Studies with the tobacco mosaic viruses (한국산 연초 "바이러스"에 관한 연구)

  • Studies with the Tobacco Mosaic Viruses; W. S Kim, and So, I Y., (Dept. of biology Sung Kyun Kwan Univer. Seoul, Korea.). Using the common strain of tobacco mosaic virus (TMV) which was sent from the Dept. of Plant Pathology, University of Wisconsin, U.S.A. as control virus, a possible new strain of tobacco mosaic virus (SMV) was isolated from tobacco leaves collected from Tobacco Experiment Station farms as well as from various blends of manufactured Korean cigaretts. SMV was isolated by single lesion isolation method and by inoculating the virus through various species of host plants. The two viruses, TMV and SMV were indentified by the difference in symptoms, host range, serological reaction, and electron micrograpy. As the results of the above experiment the author believes the virus isolate SMV is a different strain of TMV. The experimental evidences that SMV belongs to the TMV group are as follows; 1. Both viruses produced local necrotic lesions on Nicotiana glutimosa L. 2. Both showed a dilution end point of $10^8$. 3. Aphid transmission was failed with the viruses. 4. Both had an isoelectric point around pH 3.3. 5. Two viruses were serological reactive. 6. The size of the virus particles was around 270-300mu as they were observed under the electron microscope. The virus SMV, however, is different from the common strain of TMV and the experimental evidences are as follows; 1. SMV produced quite different symptoms from TMV on various host plants like tobacoo(Nicotiana tabacum L., White Burley), Nicotiana rustica L., Chenopodium Koreanse Nakai. Bata vulgaris L., and Datura tatula L., SMV produced distinct local lesions on these host plants whereas TMV incited largely mosaic diseases. 2. The serological titers obtained from the heterologous combinations were lower than those from homologous combinations of antigens and antiser.

A Study on the Variation of Color and its Components of the Flue-Cured Tobacco Leaf During the Process of Curing (Flue -cured 잎담배의 건조과정중 색상과 색소성분 변화에 관한 연구)

  • Boring the process of curing of the tobacco ( Nicotiana tabacum L. clicks) leaf, the and other fundamental components of the tobacco leaf was observed The amount of total chlorophyll was reduced from 2902$\mu\textrm{g}$/gr to 1, 131$\mu\textrm{g}$/gr as a function of curing time. In the case of total carotene, the concentration was increased from 373-712$\mu\textrm{g}$/gr. to 3, 003~2, 215$\mu\textrm{g}$/gr. up to 70 hours of curing period. However, after 70 hours of curing, the concentration of total carotene stared to be decreased up to 1, 050~1, 039$\mu\textrm{g}$/gr. Simultaneously, the greenish yellow color becomes to be close an intermediate color between yellow and orange. The amount of total nicotine and nitrogen (2~2%) in the green harvested leaf was essentially identical after the curing process. However, the amount of total sugar was enhanced from 17%, in the green harvested leaf, to 35% at the time of 70 hours curing. At the end of the curing, the amount of total sugar was again reduced up to 16%. The correlation coefficient (r) between the axis of the color coordinate and the ratio of chlorophyll/carotene was estimated to be r=-0.96. This result indicates a linear relationship between those two parameters and the following linear equation is obtained as y=28.87-68.38 x. From the above equation, x=0.442 could be estimated when chlorophyll was completely decomposed (ylongrightarrowO). As a consequence of this result, we can suggest the tobacco leaf of Hicks belongs to the high quality level.

Evaluation of Biomass and Nitrogen Nutrition of Tobacco under Sand Culture by Reflectance Indices of Ground-based Remote Sensors (지상원격측정 센서의 반사율 지표를 활용한 사경재배 연초의 생체량 및 질소영양 평가)

  • Remote sensing technique in agriculture can be used to identify chlorophyll content, biomass, and yield caused from N stress level. This study was conducted to evaluate biomass, N stress levels, and yield of tobacco (Nicotiana tabacum L.) under sand culture in a plastic film house using ground-based remote sensors. Nitrogen rates applied were 40, 60, 80, 100, 120, and 140 percent of N concentration in the Hoagland's nutrient solution. Sensor readings for reflectance indices were taken at 30, 35, 40, 45, 50 and 60 days after transplanting(DAT). Reflectance indices measured at 40th DAT were highly correlated with dry weight(DW) of tobacco leaves and N uptake by leaves. Especially, green normalized difference vegetation index(gNDVI) from spectroradiometer and aNDVI from Crop Circle passive sensor were able to explain 85% and 84% of DW variability and 85% and 92% of N uptake variability, respectively. All the reflectance indices measured at each sampling date during the growing season were significantly correlated with tobacco yield. Especially the gNDVI derived from spectroradiometer readings at the 40th DAT explained 72% of yield variability. N rates of tobacco were distinguished by sufficiency index calculated using the ratio of reflectance indices of stress to optimum plot of N treatment. Consequently results indicate that the reflectance indices by ground-based remote sensor can be used to predict tobacco yield and recommend the optimum application rate of N fertilizer for top dressing of tobacco.

