Food allergies have become a serious health concern in the past two decades, especially in developed countries. Foods associated with allergies include vegetables, some fruits, shellfish, wheat, egg, chicken, and nuts. To describe the specific fundamentals, etiological factors, and clinical manifestations, we analyzed the different physical frequency on spleen index in sensitized and regular exercise-trained mice. We also conducted a proliferation assay of lymphocytes to OVA, ROS, ASAS, and we determined the cytokine levels. Female BALB/c mice were bred in the animal laboratory of the P and D university under controlled conditions ($22{\pm}2^{\circ}C$, RH 45-55%, and a 12-hour photoperiod). The animals were 6 weeks old at the start of the study and were fed a standard commercial chow diet from 09:00 to 15:00 for the 8-week study period. All animals had access to distilled deionized water ad libitum. They were divided into four groups: a control group (S; control sensitized, n=25), a low-frequency training group (F2, n=25), a mid-frequency training group (F3, n=25), and a high-frequency training group (F5, n=25) following the treatment of exercise time per week. The results were as follows: The mice spleen index showed the highest grade in the F5 group compared with the other groups; this level showed in an exercise frequency-dependent manner. In the proliferation assay of OVA, the F5 group showed the highest grade compared with the other groups; this level was also showed in an exercise frequency-dependent manner. Peritoneal ROS and ASAS showed a statistically significant increase in the F5 group and decreased in the F2 group compared with the S group. However, there were no significant differences in the F3 group. The highest level of IL-4 was found in the F5 group compared with the other groups. However, the highest level of INF-${\gamma}$ was in the F2 group. The results suggest that FDEIA is positively correlated with the frequency of exercise due to the direct effect of physical exercise on peritoneal ROS and the cytokine profile. Further research is needed on the specific mechanism underlying the combined effects of exercise intensity and frequency on physical-induced allergy anaphylaxis.

Although active systemic anaphylaxis and passive cutaneous anaphylaxis have been empolyed to study anaphylactic hypersensitivity, it is difficult and time-consuming to induce these reactions in experimental animals. In recent, Jun et al have found a simple method to induced anaphylactic hypersensitivity such as anaphylactic shock(AS) and cutaneous reaction(CR) using compound48/80. Cortex mori (Morus alba L.), the root bark of mulberry tree has been used as an antiphlogistic, diuretic, and expectorant in herbal medicine. The purpose of this study was to determine whether the methanol extract of Cortex mori could inhibit the compound 48/80-induced AS and CR. To induce AS, various doses of compound 48/80 (5, 7.5, 10, 15$\mu\textrm{g}$/gm B.W.) were injected intraperitoneally (i.p.) into ICR mice. The animals were pretreated by three injection(i.p.) of Cortex mori before compound 48/80 administration. Peripheral blood was collected from the right ventricle to estimate the level of serum histamine at 15 minutes after the injctin(i.p.) of various concentration of compound48/80. Mortility rate, mean death time and mesenteric mast cell degranulation rate were evaluated over a 72 hour period. To estimate the effect of Cortex mori on compound 48/80-induced cutaneous reaction, various doses of compound 48/80 with or without Cortex mori were injected intradermally(i.d.) into the shaved flank of Sprague-Dawley rats, and the blue cutaneous patchs induced by Evans'blue injection at the compound 48/80 alone and Cortex mori plus compound 48/80 injection sites were observed. As a Parameter of these reactions, the levels of histamine in the supernatant, calcium uptake and intracellular CAMP of RPMC were measured. supernatant, 1)compound 48/80-induced mortility rate, mean death time, mesenteric mast cell degranulation rate, and serum histamine level in ICR mice were significantly inhibited by pretreatment of Cortex mori, 2) cutaneous reaction inducd by compound48/80 was well developed in Sprague-Dawley rat, but Cortex mori inhibited the compound 48/80-induced blue patch formation remarkably, 3) the compound 48/80-induced degranulation, histamine release and calcium uptake of RPMC pretreated with Cortex mori were significantly inhibited, compared to those of control without Cortex mori pretreatment, and 4)the level of cAMP of RPMC was reduced bythe increased concentration of compound 48/80, pretreatment of Cortex mori not only inhibited the compound 48/80-induced reduction of CAMP but also significantly increased the level of cAMP naturally, from the above results, it is suggested that Cortex mori has an some substances with an ability to inhibits the compound 48/80-induced AS,CR, and mast cell activation.

