Biocontrol Activity of Acremonium strictum BCP Against Botrytis Diseases

  • Biological control activity of Acremonium strictum BCP, a mycoparasite on Botrytis cinerea, was examined against six plant diseases such as rice blast, rice sheath blight, cucumber gray mold, tomato late blight, wheat leaf rust, and barley powdery mildew in growth chambers. The spore suspension of strain BCP showed strong control activities against five plant diseases except against wheat leaf rust. On the other hand, the culture filtrate of A. strictum BCP was effective in controlling only cucumber gray mold and barley powdery mildew. Further in vivo biocontrol activities of A. strictum BCP against tomato gray mold were investigated under greenhouse conditions. Control efficacy of the fungus on tomato gray mold increased in a concentration-dependent manner. Treatment of more than $1{\times}10^6$ spores/ml significantly controlled the disease both in tomato seedlings and in adult plants. The high disease control activity was obtained from protective application of the strain BCP, whereas the curative application did not control the disease. Foliar infections of B. cinerea were controlled with $1{\times}10^8$ spores/ml of A. strictum BCP applied up to 7 days before inoculation. In a commercial greenhouse, application of A. strictum BCP exhibited the similar control efficacy with fungicide procymidone (recommended rate, $500{\mu}g/ml$) against strawberry gray mold. These results indicate that A. strictum BCP could be developed as a biofungicide for Botrytis diseases under greenhouse conditions.

  • In order to study the genetic system of the blast resistant varieties, the conidial suspension of mutant races of T-2$^{+t}$, N-2$^{+t}$, C-8$^{+t}$ was inoculated at 4-5 leaf stage by injector for F_2 seedlings from the crosses between seven resistant varieties and four maker lines easily detectable at seedling stage. The results are summarized as follows; 1. The fertility of cross between Semi-dwaf testers and Indica resistant varieties except Carreon was about 74 percents. 2. The segregation modes of resistance varied with varieties and blast races. However, the resistance was expressed as dominance in all cases. Tetep, Tadukan and Carreon showed more complicated segregation for resistance than that of the bred lines. 3. For blast races used, four segregation ratios such as 3:1, 9:7, 13:3 and 37:27 were found in the Tatukan, Tetep, and IR747, and three segregation ratios such as 3:1, 13:3 and 15:1 in the Carreon, and two segregation ratios of 3:1 and 13:3 with Suweon 287, Suweon 288, and Iri342. 4. In the segregation of the resistance to the each races, the ratios of 3:1, 13:3, 15:1 were fitted to T-2$^{+t}$, and the ratios of 3:1, 13:3, 9:7 and 37:27 to N-2$^{+t}$ and C-8$^{+t}$. 5. Suweon 287, Suweon 288 and Iri342 carried one simple dominant gene and inhibitor gene was considered in some cross combinations. Meanwhile Tadukan, Tetep and IR747 seemed to carry one to three resistant genes, and in some cross combinations, the expression of these genes were simple dominant, inhibiting, duplicating and complimentary action. 6. Resistance genes to blast races, T-2$^{+t}$, N-2$^{+t}$ and C-8$^{+t}$ in the Tadukan, Tetep, Carreon, Suweon 287, Suweon 288 and Iri342 were found to be independent with the linkage group of II(lg), VIII(la), XI(bc), and XII(gl).bc), and XII(gl).

Genetic Stability of Magnaporthe oryzae during Successive Passages through Rice Plants and on Artificial Medium

  • Genetic instability of the rice blast fungus Magnaporthe oryzae has been suggested as a major factor underlying the rapid breakdown of host resistance in the field. However, little information is available on the mechanism of genetic instability. In this study, we assessed the stability of repetitive DNA elements and several key phenotypic traits important for pathogenesis after serially transferring two isolates though rice plants and an artificial medium. Using isolate 70-15, we obtained a total of 176 single-spore isolates from 10 successive rounds of culturing on artificial medium. Another 20 isolates were obtained from germ tubes formed at the basal and apical cells of 10 three-celled conidia. Additionally, 60 isolates were obtained from isolate KJ201 after serial transfers through rice plants and an artificial medium. No apparent differences in phenotypes, including mycelial growth, conidial morphologies, conidiation, conidial germination, appressorium formation, and virulence, or in DNA fingerprints using MGR586, MAGGY, Pot2, LINE, MG-SINE and PWL2 as probes were observed among isolates from the same parent isolate. Southern hybridization and sequence analysis of two avirulence genes, AVR-Pita1 and AVR-Pikm, showed that both genes were also maintained stably during 10 successive generations on medium and plants. However, one reversible loss of restriction fragments was found in the telomere-linked helicase gene (TLH1) family, suggesting some telomere regions may be more unstable than the rest of the genome. Taken together, our results suggest that phenotype and genotype of M. oryzae isolates do not noticeably change, at least up to 10 successive generations on a cultural medium and in host plants.