Influence of Drought on Leaf Growth and Water Potential in Tobacco (한발이 담배 잎의 생장과 수분 포텐셜에 미치는 영향)

  • The experiment which imposed the water stress to tobacco(Nicotiana tabacum L.) plant was carried at the late of maximum growth period. In order to know the influence of drought stress on the growth and developmemt of tobacco leaves of different position and to elucidate the physiological response of plant to various soil water content, stomatal conductance, and leaf water potential were measured. The drought stress at the maximum growth period negatively affected to the overall growth characteristics of shoot. The response of the growth was small at the middle and the lower leaves, and great at the upper leaves. The relative water content of upper, middle, and lower leaves at the fifth day after treatment were 74, 64, and 59%, respectively, as soil water content was reduced by 4.3%. This suggested that the wilting point of tobacco leaf was about 75%. The leaf water potential was -0.58 MPa in control and dropped to -1.20 MPa at the fifth day after treatment. This indicated that wilting of leaf may occur at the condition in which the difference of water potential between treatment and control, well watered, was greater than about 20%. Stomatal conductance at the fifth day after treatment dropped from 12 mol /$\textrm{m}^2 sec^{-1}$ to 0.8 mol /$\textrm{m}^2 sec^{-1}$ in the middle and the upper leaves. Stomatal conductance of lower leaves already matured were not affected highly by drought stress at the maximum growth period, but maturing leaves, middle and upper leaves, were highly affected by limitation of soil water.

Acquisition of Thermotolerance in the Transgenic Plants with BcHSP17.6 cDNA (BcHSP17.6 cDNA의 도입에 의한 형질전환된 식물의 내열성 획득)

  • Recombinant plasmid, pBKH4, containing NPT II and P35S-BcHSP17.6 was constructed by ligation of Bum H I -digested pBKSl-l and BcHSP 17.6 (thermotolerance gene) 6om pBLH4. The tobacco leaf disc was cocultivated with transformed Agmbacterium tumefaciens bearing pBKH4 for 24 hours and transformed shoots were selected on MS-n/B medium containing $100\;{\mu\textrm{g}}/ml$ of kanamycin. Heat-killing temperature of Nicotima tabacum was $50^{\circ}$ for >15min, and transformed tobacco plants with BcHSP17.6 cDNA exhibited thermotolerance at the heat-killing temperature. The transgenic plants were analyzed by Southern blot hybridization with the probe of ${\alpha}^{_32}P$ labelled BcHSP17.6 cDNA. Transcription and expression level of BcHSP17.6 cDNA were also continued by Northern blot analysis and Ouchterlony double immunodiffusion assay. In this study, we suggest that the BcHSP17.6 cDNA introduced to tobacco plant is related to thenuoto-lerance and 17.6-kD LMW HSP acts as a protector from heat damage in plants.

  • Production of human granulocyte-macrophage colony stimulating factor (hGM-CSF) by Nicotiana tabacum cell suspension culture was studied in Murashige and Skoog (MS) medium with sucrose as a carbon source, ammonium nitrate and potassium nitrate as nitrogen sources, potassium dihydrogen phosphate and sodium dihydrogen phosphate hydrate as phosphate sources, respectively. Optimum concentrations for carbon, nitrogen, phosphate was determined to enhance the production of hGM-CSF. Cell growth was better at high initial sucrose concentration (60 g/L), high initial nitrogen concentration (121.04 mM). Maximum cell density (18.28 g/L) was obtained at 60 g/L of sucrose after 14 days. Cell growth was not so good at low initial sucrose concentration 00 g/L), but the highest hGM-CSF production was obtained at the latter half of exponential phase. hGM-CSF production increased about 3 fold at initial phosphate concentration of 4.96 nM

