Antidiabetic Effect of Korean Red Ginseng by Puffing Process in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐에서 팽화가공 처리한 홍삼의 항당뇨 효과)

  • Antidiabetic effect of Korean red ginseng (RG) processed by puffing in streptozotocin (STZ)-induced diabetic (DM) rats was investigated. Five week-old SD rats were divided into four groups; normal control (NC) group, DM group, red ginseng (RG) group and puffed red ginseng (PG) group. The RG and PG groups were orally provided with RG or PG dissolved in water (500 mg/kg) respectively for seven weeks after single injection of STZ (50 mg/kg, i.v.) followed by identification of DM. NC group received saline vehicle instead of STZ. At the end of feeding of RG or PG, the changes of fasting blood glucose, serum insulin and amylase level and serum lipid profiles were evaluated. Also, oral glucose tolerance test (OGTT), comet assay and histopathological examination were performed. At 7th week, the fasting blood glucose levels of the RG and PG groups were reduced compared to the DM group by 11.54% and 20.22%, respectively. The result of OGTT did not show significant differences among DM and two red ginseng groups. While serum insulin and TG levels were predominantly improved in PG group (p<0.05), serum amylase level was increased in RG group. Alkaline comet assay for checking the oxidative damage of DNA showed that TL (tail length, ${\mu}m$) and TM (tail moment) in the blood lymphocyte of PG group significantly decreased in contrast with DM group. Histopathological results of pancreas showed that destruction of exocrine as well as endocrine might be cured by the administration of RG and PG. These results suggest that PG could exert more protection against STZ-induced toxicity than RG group.

Genotype Frequencies of the Sex-Linked Feathering and Their Phenotypes in Domestic Chicken Breeds for the Establishment of Auto-Sexing Strains (자가성감별 계통 조성을 위한 국내 토종 닭의 깃털 조만성 양상과 유전자형 빈도)

  • The method of sexing based on differences in the rate of feather growth provides a convenient and inexpensive approach. The locus of feather development gene (K) is located on the Z chromosome and can be utilized to produce phenotypes that distinguish between the sexes of chicks at hatching. To establish the auto-sexing native chicken strains, this study analyzed the genotype frequency of the feathering in domestic chicken breeds. The method of classification of slow- and rapid-feathering chickens was also investigated. In the slow-feathering chicks, the coverts were either the same length or longer than the primary wing feathers at hatching. However, the rapid-feathering chicks had the primary wing feathers that were longer than the coverts. The growth pattern of tail feather also distinctively differed between the rapid- and slow-feathering chicks after 5-days. The accuracy of wing feather sexing was about 98% compared with tail sexing. In domestic chicken breeds, Korean Black Cornish, Korean Rhode Island Red, and Korean Native Chicken-Red had both dominant (K) and recessive ($k^+$) feathering genes. The other breeds of chickens, Korean Brown Cornish, Ogol, White Leghorn, Korean Native Chicken-Yellow, -Gray, -White and -Black had only the recessive feathering gene ($k^+$). Consequently, feather sexing is available using the domestic chicken breeds. Establishing the maternal stock with dominant gene (K-) and paternal stock with recessive gene ($k^+k^+$), the slow-feathering characteristic is passed from mothers to their sons, and the rapid-feathering characteristic is inherited by daughters from their fathers.

