Expression of OsPTs-OX Transgenic Rice in Phosphate-Deficient Condition (무인산 조건에서 OsPTs 유전자 도입 형질전환벼의 인산흡수 반응)

  • It needs to develop high phosphate-available rice that is able to minimize environmental pollution caused by phosphate fertilizer. Then we have transformed 4 rice transporter genes, OsPT(Oryza sativa Phosphate Transporter)1, OsPT4, OsPT7 or OsPT8, to rice (Oryza sativa cv. Dongjin) via Agrobacterium-mediated transformation. We tested adaptation in the P-deficient condition of Dongjin (parental) and each transgenic line in the pot and the field conditions. Definite physiological changes have been observed in OsPTs transgenic lines including culm length, root formation and heading date. Phosphate uptake at harvesting stage was about three times higher in OsPT1-OX (overexpression) and OsPT4-OX than in Dongjin (wt) without P application. There are no variations in total phosphate-content of brown rice of OsPT1-OX in spite of high phosphate uptake. Practically the expression of OsPT1 has contributed to stabilize grain production without P fertilization in rice cultivation than Dongjin.

Studies for Development of Freeze - Flow Process (Freeze - Flow Process 개발을 위한 기초연구)

  • To develope a process by which liquid foods can be stored in the liquid state at the frozen storage temperature, suitable cryoprotectants were selected. Orange juice, chosen as an example of liquid foods, was stored with combined cryoprotectants at $-15^{\circ}C$, and quality changes of orange juice during storage were evaluated. Among 7 cryoprotectants tested, NaCl solution had lower initial freezing point than others, and initial freezing points of glucose, fructose, glycerol, propylene glycol and citric acid were close to each other. Considering flavor quality of orange juice, cryoprotectants suitable for reducing freezing point of orange juice were glucose, fructose, glycerol, and citric acid. Combined cryoprotectants for reducing freezing point of 3 and 4 folds concentrated orange juice to $-15^{\circ}C$ consisted of 10% glucose, 8% frutose, 4.6% glycerol and 3% citric acid, and 5.5% glucose, 4.5% fructose, 4.6% glycerol, and 3% cirtric acid, respectively. When destruction of ascorbic acid, sedimentation volume and sensory flavor score of orange juice stored with combined cryoprotectants at $-15^{\circ}C$ and the control stored at $-18^{\circ}C$ were compared, there were no significant differences. These results indicated that liquid foods with suitable combined cryoprotectants could be stored at $-15^{\circ}C$ or below in the liquid state without adverse effect on quality of the stored products.

A Phenology Modelling Using MODIS Time Series Data in South Korea (MODIS 시계열 자료(2001~2011) 및 Timesat 알고리즘에 기초한 남한 지역 식물계절 분석)

  • This study aimed to analyze spatio-temporal trends of phenological characteristics in South Korea by using MODIS EVI. For the phenology analysis, we had applied double logistic function to MODIS time-series data. Our results showed that starting date of phenology seems to have a tendency along with latitudinal trends. Starting date of phenology of Jeju Island and Mt. Sobeak went back for 0.38, 0.174 days per year, respectively whereas, Mt. Jiri and Mt. Seolak went forward for 0.32 days, 0.239 days and 0.119 days, respectively. Our results exhibited the fluctuation of plant phonological season rather than the change of phonological timing and season. Starting date of plant phenology by spatial distribution revealed tendency that starting date of mountain area was late, and basin and south foot of mountain was fast. In urban ares such as Seoul metropolitan, Masan, Changwon, Milyang, Daegu and Jeju, the phonological starting date went forward quickly. Pheonoligcal attributes such as starting date and leaf fall in urban areas likely being affected from heat island effect and related warming. Our study expressed that local and regional monitoring on phonological events and changes in Korea would be possible through MODIS data.

Quality Attributes of Fresh-Cut Green Onion as Affected by Rinsing and Packaging (절단 대파의 품질특성에 미치는 세척 및 포장재의 효과)

  • Quality attributes of fresh-cut green onion(Allium fistulosum L.) as affected by rinsing and packaging were investigated in terms of flesh weight, color, viable cell counts, sensory properties during storage at $10^{\circ}C$. Fresh green onions were trimmed, cut, and rinsed with cold water(approximately $5^{\circ}C$) as well as chlorine solution(100 mg/L) and then packaged in low density polyethylene film pouches of $63\;{\mu}m$ thickness. Rinsing treatments with cold water or chlorine solution did not significantly influence changes in microbial populations but sensory characteristics, resulting in cut green onions of better visual quality as compared to the control without rinsing. Fresh-cut green onions were also rinsed with cold water and packaged in sealed bags of low density polyethylene films with different thickness(22, 36, $63\;{\mu}m$), and stored at $10^{\circ}C$ for 18 days. Thickness of polyethylene film was a significant factor for microorganisms populations and sensory attributes. Mesophilic aerobic bacterial count after 13 days for the control, packed in punched film bags, was $3.07{\times}10^6}$ CFU/g, while those for samples in hermetically sealed bags ranged only $1.74{\sim}2.02{\times}10^5}$ CFU/g. Gas composition within the sealed packages changed from normal air to about $1.3{\sim}5.4%\;O_2$ and $4.0{\sim}8.0%\;CO_2$ after 13 days of storage. Particularly, the visual sensory quality of cut green onion samples was retained better in polyethylene film bags of $63\;{\mu}m$ thickness(gas transmission rate: 600 $O_2\;mL/day{\cdot}m^2{\cdot}atm;\;2,500\;CO_2\;mL/day{\cdot}m^2{\cdot}atm$) than in the others.