The purpose of this study was to evaluate the effects of several types of bibimbab (a Korean traditional meal of mixed rice with assorted vegetables), on various immune activities. Compared to control animals in a mouse model (given hamburgers), the oral administration of a portion of bibimbab containing wild plants significantly increased splenic B/T, thymic Th lymphocyte subpopulations, serum IFN-${\gamma}$ production, and enhanced hemagglutination titers up to 300%. Also, a consumption of mushroom-bulgogi bibimbab and Jeonju-style bibimbab markedly decreased compound 48/80-induced systemic anaphylaxis (immediate hypersensitivity), while bibimbab with wild plants inhibited SRBC-induced delayed type hypersensitivity. These results suggest that bibimbab with wild plants both up-regulate on immune activities and have anti-allergenic properties.

Shini-San has been used for treatment of allergic disease in Korea. However, its effect in experimental models remains unknown. The mast cell plays a pivotal role in initiating al1ergic response by secreting intracytoplasmic granular mediators such as histamine. The present report describes an inhibitory effect of Shini-San on mast cell-mediated immediate-type al1ergic reactions. Topical application of compound 48/80 can induce an ear swelling response in normal ($WBB6F_1-+/+$) mice but not in congenic mast cell-deficient $WBB6F_1-W/W^v$ mice. Shini-San inhibited concentration dependent mast cell-dependent ear swelling response induced by compound 48/80 in normal mice. Shini-San inhibited concentration-dependent passive cutaneous anaphylaxis induced by anti-dinitrophenyl (DNP) immunoglobulin E (IgE) in rats by topical application. Shini-San also inhibited in concentration-dependent fashion the histamine release from the rat peritoneal mast cells by compound 48/80 or anti-DNP IgE. Moreover, Shini-San had a significant inhibitory effect on compound 48/80-induced systemic anaphylactic reaction. These results indicate that Shini-San inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.

The immature fruits of Poncirus trifoliata L. or Ponciri fructus (PF), well known as 'Jisil' in Korea, have been used against allergic diseases for generations, and still occupy an important place in traditional Oriental medicine. Anti-allergic effects of this fruit have been investigated in a few experimental models. Immunoglobulin E (IgE) is the principal immunoglobulin involved in immediate hypersensitivities and chronic allergic diseases. The effect of an aqueous extract of PF on in vivo and in vitro IgE production was investigated. PF dose-dependently inhibited the active systemic anaphylaxis and serum IgE production induced by immunization with ovalbumin, Bordetelia pertussis toxin and aluminum hydroxide gel. PF strongly inhibited interleukin 4 (IL-4)-dependent IgE production by lipopolysaccharide-stimulated murine whole spleen cells. In the case of U266 human IgE-bearing B cells, Ponciri fructus also showed an inhibitory effect on the IgE production. On the other hand, mast cell hyperplasia can be causally related with chronic inflammation. Stem cell factor (SCF), the ligand of the c-kit protooncogene product, is a major regulator and ohernoattractant of mast cells. Ponciri fiuctus (1 mg/mL) significantly inhibited the SCF-induced migration of rat peritoneal mast cells (RPMCs). RPMCs exposed to SCF (50 ng/mL) resulted in a drastic shape change with a polarized morphology while the cells exposed to Ponciri fructus (1 mg/mL) remained resting, with little or no shape alteration. The drastic morphological alteration and distribution of polymerized actin were blocked by pretreatment with Ponciri fructus. In addition, Ponciri fructus inhibited both TNF-alpha and IL-6 secretion from RPMCs stimulated with SCF. These results suggest that Ponciri fructus has an anti-allergic activity by inhibition of IgE production from B cells. These findings also provide evidence that Ponciri fructu inhibits chemotactic response and inflammatory cytokines secretion to SCF in mast cells.

