Molecular genetic markers were genotyped used to detect chromosomal regions which contain economically important traits such as growth traits in pigs. Three generation resource population was constructed from a cross between the Korean native boars and Landrace sows. A total of 193 F2 animals from intercross of F1 were produced. Phenotypic data on 7 traits, birth weight, body weight at 3, 5, 12, 30 weeks of age, live empty weight were collected for F2 animals. Animals including grandparents (F0), parents (F1), offspring (F2) were genotyped for 194 microsatellite markers covering from chromosome 1 to 18. Quantitative trait locus analyses were performed using interval mapping by regression under line-cross model. To characterize presence of imprinting, genetic full model in which dominance, additive and imprinting effect were included was fitted in this analysis. Significance thresholds were determined by permutation test. Using imprinting full model, four QTL with expression of imprinted effect were detected at 5% chromosome-wide significance level for growth traits on chromosome 1, 5, 7, 13, 14, and 16.
We analyzed the contents of 12 types of minerals such as calcium in muscle from various species. Thereafter we observed changes of the concentrations according to age, part and sex in major domestic animals. The concentrations of calcium, iron, sodium, potassium, magnesium and zinc were high respectively whilst the content of cobalt, chromium, copper, manganese, molybdenum and nickel were low respectively in the muscle. The concentration of calcium was high in duck and dog but low in pig and horse. Also high level of sodium content was detected in dog and the content of potassium, iron showed high level in horse compared with the content in other animals. In 6 types of microminerals as cobalt, the level of muscle were no more than 1 ppm showing very low content in all animal, but $2.99{\pm}0.85$ppm of copper in duck was an exceptional case. According to the age some species showed small range of variation centering on macrominerals and there was no remarkable change in microminerals. Distribution of minerals was different according to the part and the variation was very diverse compared with other factors such as age and sex. Additionally, the content of minerals in muscle was higher in female than in male chicken and duck. In conclusion, the difference of the content of minerals according to the species was mainly focused on macrominerals.
The objective of this study was to evaluate the usefulness of detection of PRRSV and PRRSV-specific antibodies in oral fluids for monitoring of PRRSV infection in endemic farms. The level of PRRSV and anti-PRRSV antibodies in serum and oral fluids was evaluated in five age groups of pigs (6, 9, 12, 16 weeks of age and gilts). The samples (25 serums and 5 oral fluids/per a farm) were collected from 5 different farms endemically infected by PRRSV. Both serum and oral fluid samples were tested for PRRSV by quantitative reverse-transcription polymerase chain reaction (qRT-PCR) and for anti-PRRSV antibodies by two commercial PRRSV ELISA kits. ELISA mean s/p ratios (2.98 vs 1.63) and positive rate (84.0% vs 68.8%) of the oral fluid samples showed significantly higher levels but had similar patterns to the seroprofile of the blood samples. The PRRSV positive rate of oral fluid and serum samples was 40.0% and 44.0% respectively. In conclusion, the use of oral fluids for PRRS monitoring in endemic farms is strongly recommended.
In this study, we investigated the effects of mixing on the aggressive behavior of commercially housed pigs. The behavioral patterns of 36 groups of pigs (a total of 360 animals) were observed over 3 consecutive days directly after weaning ($25{\pm}1.2$ days of age), and 25 and 50 days later with the aid of video technology. Fight latency and total duration and frequency of fighting were significantly different among the age groups. The aggressive behaviors decreased in 75-day old pigs if compared to 25- and 50-day old animals. Moreover, dominance index (DI) was higher in 25-day old and lower in 75-day old pigs. A comparison of dominant (DI>0) and submissive (DI<0) pigs showed significant differences (p<0.05) for major aggressive behaviors in all age groups. Dominant pigs were involved in more aggressive interactions, had longer fights, and initiated more fights than submissive pigs. Post-mixing aggressive behavior was altered by previous experience of mixing. Aggressive behavior and DI are suitable methods for analyzing the effects of mixing on commercially housed growing pigs.