Identification and Annual Change of Races of Pyricularia oryzae in Korea (한국에 있어 벼도열병균레이스의 분화 및 년차적변동)

  • One thousand forty-four isolates of Pyricularia oryzae obtained during the period 1962-1974 front blast-infected rice samples from all rice-growing areas of Korea were tested on the Japanese differential set of twelve rice varieties. Twenty-seven races of the fungus were differentiated. The number of races falling in groups T,C and N were 4, 15 and 8, respetively. Since only Japonica type varieties had been cultivated widely before 1966, N-race groups were found to be most Prevalent with a frequency of about $85\%$ of the total isolates. After 1966, T-and C-race groups gradually increased to $21\%$ and $42\%$ respectively, as a result of the widespread cultivation of Tongil bred from a cross between IR8 x(Yukara x T(N)1. Before 1966, a few T, and C-race groups occurred only in the Suweon region; since 1966, most of the races differentiated have been uniformly distributed throughout the country regardless of region. The most prevalent races in Korea were N-2 and C-8. The variety Tetep was resistant to all races previously identified in Korea, but a new race (T-d) to which Tetep is susceptible has been found each year since 1973. It appears likely that races capable of attacking Indica types of rice will become increasingly prevalent and a programme of continuous screening is needed to detect new races as soon as they appear.

Efficient Target-Site Assay of Chemicals for Melanin Biosynthesis Inhibition of Magnaporthe grisea

  • A rapid and efficient assay to determine melanin biosynthesis inhibition of Magnaporthe grisea, a causal agent of the rice blast, by chemicals was developed. Wells in 24-well plates were loaded with spore suspension of the fungus and three known melanin biosynthesis inhibitors of KC10017, tricyclazole, and carpropamid. Subsequent color changes of mycelia and culture media in the wells were observed 7 days after incubation. The wells treated with KC10017 (an inhibitor of polyketide synthesis step and/or pentaketide cyclization step) became colorless, whereas tricyclazole (an inhibitor of 1, 3, 8-trihydroxynaphthalene reductase) or carpropamid (an inhibitor of scytalone dehydratase)-treated wells exhibited red color. They did not show any inhibitory effect on fungal growth. The inhibition of reaction steps prior to 1, 3, 6, 8-tetrahydroxynaphthalene formation was easily determined by colorless medium and mycelia. However, it was impossible to distinguish between inhibition of reduction steps and inhibition of dehydration steps by colors of the cultures. It was accomplished through HPLC analysis of the melanin biosynthesis-involving pentaketide metabolites accumulated by the inhibitors. Through screening of a number of synthetic chemicals using the in vitro assay, we could find a novel chemical group of melanin biosynthesis inhibitor.

  • Magnaporthee grisea causes the devastating blast disease of rice. Entensive research has been conducted on infection mechanisms, particularly on appressorium formation and penetration, of this fungus during the last decade. However, the role(s) of cell-wall-degrading enzymes (CWDEs) on pathogenesis is not clearly demonstrated at molecular level. Many CWDES in plant pathogenic fungi including M. grisea are redundant; that is, there are multiple genes encoding enzymes with a similar or overlapping spectrum of activities. It is laborious to isolate all of the genes encoding related enzymes and to construct mutants lacking all 9f them. Thus, we considered alternative strategies to address the role of CWDEs in pathogenesis. Since expression of CWDE genes Is repressed by a simple sugar, as the first step, we cloned a Snfl (sucrose non-fermenting) gene (MgSnf1) from M. grisea. The predicted amino acid sequence showed a high identity with other Snf1 genes from various fungi. To elucidate molecular function of MgSnf1, a transformant lacking MgSnf1 was created by targeted gene replacement. En glucose, sucrose, and xylan the MgSnf1 mutant grew normally but in pectin and complex media, it grew slower than wild type. Expression of various CWDEs in MgSnf1 mutant was investigated and found that expression of some CWDEs is repressed. However, no significant difference was observed in conidial germination, appressorium formation, and pathogenicity in MgSnf1 mutant. However, MgSnf1 functionally complemented a yeast MgSnf1 mutant. These results suggest that MgSnf1 is involved in regulation of CWDEs and MgSnf1 is dispensable in pathogenicity of M. grisea.

