The Cryoprotective Effect on Frozen-thawed Boar Semen of Egg Yolk Low Density Lipoproteins

  • In order to protect the spermatozoa against cold shock, hen egg yolk is widely used as a cryoprotective agent in semen freezing extenders for domestic animals. The protective action of yolk is largely presumed to be due to low density lipoproteins (LDL). The effects of LDL on sperm quality of bull and northern pike (Esox lucius) after freezing-thawing have been reported, but no study has been made to evaluate the effect of LDL on boar sperm motility and other characteristics. The experiment was carried out to investigate the effect of LDL on the freezing of boar sperm in 0.25 ml straws. The aim was to evaluate the quality of boar spermatozoa cryopreserved in the presence of LDL. Motility of semen cryopreserved in LDL was analyzed and compared to semen cryopreserved with Tris-citric acid-glucose (TCG) and Tris-citric acid-fructose (TCF), two basic freezing extenders containing egg yolk. Similarly, acrosome and plasma membrane integrity were also evaluated and compared to semen cryopreserved with TCG and TCF. Analysis of sperm quality after freeze-thaw showed that the motility, acrosome and plasma membrane integrity were improved with LDL in the extender, as compared to the TCG and TCF. The highest post-thaw integrity of acrosome and plasma membrane and motility were obtained with 9% LDL (w/v). Consequently, the optimum LDL concentration in the extender was 9%. It is also suggested that the concentration of LDL addition is important for the effect on boar sperm protection during freezing and thawing. The percentage of motile spermatozoa was significantly higher after freezing in 9% LDL than in TCG and TCF 54.4% versus 30.4% and 30.1% (p<0.05), respectively. The integrity of acrosome and plasma membrane were also significantly higher at 70.3% and 50.5% respectively with semen frozen in 9% LDL extender compared to TCG at 37.8% and 30.3% and TCF at 36.4% and 29.9%, respectively (p<0.05),. In conclusion, we propose that extender containing LDL extracted from hen egg yolk could be used as a cryoprotective media with a better efficiency than TCG and TCF. LDL improved boar semen quality, allowing better spermatozoa motility, acrosome and plasma membrane integrity after the freeze-thaw process. Furthermore, we found out that the extender with 9% LDL concentration significantly enhanced motility, acrosome and plasma membrane integrity of boar sperm after freezing and thawing.

  • Recently, construction disasters in thawing season are increasing due to the ground collapse and it is related to the improper compaction during winter season. Compaction at sub-zero temperature reduces the compaction effect and the research of mechanical properties of thawed soil after winter compaction can be used as useful data to understand the behavior of the ground in the thawing season. On the other hand, the research interest in the unsaturated soil mechanics has been increasing in the field of the geotechnical engineering. Therefore, it is expected that the research of unsaturated characteristics under the compaction of sub-zero temperature and freezing & thawing condition provides information to the researchers in the related fields. Therefore, in this research, unsaturated soil-water characteristics test and unsaturated uniaxial compression test were conducted on the specimens compacted at sub-zero temperature and continuous freezing & thawing condition to investigate change of unsaturated characteristics and matric suction. Based on the test results, the change of matric suction and the decrease of strength and stiffness were observed with the freezing & thawing conditions. Especially in case of the weathered soil, the strength and matric suction were significantly reduced with lower temperature and more repetition of freezing & thawing cycles. This result implies that compaction of sub-zero temperature and freezing & thawing cycles will have a considerable influence on the stability of the ground.

Changes of Characteristics in Red Pepper by Various Freezing and Thawing Methods (홍고추의 저장온도 및 해동조건에 따른 물리화학적 특성 변화)

  • The development of an effective long-term storage protocol for harvested fresh pepper is urgently required to increase the market for pepper products. The protocol must minimize quality loss, so that the product may be used either as a spice or as a raw material for processed pepper products, both in the home and in food processing plants. We investigated the optimum size of pepper fruits, freezing temperatures, storage periods, and thawing methods, to establish an optimum storage protocol. This study was conducted not only to develop freezing and thawing methods for long term storage of harvested red pepper, but also to develop processed pepper products utilizing the stored pepper. We aimed to expand the pepper products market and to increase the incomes of pepper growers. Whole red pepper, sliced red pepper, and crushed red pepper were frozen and stored at $-5^{\circ}C,\;-20^{\circ}C,\;or\;-40^{\circ}C$. The soluble solid content and the vitamin C level showed maximal stability at $-40^{\circ}C$, although total free sugars decreased on storage at all temperatures tested. Such Changes were more marked at $-5^{\circ}C$ than at the other(lower) temperature tested. The vitamin C content of whole red pepper was higher than that of sliced red pepper or crushed red pepper. Room-temperature thawing resulted in twice the drip loss seen on low temperature($5^{\circ}C$) thawing or microwave oven thawing. Brown discoloration was a serious problem with room temperature thawing. Total free sugars were higher in samples thawed at low temperature or in the microwave oven, compared to the level seen after room-temperature thawing. pepper samples thawed at low temperature scored higher in sensory tests than samples thawed at room temperature.