인지질의 기능적 특성을 조사하기 위하여 체중이 $180{\sim}220g$ 되는 Sprague-Dawley계 암컷 랫드를 사용하여 대조군(식이 제한군)과 청둥오리 인지질의 투여군으로 실험하였다. 대조군은 전 실험기간 동안 먹이를 주었고 청둥오리 인지질 첨가군은 임신 중에 임의로 주고 수유 3주 동안 제한사료를 주었다. 생후 3주에 새끼 쥐들을 어미 쥐로부터 분리시켰으며, 이후 4주 동안 임의로 사료를 먹도록 하였다. 각 군에서 0, 1, 2, 3, 5 및 7주에 새끼 쥐의 체중을 측정하고 희생하여 뇌 무게, 뇌의 protein, phospholipid, cholesterol 함량을 측정하였다. 그 결과 체중은 생후 1, 2, 3주에 대조군 및 인지질 첨가군 간에 체중이 증가하여 매우 유의적인 차이를 나타내었고, 생후 5주까지 그 차이가 나타났으나, 후기에는 유의적인 차이를 보이지 않았다. 뇌 무게는 생후 2, 3, 7주에 증가하여 유의적인 차이를 나타냈다. 뇌의 protein 함량은 생후 2주부터 대조군과 인지질 첨가군 사이에 증가하여 유의적인 차이를 나타내었으며, 그 차이는 식이회복 기간 중에도 나타냈다. 뇌의 phospholipid, cholesterol 함량은 생후 2, 3주에 두 군 사이에 유의적인 차이를 나타냈으며, 식이 회복 후기인 생후 7주에는 phospholipid(PL) 만이 점차적으로 증가하였다. 쥐를 대상으로 한 수동 회피 검사에서는 대조군에 비하여, Duck-PL 및 Pig-PL 첨가군의 수동 회피율이 우수하였다.
During 2 years from Octorber 1992 to April 1994, prevalence of general respiratory diseases and atrophic rhinitis in the pig herds located in the Western Chungnam was investigated, and isolation of B. bronchiseptica was attempted for the pigs manifested with the clinical signs of atrophic rhinitis(AR). The isolates were characterized and identified in aspects of biochemical properties, antigenicity, drug sensitivity and pathogenicity. The results obtained through the experiments are summarized as follows; 1. During 2 years of investigation, the overall prevalence of the general respiratory diseases in the pi8 herds in Western Chungnam was 35.3%, consisting of 35.1% in the pig farms and 38.8% in a slaughter house. The prevalence by age groups accounts for 9.2% in adults, 44.7% in rearings and 25.3% in sucklings. By farm size, The highest prevalence of 56.5% was observed in the smallest farm with 1 to 200 heads. 2. The prevalence of clinical cases of artrophic rhinitis was recorded by 12.7% in the group that is the sows and piglets vaccinated, 28.9% in the group that is the sows only vaccinated and 39.8% in the group of the non-vaccinated groups. In the slaughter house, 53(24.8%) of 214 pigs examined exhibit the AR lesions. 3. A total of 189 strains of B. bronchiseptica were isolated from the pig herds. Isolation rates were 12.6% in the group that is the sows and piglets vaccinated, 34.1% in the group that is the sows only vaccinated and 45.7% in the group of the non-vaccinated groups. Isolation rate in the specimen from the slaughter house was 93( 43.5% ) of 214 pigs examined. Of the AR-non-vaccinated group, the piglets aged bet- ween 61 to 90 days revealed the highest isolation rate of 58.5%. 4. The titers of antibody against B. bronchiseptica were measured by tube agglutination test. The group that is the sow and piglet-vaccinated showed the highest titer of 640-2, 560 in sow and 640longrightarrow5, 120 in piglet. The group that is the sows only-vaccinated revealed 640-2, 560 in sows and 640-1, 280 in piglets. Both of the vaccinated groups showed 100% positive reaction. The group of the non-vaccinated sho-wed relatively lower titer of 0-1, 280 in both of sows and piglets. The positive rate of the sera obtained from the slaughter house was 53.3% with the antibody titer of 0-1, 280. 5. Biochemical and serological properities of 189 isolates were very similar to those of the reference B. bronchiseptical phase I type, indicating that most of isolates are B. bronchiseptica phase I type. 6. In antimicrobial drug susceptibility, 87.3% of 189 isolates was susceptible to chloramphenicol, 79.9%, to amikacin, 64.6%, to cephalothin and less than 35.4% to others. 7. In agar-gel immunodiffusion and SDS-PAGE analysis, the isolates presented the identical antigenicity and protein profiles to the reference standard strains. 8. The whole cells and bacterial filtrates of the isolates were inoculated to guinea pigs and mice. The isolates showed the hish pathogenicity and dermonecrotoxiciy.

