The seasonal changes were evaluated in the soil microbial populations by selected media in an organic farming system (OFS) with no-till management compared to those in a conventional farming system (CFS) with tillage and synthetic amendments in a flooded paddy from 2009 to 2010. The populations of aerobic bacteria and fungi in the OFS were significantly higher than those in the CFS at the harvesting stages, whereas those of Gram-negative bacteria was significantly higher in the OFS than in the CFS before the submerging stages. In addition, populations of aerobic bacteria, Gram-negative bacteria, and fungi tended to rapidly decreased after the submerging stages may be due to insufficient oxygen. Gram-negative bacteria should be considered as potential factor responsible for the microbial population differentiation observed between the OFS and the CFS in flooded paddy fields.

A fungal strain, designated PTT-2, was isolated from the bark of the trunk of a persimmon (Diospyros kaki) tree in Cheongdo, Korea. The isolate showed morphological similarities with Leptosphaerulina saccharicola. Strain PTT-2 had more rapid growth on potato dextrose agar medium than on oatmeal agar, malt extract agar, and synthetic nutrient poor agar media, with colony sizes of 53.8 mm, 49.8 mm, 48.4 mm, and 28.1 mm after 7 days at $25^{\circ}C$ temperature, respectively. Strain PTT-2 produced ascospores, which had irregular wavy edges, oblong to ellipsoidal shape, hyaline appearance and $23.6{\times}10{\mu}m$ size. The black ascomata were developed on PDA medium, and asci were recorded. A BLAST search of the internal transcribed spacer (ITS) region, TEF1-${\alpha}$ and RPB2 gene sequences revealed that strain PTT-2 showed more than 99% nucleotide similarity with a strain of Leptosphaerulina saccharicola previously reported from Thailand. A neighbor-joining phylogenetic tree was constructed by concatenating the above-mentioned sequences, and showed that strain PTT-2 clustered in the same clade with L. saccharicola. Based on these findings, this is the first record of Leptosphaerulina saccharicola occurring in Korea.

The identification of compounds with anti-senescence activity in cell culture system is a first step in aging research. Given that pyruvate can be used energy source by conversion to acetyl-CoA in mitochondria, and protects cultured cell from various stress-induced cell damage and cell death, synthetic media (e.g., DMEM) often includes 1 mM pyruvate, which is very higher than the pyruvate concentration in human blood (approximately 30 M). However, the use of medium containing high concentration of pyruvate is not suitable for screening anti-senescence compounds, because pyruvate also protects against the cellular senescence of primary human dermal fibroblasts (NHDFs) through NAD+ generated during conversion to lactate. In this study, four extracts, i.e., Sprouted seed and fruit complex, Poncirus trifoliata fruit extract, Jaum balancing complex, and Prunus mume extract were used for evaluation of different anti-senescence effect in the absence or presence of 0.1 mM pyruvate, similar to the physiological pyruvate concentration. The senescence in NHDFs cultured with DMEM in the presence of 0.1 mM pyruvate (approximately the physiological concentration in human blood) is accelerated, as observed in pyruvate deprivation conditions. The cytotoxicity of the Poncirus trifoliata fruit extract was protected by pyruvate, and Jaum balancing complex and Prunus mume extract had anti-senescence activity in the presence of 0.1 mM pyruvate, but not in the absence of pyruvate. Given that pyruvate is a powerful protector against both cytotoxicity and cellular senescence, the screening of candidate agents for anti-senescence in high pyruvate conditions using an in vitro cell culture system is not valid. Therefore, we recommend the use of a low concentration of pyruvate to evaluate the anti-senescence effects of candidates, which is more similar to in vivo aging conditions than excessive stress-induced senescence models, to exclude the effect of excessive pyruvate in vitro.
Natural and synthetic forms of calcium phosphate cement (CPC) have been widely used in bone repair and augmentation. The major challenge of injectable CPC is to deliver the cells without cell death in order to regenerate new bone. The study objective was to investigate for the potential of stem cell-laden gelatin fibers containing injectable, nanocrystalline CPC to function as a delivery system. Gelatin noddle fiber method was developed to delivered cells into nCPC. Experimental groups were prepared by mixing cells with nCPC, mixing cell-laden gelatin fibers with nCPC and mixing cell-laden gelatin fibers containing BMP-2 with nCPC. Media diffusion test was conducted after dissolving the gelatin fibers. SEM examined the generated channels and delivered cell morphology. Fibers mixed with nCPC showed physical setting and hardening within 20 min after injection and showed good shape maintenances. The gelatin fibers mixed nCPC group had several vacant channels generated from the dissolved gelatin. Particularly, proliferation and attachment of the cells were observed inside of the channels. While live cells were not observed in the cell mixed nCPC group, cells delivered with the gelatin fibers into the nCPC showed good viability and increased DNA content with culture. Cell-laden gelatin fiber was a novel method for cell delivery into nCPC without cell damages. Results also indicated the osteogenic differentiation of gelatin fiber delivered cells. We suggest that the cell-laden gelatin fibers mixed with nCPC can be used as an injectable cell delivery vehicle and the addition of BMP-2 to enhances osteogenesis.
Steroids are a class of compounds with cyclopentane polyhydrophenanthrene as the parent nucleus, and they usually have unique biological and pharmacological activities. Most of the biosynthesis of steroids is completed by a series of enzymatic reactions starting from cholesterol. Synthetic biology can be used to synthesize cholesterol in engineered microorganisms, but the production of cholesterol is too low to further produce other high-value steroids from cholesterol as the raw material and precursor. In this work, combinational strategies were established to increase the production of cholesterol in engineered Saccharomyces cerevisiae RH6829. The basic medium for high cholesterol production was selected by screening 8 kinds of culture media. Single-factor optimization of the carbon and nitrogen sources of the culture medium, and the addition of calcium ions, zinc ions and citric acid, further increased the cholesterol production to 192.53 mg/l. In the 5-L bioreactor, through the establishment of strategies for glucose and citric acid feeding and dissolved oxygen regulation, the cholesterol production was further increased to 339.87 mg/l, which was 734% higher than that in the original medium. This is the highest titer of cholesterol produced by microorganisms currently reported. The fermentation program has also been conducted in a 50-L bioreactor to prove its stability and feasibility.
In this paper, we propose a gaze estimation network in which eye landmark position detection and gaze direction vector estimation are integrated into one deep learning network. The proposed network uses the Stacked Hourglass Network as a backbone structure and is largely composed of three parts: a landmark detector, a feature map extractor, and a gaze direction estimator. The landmark detector estimates the coordinates of 50 eye landmarks, and the feature map extractor generates a feature map of the eye image for estimating the gaze direction. And the gaze direction estimator estimates the final gaze direction vector by combining each output result. The proposed network was trained using virtual synthetic eye images and landmark coordinate data generated through the UnityEyes dataset, and the MPIIGaze dataset consisting of real human eye images was used for performance evaluation. Through the experiment, the gaze estimation error showed a performance of 3.9, and the estimation speed of the network was 42 FPS (Frames per second).

