Nematicidal Activity of Bikaverin and Fusaric Acid Isolated from Fusarium oxysporum against Pine Wood Nematode, Bursaphelenchus xylophilus

  • Pine wood nematode, Bursaphelenchus xylophilus, causes pine wilt disease in a number of Pinus species, which is one of the most serious plant diseases in forest, Korea. In the course of a search for nematicidal substances from endophytic fungi, Fusarium oxysporum EF119 out of the 23 fungal strains tested showed the strongest activity to B. xylophilus. Two nematicidal substances were isolated and identified as bikaverin and fusaric acid. Fusaric acid showed somewhat higher nematicidal activity against B. xylophilus than bikaverin; fusaric acid and bikaverin, at $100{\mu}g/ml$, killed B. xylophilus with mortality values of 50% and 43%, respectively. In addition, both compounds acted synergistically. This is the first report on the nematicidal activity of bikaverin and fusaric acid.

신갈나무로부터 유래된 추출물의 Botrytis cinerea 균주에 대한 항균활성 (Antifungal Activity of the Quercus Mongolica Extracts Against Botrytis cinerea)

  • 목질자원으로부터 Botrytis속 곰팡이병에 대하여 항균활성을 나타내는 추출물을 탐색하고자 본 연구는 시도되었다. 신갈나무를 고압 전처리하고 이로부터 열수, 메탄올, 에탄올을 이용하여 목질 추출물을 획득하였다. Botrytis속 곰팡이 3종은 $20^{\circ}C$$25^{\circ}C$의 생장온도에서 최적 생장을 하였으며 pH 5.0과 7.0에서 가장 양호한 균사 생장을 나타냈다. 열수 추출물이 Botrytis속에 대하여 가장 우수한 항균활성을 나타내는 것을 확인하였다. 열수추출물로부터 항균물질을 분리하기 위하여 n-hexane : chloroform : ethyl acetate : formic acid (12 : 17 : 8 : 0.2, v/v/v/v)를 용제로 하는 silica-gel column chromatography와 TLC를 사용하였으며, 3종 추출물들은 HPLC 분석을 통하여 화합물을 확인하였으며 UV Spectrophotometer를 이용하여 열수 추출물로부터 6개의 fraction group을 분리 하였다. 6개의 fraction group의 항균활성 test결과, fraction group I과 II는 Botrytis cinerea에 대하여 가장 높은 항균활성을 나타냈다. Fraction group의 HPLC 분석결과, 3개의 유효물질을 확인하였으며 이 3가지 물질이 항균활성에 영향을 미친다고 추측된다.

온대낙엽수림에서 초식곤충의 계절과 고도에 따른 먹이활동 양상 연구 (Seasonal and Elevational Pattern of Herbivore's Feeding Activity in Temperate Deciduous Forest)

  • 본 연구는 온대활엽수림에서 초식곤충의 먹이 활동이 계절 및 고도에 따라 어떻게 변하는 가를 알아보기 위하여 수행되었다. 조사는 지난 3년간(2011~2013) 지리산국립공원의 고도가 다른 세 지역(피아골, 시암재, 노고단)에서 이루어졌다. 조사방법은 각 지역에 서식하는 수종(3종)을 선정하여 4월부터 6월까지 1~2주마다 초식곤충의 식흔 활동을 식흔지수를 이용하여 관찰하였다. 조사 결과 고도가 가장 낮은 피아골(${\approx}300m$)에서 가장 먼저 식흔이 나타났고, 중간고도인 시암재(${\approx}900m$)에서는 가장 많은 식흔이 조사되었다. 고도가 가장 높은 노고단(${\approx}1300m$)에서는 식흔이 가장 늦게 나타났지만 짧은 시간에 급속히 식흔의 총량이 증가하였다. 또한 기존에 발표된 연구를 토대로 한 결과 초식곤충의 활동시기는 조류(곤줄박이)의 첫 산란일과 거의 비슷한 것으로 나타나 식물-곤충-2차 소비자(조류)와 밀접한 관련을 보여준다. 기후 환경이 변함에 따라 초식곤충 활동의 기간이 바뀌게 될 것이며 이러한 먹이사슬의 변동을 감시할 수 있는 지속적인 장기 생태 모니터링이 필요하다.

Differential Expression of a Chimeric nos-npt II Gene in 9 Years Old Hybrid Poplars (Populus koreana x P. nigra)

  • The expression of a chimeric transgene (nos-npt II) has been examined in 9 years old transgenic poplars (Populus koreana x P. nigra) growing in a nursery. The expression of the gene in twenty six independentely transformed plants were examined by 1) enzyme (NPT II) assay, 2) RT-PCR, and 3) resistance to kanamycin. High NPT II activities in young leaves of all the transformed plants were found even without a selection pressure for antibiotics for 9 years. However, the activity varied with the positions of leaves in the stem in that young leaves showed higher activity than did mature tissues. When leaf segments were cultured in the presence of 150 mg/l kanamycin, only those from young leaves produced vigorously growing callus. However, as in the case of NPTII assay, the leaf segments from mature leaves did not form callus well on the media. RT-PCR with nptII specific primers also showed that amplification products were observed only when RNAs from young tissues were used. The total RNA gel showed that while RNA in young leaves are relatively stable and in a large quantity, those in old leaves were mostly degraded. All the above results suggest that the gene is transcriptionally active only in young tissue even though it is attached to a constituitive promoter. Therefore, the expression of foreign gene in poplar plants seemed to be affected by the metabolic state of the cells and thus vary greatly with the developmental stages and the age of tissue.

