Characterization of Molecular Composition of Bacterial Melanin Isolated from Streptomyces glaucescens Using Ultra-High-Resolution FT-ICR Mass Spectrometry

  • In this study, the chemical composition of bacterial melanin isolated from the Streptomyces glaucescens strain was elucidated by ultra-high-resolution Fourier transform ion cyclotron resonance (FT-ICR) mass spectrometry. Ultra-high-resolution mass profiles of the microbial melanin product were acquired using a 15 Tesla FT-ICR mass spectrometer in positive and negative ion modes via electrospray ionization to obtain more complete descriptions of the molecular compositions of melanin-derived organic constituents. A mass resolving power of 500,000 (at m/z 400) was achieved for all spectra while collecting 400 scans per sample with a 4 M transient. The results of this analysis revealed that the melanin pigment isolated from S. glaucescens predominantly exhibits CHON and CHO species, which belong to the proteins class of compounds, with the mean C/O and C/N ratios of 4.3 and 13.1, thus suggesting that the melanin could be eumelanin. This analytical approach could be utilized to investigate the molecular compositions of a variety of natural or synthetic melanins. The compositional features of melanins are important for understanding their formation mechanisms and physico-chemical properties.

  • This study is including the independent project, "building children's library of the town" that is conducted by a local resident. The study object is the contents of the "design workshop for library planning" and the general process during one year since the opening. This study is delivering "information of children's library" through the work-shop and is conducting the role of a professional space planner by measuring the blueprint. Thus, the study could be conducted with synthetic viewpoint on each library. The followings are the results from the study. First, there were various space operations and a variety of programs in the case library that is independently managed. This is directing to "the constructive and active space usage" by the users and finally results in positive circulation. Secondly, there is a change through the process of execution and operation in the library construction plan. There are needs for the flexible space expansion and adjustment, modifiable furniture layout and design and various keeping areas because there is on-going tendency of corresponding space change. The third, the author could draw more practicable consideration points for space planning by looking at the trend of space change in Transactionist viewpoint. Thus, this study is very meaningful in terms of planning of sustainable public areas.

FASIM: Fragments Assembly Simulation using Biased-Sampling Model and Assembly Simulation for Microbial Genome Shotgun Sequencing

  • We have developed a program for generating shotgun data sets from known genome sequences. Generation of synthetic data sets by computer program is a useful alternative to real data to which students and researchers have limited access. Uniformly-distributed-sampling clones that were adopted by previous programs cannot account for the real situation where sampled reads tend to come from particular regions of the target genome. To reflect such situation, a probabilistic model for biased sampling distribution was developed by using an experimental data set derived from a microbial genome project. Among the experimental parameters tested (varied fragment or read lengths, chimerism, and sequencing error), the extent of sequencing error was the most critical factor that hampered sequence assembly. We propose that an optimum sequencing strategy employing different insert lengths and redundancy can be established by performing a variety of simulations.

  • Because of the changes of a variety of aspects of disease theory alternative to standard medicine, demand for traditional medicine has increased. This study was undertaken to grasp and compare traditional-medicine administrative structure, functions and role between Korea and China. According to our research, both nations promote policy related to traditional-medicine, yet there are various differences because of each nation's scale, political structure, medical system, etc. Especially, compare to the Korean government, the Chinese government not only has paid more attention to traditional medicine but also established and promoted a synthesis development strategy for a long time. Additionally, there is weakness in Korea's traditional medicine policy compared to the Chinese's Synthetic development-strategy, which is the Oriental-policy Department and Korea Food&Drug administration. Therefore, to make up for the weakness, they will have to organize their role better and make a long-term development-strategy plan for traditional medicine.

  • In contemporary architecture, there are developing aspects of variety design in material, image and form. These various architectural trends even change classical media and this concept is based on the background of current idea - digital, information-media, imagination with paradigm. So this study aims to show the present status of the expressions in the contemporary architecture of Korea under its media situation. Namely usual thoughts for design have to be changed because our life is surrounded by many kinds of powerful media signs and various mixed cultural circumstances. And like this, we know for study about the relation between media and current architecture, still more Korean present condition. In chapter one, the background, purpose and method of study are explained. In chapter two, the general concept of media is inquired in this study. In the process of searching for the formingbasic background, the characteristic of this study is understood. In chapter three, this study inquires the aspect of media expression in modern architecture. In chapter four, according to chapter three, this study shows the present status of media expressions in the contemporary architecture of Korea. In chapter five, a synthetic conclusion is presented.