  • This paper reviews the most important steps that have generated consistent progress in principles and developmental progress of embryo cryopreservation, and also study on freezing procedure and its application by conventional method and current improved method for freezing procedure and its appilcation of embryo cryopreservation in farm animals. Four were of particular interest: 1.The transport of water across the ccli membrane (zona pellucida) during freezing and thawing accordinglyplays a role in determing whether the celi survives. This movement of water is controlied mainly by extracellular phase changes and by the nature and concentration of any cryoprotective agent present. Therates of cooling, freezing and warming, and the intervals over which they are applied are further decisi've factors in determining whether a cryopreservation procedure allows survival after thawing. 2.The first successful deep freezing experiments with sheep morula and blastocysts during the seventies were based on the early procedures used for mouse embryos.Current research during the eighties is developed with the aim of simplifying and improving current procedures such as one-step dilution and rapid or ultra-rapid cooling by using the model of laboratory animals. 3.The conventional method for the embryo cryopreservation is described. An alternative to this method which may result in high survival and also in reducing of the freezing and thawing time is done by combing a permeable cryoprotectant such as glycerol, DMSO or propanediol and a non-permeable compound such as sucrose, trehalose, raffinose or lactose. 4.Finally a different approach to the preservation of embryos, named vitrification, is introduced. This procedure depends upon the ability of concentrated solutions of cryoprotective agents such as glycerol and propanediol to supercool to very low temperature (-196$^{\circ}C$) during rapid cooling before solidifying without formation of ice. However, more complete data are necessary for successful vitrification of blastocysts.

A Study on the Strength Degradation of Weathered Granite Soil by Freezing and Thawing (동결융해에 의한 화강풍화토의 강도저하에 관한 연구)

  • The property of weathered soil cannot but have a possibility of having a limit when its precise approaching due to the various weathering environment, and the peculiarity of its conduct affected by the weathering degree and effect factors. Especially most domestic or international researches are concentrated on the sedimentary soil, trying to analyze the mechanical behavior of weathered soil from the viewpoint of sedimentary soil. Therefore, it's difficult to judge if those results meet the actual conduct of weathered soil. This study suggested a way of weathering degree changing experiment as an early stage in an experiment of artificially changing weathering degree. In order to find out the property of strength change by the change of weathering degree, indoor mechanical experiment was made using soil sample after freezing and thawing. Under the weathering degree, characteristic change is watched by country rock and region of weathered granite soil.

Effects of Different Infusion Frequency of Liquid Nitrogen on Human Embryo Development and Pregnancy Rates after Freezing and Thawing (인간 배아 동결 해빙시 액체질소의 분사속도가 배아 발달 및 임신에 미치는 영향)

  • Objective : To investigate the efficacy of high infusion frequency of liquid nitrogen on pregnancy in human embryo after freezing and thawing. Materials and Methods: 150 infertile patients underwent 162 consecutive thawing-ET cycles. In the high infusion frequency group (Group A), 47 patients (50 cycles) underwent cryopreservation with high infusion frequency of liquid nitrogen. In the low infusion frequency group (Group B), 103 patients (112 cycles) underwent cryopreservation with low infusion frequency of liquid nitrogen. We analyzed the clinical characteristics, fertilization rates, development of embryo, good quality embryo ratio, implantation rates, and pregnancy rates between these two groups. Results: There was no difference between the groups with regard to clinical characteristics (mean age, infertility duration, infertility factors, hormone profile), mean number of oocyte retrieval, fertilization rates, and mean embryo number of transfers. The survival rates in group A was 64.9% (228 of 350 embryos), and among the 228 embryos 190 embryos (83.3%) which progressed to the two- to eight-cell stage. After thawing, the embryo numbers were 65 (34.2%), 29 (15.3%), 35 (18.4%), and 37 (19.5%) of grades 1, 2, 3, and above 4, respectively. The survival rates in group B was 63.8% (482 of 755 embryos), and among the 482 embryos 465 embryos (96.5%) which progressed to the two- to eight-cell stage. After thawing, the embryo numbers were 106 (22.8%), 94 (20.2%), 89 (19.1%), and 112 (24.1%) of grades 1, 2, 3, and above 4, respectively. There was no difference in embryo quality change after the freezing-thawing procedure between the groups. Implantation rates (31.1% vs. 34.3%) were not significant. However hCG positive rates in group A (40%) were higher than group B, but not statistically significant. Clinical pregnancy rate (26% vs. 25.9%), on going pregnancy rates (>20 weeks) were not significant (26% vs. 25%). Conclusion: We compared embryo quality change, survival rates, and pregnancy rates between high infusion frequency group and low infusion frequency group and the results were similar between the two groups. Therefore, high infusion frequency of liquid nitrogen for cryopreservation is a worthy method to preserve in human embryos.