Botulinum toxins are the most potent toxins known to mankind. Botulinum toxin acts by blocking the cholinergic neuromuscular or the cholinergic autonomic innervation of exocrine glands and smooth muscles. Seven distinct antigenic botulinum toxins (A, B, C, D, E, F and G) produced by different strains of Clostridium botulinum have been described and only A and B type of botulinum toxins were clinically used. Toxins were consisted of a heavy chain with a molecular weight of 100 kD and a light chain with a molecular weight of 50 kD. Toxins are bound with an astounding selectivity to glycoprotein structures located on the cholinergic nerve terminal. Subsequently light chain of toxin is internalized and cleaves different proteins of the acetylcholine transport protein cascade transporting the acetylcholine vesicle from the intracellular space into the synaptic cleft. After a decade of therapeutic application of the toxin, no anaphylaxis or deaths have been reported and systemic adverse effects have not been reported so far. However the toxin's immunologic properties can lead to the stimulation of antibody production, potentially rendering further treatments ineffective. Botulinum toxin is a safe and effective treatment. Use of botulinum toxin in clinical medicine has grown exponentially in recent years, and many parts of the human body are now being targeted for therapeutic purposes.
Vitamin E, which has its advocates in the treatment of diabetes mellitus. autoimmune disease, cancer and peripheral vascular and thromboembolic disease, has now been alleged to have a powerful antioxident effect and to affect various biological activities such as fertility factor, inhibition of human platelet aggregation and stabilization of biological membranes. The present study was designed to test whether vitamin I(alpha-tocopherol) can : (1) enhance the hemagglutinin response to sheep red blood cells (SRBC), (2) modulate Arthus and delayed type hypersensitivity(DTH) to SRBC and contact hypersensitivity to dinitrofluorobenzene (DNFB). (3) enhance the mitogenic response of murine splenocyte, (4) decrease the recovery of Cryptococcus neoformans from brain, lung, liver, spleen and kidney of infected mice and (5) have an inhibitory or enhancing effect on the induction of active systemic anaphylaxis(ASA) induced by chicken-gamma globulin (CGG) in mice. Mice were given either intramuscular injections of 0.3ml (300mg) of vitamin I before immunization or were infection for 10 consecutive days or were given by vitamin I esophageal intubation, 0.1ml(100mg), for 20 days before sacrifice for the mitogenic response experiments. It was found that vitamin E treated mice showed a significant enhancement in hemagglutinin response, Arthus reaction and DTH to SRBC and contact hypersensitivity to DNFB. There was no significant difference in the mitogenic response to phytohemagglutinin(PHA), but the response to concanavalin A(ConA) or pokeweed mitogem(PWM) was increased in vitamin E-treated mice. Interestingly, the vitamin E administration before C. neoformans infection decreased significantly the recovery of C. neoformans from brain lung, liver, spleen and kidney of the infected mice as compared with that of the control mice, strongly suggesting that vitamin E pretreatment may increase the resistance of mice to the fungal infection. Unexpectedly, vitamin E administration enhanced the production of CGG -induced ASA. Taken together, it can be concluded that vitamin I administration may in-crease the humoral and cellular immune response and resistance. to C. neoformans infection, but enhance the induction of ASA to CGG. Further studies are necessary to clarify the underlying mechanism accounting for these effects.