  • The authors of this paper aim to make a record of the construction process, its symbolic meaning, and the changes in the status of the landscape plants at the Pasanseodang according to the Report on the Pasanseodang written by Park Gyu-hyun in 1874. First, the construction of Samgahun Pavilion, which is located in Myo-ri, Habin-myun, Dalsung-gun, Daegu, took about 90 years and spanned the lifetimes of Park Sungsoo, an 11th-generation descendant of Park Paengnyun (1417~1456) through to Park Kyuhyun, a 14th-generation descendant. It was called the shape of dragon, with its head facing the tail (回龍顧尾形), in feng shui. Second, the village of Pahwoe was founded in 1769, the 45th year of the reign of King Yeongjo, by Park Sungsoo for the purpose of socializing with his friends at his thatched home, and was named after his own courtesy name (Samgahun). Park Kwangseok, the second son of Park Sungsoo, built the sarangchae in 1826 and the anchae in 1869 after his marriage (in 1783). Then, Park Kyuhyun, the grandson of Park Kwangseok, built the pond and planted it with lotus flowers, and built the Hayeopjeong in 1874. The Pasanseodang, as the precursor of the Hayeopjeong, may be related with the name of the hillside region behind Samgahun. Third, a quadrangular-shaped pond with a length of 21m and a width of 15m was also built and planted with lotus flowers. In the center of the pond is a small round island that reflects the world view of the Chosun dynasty, i.e. that the sky is round and the landmass is quadrangular. Meanwhile, the name of the Hayeopjeon reflects the value system of aristocrats who lived a life of leisure and artistic indulgence. They called the eastern room "Yeeyeonhun" (怡燕軒) and the western room "Mongyangjae" (蒙養齋), names which embody their wishes for a good life as a member of the nobility and a bright future for one's descendants. Fourth, in Confucian terms, the authors infer the points of view reflected in the kinds of trees that were planted according to Confucian norms (pine tree, lotus, bamboo), the living philosophy of sustainability (willow), the ideology of seclusion and the search for peace of mind (bamboo), and relief efforts for the poor and a life of practicality (chestnut, oak, wild walnut, lacquer). The authors assert that this way of planting trees was a highly effective design feature of landscape architecture that drew on the locational and symbolic significance of the Seodang. Fifth, the majority of the trees that were initially planted withered and were replaced with different species, except for the locust and lotus, at this point. Nevertheless, a review of the process of construction, symbolic meaning, and original architectural landscape of the Samgahun is of value in demonstrating the extended symbolic meaning of their descendants in terms of the practical loss of the function of the Seodang, the values of Feng Sui (red in the east, white in the west, based on the principles of Feng Sui), the function of repelling evils spirits (kalopanax, trifoliate orange), aesthetic and practical values (sweetbrier, apricot, pear, peach, and oriental oak trees), and the prosperity of the family and the timeless value of honest poverty (silk, crape myrtle, and yew trees).

  • Rockfish, Sebastes inermis is a ovoviviparous fish distributed around the coastal waters of Korea and Japan. Naturally beared larvae obtained from female fish kept in the aquarium were reared for three months in the laboratory. The naturally healed larvae were 6.05mm in average total length(TL) with $8+18{\sim}19=26{\sim}27$ myomeres. Melanophores were distributed on the top of head, around the intestine tube and on the dorsal and ventral margins of the tail, but none on the pectoral fins. The larva(7.1mm in TL) consumed all the yolk and oil globule, in 7 days after bearing, and jaw bones were more rapidly ossified than vertebrae and cranium. In 21 days after bearing, the larvae averaged 8.50mm in TL and the caudal notochord flex $45^{\circ}$. Fin development in S. inermis larvae proceeds in the following sequence: caudal, pectoral, dorsal, anal and ventral fin. Individuals $17.6{\sim}20.8mm$ in TL($65{\sim}69$ days after bearing) are regarded as the juvenile stage. All fins bones was nearly completed in this stage.

A Study of the Intersection in Reduce Car Accidents for Traffic Signal Light to Supplement (교차로 사고 감소를 위한 신호등 보완에 관한 연구)

  • Three types of traffic signal systems are two-color signal systems that flash red and green and are mainly used on crosswalks, next, three-color equalization systems mainly used at T-shaped intersections with red and yellow lights and a green arrow, and third, four-color intersections that generally have red, yellow and green colored lights and a green arrow. In what is known as the "dilemma zone" area, a driver collects information that influences his/her decision whether to stop, speed, tail, interrupt, or violate a traffic light, depending on the intersection width, vehicle speed, cognitive response time and reference yellow signal time. This study examined the impact of changes in the length of the dilemma zone areas based on changes in yellow signal times, the speed of the intersection passages, and signal lamps. Downward adjustments of 50km/h and 60km/h affected yellow signal time. The yellow signal time increased by 0.1 to 2.3[s] due to this effect and the dilemma zone area increased by 1.22 to 26[m]. The driver of the dilemma zone could quickly decide to reduce the time remaining of the straight (3color, 4color) green signal to reduce the potential of a traffic accident at the intersection traffic. Safe entry of red (LED palm) and left-turn signals for entering flashed at the intersection and operated at midnight.

  • Tetragomphius procyonis Baylis & Daubney, 1923 were obtained from the pancreatic duct of a naturally infected Eurasian badger, Meles meles, which was submitted to animal hospital for parasitic diagnosis from Gyeryongsan National Park in Korea. The hookworms were examined by light and scanning electron microscope. The length of body measured male 15.0-18.8 mm, female 21.5-25.5 mm, respectively. In both sex, the ventral cutting plates of oral margin are much reduced and elongated latero-dorsally, the dorsal cutting plate is located long follow doral margin of the oral opening. The buccal capsule is cup-shaped and thicken with four cusped tooth at its base. The copulatory bursa has elongated ventral lobes and their large rays are parallel, while the dorsal lobe with its supporting rays is slightly split in two. The slender spicules are filariform and very long (8.7-9.3 mm), and their tips are fused together. The hookworm has following characters: dorsal cone on the both sex, gubernaculum on the male and terminal spine on the female tail absent; vulva is opening in the juction of the fourths and fifths of the body; dorsal ray with two long stems. The eggs from the uterus are 16 cell stages and those from the rectal feces, 32 cell stages. The size of eggs is 77.48-83.45 ${\times}$ 50.75-63.38 ${\mu}m$.