  • Ascorbic acid is a great antioxidant and helps protect the body against pollutants. GDP-mannose pyrophosphorylase (GMPase) is a key enzyme in manufacturing GDP-mannose, a glycosyl donor for ascorbate and cell wall biosynthesis as well as for protein glycosylation. In this study, we described molecular cloning of a full-length cDNA from Potato (Solanum tuberosum L. cv. Jasim), using tuber. The cDNA isolated encoded a GDP-mannose pyrophosphrylase. The nucleotide sequence of pGMPC showed about 95%, 89% and 80% homology with S. tuberosum (AF022716), N. tabacum (AB066279) and A. thaliana (AF076484) cDNAs clone known as GMPase, respectively. We detected the expression of GMPase using RT-PCR. The highest expression of GMPase was found in stems, and the largest amount of ascorbic acid was also presented in stems. In contrast, the leaf showed minimal level of GMPase transcript and ascorbic acid content. We propose that GMPase expression patterns were similar to the changes of ascorbic acid content in the leaves treated with diverse stresses.

Screening Method for Photosynthetic Electron Transport Inhibitors Using Photoautotrophic Cultured Cells (광학적 자가영양 배양세포를 이용한 광합성 전자전달억제자의 간이검정방법)

  • To investigate a simple and rapid screening method for photosynthetic inhibitory herbicides, responses of tobacco(Nicotiana tabacum L.) and liverwort(Marchantia polymorpha L.) PA(photoautotrophic) cells to various commercial herbicides with different modes of action and leaf extracts of four weed species were compared. PET (photosynthetic electron transport) inhibitory type of herbicides has greater inhibitory effect in liverwort photoautotrophic cells than the photomixotrophic and heterotrophic cultured cells. Similary, PET inhibitory type of herbicides inhibited the oxygen evolution more in liverwort PA cells than the other type of herbicides. Based on oxygen evolution, 60% inhibition was observed by the 10% aqueous extracts of Polygonum hydropiper, while there was 100% inhibition by the 10% methanol extracts of Polygonum hydropiper. This assay gave well correlated results to the Hill reaction data using isolated thylakoids. Thus liverwort photoautotrophic cells might be suitable materials for rapid screening method for photosynthetic inhibitory herbicides.

Enhanced Production of hGM-CSF by Immobilized Transgenic Plant Cell Cultures (형질전환된 식물세포에서 고정화 방법을 통한 hCM-CSF의 생산성 증대 연구)

  • Plant cell immobilization can protect plant cells from shear forces and increase the stability of gene. An additional advantage of immobilization is the easiness for performing continuous culture with cell recycling. Therefore plant cell immobilization can overcome the limitations of plant cell applications. In addition, target protein should be selected from pharmaceutical proteins to get rid of low expression level problem. The enhanced production of human granulocyte-macrophage colony-stimulating factor (hGM-CSF) was investigated in immobilized Nicotiana tabacum suspension cell cultures. When the cells were immobilized in polyurethane foam, specific production of hGM-CSF was higher than that in alginate bead immobilization. Optimum continuous culture condition was the addition of 60 g/L sucrose in growth media with exchanging media every 6 day. Under the same condition, specific hGM-CSF production was 7 times higher in a 500-mL spinner flask than that in 100-mL Erlenmeyer flasks. Therefore, development of an effective immobilization process would be possible when the advantage of easy cell recycling was used. Consequently, enhanced production of target proteins could be possible in immobilized continuous cultures when the advantages of immobilization were applied.

Identification of Korean Ginseng Cytochrome P450 gene and Its Characterization by Transformation System (고려인삼 유래 Cytochrome P450 유전자의 동정 및 형질전환에 의한 특성검정)

  • Triterpenoid saponins were synthesized in Panax ginseng C.A. Meyer via the isoprenoid pathway by cyclization of 2,3-oxidosqualene to give primarily oleanane (beta-amyrin) or dammarane triterpenoid skeletons. The triterpenoids are backbone and undergoes various modifications (oxidation, substitution and glycosylation), mediated by cytochrome P450 (CYP)-dependent monooxygenases, glycosyltransferase and other enzymes. This is likely to be due in part to the complexity of the molecules and the lack of pathway intermediates for biochemical studies. A cDNA clone encoding a putative CYP gene was isolated from flower bud of ginseng and transformed into the plant(Nicotiana tabacum cv. Xanthi) and confirmed by PCR analysis. The CYP gene (PgCYP) contained an open reading frame(ORF) encoding mature protein of 500 amino acids. The expression of PgCYP were investigated in transgenic tobacco by reverse transcriptase-polymerase chain reaction (RT-PCR).