Gamitonggyutang has been used for treatment of rhinitis in oriental medical science. It is reported that Gamitonggyutang has a good effect on sinusitis in clinical medicine. So this study analysed the effect of Gamitonggyutang on anti-inflammatory, analgenic, anti-Allergic effect and Antibacterial Activity. The result were summerised as follows; 1. Gamitonggyutang extract decreased the edema indused by Carregennin at 200mg/kg and 400mg/kg. 2. Gamitonggyutang extract decreased the protein exudation indused by CMC-pouch at 200mg/kg and 400mg/kg. 3. Gamitonggyutang extract showed the ataralgesia at 800mg/kg by measurement of writhing syndrome, paw licking time and escape time induced by the i.p. infection of acetic acid and hot plate. 4. Gamitonggyutang extract decreased the effluent of vascular permeability indused by Evans blue at 600mg/kg and 800mg/kg. 5. Gamitonggyutang extract decreased the acute edema indused by Carregennin about 2 and 4 hours but didn't show useful effect. 6. Gamitonggyutang extract decreased the death rate, resulted from the effect of active systemic anaphylaxis reaction indused by CGG, but didn't show useful effect.. 7. Gamitonggyutang extract showed the growth inhibitory effect of each bacterias at $52mg/m{\ell}$. 8. Gamitonggyutang extract suppressed the growth of Streptococcus mutans 10449, and showed the supression of acid fabrication in case of 1:10 more than 1:100.
The immediate-type allergic reaction (anaphylaxis) is involved in many allergic diseases such as asthma, allergic rhinitis, and sinusitis. The discovery of drugs for the treatment of immediate-type allergic diseases is a very important subject in human health. In this study, we investigated the effect of Rubus coreanus Miq.(Rosaceae) unripe fruits (RCF) on mast cell-mediated allergic reaction and inflammatory cytokine secretion. RCF inhibited compound 48/80-induced systemic reactions in mice. RCF attenuated immunoglobulin (Ig) E-mediated local allergic reactions. In addition, RCF dependently reduced histamine release from rat peritoneal mast cells local allergic reactions. In addition, RCF dependently reduced histamine release from rat peritoneal mast cells activated by compound 48/80 or IgE. Furthermore, RCF decreased the phorbol 12-myristate 13-acetate plus calcium ionophore A23187-stimulated tumor necrosis factor $(TNF)-{\alpha}$ and interleukin (IL)-6 secretion in human mast cells. Our findings provide evidence that RCF inhibits mast cell-derived immediate-type allergic reactions.
Objectives : This experimental research has been done to study the effects of herbal-acupuncture with Juglandis Semen on the anti-allergic reaction. Methods : ICR mice were injected intraperioneally with compound 48/80 to induce the immediately type hypersensitivity (anaphylaxis). ICR mice were sensitized by PC and SRBC to induce the delayed type hypersensitivity. Sprage-Dawley rats were injected subcutaneously with egg albumin to measure the WBC. ICR mice and Sprage-Dawley rats were injected with Juglandis Semen herbal-acupuncture for the anti-allergic reaction test. Results : 1. In the restraint effect of systemic anaphylactic reaction, the mortality of mice decreased 20% each before 10 minutes and after 5 minutes. 2. In the contact dermatitis induced by PC, both Treat Ⅰand Treat Ⅱ group showed significant(p<0.01, p<0.05) decrease in comparison with the Control group, and also Treat Ⅲ group showed significance(p<0.005). 3. In the delayed type hypersensitivity in mice challenged by SRBC, Treat Ⅰ group showed significant(p<0.05) decrease in comparison with the Control group, and there was significance(p<0.05, p<0.05) in Treat Ⅲ and Treat Ⅳ group. 4. The change of the WBC in allergic rats induced by egg albumin showed significant(p<0.01, p<0.05, p<0.05) decrease in Treat Ⅰ, Treat Ⅱ and Treat Ⅲ group. Conclusions : The restraint effects of Juglandis Semen herbal-acupuncture on the anti-allergic reaction showed significance statistically. And also allergic group treated with $BL_{17}$ acupuncture (Treat Ⅲ group) showed same results.
