Molecular Monitoring of Eukaryotic Plankton Diversity at Mulgeum and Eulsukdo in the Lower Reaches of the Nakdong River (낙동강 하류 물금과 을숙도 수환경의 진핵 플랑크톤 종조성에 대한 분자모니터링)

  • We have studied the eukaryotic plankton species diversity to compare the community structure of fresh and brackish waters in the lower reaches of the Nakdong River using metagenomic methods. We constructed 18S rDNA clone libraries of total DNAs extracted from environmental water samples collected at Mulgeum (MG100929, fresh) and Eulsukdo bridge (ES, brackish). Through the steps of colony PCR, PCR-RFLP, sequencing and similarity analysis, we discovered the diverse species composition of eukaryotic plankton. Total 338 clones (170 at MG100929 and 168 at ES) were analyzed, and then we found 74 phylotypes (49 for MG100929 and 25 for ES). From the phylogenetic analysis, we confirmed various eukaryotic plankton of broad range of taxonomic groups, including Stramenopiles, Cryptophyta, Viridiplantae, Alveolata, Rhizaria, Metazoa, and Fungi. We also found several unreported species in Korea and candidates of new taxonomic entities at levels higher than genus. Especially, the cryptic species diversity including unreported phylotypes of Pirsonia (Stramenopiles) and Perkinsea (Alveolata) suggests that the molecular monitoring method can produce new informative biological data in monitoring the changes in the Nakdong River Mouth ecosystem.

Bacterial Community Composition and Diversity of a Full-Scale Integrated Fixed-Film Activated Sludge System as Investigated by Pyrosequencing

  • The integrated fixed-film activated sludge (IFAS) system is a variation of the activated sludge wastewater treatment process, in which hybrid suspended and attached biomass is used to treat wastewater. Although the function and performance of the IFAS system are well studied, little is known about its microbial community structure. In this study, the composition and diversity of the bacterial community of suspended and attached biomass samples were investigated in a full-scale IFAS system using a high-throughput pyrosequencing technology. Distinct bacterial community compositions were examined for each sample and appeared to be important for its features different from conventional activated sludge processes. The abundant bacterial groups were Betaproteobacteria (59.3%), Gammaproteobacteria (8.1%), Bacteroidetes (5.2%), Alphaproteobacteria (3.9%), and Actinobacteria (3.2%) in the suspended sample, whereas Actinobacteria (14.6%), Firmicutes (13.6%), Bacteroidetes (11.6%), Betaproteobacteria (9.9%), Gammaproteobacteria (9.25%), and Alphaproteobacteria (7.4%) were major bacterial groups in the attached sample. Regarding the diversity, totals of 3,034 and 1,451 operational taxonomic units were identified at the 3% cutoff for the suspended and attached samples, respectively. Rank abundance and community analyses demonstrated that most of the diversity was originated from rare species in the samples. Taken together, the information obtained in this study will be a base for further studies relating to the microbial community structure and function of the IFAS system.

An Introduction to the Chinese Soil Taxonomic Classification System (중국(中國) 토양(土壤) 계통(系統) 분류(分類) 안(案)의 소개(紹介) 및 고찰(考察))

  • 중국(中國)은 1991년에 새로운 토양계통분류법(土壤系統分類法)을 1차(次) 시안(試案)으로 확정 발표하였다. 본 체계는 미국의 Soil Taxonomy 체계와 비슷한 형태계량적규준(形態計量的規準)(Morpho-metric definition)으로 되어 있고 전산(電算)프로그램화(化)도 가능한 체계로 되어 있다. 분류체계는 목(目)(Order, 토망(土網)), 아목(亞目)(Suborder, 아망(亞網)), 군(群)(Group, 토류(土類)), 아군(亞群)(Subgroup, 아류(亞類)), 속(屬)(Family, 토속(土屬)), 통(統)(Series, 토종(土種))으로 되어 있고, 현재 13개 목망(目網), 33개 아목(亞目), 77개 군(群), 301개 아군(亞群)으로 분류되어 있다. 특징적표층(特徵的表層)은 유기표층(有機表層) 등(等) 8종, 진단표하층(診斷表下層)(차표층(次表層))은 표백층(漂白層) 등(等) 10종, 기타 특징토층(特徵土層)은 석회적표층(石灰的表層) 등(等) 12종, 그리고 암성(岩性) 등(等) 23종의 토양 생성적(生成的) 특징(特徵) 또는 현상(現像)을 분류기준으로 쓰고 있다. 분류상의 특징은 고위도(高緯度)나 고산지대(高山地帶)의 영구동결층(永久凍結層)을 강조하여 아목수준(亞目水準)에서 분류하였고 논토양이나 관개퇴적토(灌漑堆積土), 영양분(營養分)이 심층(深層)까지 집적(集積)된 채전(菜田) 등(等) 인위적(人爲的) 영향(影響)을 크게 받은 토양을 최고차(最高次) 분류단위(分類單位)인 목수준(目水準)에서 독립(獨立)시키고 있을 뿐 아니라 인위적인 교란(攪亂)을 받았으나 아직 단면(斷面)의 발달이 없는 토양을 초육토(初育土)의 아목수준(亞目水準)에서 구분하고 있다.