Electron Donating Ability of MeOH Extracts from Three Korean Mistletoes

  • This experiment was accomplished to investigate antioxidative activity of Korean mistletoe by organic solvents partitioning of methanol extact of 3 Korean mistletoes, fractionationing on column chromatography, and evaluation the reduction of a free radical ${\alpha}$,${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl. Butanol partition of Loranthus yadoriki MeOH extract showed higher electron donating ability than ${\alpha}$-tocopherol. It was thought phenolic compounds including gallic acid account for antioxidative activity, on execution sub-fractionation, electron donating ability evaluation, and GC/MS analysis, but further studies on what are major actives must be investigated exactly.

제주 곶자왈 숲 자연 놀이 체험 학습이 초등학생의 환경 감수성 변화에 미치는 영향 (Effects of the Nature Game Experience Learning on Schoolchild's Environmental Sensitivity Change in Jeju Gotjawal Forest)

  • The aims of this study are to develop and apply a Nature Game program for the ecological experience learning in Jeju Gotjawal forest and analyze the environmental sensitivity change of the students. The results obtained in 4th graders of A elementary school of Seogwipo-city are as follows: It was found that attention, susceptibility and practice will of environment protection for forest environment of the experimental class were significantly higher than those of the comparison class. And it was found that interest and curiosity into forest environment were also elevated in the results of the qualitative evaluation, suggesting that we could get the effect of developed Nature Game program on ecological experience learning. If various ecological experience learning programs considering the regional-specific characteristics are developed to cause students' interest and curiosity, students would come to participate in the protection activity of environment aggressively a little more.

Cloning and Expression of cDNA Encoding a Cysteine Protease Inhibitor from Clamworm and Its Possible Use in Managing Anoplophora glabripennis Motschulsky (Coleoptera: Cerambycidae)

  • A cDNA encoding a cysteine protease inhibitor (CPI) was isolated from the cDNA library of clamworm Perinereis aibuhitensis Grube. The deduced amino acid sequence analysis showed that the protein had 51%, 48%, and 48% identity with Zgc:153129 from Danio rerio, cystatin B from Theromyzon tessulatum, and the ChainA, stefin B tetramer from Homo sapiens, respectively. The gene was cloned into the intracellular expression vector pET-15b and expressed in Escherichia coli. The recombinant CPI (PA-CPI) was purified by affinity chromatography on Ni-charged resin and ion-exchange chromatography on DEAE-Sepharose FF. The relative molecular mass of PA-CPI was 16 kDa as deduced by SDS-PAGE. Activity analysis showed that the recombinant protein could inhibit the proteolytic activity of papain. A constitutive and secretive expression vector was also constructed, and the cDNA encoding CPI was subcloned into the vector for extracellular expression. Western blotting analysis results showed that the PA-CPI was secreted into the medium. Bioassay demonstrated that E. coli DH5${\alpha}$ harboring pUC18ompAcat-CPI showed a significant difference in mortality to the Asian longhorned beetle Anoplophora glabripennis compared with untransformed E. coli DH5${\alpha}$ and control.

Neuroprotective effect of Aster yomena ethanolic extract in HT-22 and SK-N-MC cells based on antioxidant activity

  • The antioxidant potentials of ethanolic extracts derived from Aster yomena (A. yomena) were evaluated by assessing their total phenolic and flavonoid contents and radical scavenging activities. Our findings revealed that the 60% ethanolic extract of A. yomena exhibited the most robust antioxidant properties among all extracts tested. Specifically, the IC50 values for the 2,2'-azino-bis (3-ethyl benzothiazoline-6-sulfonic acid) and 1,1-diphenyl-2-picrylhydrazyl radical scavenging activities of the 60% ethanolic extract from A. yomena were determined to be 1,640.30 ㎍/mL and 2,655.10 ㎍/mL, respectively. Moreover, the inhibitory effect on malondialdehyde increased with the 60% ethanolic extract from A. yomena. To assess the neuroprotective effects, we examined the impact of the 60% ethanolic extract from A. yomena against H2O2-induced cytotoxicity in HT-22 (mouse hippocampal neuronal cell line) and SK-N-MC (human neuroblastoma cell line) cells. The results demonstrated a significant improvement in cell viability and reduced intracellular oxidative stress. Furthermore, the major bioactive compounds present in the 60% ethanolic extract from A. yomena were identified as chlorogenic acid and rutin through high-performance liquid chromatography (HPLC) analysis.

Classification of cardiotocograms using random forest classifier and selection of important features from cardiotocogram signal

  • In obstetrics, cardiotocography is a procedure to record the fetal heartbeat and the uterine contractions usually during the last trimester of pregnancy. It helps to monitor patterns associated with the fetal activity and to detect the pathologies. In this paper, random forest classifier is used to classify normal, suspicious and pathological patterns based on the features extracted from the cardiotocograms. The results showed that random forest classifier can detect these classes successfully with overall classification accuracy of 93.6%. Moreover, important features are identified to reduce the feature space. It is found that using seven important features, similar classification accuracy can be achieved by random forest classifier (93.3%).

Studies about Monoamine Oxidase Inhibitory Activities of Korean Green Tea (Teae sinensis L.) Harvested from Different Time and Location

  • This study was designed to investigate the nervous sedative effects of green tea. The sedative effect was evaluated by examination of Monoamine oxidases (MAOs) inhibitory activity in vitro in the brain and liver of rat fed on green tea cultivated and harvested from the different regions and periods. It showed that methanol extracts of green tea inhibited significantly the brain MAO-A activity. Especially late harvested green tea extracts showed potential inhibitory activity. The liver MAO-B activity was also inhibited by all of the green tea extracts with strong intensity. This study confirmed that major compounds of green tea such as catechin, epigallocatechin-3-gallate (EGCG) and L-theanine, which were well known for the main bioactive components in the tea plants, were not associated with the MAO inhibitory activities of green tea. These results suggested that a MAO inhibition activity comes from other minor tea components we have to search in the future.