The present study investigated the effects of varying durations of nursery diets differing in percentages of milk products on growth performance of pigs during the nursery phase (NP) and subsequent grow-finish phase (GFP) to find the feasibility of reducing the use of nursery diets containing costly milk products. A total of 204 21-d-old weanling female and castrated male pigs were subjected to one of three nursery phase feeding programs differing in durations on the NP 1 and 2 and GFP diets containing 20%, 7%, and 0% lacrosse and 35%, 8%, and 0% dried whey, respectively, in 6 pens (experimental units) for 33 d: HIGH (NP 1, 2 and 3 diets for 7, 14, and 12 d), MEDIUM (NP 2 and 3 for 14 and 19 d), and LOW (NP 2 and 3 and GFP 1 for 7, 14, and 12 d). Subsequently, 84 randomly selected pigs [14 pigs (replicates)/pen] were fed the GFP 1, 2 and 3 diets during d 54-96, 96-135, and 135-182 of age, respectively. The final body weight (BW) and average daily gain (ADG) of nursery pigs did not differ among the HIGH, MEDIUM, and LOW groups (14.8, 13.3, and 13.7 kg in BW and 273, 225, and 237 g in ADG, respectively). The average daily feed intake during the nursery phase was greater (p < 0.01) in the HIGH group than in the MEDIUM and LOW groups, whereas the gain:feed ratio did not differ across the treatments. The BW on d 182 and ADG during d 54-182 were greater in the HIGH and MEDIUM groups vs. the LOW group (110.0, 107.6, and 99.6 kg in BW, respectively; p < 0.01). The backfat thickness and carcass grade at slaughter on d 183 did not differ across the treatments. In conclusion, the MEDIUM program may be inferior to the commonly used HIGH program in supporting nursery pig growth. Nevertheless, the former appears to be more efficient than the latter in production cost per market pig whereas the LOW program is thought to be inefficient because of its negative effect on post-nursery pig growth.
The primary aim of this study was to investigate biosecurity practices in pig farms and to determine the major risk factors associated with PCV2 infection for a sampled swine population in Korea. To this end, we analyzed data from a cross-sectional study of 296 farrow-to-finish farms, which was conducted between March and September 2014 to explore the prevalence of swine disease at farm level. Face-to-face interviews by on-site visit of trained veterinarians were conducted with the farm owners or managers using a standardized questionnaires with information about basic demographical data and management practices. Farms were classified as negative or positive through the use of infection profiles that combined data on serological testing including PCR antigen test result, antibody titer and sero-conversion pattern at each age category taking into account vaccination status. Data were analyzed using multivariate ordinal logistic regression. Results from this study indicated that biosecurity level of the farms was considered not good given low compliance of the biosecurity programs and facilities in the farm: off-site removal of dead stocks (7%), off-site location of storage facility for incoming feeds (12.6%), off-site pick-up location for finishers (19.3%), restrictions on feed supplier vehicles for farm entrance (19.6%), restriction of finisher trucks entering the farm (22.4%), and restriction on manure disposal trucks entering the farm (26.4%). In the final model (n = 255), allowance of finisher truck driver to the pig unit had increased risk of infection (OR = 2.4, 95% CI 1.22-4.67) whereas farms with a sign forbidding the entrance had decreased risk of infection (OR = 0.19, 95% CI 0.10-0.58). Further comprehensive research with larger sample size is required to better understand the multifactorial characteristic that some predisposing risk factors that were not available in this study. To the best knowledge of the authors, this was the first study to use empirical data to report risk factors associated with PCV2 infection in the Korean pig farms. Results from the current study could be used to decide optimal biosecurity measures to reduce the impact of PCV2 infection to farmers and policy makers.