  • Naturally beared larvae obtained from female fish kept in the aquarium were reared for 45 days in the laboratory. The naturally beared larvae were 3.19 mm in average total length (TL) with 9+15~16=24~25 myomeres. Melanophores were distributed on the dorsal margin of intestine tube, ventral margins of the tail and around the oil globule, but none on the top of head and on the pectoral fins. The larva (3.35 mm TL) consumed all yolk and oil globule in 5 days after bearing, and jaw bones and clavicle began to be ossified rapidly. In 22 days after bearing, the larvae averaged 6.30 mm TL and the caudal notochord flex $45^{\circ}C$. Fin development in S. marmoratus larvae proceeds in the following sequence : caudal, pectoral, dorsal, anal and ventral fin.

Novel Low-Temperature-Active Phytase from Erwinia carotovora var. carotovota ACCC 10276

  • A phytase with high activity at low temperatures has great potential for feed applications, especially in aquaculture. Therefore, this study used a degenerate PCR and TAIL PCR to clone a phytase gene from Erwinia carotovora var. carotovota, the cause of soft rot of vegetables in the ground or during cold storage. The full-length 2.5-kb fragment included an open reading frame of 1,302 bp and encoded a putative phytase of 45.3 kDa with a 50% amino acid identity to the Klebsiella pneumoniae phytase. The phytase contained the active site RHGXRXP and HD sequence motifs that are typical of histidine acid phosphatases. The enzyme was expressed in Escherichia coli, purified, and displayed the following characteristics: a high catalytic activity at low temperatures (retaining over 24% activity at $5^{\circ}C$) and remarkably thermal lability (losing >96% activity after incubation at $60^{\circ}C$ for 2 min). The optimal phytase activity occurred at pH 5.5 and ${\sim}49^{\circ}C$, and the enzyme activity rapidly decreased above $40^{\circ}C$. When compared with mesophilic counterparts, the phytase not only exhibited a high activity at a low temperature, but also had a low $K_m$ and high $k_{cat}$. These temperature characteristics and kinetic parameters are consistent with low-temperature-active enzymes. To our knowledge, this would appear to be the first report of a low-temperature-active phytase and its heterogeneous expression.

  • An important problem in frequency analysis is the estimation of the quantile for a certain return period. In frequency analysis an assumed probability distribution is fitted to the observed sample data to estimate the quantile at the upper tail corresponding to return periods which are usually much larger than the record length. In most cases, the selection of an appropriate probability distribution is based on goodness of fit tests. The goodness of fit test method can be described as a method for examining how well sample data agrees with an assumed probability distribution as its population. However it gives generally equal weight to differences between empirical and theoretical distribution functions corresponding to all the observations. In this study, the modified Anderson-Darling (AD) test statistics are provided using simulation and the power study are performed to compare the efficiency of other goodness of fit tests. The power test results indicate that the modified AD test has better rejection performances than the traditional tests. In addition, the applications to real world data are discussed and shows that the modified AD test may be a powerful test for selecting an appropriate distribution for frequency analysis when extreme cases are considered.

Morphological Development of the Larvae and Juveniles of Halfbeak Fish, Hyporhampus sajori (Temminck et Schlegel) (학공치 Hyporhampus sajori (Temminck et Schlegel) 자치어(仔稚漁)의 형태 발달)

  • Observation of morphological changes during larval and juvenile stages of the halfbeak fish, Hyporhampus sajori (Temminck et Schlegel), was made based on samples of fertilized eggs collected on the shores of Youngil Bay, Phohang-shi, Korea, on May 27, 1991, and incubated in the laboratory. During the incubation period, water temperature fluctuated between $17.6^{\circ}C$ and $23.2^{\circ}C$. The newly hatched larvae were elongated and 7.0~8.50 mm in total length (TL) with 40~42+17~18 = 59~60 myotomes. Numerous melanophores were on the head, mid-dorsal part of the body, intestine, and the tail part of the notochord. Three days after hatching the larvae had attained 8.60~10.90 m in TL, had completely absorbed the yolk, and the caudal notochord was flexed $45^{\circ}$ upward. At this time, it was attained postlarvae stage. Thirty-seven days after hatching the postlarvae were 32.37~44.95 mm in TL and had reached the juvenile stage. All fins were formed with a complete set of fin rays with the following counts: dorsal fin rays 16~17; pectoral f in rays 6; ventral fin rays 12~14; caudal fin rays 11~12+11~12 = 22~24.