  • Contemporary architecture is developing aspects of variety design in form and material. The various architectural trends even change classical materiality. And like this, we know for study about the relation between dematerialization and current architecture, still more Korean present condition. In chapter one, the background, purpose and method of study are explained. In chapter two, this study inquires the general concept of dematerial. In the process of searching for the forming basic background this study understands the characteristic. In chapter three, this study inquires the aspect of dematerialized expression in modern architect. In chapter four, with characteristics drawn in chapter three, this study show the present status of the expressions in the contemporary architecture in Korea under its dematerialized situation. In chapter five, a synthetic conclusion is presented.

Biodergradation of Reactive Dyes Using Multistage Rotating Disc Contactor Immobilized by Aspergillus sojae B-10 (회전원판 반응조에 고정화시킨 Aspergillus sojae B-10에 의한 반응성 염료의 탈색)

  • Dyes are released into the environment as industrial wasterwater. Dyes are considered to be a pollution problem because of the wide spread into environment with a variety of colors. Continuous biodegradation of reactive dyes such as Rifacion Red H-3EB, Rifazol Blue BT, Rifacion Yellow P4G and Rifacion Brown RT were demonstrated using multistage rotating disc contactor immobilized by Aspergillus sojae B-10. Aspergillus sojae B-10 was cultivated the optimal medium containing 2.0% glucose, 0.08% $NaNO_3$, 0.1% $KH_2PO_4$ and 0.5% $MgSO_4\cdot 7H_2O$, pH 5 at 32$\circ$C. Mycelium of Aspergillus sojae B-10 were guck to the rotating disc for 10 days until steady state. For continuous biodegradation of reactive dyes by using rotating disc contactor, it was most effective biodegradation in the medium containing 1,000 ppm each dyes at the medium feeding rate of 20 ml per hour. Under the conditions biodegradation of each dyes on 2, 4 and 6 days were 20~50%, 75% and 96%, respectively. Therefore, practical application of reactive dyes were carried out at the feeding rate of 20 ml/h as synthetic wasterwater containing 500 ppm of mixture reactive dyes. It was found the highest levels of 94% biodegradation during 20 days.

Inhibitory Action of Minocycline on Lipopolysaccharide-Induced Release of Nitric Oxide and Prostaglandin E2 in BV2 Microglial Cells

  • Microglia are the major inflammatory cells in the central nervous system and become activated in response to brain injuries such as ischemia, trauma, and neurodegenerative diseases including Alzheimer's disease (AD). Moreover, activated microglia are known to release a variety of proinflammatory cytokines and oxidants such as nitric oxide (NO). Minocycline is a semi-synthetic second-generation tetracycline that exerts anti-inflammatory effects that are completely distinct form its antimicrobial action. In this study, the inhibitory effects of minocycline on NO and prostaglandin E$_2$ (PGE$_2$) release was examined in lipopolysaccharides (LPS)-challenged BV2 murine microglial cells. Further, effects of minocycline on inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression levels were also determined. The results showed that minocycline significantly inhibited NO and PGE$_2$ production and iNOS and COX-2 expression in BV2 microglial cells. These findings suggest that minocycline should be evaluated as potential therapeutic agent for various pathological conditions due to the excessive activation of microglia.

  • SAR is a Radar to obtain the video information using a radio wave. Platform emit the radio wave, depending backscattered waves returned from the target object the signal to the distance, to create a topographical map is recorded in two-dimensional image. In this paper, through a simulation to apply a variety of window in the SAR image processing for SAR image recovery is to study the application effect of the window, as a result, at the side of the signal of the SNR, Flattop window to improve the best performance it was confirmed to show.

Change in Photosynthesis, Proline Content, and Osmotic Potential of Corn Seedling under High-Saline Condition

  • To identify salt-tolerance characteristics of corn seedling was treated in solution of 0, 50 and 100 mM NaCl of hydroponic cultivation. In photosynthesis of corn seedling, there was no large difference between 50mM and 0 mM NaCl solution, however, in 100 mM NaCl solution, the tolerance gradually decreased to $76\%,\;49\%,\;and\;31\%$ after one day, four days, and seven days, respectively, in comparison to 0 mM NaCl solution. Osmotic potential of corn in seedling period was significantly decreased with increasing saline level, however, free proline content in the plant on the ground was significantly increased with increasing saline level and with the lapse of time. In terms of correlation among major characteristics, there was a highly significant positive difference between osmotic pressure potential and photosynthesis, However, highly negative correlation was found between osmotic pressure potential and free proline content. In addition, it was expected that young seedling of corn with saline tolerance may be utilized in the transplantation in salt-accumulated land. Based on above-shown result, in terms of saline tolerance of Chalok-2 variety, growth suppression was serious with 100mM NaCl solution. However, growth was expected that seedling growth would be favorable under 50 mM NaCl solution.