Sharing of online videos via internet is an emerging and important concept in different types of applications like surveillance and video mobile search in different web related applications. So there is need to manage personalized web video retrieval system necessary to explore relevant videos and it helps to peoples who are searching for efficient video relates to specific big data content. To evaluate this process, attributes/features with reduction of dimensionality are computed from videos to explore discriminative aspects of scene in video based on shape, histogram, and texture, annotation of object, co-ordination, color and contour data. Dimensionality reduction is mainly depends on extraction of feature and selection of feature in multi labeled data retrieval from multimedia related data. Many of the researchers are implemented different techniques/approaches to reduce dimensionality based on visual features of video data. But all the techniques have disadvantages and advantages in reduction of dimensionality with advanced features in video retrieval. In this research, we present a Novel Intent based Dimension Reduction Semi-Supervised Learning Approach (NIDRSLA) that examine the reduction of dimensionality with explore exact and fast video retrieval based on different visual features. For dimensionality reduction, NIDRSLA learns the matrix of projection by increasing the dependence between enlarged data and projected space features. Proposed approach also addressed the aforementioned issue (i.e. Segmentation of video with frame selection using low level features and high level features) with efficient object annotation for video representation. Experiments performed on synthetic data set, it demonstrate the efficiency of proposed approach with traditional state-of-the-art video retrieval methodologies.

Cultured meat is a potential sustainable food generated by the in vitro myogenesis of muscle satellite (stem) cells (MSCs). The self-renewal and differentiation properties of MSCs are of primary interest for cultured meat production. MSC proliferation and differentiation are influenced by a variety of growth factors such as insulin-like growth factors (IGF-1 and IGF-2), transforming growth factor beta (TGF-β), fibroblast growth factors (FGF-2 and FGF-21), platelet-derived growth factor (PDGF) and hepatocyte growth factor (HGF) and by hormones like insulin, testosterone, glucocorticoids, and thyroid hormones. In this review, we investigated the roles of growth factors and hormones during cultured meat production because these factors provide signals for MSC growth and structural stability. The aim of this article is to provide the important idea about different growth factors such as FGF (enhance the cell proliferation and differentiation), IGF-1 (increase the number of myoblasts), PDGF (myoblast proliferation), TGF-β1 (muscle repair) and hormones such as insulin (cell survival and growth), testosterone (muscle fiber size), dexamethasone (myoblast proliferation and differentiation), and thyroid hormones (amount and diameter of muscle fibers and determine the usual pattern of fiber distributions) as media components during myogenesis for cultured meat production.
3D Object Detection using deep neural network has been developed a lot for obstacle detection in autonomous vehicles because it can recognize not only the class of target object but also the distance from the object. But in the case of 3D Object Detection models, the detection performance for distant objects is lower than that for nearby objects, which is a critical issue for autonomous vehicles. In this paper, we introduce a technique that increases the performance of 3D object detection models, particularly in recognizing distant objects, by generating virtual 3D vehicle data and adding it to the dataset used for model training. We used a spherical point tracing method that leverages the characteristics of 3D LiDAR sensor data to create virtual vehicles that closely resemble real ones, and we demonstrated the validity of the virtual data by using it to improve recognition performance for objects at all distances in model training.
Recently, as VR (Virtual Reality) technology has been in the spotlight, 360° panoramic images that can view lively VR contents are attracting a lot of attention. Image stitching technology is a major technology for producing 360° panorama images, and many studies are being actively conducted. Typical stitching algorithms are based on feature point-based image stitching. However, conventional feature point-based image stitching methods have a problem that stitching results are intensely affected by feature points. To solve this problem, deep learning-based image stitching technologies have recently been studied, but there are still many problems when there are few overlapping areas between images or large parallax. In addition, there is a limit to complete supervised learning because labeled ground-truth panorama images cannot be obtained in a real environment. Therefore, we produced three fisheye images with different camera centers and corresponding ground truth image through carla simulator that is widely used in the autonomous driving field. We propose image stitching model that creates a 360° panorama image with the produced fisheye image. The final experimental results are virtual datasets configured similar to the actual environment, verifying stitching results that are strong against various environments and large parallax.