  • Repair materials which applied to the concrete structures may have different quality characteristics depending on the environmental factors. Evaluation on durability of domestic repair materials have not yet secured enough quality performance on durability, mainly due to the lack of test methods resulted from various environmental factors. In this study, we carried out the tests on freezing and thawing resistance of domestic repair materials with different environmental factors applied under BS EN 13687, and analyzed the results by comparing with Korea's national test standards(KS F 4716). The results indicate that after the repetition of dry and wet conditions and the test on freezing and thawing with salt immersion resistance bond strength might show great difference depending on the type of repair materials and the size of sample. For securing better quality performance of repair materials, it is required to establish various standards on the test methods of freezing and thawing resistance with different environmental factors applied.

Flexural Strength and Durability Evaluation by Freezing and Thawing Test of Repaired Reinforced Concrete Beams (보수보강을 실시한 철근콘크리트 보의 동결융해시험을 통한 휨강도 및 내구성 평가)

  • This paper presents experimental investigations about flexural strength and durability of reinforced concrete beams repaired using ductile fiber reinforced cementitious composite (DFRCC) and carbon fiber sheet through freezing and thawing test. Total 14 RC beams of $100{\times}100{\times}400mm$ size were tested by 3-point bending and freezing and thawing test by KS F 2456. The beams were reinforced using 3D10 steels on both the tensile and compressive sides, and repaired on 3 sides expect on top cycle. Test results showed that the beams repaired using fiber carbon sheet revealed about 15% higher values of flexural strength compared than the cases of DFRCC motar. On the other hand, the results did not showed meaningful differences in the aspect of durability. For further research, consideration of the steel interference effect and real old specimens such as taken from real deteriorated structures are needed to be tested after repairing with DFRCC and carbon fiber sheet.

  • This paper presents experimental investigations about compressive strength and durability of reinforced concrete compressive members repaired using ductile fiber reinforced cementitious composite (DFRCC) and carbon fiber sheet through freezing and thawing test. Total 24 RC specimens of 100x100x400mm size were tested by compressive strength test and freezing and thawing test by KS F 2456. The specimens were reinforced using 4D10 steels and repaired on 4 sides expect on top cycle. Test results showed that the specimens repaired using fiber carbon sheet revealed about 5% higher values of the compressive strength compared than the cases of DFRCC motar. On the other hand, the resurts did not showed meaningful differences in the aspect of durability. For further research, considerations of the steel interference effect and real old specimens such as taken from real deteriorated structures are needed to be tested after repairing with DFRCC and carbon fiber sheet.

Effect of Temperature Abuse on Quality and Metabolites of Frozen/Thawed Beef Loins

  • The objective of this study was to examine the effect of temperature abuse prior to cold storage on changes in quality and metabolites of frozen/thawed beef loin. The aerobic packaged samples were assigned to three groups: refrigeration (4℃) (CR); freezing (-18℃ for 6 d) and thawing (20±1℃ for 1 d), followed by refrigeration (4℃) (FT); temperature abuse (20℃ for 6 h) prior to freezing (-18℃ for 6 d) and thawing (20±1℃ for 1 d), followed by refrigeration (4℃) (AFT). FT and AFT resulted in higher volatile basic nitrogen (VBN) values than CR (p<0.05), and these values rapidly increased in the final 15 d. Cooking loss decreased significantly with an increase in the storage period (p<0.05). In addition, cooking loss was lower in the FT and AFT groups than in the CR owing to water loss after storage (p<0.05). A scanning electron microscope (SEM) revealed that frozen/thawed beef samples were influenced by temperature abuse in the structure of the fiber at 15 d. Metabolomic analysis showed differences among CR, FT, and AFT from partial least square discriminant analysis (PLS-DA) based on proton nuclear magnetic resonance (1H NMR) profiling. The treatments differed slightly, with higher FT than AFT values in several metabolites (phenylalanine, isoleucine, valine, betaine, and tyrosine). Overall, temperature abuse prior to freezing and during thawing of beef loin resulted in accelerated quality changes.
