheral blood mononuclear cells (PBMC) in each round to preclude RNAs that recognize the common cellular components on the surface of normal and cancer cells. The precluded RNAs were used for the selection of Jurkat T cell-specific aptamers, and the specific RNAs were then gradually enriched from start to the following selections. After 16 rounds of the subtractive SELEX, the selected aptamers were found to preferentially bind to Jurkat T cells, but not to the normal PBMC, evidenced by fluorescence-activated cell sorting analysis. Thus, the subtractive SELEX can be used to identify ligands to cancer-specific biological markers without prior knowledge of the nature of markers. The aptamers could be applied to specific cell sorting, tumor therapy, and diagnosis, and moreover, to find cancer cell-specific markers.

  • In this paper, we improve pre-processing method using markers and propose accurate lattice extraction of elemental image array using minimal markers to correct distortion. We propose a method that almost equal performance of existing method and be more simple calculation. For our method, we did computational experiments and compared with reconstructed images.