The Changes of Plasma Atrial Natriuretic Peptide Concentrations During Waking and Sleep in Patients with Obstructive Sleep Apnea Syndrome (폐쇄성 수면 무호흡증후군 환자에서 각성시와 수면중의 혈중 Atrial Natriuretic Peptide 농도 변화)

  • Objectives : Patients with obstructive sleep apnea syndrome(OSAS) often complain of nocturnal enuresis. There are a few reports that OSAS patients have altered renal function, and there are some evidences that the increased release of atrial natriuretic peptide(ANP) may be involved in the pathogenesis of nocturnal urinary symptoms of OSAS patients. In this study, we measured plasma ANP concentrations during waking and sleep in OSAS patients and normal controls to investigate whether there were differences of ANP concentrations between OSAS patients and normal subjects. Methods : 27 patients with OSAS and 10 normal subjects were studied. All subjects underwent a full-night polysomnographic study. Venous blood samples were separately drawn during waking and sleep. Plasma ANP concentrations were measured using radioimmunoassay. Results : In OSAS patients, ANP concentrations during sleep($122.9\;{\pm}\;29.9pg/ml$) were significantly higher than ANP concentrations during waking($60.2\;{\pm}\;5.8pg/ml$)(p < 0.05). However, in normal subjects, there was no significant difference between ANP concentrations during waking($59.2\;{\pm}\;5.7pg/ml$) and sleep($69.6\;{\pm}\;3.0pg/ml$)(p > 0.05). There was no significant difference of ANP concentrations during waking between OSAS patients($60.2\;{\pm}\;5.8pg/ml$) and normal controls($59.2\;{\pm}\;5.7pg/ml$)(p > 0.05), and also there was no significant difference during sleep between OSAS patients($122.9\;{\pm}\;29.9pg/ml$) and normal subjects($69.6\;{\pm}\;3.0pg/ml$)(p > 0.05). Plasma ANP concentrations during sleep showed significant positive correlations with apnea index(r = 0.3846, p < 0.05) and respiratory disturbance index(r = 0.3939, p < 0.05) in OSAS patients. Conclusion : These data suggest that, in OSAS patients, plasma ANP concentrations during sleep are significantly higher than plasma ANP concentrations during waking, and there is a positive correlation between the plasma ANP concentration during sleep and the severity of sleep apnea.

Changes in Microbial Contents and Volatile Basic Nitrogen of Pheasant Meat Products during Storage (꿩고기 가공제품의 저장중 미생물과 휘발성 염기태질소의 변화)

  • To evaluate the storage characteristics of pheasant meat products, contents of aerobic, anaerobic and food poisoning bacteria as well as VBN were measured during storage at various temperatures. 1. During the storage Escherichia coli and food poisoning bacteria including Salmonella, Shigella and Staphylococcus were not detected from any of the pheasant meat products. 2. Total plate counts of aerobic and anaerobic bacteria increased with storage temperature, showing more than $10^6CFU/g$ of most pheasant meat products within 5~10 days of storage at $20^{\circ}C$ and $30^{\circ}C$. However, frank sausage, loin ham, pressed ham and salad showed less than $10^5CFU/g$ in 20~30 days of storage at $20^{\circ}C$. 3. When stored at $10^{\circ}C$, smoked product, electric roasted product and pressed ham showed the bacterial counts of more than $10^4CFU/g$ within 10 days of storage. Frank sausage, loin ham and salad, however, showed less than $10^3CFU/g$ in 10 days of storage at $10^{\circ}C$. 4. VBN contents of smoked product, electric roasted product and frank sausage exceeded edible limit of 20 mg%, showing more than 40 mg% and 80 mg% within 5 days and 10 days, respectively, of storage at $10^{\circ}C$. In contrast, loin ham, pressed ham and salad had the VBN of less than 20 mg% in 10 days of storage at $10^{\circ}C$. In summary, while pheasant meat products in general appear to be prone to microbial growth, loin ham and salad are thought to have a longer storage period than others, showing about 10 days of preservation at $10^{\circ}C$. Products other than loin ham and salad are suggested to be stored frozen or refrigerated at below $10^{\circ}C$.

