Anti-inflammatory mechanism of bee vemon in Raw 264.7 cells and Synoviocyte

  • Bee venom (BY) has been utilized to relieve pain and to treat inflammatory diseases such as rheumatoid arthritis (RA). However, the molecular mechanism by which BV-induced anti-arthritis effect has been not reported yet. Therefore, in the present study we investigated anti-inflammatory effect of BV in a murine marcrophage cell line Raw 264.7 cell and synoviocyte obtained from RA patients. The present data showed that BV has a preventive effect on lipopolysaccharide (LPS) and sodium nitroprusside (SNP) induced induction of COX-2, cPLA2 and iNOS. (omitted)

  • Melittin,cationic 26-amino acid, is the principal component of the bee venom (BV) which has been used for treatment of inflammatory disease such as arthritis rheumatism NF-kB is activated by subsequent release of inhibitory IkB via activation of a multisubunit IkB kinase (IKK). We previously found that melittin bind to the sulfhydryl group of p50, a subunit of NF-kB. Since sulfhydryl group is present in kinase domain of IKKa and IKKb, melittin could modify IKK activity by protein-protein interaction. We therefore examined effect of melittin on IKK activities in sodium nitroprusside (SNP)-stimulated synoviocyte obtained from RA patients. Melittin suppressed the SNP-induced release of IkB resulted in inhibition of DNA binding activity of NF-kB and NF-kB-dependent luciferase activity. Consistent with the inhibitory effect on NF-kB activation, IKKa and IKKb activities were also suppressed by melittin. Surface plasmon resonance analysis realized that melitin binds to IKKa $(Kd\;=\;1.34{\times}10-9M)$ and IKKb$(Kd\;=\;1.0{\times}10-9M)$. Inhibition of IKKa and IKKb resulted in reduction of the SNP-induced production of inflammatory mediators NO and PGE2 generation. The inhibitory effect of melittin on the IKKs activities, binding affinity of melittin to IKKs, and NO and PGE2 generation were blocked by addition of reducing agents dithiothreitol and glutathione. In addition, melittin did not show inhibitory effect in the transfected Synoviocytes with plasmid carrying dominant negative mutant IKKa (C178A) and IKKb (C179A). These results demonstrate that melittin directly binds to sulfhydryl group of IKKs resulting in IkBrelease, thereby inhibits activation of NF-kB and expression of genes involving in the inflammatory responses.

  • Objective : The purpose of this study was investigation how the bee venom(BV) prevents inflammation in human cell. Methods : we induced inflammation on human synoviocyte cell by lipopolysaccharide(LPS) and sodium nitroprusside(SNP), treated the bee venom and melittin on this cell, surveyed the expression of Nisotric oxide(NO), inducible nitric oxide synthase(iNOS), Cyclooxygenease-2(COX-2), cytolic phospholipase $A_2(cPLA_2)$, Prostaglandin $E_2(PGE_2)$ and nuclear factor-${\kappa}B$(NF-${\kappa}B$), and got below conclusions. Results : Compared with control 1. Expressions of LPS-induced $PGE_2$(BV 1, $5{\mu}g/m{\ell}$) and SNP-induced PGE2(BV 0.5, 1, $5{\mu}g/m{\ell}$)were decreased significantly. 2. Expressions of LPS-induced NO(BV 0.5, 1, $5{\mu}g/m{\ell}$) and SNP-induced NO(BV 1, $5{\mu}g/m{\ell}$)were decreased significantly. 3. Expressions of LPS-induced COX-2(BV 1, $5{\mu}g/m{\ell}$) and SNP-induced COX-2(BV $5{\mu}g/m{\ell}$)were decreased significantly. 4. Expressions of LPS-induced iNOS(BV 0.5, 1, $5{\mu}g/m{\ell}$) and SNP-induced iNOS(BV $5{\mu}g/m{\ell}$) were meanless by all dose. 5. Expressions of LPS-induced $cPLA_2$(BV 1, $5{\mu}g/m{\ell}$) and SNP-induced cPLA2(BV 1, $5{\mu}g/m{\ell}$)were decreased significantly. 6. Expressions of LPS-induced NF-${\kappa}B$(BV $5{\mu}g/m{\ell}$, melittin $5{\mu}g/m{\ell}$) and SNP-induced NF-${\kappa}B$(BV 0.5, 1, $5{\mu}g/m{\ell}$, melittin 5, $10{\mu}g/m{\ell}$)were decreased significantly. 7. Expressions of LPS-induced NF-${\kappa}B$ binding activity (BV $1{\mu}g/m{\ell}$, melittin $5{\mu}g/m{\ell}$, melittin $5{\mu}g/m{\ell}$+ DTT 20mM) were decreased significantly. Conclusion : The bee venom treatments on synoviocyte showed significant changes in LPS and SNP induced NO, iNOS, COX-2, cPLA2, PGE2 and NF-${\kappa}B$, these results suggest that bee venom is effective to inflammations and establish the process of bee venom therapy, so we expect active use of bee venom to control the inflammation.