The study of pigs as a human disease model has been conducted in neuroscience. But the morphological development of pig brain by using MRI is rare. The purpose of this study is to determine whether cerebellum maintains consistent proportion to other brain regions in aging. Clinically healthy sixteen micropigs, 1, 2, 4, and 8 months were studied. The micropigs were anesthetized with isoflorane. MRI was acquired using a 0.3T system. To figure out development of ratio that allowed identification of normal cerebellum size, we measured the area of the cerebellum, brainstem, and forebrain from the mid-sagittal brain images on T1W. Mid-sagittal cross-sectional area (CSA) of total brain, forebrain, brainstem, and cerebellum were expressed as absolute values and also as percentages which were compared between the four age groups of micropigs for the purpose to define the effect of age on brain morphometry. It was found that there was not a significant difference in the percentage of the brain occupied by an individual region between groups although the absolute CSA differed significantly among age groups. There was no effect of age on the ratio between the cerebellum and total brain in 4 age groups. The normal size of cerebellum changes during brain development maintained a consistent ratio to other brain regions in normal micropigs. The ratio of CSA quantified on the mid-sagittal MR images offers a suitable method to detect presence of cerebellar anomalies in micropigs.
G protein-coupled receptor 43 (GPR43) is a newly-discovered short-chain free fatty acid receptor and its functions remain to be defined. The objective of this study was to investigate the function of GPR43 on lipolysis. We successfully cloned the GPR43 gene from the pig (EU122439), and measured the level of GPR43 mRNA in different tissues and primary pig adipocytes. The expression level of GPR43 mRNA was higher in adipose tissue and increased gradually with adipocyte differentiation. Then we examined GPR43 mRNA level in different types, growth-stages and various regions of adipose tissue of pigs. The results showed that the expression level of GPR43 mRNA was significantly higher in adipose tissue of obese pigs than in lean pigs, and the expression level also gradually increased as age increased. We further found that the abundance of GPR43 mRNA level increased more in subcutaneous fat than visceral fat. Thereafter, we studied the correlation between GPR43 and lipid metabolism-related genes in adipose tissue and primary pig adipocytes. GPR43 gene had significant negative correlation with hormone-sensitive lipase gene (HSL, r = -0.881, p<0.01) and triacylglycerol hydrolase gene (TGH, r = -0.848, p<0.01) in adipose tissue, and had positive correlation with peroxisome proliferator-activated receptor $\gamma$ gene ($PPAR_{\gamma}$, r = 0.809, p<0.01) and lipoprotein lipase gene (LPL, r = 0.847, p<0.01) in adipocytes. In addition, we fed different concentrations of docosahexaenoic acid (DHA) to mice, and analyzed expression level changes of GPR43, HSL and TGH in adipose. The results showed that DHA down-regulated GPR43 and up-regulated HSL and TGH mRNA levels; GPR43 also had significant negative correlation with HSL (low: r = -0.762, p<0.01; high: r = -0.838, p<0.01) and TGH (low: r = -0.736, p<0.01; high: r = -0.586, p<0.01). Our results suggested that GPR43 is a potential factor which regulates lipolysis in adipose tissue, and DHA as a receptor of GPR43 might promote lipolysis through down-regulating the expression of GPR43 mRNA.