  • Objectives: This clinical study evaluated the effect of light activation on the whitening efficacy and safety of in-office bleaching system containing 15% hydrogen peroxide gel. Materials and Methods: Thirty-three volunteers were randomly treated with (n = 17, experimental group) or without light activation (n = 16, control group), using Zoom2 white gel (15% $H_2O_2$, Discus Dental) for a total treatment time of 45 min. Visual and instrumental color measurements were obtained using Vitapan Classical shade guide and Shadepilot (DeguDent) at screening test, after bleaching, and 1 month and 3 month after bleaching. Data were analyzed using t-test, repeated measure ANOVA, and chi-squared test. Results: Zoom2 white gel produced significant shade changes in both experimental and control group when pre-treatment shade was compared with that after bleaching. However, shade difference between two groups was not statistically significant (p > 0.05). Tooth shade relapse was not detected at 3 months after bleaching. The incidence of transient tooth sensitivity was 39.4%, with being no differences between two groups. Conclusions: The application of light activation with Zoom2 white gel system neither achieved additional whitening effects nor showed more detrimental influences.

Surgical Outcome of Patients with Ischemic Cardiomyopathy Selected by the Results of Myocardial Viability by Preoperative F-18 FDG PET (F-18 FDG 양전자단층촬영을 이용한 생존심근 평가 후 수술한 허혈성 심근병증 환자의 수술 결과)

  • Purpose: We investigated the operative outcome after bypass surgery in patients selected using viability criteria on F-18 FDG PET. Materials and Methods: Rest-24hr delay redistribution imaging of Tl-201 SPECT and F-18 FDG PET were performed in 11 patients. Seven of these 11 patients (6 men, 1 woman) were evaluated to have viable myocardium by F-18 FDG PET. Changes in symptoms and left ventricular ejection fraction (LVEF) after operation were evaluated. Results: In seven of 11 patients, a significant amount of viable myocardium was found on F-18 FDG PET and Tl-201 SPECT. Severity of both chest pain and dyspnea improved markedly in all patients. Mean LVEF improved from 22% to 32%. Conclusion: F-18 FDG PET could be used to select the patients who will benefit from coronary artery bypass surgery.

Effectiveness of Home Economics Class on self-esteem and stress of middle school students (가정과수업이 중학생의 자아존중감과 스트레스에 미치는 효과)

  • This study investigated the effectiveness of home economics class on self-esteem and stress of middle school students. Data were collected from self-reported inventory of the middle school students in Y city of Gyeonggi province and 165 data copies were used for analyses. Data were analyzed for frequency, percentage, mean, standard deviation, Cronbach's ${\alpha}$, t-test, corelation and dependent t-test using SPSS/PC 18.0 program. The results were as follows. First, the average of positive self-esteem was 2.74 and that of negative self-esteem was 1.90 on a 4 Likert scale. The range of stress level was 1.88~2.48 on a 5 Likert scale. Life satisfaction was 3.90 and helpfulness of home economics class was 3.89 on a 5 point Likert scale, which means moderately high. Second, examining differences in variables according to gender, there were statistically significant differences in stress regarding family for both pre- and post-tests. Third, helpfulness of home economics class had positive correlation with positive self-esteem and life satisfaction and had negative correlation with negative self-esteem and stress, which means that home economics class enhance life satisfaction and positive self-esteem and lessen negative self-esteem and stress of middle students. Fourth, as a result of comparison of pre and post-tests, positive self-esteem, life satisfaction, and helpfulness of home economics class increased statistically after home economics classes, while stress regarding school life decreased statistically. Even though students' growth and changes resulting from learning other subjects were not controlled, it was meaningful that effectiveness of home economics class were examined and helpful information for enhancing perception of home economics education were provided.

Influence of Ultrasonification on Extraction Yield and Chemical Property of Green Tea Infusion (초음파 처리가 녹차 침출액의 추출 수율 및 화학적 특성에 미치는 영향)

  • The objective of this work was to investigate the influence of ultrasonification on extraction yield and chemical properties of green tea infusion. Changes in total soluble matter(TSM), vitamin C, total phenolic compounds, flavonols, catechins, caffeine, free amino acids contents in green tea infusion(GTI) influenced by ultrasonification at $60^{\circ}C$ of extraction temperature for 1, 5, 30, and 60 min were investigated. The amount of infused TSM increased about 5.3% by ultrasonification for 60min. Vitamin C contents also increased 0.21, 0.16, 0.31 mg/g from 1 to 30 min by ultrasonification. However, vitamin C decreased from 2.47 to 2.22 mg/g at 60min. Total phenol compounds contents increased about 10~13 mg/g on all extraction times by ultrasonification. Flavonols such as, myricetin, quercetin, kaempferol were increased to doubled contents as an influence of ultrasonification. Catechins such as, EGCG, EGC, ECG, EC, (+)-C and caffeine contents showed same tendency as the results of vitamin C. On the other hand, result of free amino acids showed different tendency. All amounts of free amino acids did not increase by ultrasonification. Consequently, content of bioactive compounds such as, vitamin C, total phenolic, flavonols and catechins in green tea infusion were influenced by ultrasonification.