In Vitro Effects of Bupivacaine in Cell Proliferation and Matrix Metalloproteinase of Cultured Fibroblast Like Synoviocytes from Rheumatoid Arthritis from Rheumatoid Arthritis (부피바카인이 류마티스 관절염환자의 섬유모세포양 활막세포 배양시 세포증식과 금속단백분해효소 생산에 미치는 실험실적 영향)

  • Background: Intraarticular local anesthetic injection has been therapeutically applied for pain control in various arthritis patients. However, little physiologic effects of local anesthetics on their tissue were known. This study was conducted to determine its effects on the cell proliferation and matrix metalloproteinases (MMP) production of cultured fibroblast like synoviocytes (FLS) derived from synovial tissues of rheumatoid arthritis patients. Methods: Bupivacaine with varying concentrations 0 (control), 0.1, 0.25, 0.5% was applied to experimental cell groups growing as monolayers in culture plates for varying durations 0 (control), 30, 90, 180 seconds in the presence and absence of interleukin-$1\beta$. Results: No statistical significances were noted in thymidine incorporation between 0, 30, 90 and 180 seconds exposure groups with 0.5% bupivacaine after 1 day and 2 days. Thymidine incorporation between 0, 0.1, 0.25, 0.5% exposure groups 1 day and 2 days after 90 seconds exposure did not show any differences. After exposure to bupivacaine, there were statistically significant increases in MMP-1 (p=0.025) and MMP-3 productions (p=0.000) of FLS in the absence of IL-$1\beta$, but no differences among the groups in the presence of IL-$1\beta$. Conclusion: We concluded that in this short-term in vitro study, bupivacaine does not have injurious effect on cultured rheumatoid arthritic joint tissues. The long-term effect cannot be known from this investigation.

  • Synovial joint of BALB/C mice were injeced with Lipopolysaccharide(LPS) were observed to investigate the morphological changes of synovial capsule caused by rheumatoid arthritis(RA). The RA on female Balb/c mice were induced by LPS injection, as dose of $300{\mu}{\ell}/kg$, into synovial cavity of knee joint. And then these specimen were fixed in 10% neutral buffered formalin and were decalcificated in EDTA solution for 4 weeks. The hyperplasia of synovium were appeared in synovial membrane. The filopodia of phagocytic like synoviocyte(type I synoviocyte) projected into synovial cavity and the number of fibroblast like synoviocyte(type II synoviocyte) with well-developed endoplasmic reticulum were increased in synovium. In fibrous membrane, the fibrosis induced by synthesis of collagen fiber were enlarged to all fibrous membrane, and the number of fibroblast were increased. A great number of inflammation component cell as lymphocyte and neutrophil leukocyte were infiltrated around capillary and the degranulate typed mast cell were increased. As results indicated that the hyperplasia of synovium induced by LPS, subsequently to cause the fibrosis, infiltration of imflammation component cell, and increase of degranulated type mast cell as same as symptoms of RA.

  • Gamimahaenggamsuk-tang (GMHGST) is used for treatment of inflammatory diseases including rheumatoid arthritis (RA). Here, regulatory activity of GMHGST on RA-mediated inflammatory responses was investigated in cultured human fiDroblast-like synoviocytes (FLS), Levels of mRNAs encoding for inflammatory cytokines such as $IL-1{\beta}$, IL-6 and IL-8 and NOS-II enzyme, which had been induced by $TNF-{\alpha}$ and $IL-1{\beta}$ cotreatment, were decreased to the similar levels as those in cells treated with anti-inflammatory agent MTX. mRNA expressions of matrix metalloproteinase-3 (MMP-3) and tissue inhibitor of metalloproteinases (TIMPs) as well as intercellular adhesion molecule (ICAM) were also downregulated by increasing doses of GMHGST in activated FLS. Moreover, GMHGST appeared to protect cells by decreasing NO levels, and inhibited cell proliferation which had been induced by inflammatory stimulation by $TNF-{\alpha}$ and IL-1. These results suggest that GMHGST is effective as an inhibitory agent for regulating inflammatory responses in activated FLS.

An Immuno-Electronmicroscopic Study on the Synoviocytes in the Knee Joint of the Human (인체 무릎관절 윤활세포에 관한 면역전자현미경적 연구)

  • This study was designed to observe the ultrastructure of synoviocytes which are concerned with phagocytic function in the knee joint of the human. The synovia were dissected and were fixed for two hours in 0.2% glutaraldehyde and 4% paraformaldehyde solution and processed and finally infused in 2.3 M sucrose and 20% PVP solution. The tissues were cut with the cryoultramicrotome and labelled with primary antibodies (anti-tubulin, anti-vimentin) and secondary antibody-6 nm colloidal gold particles. The tissues were observed under transmission electronmicroscope. The results were followings. 1. In phagocytic synovial cells, the distributions of tubulin were cytoplasm, especially around vacuoles. 2. In phagocytic synovial cells, the distributions of vimentin were cytoplasm. 3. Both tubulin and vimentin were not located inside of vacuoles. On the basis of above findings, it is obvious that the phagocytic functions are concerned with tubulin, and the phagocytic synovial cells contain vimentin.