Antibiotic and Phytotoxic Activities of Ophiobolins from Helminthosporium Species

  • Twenty isolates of Helminthosporium species were obtained from various grass plants and tested for controlling efficacy on the development of plant diseases. An isolate of Helminthosporium sp. TP-4 was chosen and six antibiotic substances were purified from cultures of the fungus by repeated silica gel column chromatography and preparative thin-layer chromatography. They were identified as ophiobolin a, 6-epiophiobolin A, 3-anhydroophiobolin A, 3-anhydro-6-epiophiobolin A, iphiobolin B, and iphiobolin I mainly by mass spectrometry and nuclear magnetic resonance spectrometry. Ophiobolins inhibited the growth of a grampositive bacterium Streptomyces griseus, but were not active against gram-negative bacteria. They also showed an antifungal activity. In in vivo tests, iphiobolin B exhibited potent controlling activities against rice blast, tomato late blight, and wheat leaf rust with control values more than 90% and 70% at concentration of $500\mu\textrm{m}$/ml and 100 ${\mu}{\textrm}{m}$/ml. Ophiobolin A and 6-epiophiobolin A controlled the development of wheat leaf rust more than 80% at concentrations of 100 /ml and $500\mu\textrm{m}$/ml respectively. 3-Anhydro-6-epiophiobolin A was not active against any plant disease. On the other hand, the A-series ophiobolins other than 3-anhydroophiobolin A showed stronger phytotoxic activity in a leaf-wounding assay using 8 plant species than those of 3-anhydroophiobolin A, ophiobolin B, and ophiobolin I. The results indicate that there is little correlation between antifungal activity and phytotoxicity of ophiobolins.

Prediction and Annotation of ABC Transporter Genes from Magnaporthe oryzae Genome Sequence (벼도열병균 게놈서열로부터 ABC transporter 유전자군의 예측 및 특성 분석)

  • Magnaporthe oryzae is destructive plant-pathogenic fungus and causes rice blast. The pathogen uses several mechanisms to circumvent the inhibitory actions of fungicides. ATP-binding cassette (ABC) transporters are known to provide protection against toxic compounds in the environment. PC facilitated bioinformatic analysis, particularly with respect to accessing and extracting database information and domain identification. We predicted ABC transporter genes from the M. oryzae genome sequence with computation and bioinformatics tools. A total of thirty three genes were predicted to encode ABC transporters. Three of thirty three putative genes corresponded to three known ABC transporter genes (ABC1, ABC2 and ABC3). Copy numbers of the ABC transporter genes were proven by Southern blot analysis, which revealed that twenty genes tested exist as a single copy. We amplified the DNA complementary to RNA corresponding to eleven of these by reverse transcriptase polymerase chain reaction.

An Histopathological Investigation on the Resistant Tong-Il Cultivar by Inoculating Rice Blast Fungus, Pyricularia oryzae Cav. (수도(水稻) 도열병(稻熱病)에 저항성(抵抗性)인 통일품종(統一品種)에 대한 침입생리(侵入生理)에 관(關)한 연구(硏究))

  • In order to investigate histopathologically the nature of varietal resistance and infection process of the rice bast fungus, Pyricularia oryzae. this experiment was undertaken by using the resistant cultivar Tongil and susceptible cultivars Norin No.6 and Jinheung in 1973 to 1974. 1) It was found that appressorium of the fungus forms not only at 4 hours after incubation under the favorable conditions but also peak of appressorial formation is at 48 hours treatment. Physical stimulus known to be a definite factor for appressorial formation. The optimum temperature range for appressorial infection was at $24^{\circ}C$ to $28^{\circ}C$, and pH was between 4.8 to 8.0 with 6.8 as the optimum. 2) Although percent of appressorial formation on the leaves of resistant Tongil and susceptiible Norin No.6 were only slightly different, there was a remarkable difference between resistant and susceptible cultivars with regard to percent of hyphal infection and index for hyphal extension. Index of hyphal extension was 1.6-2.7 in Tonyil. while in susceptible cultivar was 3.4-6.6. The rate of discoloration of infected cells, a indication of hypersensitivity, was greater in the resistant than in susceptible cultvar. 3) Therefore, it could be concluded that resistance of Tongil cultivar may be attributable to a higher degree of mechanical barriers as well as a higher level of antifungal substance accumulation.

  • Effect of percent penetration and hyphal growth on lesion formation was evaluated at 23/15C (day/night), 29/21C regimes and greenhouse condition (33-23C) using three rice cultivars, Aichi-asahi, Toyotama and Yamabiko with 6 isolates of Pyricularia oryzae. Percent penetration at 72hr and 96hr after inoculation was highly significant with lesion formation at any temperature regime. At the same time, number of appressoria that exceeded hyphal growth value 11 was also highly significant with lesion formation. However, number of appressoria that exceeded hyphal growth value 30 was significant only at 72hr after inoculation at 29/21C regime. In general, statistical fitness of the model was higher at 23/15C regime than at 29/21C regime or a greenhouse condition. These series of phenomenon were more apparent in a susceptible cultivar Aichi-asahi.