  • 50 specimens of higher fungi were collected on Mt. Sobaek and Andong areas from September 5, 1974 to July 30, 1975. These fungi were identified by Imazeki and Hongo's classification key based on Singer's taxonomic system. Nine species of them were newly recognized to occur in Korea; Hygrophorus pantoleucus Hongo. Marasmius prasiosmus (Fr.) Fr. Conocybe lactea (Lange) Metrod, Pholiota spumosa(Fr.) Sing., Crepidotus sulphurinus Imaz. et Toki, Pantoleucus retipes (Berk. et Curt.) Sing., Calodon zonatus(Fr.) $Qu{\acute{e}}l$, Patella scutellata (St. Amans) Morgan and Peziza badia Pers. ex Merat.

Macrofungal Survey of the Tian Shan Mountains, Kyrgyzstan

  • The Tian Shan mountain system is one of the large mountain ranges located in Central Asia. This region is globally recognized as mountain ranges, offering inestimable wealth in fauna and flora with significant biodiversity values. We surveyed macrofungal diversity of Tian Shan in Kyrgyzstan from 2016 to 2018. A collection of macrofungi was made, and these were subjected to sequence comparisons and phylogenetic analysis to ensure the identity of the collected macrofungi. Of those collected, 95 out of 100 specimens were successfully sequenced and compared with those of other related species retrieved from GenBank. The sequenced specimens were classified into 2 phyla, 8 orders, 24 families, 47 genera, and 57 species, based on current taxonomic concepts (combining morphology and phylogeny). To the best of our knowledge, this study provides the first well-documented checklist and phylogenetic analysis of macrofungi recovered from the Tian Shan mountains in Kyrgyzstan.

  • Rapid and reliable identification of microorganisms is a key for tracing the relationship between the target bacteria and related infectious diseases. Various identification methods such as classical phenotypic analysis, numerical taxonomic analysis, and DNA sequencing have been widely used to classify microorganisms in milk and dairy products. Matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometry (MS) identifies targeted bacteria in milk and milk products. Several studies have demonstrated that MALDI-TOF MS identification is an efficient and inexpensive method for the rapid and routine identification of isolated bacteria. MALDI-TOF MS could provide accurate identification of bacteria in milk and milk products at the serotype or strain level and enable antibiotic resistance profiling within minutes.

Reproductive Isolation between Moroco oxycephalus and M. lagowskii (Pisces; Cyprinidae) in Korea

  • To clarify taxonomic status of the two sibling species, Moroco oxycephalus and M. lagowskii reproductive isolation mechanisms were investigated at sympatric area located in Kansung-up, Kosung-gun, Kangwon-do, Korea. Genetic analysis was performed to reveal mating system and intensity of Hybridization between the two species. The frequencies of hybrids were increased since 1989, and then the observed hybrid frequencies ($H_O$) did not significantly differ from the expected hybrid ($H_E$) in 1998 and 1999. However, based on histological analysis of two parents and their hybrid s gonads, the hybridizations between M. oxycephalus and M. lagowskii produced mostly fertile females but sterile males in accordance with Haldane s rule. Although it was suspected that pre- and postmating isolation mechanisms were affected between the two species, M. oxycephalus and M. lagowskii seemed to be strongly isolated with microhabitat at sympatry until 1997. Since 1998, hybrid frequencies were increased by habitat disturbance. However, their hybrid frequencies would be reduced by postmating isolation mechanisms. Therefore, the two species are considered to be distinct species recently diverged.

  • It is considered that comets near the ecliptic plane have been injected into inner solar system from Kuiper-belt. Some of them are still active while others are dormant with no detectable tails and comae. These dormant comets have eccentric and/or inclined orbits, which are parameterized by Tisserand parameter TJ<3. In addition, dormant comets can be differentiated from asteroids based on the albedo, because they have low albedo (the geometrical albedos pv<0.1). The conditions of TJ<3 and pv<0.1 have been used as a criteria to discriminate dormant comets from asteroids.   • The plant origines of Atractylodis Rhizoma Alba and Atractylodis Rhizoma are genus Atractylodes*Compositae). Their origines in pharmacopoeia are slightly different among Korea, Japan, North Korea and China. The species of Atractylodis Rhizoma Alba are Atrctylodes japonica $K_{OIDZ}$. A. ovata $T_{HUNB}$.. Atractylodis Rhizoma are A. lancea DC., A. chinensis DC. and A. koreana $K_{ITM}$. But the texonomic genealogy of Atractylodis Rhizoma Alba is different from that of Atractylodis Rhizoma. The herbal origines of Atractylodis Rhizoma Alba and Atractylodis Rhizoma were not different at the early days. Since Tao-Hong-Jing(陶弘景) distinguished between Atractylodis Rhizoma Alba and Atractylodis Rhizoma at the very beginning, theri medicinal usages are different, Atractylodis Rhizoma have been used to remove pathogenic dampness in the digestive organs and Atractylodis Rhizoma Alba have been used to invigorate the digestive system and replenish qi. In additions, the figures and historical records of herbal appearance are different between Atractylodis Rhizoma Alba and Atractylodis Rhizoma. The diagnostics of them are in accord with the texonomic genealogies.

  • In this paper, three species of Russula previously not recorded in Korea were found in Suweon, Mt. Chiri, Taegwanryung, Kwangneung, and etc., during 1977. The authors classified 32 species of Russula including 3 unrecorded species into 4 subgenera acording to morphological and cytological characteristics by S. Ito's taxonomic system, and proposed to designate the 3 unrecorded species of Russula and 4 sub genera, ((Compactae, Ingratae, Rigidae, and Fragilis) in Korean common name.
