Amino Acids Analysis of Silk Fibroins among Varieties (가잠견사 Fibroin의 품종별 Amino산 분석)

  • This report is to observe the amino acid composition at silk fibroins among varieties by using paper chromatography method and to study how they may concern with the genetical problem and the biochemical synthesis of the fibroin in silkgland. The results reached are as follows: 1. The amino acid composition of silk fibroin among varieties were observed as similar with each other on the paper chromatogram pictures. 2. In spite of the results of the amino acid composition analysis was similar with other workers results, there was some difference among varieties in numerical analysis. 3. The amino acid composition was considered to concern somewhat with the genetic problem of silkworm. 4. Some nutritive amino acids are considered to related with the worm growth more than the silk fibroin. 5. The paper chromatography of this report showed an unknown amino acid which was not found before in specific species silk fibroin and it is imagined as Hydroxyproline in view of other worker's graphy result. 6. The biochemical synthesis mechanism of silk fibroin in the silk gland, neither Stepwise process nor Template, could stand with the results of this paper satisfactory one side.

  • Tetraethoxysilane(TEOS) as a silica precursor and hydrochloric acid as an acid catalyst have been used in a surfactant-template synthesis of micrometer-sized mesoporous silica microspheres based on the macroemulsion technique. Increase in the concentration of tetraethoxysilane of the reaction mixture has a serious destructive effect on the particle shape and pore structure. As the acid concentration increases, relatively small microspheres are formed without destroying their spherical morphology of the particles as well as the pore structures. However, due to the attractive interaction between particles in an acidic condition, strong silica agglomerates are formed, and therefore are subject to a post-ultrasonic treatment to separate into an individual single particle.

  • R&D of Nanofiber membrane has been carried out in the various fields, gas, water treatment, energy, and etc, with the continuous growth of membrane technology. There are several preparation methods for nanofiber, i.e. drawing, template synthesis, phase separation, self-assembly, and electrospinning. However, an electrospinning has many advantages such as high productivity, low production cost, easy to select law material, high relative surface area, and easy to functionalize. Nanofiber has been used in the field of membrane technologies such as secondary battery and water treatment fields. For the secondary battery separator, the separators having a high power and high thermal stability can be developed with spread of nanofiber on the commercial PP or PE/PP separators. High functional membranes can be also developed by adding the functional additives like antibacterial materials in the nanofiber membrane. It can be expected the high value added with nanofiber membrane because of its diverse applications from the water treatment to the energy field and because of its various functional advantages.

Synthesis of Magneli Phases and Application to the Photoelectrochemical Electrode (마그넬리상 합성과 광전기화학셀 전극 응용)

  • Hydrothermal synthesis of highly crystalline $TiO_2$ nanorods is a well-developed technique and the nanorods have been widely used as the template for growth of various core-shell nanorod structures. Magneli/CdS core-shell nanorod structures are fabricated for the photoelectrochemical cell (PEC) electrode to achieve enhanced carrier transport along the metallic magneli phase nanorod template. However, the long and thin $TiO_2$ nanorods may form a high resistance path to the electrons transferred from the CdS layer. $TiO_2$ nanorods synthesized are reduced to magneli phases, $TixO_{2x-1}$, by heat treatment in a hydrogen environment. Two types of magneli phase nanorods of $Ti_4O_7$ and $Ti_3O_5$ are synthesized. Structural morphology and X-ray diffraction analyses are carried out. CdS nano-films are deposited on the magneli nanorods for the main light absorption layer to form a photoanode, and the PEC performance is measured under simulated sunlight irradiation and compared with the conventional $TiO_2/CdS$ core-shell nanorod electrode. A higher photocurrent is observed from the stand-alone $Ti_3O_5/CdS$ core-shell nanorod structure in which the nanorods are grown on both sides of the seed layer.

A Facile Synthesis of SAPO-34 Molecular Sieves with Microwave Irradiation in Wide Reaction Conditions

  • Various reaction conditions uding temperature, time and type and concentration of templates have been changed in order to facilely synthesize, especially with microwave (MW) heating, SAPO-34 molecular sieves. SAPO-34 molecular sieve can be synthesized rapidly with microwave irradiation from a gel containing tetraethylammonium hydroxide (TEAOH) as a template. However, other several templating molecules lead to SAPO-5 molecular sieve under microwave irradiation even though SAPO-34 is obtained by conventional electric synthesis from the same reactant gels. Moreover, SAPO-34 can be obtained more easily by increasing the TEAOH or silica concentration or by increasing the reaction temperature. SAPO-34 can be obtained within 5 min in a selected condition (high temperature of 210 $^{\circ}C$) with microwave heating, which may lead to a continuous production of the important material. SAPO-34 synthesized by microwave irradiation is homogeneous and small in size and shows acidity and a stable performance in the dehydration of methanol and 2-butanol to olefins, suggesting potential applications in acid catalysis.

The G23 and G25 Genes of Temperate Mycobacteriophage L1 Are Essential for The Transcription of Its Late Genes

  • Two lysis-defective but DNA synthesis non-defective temperature-sensitive (ts) mutants of mycobacteriophage L1, L1G23ts23 and L1G25ts889 were found to be defective also in phage-specific RNA synthesis in the late period of their growth at 42$^{\circ}C$each to the extent of 50% of that at 32$^{\circ}C$The double mutant, L1G23ts23G25ts889 showed the ts defect in phage RNA synthesis that was nearly additive of those shown individually by the two single-mutant parents. Both G23 and G25 were shown to start functioning sometimes between 30 and 45 min after infection but the former gene might be dispensable after 45 min, while the latter was not. Northern analysis also shows that at 42$^{\circ}C$>, L1G23ts23 affects RNA synthesis more strongly than L1G25ts889 from L1 DNA segments that serve as the template for late gene transcription. Among the 21 virion and 12 non-virion late proteins synthesized by L1, L1G23ts23 is defective in the synthesis of at least 9 virion and all of non-virion proteins at 42$^{\circ}C$>. In contrast, L1G25ts889 is completely defective in synthesis of all the 33 late proteins. Possible roles of G23 and G25 in the positive regulation of transcription of different sets of late genes of L1 have been discussed.

  • Due to recent advances in genome sequencing, there has been a dramatic increase in the quantity of genetic information, which has lead to an even greater demand for a faster, more parallel expression system. Therefore, interest in cell-free protein synthesis, as an alternative method for high-throughput gene expression, has been revived. In contrast to in vivo gene expression methods, cell-free protein synthesis provides a completely open system for direct access to the reaction conditions. We have developed an efficient cell-free protein synthesis system by optimizing the energy source and S30 extract. Under the optimized conditions, approximately $650{\mu}g/mL$ of protein was produced after 2h of incubation, with the developed system further modified for the efficient expression of PCR-amplified DNA. When the concentrations of DNA, magnesium, and amino acids were optimized for the production of PCR-based cell-free protein synthesis, the protein yield was comparable to that from the plasmid template.

Electrolytic Synthesis of Cobalt Nanorods without Using a Supporting Template (템플릿 없이 전해 합성된 코발트 나노 로드)

  • Cobalt nano-rods were fabricated using a template-free electrochemical-deposition process. The structure of cobalt electro-deposits strongly depends on the electrolyte composition and on the density of the applied current. In particular, as the content of boric acid increased in the electrolyte, deposits of semi-spherical nuclei formed, and then grew into one-dimensional nano-rods. From analysis of the electro-deposits created under the conditions of continuous and pulsed current, it is suggested that the distribution of the active species around the electrode/electrolyte interface, and their transport, might be an important factor affecting the shape of the deposits. When transport of the active species was suppressed by lowering the deposition temperature, more of the well-defined nano-rod structures were obtained. The optimal conditions for the preparation of well-defined nano-rods were determined by observing the morphologies resulting from different deposition conditions. The maximum height of the cobalt nano-rods created in this work was $1{\mu}m$ and it had a diameter of 200 nm. Structural analysis proved that the nano-rods have preferred orientations of (111).

  • The objective of this study was to clone a partial fragment of $\alpha$-amylase from Korean maize. We designed and synthesized an oligonucleotide probe and two kinds of PCR primers based on cDNA conserved region of $\alpha$-amylase sequences from other plants. Total RNA from 3-day-old maize seedling was used as template for 1st strand cDNA synthesis and RNA-DNA hybrid was used as template for polymerase chain reaction(PCR). The product of PCR was about 0.5 kb long and inserted into pUC19. We named this recombinant plasmid as pZM$\alpha$'. The cloned fragment was certified by Southern blot analysis using labeled synthetic oligonucleotide as probe.

Synthesis and Characterization of Methyltriethoxysilyl-Mediated Mesoporous Silicalites

  • A series of mesoporous silicalites was synthesized using different compositions of tetraethylorthosilicate and methyltriethoxysilane (MTES) as the silica source. Cetyltrimethylammonium bromide was used as the organic template. Their detailed pore structures were investigated by transmission electron microscopy, X-ray diffraction, and N2 adsorption method. The thermal properties of these silicalites were studied by thermogravimetric analysis. The increased amount of MTES destroyed mesoporous channels and reduced pore sizes from 3.4 nm to 2.8 nm in calcined silicalites. The calcined silicalite transformed completely into an amorphous state at 30% MTES loading. Methyl pending groups of MTES hindered the structural ordering of ≡Si-O- frameworks, resulting in an amorphous structure. This was caused by the insufficient formation of supramolecular assembly with the organic template. No capillary condensation step was found in MS 7/3 silicalite. The other capillary condensation steps shifted toward the lower relative pressure with increasing MTES content, indicating the reduction of pore sizes.
